Gli oli microbici stanno ricevendo sempre più attenzioni come possibile alternativa agli oli vegetali, nel processo di sostituzione dei combustibili fossili. Tuttavia, diversi aspetti necessitano di essere ottimizzati al fine di ottenere oli economicamente competitivi e con caratteristiche chimico-fisiche desiderate. In questa ricerca, sono stati utilizzati due differenti approcci per poter realizzare l’obiettivo preposto. Il primo, si è basato sull’ingegnerizzazione genetica del lievito C. oleaginous, al fine di incrementare la produttività di lipidi e modificare la composizione dei trigliceridi (TAG) sintetizzati. Un protocollo basato su una trasformazione genetica mediata da Agrobacterium è stato utilizzato per sovraesprimere la diacilglicerol trasnferasi (DGA1), l’enzima responsabile dell’ultimo step della sintesi dei TAG, e la Δ9-desaturasi, l’enzima che catalizza la conversione dell’acido stearico (C18:0) in acido oleico (C18:1). La selezione di colonie positive e l’analisi dei mutanti ottenuti ha confermato la buona riuscita della trasformazione. Il secondo approccio ha mirato a studiare l’influenza sulla crescita e sul profilo di lipidi accumulati da C. oleaginous da parte di diversi acidi grassi volatili (VFAs), una materia prima ottenibile da trattamenti di scarti industriali. A questo proposito, sono state utilizzate fermentazioni fed-batch su scala da 1-L basate su glucosio e miscele sintetiche di acido acetico e di VFAs come fonte di carbonio. L’utilizzo simultaneo di acido acetico e acidi secondari ha mostrato come sia possibile stimolare il metabolismo microbico al fine di incrementare l'accumulo di oli e ottenere una composizione chimica lipidica desiderata.

This thesis is part of the fields of Material Physics and Organic Electronics and aims to determine the charge carrier density and mobility in the hydrated conducting polymer–polyelectrolyte blend PEDOT:PSS. This kind of material combines electronic semiconductor functionality with selective ionic transport, biocompatibility and electrochemical stability in water. This advantageous material properties combination makes PEDOT:PSS a unique material to build organic electrochemical transistors (OECTs), which have relevant application as amplifying transducers for bioelectronic signals. In order to measure charge carrier density and mobility, an innovative 4-wire, contact independent characterization technique was introduced, the electrolyte-gated van der Pauw (EgVDP) method, which was combined with electrochemical impedance spectroscopy. The technique was applied to macroscopic thin film samples and micro-structured PEDOT:PSS thin film devices fabricated using photolithography. The EgVDP method revealed to be effective for the measurements of holes’ mobility in hydrated PEDOT:PSS thin films, which resulted to be <μ>=(0.67±0.02) cm^2/(V*s). By comparing this result with 2-point-probe measurements, we found that contact resistance effects led to a mobility overestimation in the latter. Ion accumulation at the drain contact creates a gate-dependent potential barrier and is discussed as a probable reason for the overestimation in 2-point-probe measurements. The measured charge transport properties of PEDOT:PSS were analyzed in the framework of an extended drift-diffusion model. The extended model fits well also to the non-linear response in the transport characterization and results suggest a Gaussian DOS for PEDOT:PSS. The PEDOT:PSS-electrolyte interface capacitance resulted to be voltage-independent, confirming the hypothesis of its morphological origin, related to the separation between the electronic (PEDOT) and ionic (PSS) phases in the blend.

Questo lavoro di tesi riguarda lo studio e l’implementazione di un algoritmo di multiple kernel learning (MKL) per la classificazione e la regressione di dati di neuroimaging ed, in particolare, di grafi di connettività funzionale. Gli algoritmi di MKL impiegano una somma pesata di vari kernel (ovvero misure di similarità) e permettono di selezionare le features utili alla discriminazione delle istanze durante l’addestramento del classificatore/regressore stesso. L’aspetto innovativo introdotto in questa tesi è stato lo studio di un nuovo kernel tra grafi di connettività funzionale, con la particolare caratteristica di conservare l’informazione relativa all’importanza di ogni singola region of interest (ROI) ed impiegando la norma lp come metodo per l’aggiornamento dei pesi, al fine di ottenere soluzioni sparsificate. L’algoritmo è stato validato utilizzando mappe di connettività sintetiche ed è stato applicato ad un dataset formato da 32 pazienti affetti da deterioramento cognitivo lieve e malattia dei piccoli vasi, di cui 16 sottoposti a riabilitazione cognitiva tra un’esame di risonanza ma- gnetica funzionale di baseline e uno di follow-up. Le mappe di con- nettività sono state ottenute con il toolbox CONN. Il classificatore è riuscito a discriminare i due gruppi di pazienti in una configurazione leave-one-out annidata con un’accuratezza dell’87.5%. Questo lavoro di tesi è stato svolto durante un periodo di ricerca presso la School of Computer Science and Electronic Engineering dell’University of Essex (Colchester, UK).

