Organotin compounds are worldwide diffused environmental contaminants, mainly as consequence of their extensive past use as biocides in antifouling paints. In spite of law restrictions, due to unwanted effects, organotin still persist in waters, being poorly degraded, easily resuspended from sediments and bioaccumulated in exposed organisms. The widespread toxicity and the possible threat to humans, likely to be organotin-exposed through contaminated seafood, make organotin interactions with biomolecules an intriguing biochemical topic, apart from a matter of ecotoxicological concern. Among organotins, tributyltin (TBT) is long known as the most dangerous and abundant chemical species in the Mediterranean Sea. Due to its amphiphilic nature, provided by three lipophilic arms and an electrophilic tin core, TBT can be easily incorporated in biomembranes and affect their functionality. Accordingly, it is known as a membrane-active toxicant and a mitochondrial poison. Up to now the molecular action modes of TBT are still partially unclear and poorly explored in bivalve mollusks, even if the latter play a not neglectable role in the marine trophic chain and efficiently accumulate organotins. The bivalve mollusk Mytilus galloprovincialis, selected for all experiments, is widely cultivated in the Mediterranean and currently used in ecotoxicological studies. Most work of this thesis was devoted to TBT effects on mussel mitochondria, but other possible targets of TBT were also considered. A great deal of literature points out TBT as endocrine disrupter and the masculinization of female marine gastropods, the so-called imposex, currently signals environmental organotin contamination. The hormonal status of TBT-exposed mussels and the possible interaction between hormones and contaminants in modulating microsomal hydroxilases, involved in steroid hormone and organotin detoxification, were the research topics in the period spent in Barcelona (Marco Polo fellowship). The variegated experimental approach, which consisted of two exposure experiments and in vitro tests, and the choice of selected tissues of M. galloprovincialis, the midgut gland for mitochondrial and microsomal preparations for subsequent laboratory assays and the gonads for the endocrine evaluations, aimed at drawing a clarifying pattern on the molecular mechanisms involved in organotin toxicity. TBT was promptly incorporated in midgut gland mitochondria of adult mussels exposed to 0.5 and 1.0 μg/L TBT, and partially degraded to DBT. TBT incorporation was accompanied by a decrease in the mitochondrial oligomycin-sensitive Mg-ATPase activity, while the coexistent oligomycin-insensitive fraction was unaffected. Mitochondrial fatty acids showed a clear rise in n-3 polyunsaturated fatty acids after 120 hr of TBT exposure, mainly referable to an increase in 22:6 level. TBT was also shown to inhibit the ATP hydrolytic activity of the mitochondrial F1FO complex in vitro and to promote an apparent loss of oligomycin sensitivity at higher than 1.0 μM concentration. The complex dose-dependent profile of the inhibition curve lead to the hypothesis of multiple TBT binding sites. At lower than 1.0 μM TBT concentrations the non competitive enzyme inhibition by TBT was ascribed to the non covalent binding of TBT to FO subunit. On the other hand the observed drop in oligomycin sensitivity at higher than 1.0 μM TBT could be related to the onset of covalent bonds involving thiolic groups on the enzyme structure, apparently reached only at high TBT levels. The mitochondrial respiratory complexes were in vitro affected by TBT, apart from the cytocrome c oxidase which was apparently refractory to the contaminant. The most striking inhibitory effect was shown on complex I, and ascribed to possible covalent bonds of TBT with –SH groups on the enzyme complexes. This mechanism, shouldered by the progressive decrease of free cystein residues in the presence of increasing TBT concentrations, suggests that the onset of covalent tin-sulphur bonds in distinct protein structures may constitute the molecular basis of widespread TBT effects on mitochondrial complexes. Energy production disturbances, in turn affecting energy consuming mechanisms, could be involved in other cellular changes. Mussels exposed to a wide range of TBT concentrations (20 - 200 and 2000 ng/L respectively) did not show any change in testosterone and estrogen levels in mature gonads. Most hormones were in the non-biologically active esterified form both in control and in TBT-treated mussels. Probably the endocrine status of sexually mature mussels could be refractory even to high TBT doses. In mussel digestive gland the high biological variability of microsomal 7-benzyloxy-4-trifluoromethylcoumarin-O-Debenzyloxylase (BFCOD) activity, taken as a measure of CYP3A-like efficiency, probably concealed any enzyme response to TBT exposure. On the other hand the TBT-driven enhancement of BFCOD activity in vitro was once again ascribed to covalent binding to thiol groups which, in this case, would stimulate the enzyme activity. In mussels from Barcelona harbour, a highly contaminated site, the enzyme showed a decreased affinity for the 7-benzyloxy-4-trifluoromethylcoumarin (BCF) substrate with respect to mussel sampled from Ebro Delta, a non-polluted marine site. Contaminant exposure may thus alter the kinetic features of enzymes involved in detoxification mechanisms. Contaminants and steroid hormones were clearly shown to mutually interact in the modulation of detoxification mechanisms. The xenoestrogen 17α-ethylenyl estradiol (EE2) displayed a non-competitive mixed inhibition of CYP3A-like activity by a preferential bond to the free enzyme both in Barcelona harbour and Ebro Delta mussels. The possible interaction with co-present contaminants in Barcelona harbour mussels apparently lessened the formation of the ternary complex enzyme-EE2-BCF. The whole of data confirms TBT as membrane toxicant in mussels as in other species and stresses TBT covalent binding to protein thiols as a widespread mechanism of membrane-bound-enzyme activity modulation by the contaminant.

