Ilex guayusa, a tree native from the Amazon Rainforest, represents an important part of the culinary traditions and folk medicine of the indigenous tribes. In fact, infusions of different parts of the tree have been used as natural remedies. Particularly, the infusion obtained by the dry leaves of guayusa is a source of phenolic compounds, which are considered as antioxidant substances and have been associated with numerous benefits for human health. Currently, the growing interest of consumers towards healthy food and drinks has led to the rapid spread of this drink. However, the scientific literature about the content of polar compounds in infusion of guayusa leaves is scarce. Therefore, the aims of the present work were to enhance the extraction conditions of phenolic compounds from guayusa leaves by infusion and to characterize them via HPLC-ESI-TOF-MS. To reach these objectives, a Box-Behnken design (BBD) was applied to test the effect of different extraction conditions (time 2, 8 and 14 min), temperature (25, 62.5 and 100 °C) and solid ratio (0.25, 0.375 and 0.50 g) on the sum of phenolic content. On the one hand, the optimal conditions were 1 min infusion, 100 °C and 0.370 g of dry leaves. On the other hand, the 99% of the nine phenolic compounds identified were phenolic acids derivatives from hydroxycinnamic acid and the 1% belonged to the flavonoid family. The major compound was dicaffeoylquinic acid (68%). The difference between the results obtained and those of other researches is probably due to the stochastic nature of the vegetable matrix samples, since their chemical composition is susceptible to multiple factors. To sum up, the use of experimental design provided greater quantity of phenolic compounds than other extraction techniques such as blanching, or only to the oxidation process. Besides, the high resolution of the TOF spectrometer allowed the characterization of new isomers of the compounds previously described.

Il presente elaborato di tesi valuta l’effetto delle condizioni di estrazione dei composti fenolici in un sottoprodotto della lavorazione industriale delle patate, la buccia, con l’obiettivo di poter valorizzare tale residuo di processo per utilizzi futuri. Al fine di ottimizzare le condizioni di estrazione, si è deciso di utilizzare il disegno sperimentale Box-Behnken (DBB) mediante la variazione di tre fattori (tempo, tipologia di solvente, rapporto campione/solvente) e per un totale di 15 determinazioni. I dati risultanti dalla determinazione dei composti fenolici totali e dell’attività antiossidante hanno messo in evidenza come la buccia di patate, sottoprodotto del processo di lavorazione industriale, sia un prodotto interessante per il proprio contenuto in composti fenolici ad elevata attività antiossidante. Tuttavia, la scelta delle condizioni ottimali di estrazione è fondamentale al fine di evitare la sottostima del contenuto in composti bioattivi, che deriverebbe da una incompleta estrazione degli stessi dalla matrice alimentare. Il disegno Box-Behnken ha permesso di stabilire le condizioni ottimali di estrazione dei composti fenolici e per questo sará utilizzato in futuro per lo screening di diversi campioni procedenti dalla produzione industriale. L’estrazione ottimale dei composti di interesse ha comunque evidenziato come la buccia di patata sia una fonte di composti fenolici ad attivitá antiossidante che potrá essere utilizzata sia come ingrediente alimentare che in campo nutraceutico.

A new study on suspension bridges has been prompted by the big disaster of the Tacoma Narrow Bridge at half its design speed. The aerodynamic instability of long-span bridges has been studied using wind tunnel tests. As a result of improved aerodynamic performance from the geometrical configuration of the bridge deck, the aerodynamic criteria for suspension and cable-stayed bridges have become well established in recent years, thereby allowing longer bridge spans to be developed. Although the Messina Strait Bridge has yet to be constructed, we are looking forward to evaluating the impact of different deck cross-sections on both aerodynamic stability and cost reduction. To further improve the aerodynamic characteristics of long-span suspension bridges, an optimized multi-box bridge deck model with two side decks for traffic lanes, two middle railway decks, and three gaps separating them has been proposed aerodynamic performance has been experimentally verified. 1:80 scale wind tunnel tests have been conducted. According to the current MIDAS Model, the first torsional and the first vertical frequency ratios are 1.27787 and 1.36[1] respectively. It is the torsional/vertical frequency ratio, combined with the deck aerodynamic properties, that determines the wind response properties of the bridge for the most dangerous possible form of aeroelastic instability. The classic flutter is caused by the coupling of torsional and vertical modes. Stabilizing cables to the deck could be a solution to this classic flutter by reducing lateral displacement of the deck and increasing frequency ratios. Stabilizing cables will be installed on the deck in three different orientations: vertical, inclined, and horizontal, with diameters of 80 cm, 60 cm, and 40 cm in each orientation respectively. An overview of the research undertaken on this topic will be presented, as well as the most important findings.

This master's thesis investigates different aspects of Dual-Active-Bridge (DAB) Converter and extends aspects further to Multi-Active-Bridges (MAB). The thesis starts with an overview of the applications of the DAB and MAB and their importance. The analytical part of the thesis includes the derivation of the peak and RMS currents, which is required for finding the losses present in the system. The power converters, considered in this thesis are DAB, Triple-Active Bridge (TAB) and Quad-Active Bridge (QAB). All the theoretical calculations are compared with the simulation results from PLECS software for identifying the correctness of the reviewed and developed theory. The Hardware-in-the-Loop (HIL) simulation is conducted for checking the control operation in real-time with the help of the RT box from the Plexim. Additionally, as in real systems digital signal processor (DSP), system-on-chip or field programmable gate array is employed for the control of the power electronic systems, and the execution of the control in the real-time simulation (RTS) conducted is performed by DSP.