One of the most diffused electronic device is the field effect transistor (FET), contained in number of billions in each electronic device. Organic optoelectronics is an emerging field that exploits the unique properties of conjugated organic materials to develop new applications that require a combination of performance, low cost and processability. Organic single crystals are the material with best performances and purity among the variety of different form of organic semiconductors. This thesis is focused on electrical and optical characterization of Rubrene single crystal bulk and thin films. Rubrene bulk is well known but for the first time we studied thin films. The first Current-voltage characterization has been performed for the first time on three Rubrene thin films with three different thickness to extract the charge carriers mobility and to assess its crystalline structure. As results we see that mobility increase with thickness. Field effect transistor based on Rubrene thin films on $SiO_2$ have been characterize by current-voltage (I-V) analyses (at several temperatures) and reveals a hopping conduction. Hopping behavior probably is due to the lattice mismatch with the substrate or intrinsic defectivity of the thin films. To understand effects of contact resistance we tested thin films with the Transmission Line Method (TLM) method. The TLM method revealeds that contact resistance is negligible but evidenced a Schottky behavior in a limited but well determined range of T. To avoid this effect we carried out annealing treatment after the electrode evaporation iswe performed a compete I-V characterization as a function of in temperature to extract the electronic density of states (DOS) distribution through the Space Charge Limited Current (SCLC) method. The results show a DOS with an exponential trenddistribution, as expected. The measured mobility of thin films is about 0.1cm^2/Vs and it increases with the film thickness. Further studies are necessary to investigate the reason and improve performances. From photocurrent spectrum we calculated an Eg of about 2.2eV and both thin films and bulk have a good crystal order. Further measurement are necessary to solve some open problems

In this work, a Hardware-in-the-loop test bench is designed. The bench is used to test the behaviour of an electronic control unit used in Maserati to control the dynamics of an air spring system. First the mathematical model of the plant has been defined, then the simulation enviroment and the test environment have been set up. The performed tests succesfully highlighted some bugs in the device under test.

This thesis focuses on the investigation and the implementation of different observers for the estimation of the roll angle of a motorbike. The central core of the activity is applying a Model-Based design in order to outline, simulate and implement the filters with the aim of a final comparison of the performances. This approach is crucially underlined among the chapters that articulate this document: first the design and tuning of an Extended Kalman Filter and a Complementary Filter in a pure simulation environment emphasize the most accurate choice for the particular problem. After this, several steps were performed in order to move from the aforementioned simulation environment to a real hardware application. In conclusion, several sensor configurations were tested and compared in order to highlight which sensor suite gives the best performances.

In recent years, energy modernization has focused on smart engineering advancements. This entails designing complicated software and hardware for variable-voltage digital substations. A digital substation consists of electrical and auxiliary devices, control and monitoring devices, computers, and control software. Intelligent measurement systems use digital instrument transformers and IEC 61850-compliant information exchange protocols in digital substations. Digital instrument transformers used for real-time high-voltage measurements should combine advanced digital, measuring, information, and communication technologies. Digital instrument transformers should be cheap, small, light, and fire- and explosion-safe. These smaller and lighter transformers allow long-distance transmission of an optical signal that gauges direct or alternating current. Cost-prohibitive optical converters are a problem. To improve the tool's accuracy, amorphous alloys are used in the magnetic circuits and compensating feedback. Large-scale voltage converters can be made cheaper by using resistive, capacitive, or hybrid voltage dividers. In known electronic voltage transformers, the voltage divider output is generally on the low-voltage side, facilitating power supply organization. Combining current and voltage transformers reduces equipment size, installation, and maintenance costs. These two gadgets cost less together than individually. To increase commercial power metering accuracy, current and voltage converters should be included into digital instrument transformers so that simultaneous analogue-to-digital samples are obtained. Multichannel ADC microcircuits with synchronous conversion start allow natural parallel sample drawing. Digital instrument transformers are created adaptable to substation operating circumstances and environmental variables, especially ambient temperature. An embedded microprocessor auto-diagnoses and auto-calibrates the proposed digital instrument transformer.

In this thesis the design of a pressure regulation system for space propulsion engines (electric and cold gas) has been performed. The Bang-Bang Control (BBC) method has been implemented through the open/close command on a solenoid valve, and the mass flow rate of the propellant has been fixed with suitable flow restrictors. At the beginning, research for the comparison between mechanical and electronic (for BBC) pressure regulators has been performed, which resulted in enough advantages for the selection of the second valve type. The major advantage is about the possibility to have a variable outlet pressure with a variable inlet pressure through a simple remote command, while in mechanical pressure regulators the ratio between inlet and outlet pressures must be mechanically settled. Different pressure control schemes have been analyzed, changing number of solenoid valves, flow restrictors and plenums. For each scheme the valve’s frequencies were evaluated with simplified mathematical models and with the use of simulators implemented on Python; the results obtained from those two methods matched quiet well. From all the schemes it was possible to observe varying frequency and duty cycle, for changes in different parameters. This results, after experimental checks, can be used to design the control system for a given total number of cycles that a specific solenoid valve can guarantee. Finally, tests were performed and it was possible to verify the goodness of the control system. Moreover from the tests it was possible to deduce some tips in order to optimize the running of the simulator.

Electric vehicles and electronic components inside the vehicle are becoming increasingly important. The software as well starts to have a significant impact on modern high-end cars therefore a careful validation process needs to be implemented with the aim of having a bug free product when it is released. The software complexity increases and thus also the testing phases is more demanding. Test can be troublesome and, in some cases, boring and easy. The intelligence can be moved in test definition and writing rather than on test execution. The aim of this document is to start the definition of an automatic modular testing system capable to execute test cycles on systems that interacts with the CAN networks and with DUT that can be touched with a robotic arm. The document defines a first version of the system, in particular the hardware interface part with the aim of taking logs and execute test in an automated fashion with the test engineer can have a higher focus on the test definition and analysis rather than execution.