The present issue analyses bluefin tuna (Thynnus thunnus) and swordfish (Xiphias glaudis) diet, caught by professional long-line fishing in the middle Adriatic Sea (Pomo pit). These species represent apex predators in pelagic environment that may play key roles in determining food web structure and ecosystem dynamics. The studies about their feedings habits, based upon stomach contents analysis, are important for the comprehension of biological and ecological interaction. Over the years, many studies have been performed on the diet of tuna and swordfish in the Mediterranean Sea. This research is based on a fairly wide number of analyzed stomach contents, in comparison with the previous ones. In this work, the analysis of 340 stomach contents of bluefin tuna caught by long-line in the central Adriatic sea confirms in general the opportunistic behaviour of this species. Finding support the hypothesis that Adriatic tuna and swordfish chase their food over a wide bathymetric zone and probably near the surface at night. No indication of food preference respect to size of predator or sample season are found. It seems that the two species are able to cohabit because their trophic niche are not overlapped, changing during the time and the vertical and horizontal space.

Tuta absoluta (Meyrick) è un lepidottero originario dell’America meridionale, infeudato a pomodoro e ad altre solanacee coltivate e spontanee. Con l’attività trofica le larve causano mine fogliari e gallerie nei frutti, con conseguenti ingenti danni alle colture. T. absoluta è stato segnalato per la prima volta in Italia nel 2008 e in Piemonte nel 2009. Pertanto le ricerche sono state condotte per rilevarne la distribuzione in Piemonte, studiarne l’andamento di popolazione in condizioni naturali e controllate, e valutare l’efficacia di differenti mezzi di lotta al fine di definire le strategie di difesa. Il monitoraggio, condotto nel 2010, ha evidenziato come T. absoluta sia ormai largamente diffuso sul territorio regionale già pochi mesi dopo la segnalazione. L’insetto ha mostrato di prediligere condizioni climatiche più miti; infatti è stato ritrovato con maggiore frequenza nelle aree più calde. Il fitofago ha raggiunto densità di popolazione elevate a partire dalla seconda metà dell’estate, a ulteriore dimostrazione che, in una regione a clima temperato come il Piemonte, T. absoluta dà origine a infestazioni economicamente rilevanti solo dopo il culmine della stagione estiva. Per definire le strategie di lotta, sono state condotte prove in laboratorio, semi-campo e campo volte a valutare la tossicità nei confronti del lepidottero di preparati a base di emamectina benzoato, rynaxypyr, spinosad e Bacillus thuringiensis Berliner. In campo è stata verificata anche l’efficacia del miride dicifino Macrolophus pygmaeus (Rambur), reperibile in commercio. In tutte le prove, è stata riscontrata una maggiore efficacia di rynaxypyr ed emamectina benzoato. In campo M. pygmaeus ha mostrato difficoltà d’insediamento ed è stato in grado di contenere efficacemente il fitofago soltanto con bassi livelli d’infestazione. Per contro è stata costantemente osservata la presenza naturale di un altro miride dicifino Dicyphus errans (Wolff), che in laboratorio ha mostrato di non essere particolarmente disturbato dalle sostanze saggiate.

