The importance of the β-amino nitroalkanes is due to their high versatility allowing a straightforward entry to a variety of nitrogen-containing chiral building blocks; furthermore obtaining them in enantiopure form allows their use in the synthesis of biologically active compounds or their utilization as chiral ligands for different uses. In this work, a reaction for obtaining enantiopure β-amino nitroalkanes through asymmetric organocatalysis has been developed. The synthetic strategy adopted for the obtainment of these compounds was based on an asymmetric reduction of β-amino nitroolefins in a transfer hydrogenation reaction, involving an Hantzsch ester as hydrogen source and a chiral thiourea as organic catalyst. After the optimization of the reaction conditions over the β-acyl-amino nitrostyrene, we tested the reaction generality over other aromatic compound and for Boc protected substrate both aromatic and aliphatic. A scale-up of the reaction was also performed.

The present work is a collaboration between the Department of Physics and Inorganic Chemistry (group of Prof. Valerio Zanotti - Mattia Vaccari, Dr. Rita Mazzoni) and the Department of Industrial Chemistry and Materials (Group of Prof. Angelo Vaccari - Dr. Thomas Pasini, Dr. Stefania Albonetti, Prof. Fabrizio Cavani), and it’s a project devoted to evaluate the activity and selectivity of the Shvo catalyst, in the selective hydrogenation of polar double bond of 5 -hydroxymethylfurfural (HMF) in homogeneous phase. The HMF is an organic compound easily obtained from biomass, which can be used as a building block for fine chemicals abd polymer production or additives for biofuels with a high calorific value. In particular, our attention turned to the production of 2.5-bishydroxymethylfuran (BHMF), an important building block for the production of polymers and polyurethane foams. This thesis has involved the development of a new synthetic methodology for the preparation of Shvo’s catalyst and the study of its catalytic activity in the reduction of HMF to BHMF. The behavior of the catalyst was monitored by studying the yield in BHMF as a function of all the reaction parameters: temperature, pressure of H2, solvent, substrate to catalyst molar ratio, concentration, time. Subsequently it was evaluated the possibility of recycling the catalyst recovering the product of extraction with water, by precipitation or performing the reaction in biphasic mixture (toluene/H2O).

Aim of the present work of thesis is to synthesize new non-noble metal based complexes to be employ in redox reactions by a metal-ligand cooperative mechanism. The need of replacing toxic and expensive precious metal complexes with more available and benign metals, has led to the development of new compounds based on cobalt and iron, which are the metals investigated in this study. A carbonyl-tetrahydroborato-bis[(2-diisopropylphosphino)ethyl]amine-cobalt complex bearing a PNP-type ligand is synthesized by a three-step route. Optimization attempt of reaction route were assessed in order to lowering reaction times and solvent waste. New cobalt complex has been tested in esters hydrogenation as well as in acceptorless dehydrogenative coupling of ethanol. Other varieties of substrates were also tested in order to evaluate any possible applications. Concerning iron complex, dicarbonyl-(η4-3,4-bis(4-methoxyphenyl)-2,5-diphenylcyclopenta-2,4-dienone)(1,3-dimethyl-ilidene)iron is synthesized by a three steps route, involving transmetallation of a silver complex, derived from an imidazolium salt, to iron complex. In order to avoid solvent waste, optimization is assessed. Studies were performed to assess activity of triscarbonyl iron precursor toward imidazolium salt and silver complexes.

In this project we researched and optimized an new synthetic route for R-Equol, a molecule that is attracting increasing interest for the medicine because of its phytoestrogenic properties and the chemoprevention of breast cancer. To reach this objective we start, from smaller building blocks, with the synthesis of Daidzein followed by a chemoselective borane reduction to obtain an olefin that will be hydrogenated enantioselectively with a commercial Ir-BARF catalyst. The increasing success of these catalysts even with this genre of substrates has already given good results with different catalysts in both e.e. and yield. For further researches we deuterate the Equol in the aliphatic O-ring and attempt a secondary synthetic route with an hydrogenation using QN-modified Pd.

The present work is part of a research project that involves the study of new ruthenium-based transition metal complexes in order to develop new metal-ligand bifunctional catalysts to employ in homogeneous catalytic systems, in particular in redox reactions such as hydrogenation and dehydrogenation through hydrogen transfer. My project is focused on the optimization of the synthesis of Ru(0) complexes that combines different ligands as tetraphenylcyclopentadienone and N-heterocyclic carbenes and the synthesis of the corresponding cationic complexes by protonation. Furthermore, it is reported the synthesis and characterization of a new cationic complex obtained by methylation of the corresponding neutral complex. All the prepared complexes were employed as catalyst precursors in the transfer hydrogenation of 4-fluoroacetophenone and their performances were investigated in relation to the type of ligands, additives and counterions. The reactivity of these ruthenium complexes was also investigated with the aim of delineate some hypothesis on the reaction mechanism, in particular employing pyridine as a trapping agent. Finally, preliminary studies on the oxidation of benzyl alcohol have been carried out.

In this work, we have examined the activity and selectivity of new catalysts for the single-stage production of methyl isobutyl ketone (MIBK, 4- methyl-2-pentanone) from acetone (both in liquid and gas phase), using a fixed bed reactor operated in the temperature range between 373 and 473 K. The main reaction pathways for the synthesis of MIBK from acetone are given in Fig.1. The first step is the self condensation of acetone to diacetone alcohol (DAA, 4-hydroxy-4-methyl-2-pentanone); the second step is the dehydration of DAA to mesityl oxide (MO, 4-methyl-3-penten-2-one); the final step is the selective hydrogenation of the carbon–carbon double bond of MO to form MIBK. The most commonly observed side reactions are over-condensations and unselective hydrogenations (also shown in Fig.1). Two types of catalysts were studied: i)Pd supported on MgO-SiO2 mixed oxides with ratio of Mg to Si, synthetized using Ohnishi’s method and ii)Pd supported on alumina doped with 5% or 10% of MgO. The different Mg-Si and Mg-Al catalysts were characterized by different techniques (XRD, BET, SEM, NH3-TPD and CO2-TPD) and tested under different conditions in the condensation of acetone to diacetone alcohol and its dehydration to mesityl oxide to enhance the activity. Palladium was chosen as metal component, and its hydrogenation activity was studied. A low hydrogenation activity negatively affects the acetone conversion and promotes the production of mesityl oxide. Hydrogenation conditions being too severe may favor the unwanted hydrogenation of acetone to 2-propanol and of MIBK to methyl isobutyl carbinol (MIBC, 4-methyl-2-pentanol) but this effect is less detrimental to the MIBK selectivity than an unsufficient hydrogenation activity.

The hydrogenation of biomass-derived molecules is a key reaction in upgrading these compounds into chemicals and fuels. The use of catalytic transfer hydrogenation, employing alcohols as hydrogen sources, offers an alternative approach to this process, avoiding the use of H2 under high pressure and precious metal catalysts. In this work, the gas-phase conversion of biomass-derived furfural into furfuryl alcohol and 2-methylfuran was studied, using methanol as the H-transfer agent and CaO-based catalysts. The results obtained with this catalyst were compared with those obtained by using MgO, which due to its basic properties and to its high surface area, at present appears to be among the best basic catalysts used for the conversion of biomass-derived molecules. Pure CaO, despite having a very low surface area, compared to MgO catalyst (5 m2/g vs. 172 m2/g), was shown to reduce furfural into its corresponding unsaturated alcohol at 350°C, thus allowing selective H-transfer from methanol to the substrate. These results highlight the potential application of the H-transfer reaction over CaO based catalysts as an efficient process for the selective reduction of biomass-derived molecules.
