In questo lavoro di tesi sono stati analizzati campioni di orate (Sparus aurata) da acquacoltura allevate con differenti diete, commerciali o innovative, con lo scopo di verificare se le diverse formulazioni dei mangimi hanno un effetto sul profilo molecolare dei pesci. A tal proposito, è stata utilizzata la spettroscopia di Risonanza Magnetica Nucleare (1H NMR) accoppiata ad un approccio metabolomico poiché, rispetto ad altre tecniche, consente la determinazione simultanea di un’ampia classe di metaboliti che caratterizzano il campione attraverso la generazione di spettri che contengono un ampio spettro di informazioni. Questo lavoro di tesi si sposa bene con l’attuale tendenza di sviluppo dell’acquacoltura, che viene vista come uno strumento interessante per fornire cibo e migliorare la sicurezza alimentare, ma allo stesso tempo esso è in accordo anche con la sempre più elevata attenzione verso sostenibilità, qualità e conformità alle richieste dei consumatori, che sono oggi le principali tendenze nel settore alimentare. Negli ultimi anni si è assistito ad un ampio sviluppo dell’acquacoltura e questo ha portato alla necessità di ricercare strategie per migliorare l’efficienza, la produttività, ma anche la sostenibilità di questi sistemi. Questo ha condotto ad un elevato aumento delle ricerche riguardanti questi aspetti dell’acquacoltura, e l’impatto che la dieta ha sul metabolismo dei pesci è uno di questi. I dati spettroscopici sono stati interpretati attraverso analisi statistica multivariata, nello specifico attraverso l’analisi delle componenti principali, PCA, per individuare eventuali differenze significative tra i tre gruppi a seguito della diversa tipologia di alimentazione. Dal presente lavoro di tesi è stato possibile concludere che i campioni vengono distribuiti e classificati in funzione della dieta. Inoltre, è stato possibile individuare i metaboliti che maggiormente determinano questa separazione.

Due to its environmental, safety, health and socio-economic impacts, marine litter has been recognized as a 21st century global challenge, so that it has been included in Descriptor 10 of the EU MSFD. For its morphological features and anthropogenic pressures, the Adriatic Sea is very sensitive to the accumulation of debris, but data are inconsistent and fragmented. This thesis, in the framework of DeFishGear project, intents to assess marine litter on beaches and on seafloor in the Western Adriatic sea, and test if debris ingestion by fish occurs. Three beaches were sampled during two surveys in 2015. Benthic litter monitoring was carried out in the FAO GSA17 during fall 2014, using a rapido trawl. Litter ingestion was investigated through gut contents analysis of 260 fish belonging to 8 commercial species collected in Western Gulf of Venice. Average litter density on beaches was 1.5 items/m2 during spring, and decreased to 0.8 items/m2 in summer. Litter composition was heterogeneous and varied among sites, even if no significant differences were found. Most of debris consisted of plastic sheets, fragments, polystyrene pieces, mussels nets and cottons bud sticks, showing that sources are many and include aquaculture, land-based activities and local users of beaches. Average density of benthic litter was 913 items/Km2 (82 Kg/Km2). Plastic dominated in terms of numbers and weight, and consisted mainly of bags, sheets and mussel nets. The highest density was found close to the coast, and sources driving the major differences in litter distribution were mussel farms and shipping lanes. Litter ingestion occurred in 47% of examined fish, mainly consisting of fibers. Among species, S. pilchardus swallowed almost all debris categories. Findinds may provide a baseline to set the necessary measures to manage and minimize marine litter in the Western Adriatic region and to protect aquatic life from plastic pollution, even accounting the possible implications on human health.

In gilthead seabream aquaculture, the feed supplies in the market is very expensive due to its high content of animal protein. In this respect, spiruline appears to be a valuable substitute to animal and vegetable protein. In this study we performed two experiments. The scope of the first one was to determine the effect of the inclusion of Spirulina platensis hydrolyzed on the physiological state and growth in juveniles of Sparus aurata. A total of 180 individuals were fed for 128 days with three different feeds: control diet, diet with 2% of hydrolyzed microalgae (Sp2), and diet with 4% of hydrolyzed microalgae (Sp4).The experimental groups were tested in triplicate (except control group that was in duplicate). Biometric parameters were registered every two or three weeks. At the end of the experiment blood samples were collected to analyze plasma metabolites. After this we tried to evaluate the anti-oxidant response in animals remained from the first experiment using a toxicological assay with sodium nitrite lasting three days. Fish were divided into control, Spi 2% and Spi 4%, all them with and without NaNO2. Even then, the plasma metabolites data were collected after 24h and 72h. At the end of the first experiment the administration of S. platensis appeared to have a negative impact on growth of S. aurata respect the control feed. Furthermore, the lactate content registered showed a significant difference between the control and the spiruline administration. In the second experiment the spiruline feed showed a glucose and a lactate content with significant differences after 72h of exposition to nitrites respect the control group due to the interaction between nitrites and treatment. S. platensis hydrolyzed 2% and 4% do not seems a good substitution for S. aurata both as a growth enhancer and improver of health metabolic pathways. Its role as a good antioxidant has not been confirmed in these experiments.