The human airway epithelium is a pseudostratified heterogenous layer comprised of cili-ated, secretory, intermediate and basal cells. As the stem/progenitor population of the airway epi-thelium, airway basal cells differentiate into ciliated and secretory cells to replenish the airway epithelium during physiological turnover and repair. Transcriptome analysis of airway basal cells revealed high expression of vascular endothelial growth factor A (VEGFA), a gene not typically associated with the function of this cell type. Using cultures of primary human airway basal cells, we demonstrate that basal cells express all of the 3 major isoforms of VEGFA (121, 165 and 189) but lack functional expression of the classical VEGFA receptors VEGFR1 and VEGFR2. The VEGFA is actively secreted by basal cells and while it appears to have no direct autocrine function on basal cell growth and proliferation, it functions in a paracrine manner to activate MAPK signaling cascades in endothelium via VEGFR2 dependent signaling pathways. Using a cytokine- and serum-free co-culture system of primary human airway basal cells and human endothelial cells revealed that basal cell secreted VEGFA activated endothelium to ex-press mediators that, in turn, stimulate and support basal cell proliferation and growth. These data demonstrate novel VEGFA mediated cross-talk between airway basal cells and endothe-lium, the purpose of which is to modulate endothelial activation and in turn stimulate and sustain basal cell growth.

It is well known that many realistic mathematical models of biological systems, such as cell growth, cellular development and differentiation, gene expression, gene regulatory networks, enzyme cascades, synaptic plasticity, aging and population growth need to include stochasticity. These systems are not isolated, but rather subject to intrinsic and extrinsic fluctuations, which leads to a quasi equilibrium state (homeostasis). The natural framework is provided by Markov processes and the Master equation (ME) describes the temporal evolution of the probability of each state, specified by the number of units of each species. The ME is a relevant tool for modeling realistic biological systems and allow also to explore the behavior of open systems. These systems may exhibit not only the classical thermodynamic equilibrium states but also the nonequilibrium steady states (NESS). This thesis deals with biological problems that can be treat with the Master equation and also with its thermodynamic consequences. It is organized into six chapters with four new scientific works, which are grouped in two parts: (1) Biological applications of the Master equation: deals with the stochastic properties of a toggle switch, involving a protein compound and a miRNA cluster, known to control the eukaryotic cell cycle and possibly involved in oncogenesis and with the propose of a one parameter family of master equations for the evolution of a population having the logistic equation as mean field limit. (2) Nonequilibrium thermodynamics in terms of the Master equation: where we study the dynamical role of chemical fluxes that characterize the NESS of a chemical network and we propose a one parameter parametrization of BCM learning, that was originally proposed to describe plasticity processes, to study the differences between systems in DB and NESS.

This PhD Thesis includes five main parts on diverse topics. The first two parts deal with the trophic ecology of wolves in Italy consequently to a recent increase of wild ungulates abundance. Data on wolf diet across time highlighted how wild ungulates are important food resource for wolves in Italy. Increasing wolf population, increasing numbers of wild ungulates and decreasing livestock consume are mitigating wolf-man conflicts in Italy in the near future. In the third part, non-invasive genetic sampling techniques were used to obtain genotypes and genders of about 400 wolves. Thus, wolf packs were genetically reconstructed using diverse population genetic and parentage software. Combining the results on pack structure and genetic relatedness with sampling locations, home ranges of wolf packs and dispersal patterns were identified. These results, particularly important for the conservation management of wolves in Italy, illustrated detailed information that can be retrieved from genetic identification of individuals. In the fourth part, wolf locations were combined with environmental information obtained as GIS-layers. Modern species distribution models (niche models) were applied to infer potential wolf distribution and predation risk. From the resulting distribution maps, information pastures with the highest risk of depredation were derived. This is particularly relevant as it allows identifying those areas under danger of carnivore attack on livestock. Finally, in the fifth part, habitat suitability models were combined with landscape genetic analysis. On one side landscape genetic analyses on the Italian wolves provided new information on the dynamics and connectivity of the population and, on the other side, a profound analysis of the effects that habitat suitability methods had on the parameterization of landscape genetic analyses was carried out to contributed significantly to landscape genetic theory.

Despite extensive studies focus mainly on sexual reproductive characteristics in tropical scleractinian species, there is limited knowledge on temperate regions. The Mediterranean is a biodiversity hotspot under intense pressure from anthropogenic impacts. Climatic models further predict that the Mediterranean basin will be one of the most impacted regions by the ongoing warming trend. This makes it a potential model of more global patterns to occur in the world’s marine biota, and a natural focus of interest for research on climate. The present research contributed to increase data on reproductive modes and sexuality of temperate scleractinian corals, highlighting their developmental plasticity, showing different forms of propagation and different responses to environmental change. For the first time, sexuality and reproductive mode in Caryophyllia inornata were determined. An unusual embryogenesis without a clear seasonal pattern was observed, suggesting the possibility of an asexual origin. Sexual reproduction of Astroides calycularis was governed by annual changes in seawater temperature, as observed for other Mediterranean dendrophylliids. Defining the reproductive biology of these species is the starting point for studying their potential response to variations of environmental parameters, on a global climate change context. The results on the influence of temperature on reproductive output of the zooxanthellate (symbiosis with unicellular algae) Balanophyllia europaea and the non-zooxanthellate Leptopsammia pruvoti suggest that the latter may be quite tolerant to temperature increase, since the zooxanthellate species resulted less efficient at warm temperatures. A possible explanation could be related to their different trophic system. In B. europaea thermal tolerance is primarily governed by the symbiotic algae, making it more sensitive to temperature changes. On the contrary, the absence of symbionts in L. pruvoti might make it more resistant to temperature. In a progressively warming Mediterranean, the efficiency on scleractinian reproduction could be influenced in different ways, reflecting their extraordinary adaptability.

Lo studio dei processi biogeochimici che avvengono all’interfaccia acqua-sedimento riveste grande importanza per comprendere quali fattori ambientali siano responsabili di un eventuale modifica nel bilancio del carbonio organico e di altri elementi maggiori o minori e può` fornire un' indicazione su quali siano le aree più sensibili a tali processi. In questo studio sono stati analizzati i meccanismi che guidano la mineralizzazione della sostanza organica in aree caratterizzate da differenti condizioni idrodinamiche, batimetriche e trofiche nel Mediterraneo centrale. In particolare sono state prelevate carote di sedimento e analizzate le acque interstiziali in siti localizzati nell'Adriatico centro-meridionale, caratterizzati da basse profondità, alti tassi di sedimentazione e elevati apporti di sostanza organica, e in siti localizzati nello Ionio centro-settentrionale, caratterizzati da profondità crescenti, minori tassi di sedimentazione e ridotti apporti fluviali. L'analisi dei processi di degradazione della sostanza organica evidenzia differenze regionali tra il bacino adriatico e quello ionico: processi di mineralizzazione ossica e subossica appaiono intensi nei sedimenti adriatici, diversamente il bacino ionico appare caratterizzato principalmente da processi di degradazione ossica della sostanza organica. Inoltre, relativamente ai flussi bentici di Carbonio Inorganico Disciolto (DIC) flussi inversi sono stati registrati nei due bacini: i sedimenti adriatici si comportano come sourse di DIC, mentre i sedimenti Ionici si comportano come dei sink di DIC suggerendo una possibile precipitazione di carbonati nel bacino ionico.

An appropriate management of fisheries resources can only be achieved with the continuous supply of information on the structure and biology of populations, in order to predict the temporal fluctuations. This study supports the importance of investigating the bio-ecology of increasingly exploited and poorly known species, such as gurnards (Osteichthyes, Triglidae) from Adriatic Sea (Mediterranean), to quantify their ecological role into marine community. It also focuses on investigate inter and intra-specific structuring factor of Adriatic population. These objectives were achieved by: 1) investigating aspects of the population dynamics; 2) studying the feeding biology through the examination of stomach contents; 3) using sagittal otoliths as potential marker of species life cycle; 4) getting preliminary data on mDNA phylogeny. Gurnards showed a specie-specific “critical size” coinciding with the start of sexual maturity, the tendency to migrate to greater depths, a change of diet from crustaceans to fish and an increase of variety of food items eaten. Distribution of prey items, predator size range and depth distribution were the main dimensions that influence the breadth of trophic niche and the relative difference amongst Adriatic gurnards. Several feeding preferences were individuated and a possible impact among bigger-size gurnards and other commercial fishes (anchovy, gadoids) and Crustacea (such as mantis prawn and shrimps) were to be necessary considered. Otolith studies showed that gurnard species have a very fast growth despite other results in other areas; intra-specific differences and the increase in the variability of otolith shape, sulcus acusticus shape, S:O ratios, sulcus acusticus external crystals arrangement were shown between juveniles and adults and were linked to growth (individual genetic factors) and to environmental conditions (e.g. depth and trophic niche distribution). In order to facilitate correct biological interpretation of data, molecular data were obtained for comparing morphological distance to genetic ones.
