Interactive theorem provers (ITP for short) are tools whose final aim is to certify proofs written by human beings. To reach that objective they have to fill the gap between the high level language used by humans for communicating and reasoning about mathematics and the lower level language that a machine is able to “understand” and process. The user perceives this gap in terms of missing features or inefficiencies. The developer tries to accommodate the user requests without increasing the already high complexity of these applications. We believe that satisfactory solutions can only come from a strong synergy between users and developers. We devoted most part of our PHD designing and developing the Matita interactive theorem prover. The software was born in the computer science department of the University of Bologna as the result of composing together all the technologies developed by the HELM team (to which we belong) for the MoWGLI project. The MoWGLI project aimed at giving accessibility through the web to the libraries of formalised mathematics of various interactive theorem provers, taking Coq as the main test case. The motivations for giving life to a new ITP are: • study the architecture of these tools, with the aim of understanding the source of their complexity • exploit such a knowledge to experiment new solutions that, for backward compatibility reasons, would be hard (if not impossible) to test on a widely used system like Coq. Matita is based on the Curry-Howard isomorphism, adopting the Calculus of Inductive Constructions (CIC) as its logical foundation. Proof objects are thus, at some extent, compatible with the ones produced with the Coq ITP, that is itself able to import and process the ones generated using Matita. Although the systems have a lot in common, they share no code at all, and even most of the algorithmic solutions are different. The thesis is composed of two parts where we respectively describe our experience as a user and a developer of interactive provers. In particular, the first part is based on two different formalisation experiences: • our internship in the Mathematical Components team (INRIA), that is formalising the finite group theory required to attack the Feit Thompson Theorem. To tackle this result, giving an effective classification of finite groups of odd order, the team adopts the SSReflect Coq extension, developed by Georges Gonthier for the proof of the four colours theorem. • our collaboration at the D.A.M.A. Project, whose goal is the formalisation of abstract measure theory in Matita leading to a constructive proof of Lebesgue’s Dominated Convergence Theorem. The most notable issues we faced, analysed in this part of the thesis, are the following: the difficulties arising when using “black box” automation in large formalisations; the impossibility for a user (especially a newcomer) to master the context of a library of already formalised results; the uncomfortable big step execution of proof commands historically adopted in ITPs; the difficult encoding of mathematical structures with a notion of inheritance in a type theory without subtyping like CIC. In the second part of the manuscript many of these issues will be analysed with the looking glasses of an ITP developer, describing the solutions we adopted in the implementation of Matita to solve these problems: integrated searching facilities to assist the user in handling large libraries of formalised results; a small step execution semantic for proof commands; a flexible implementation of coercive subtyping allowing multiple inheritance with shared substructures; automatic tactics, integrated with the searching facilities, that generates proof commands (and not only proof objects, usually kept hidden to the user) one of which specifically designed to be user driven.

The Assimilation in the Unstable Subspace (AUS) was introduced by Trevisan and Uboldi in 2004, and developed by Trevisan, Uboldi and Carrassi, to minimize the analysis and forecast errors by exploiting the flow-dependent instabilities of the forecast-analysis cycle system, which may be thought of as a system forced by observations. In the AUS scheme the assimilation is obtained by confining the analysis increment in the unstable subspace of the forecast-analysis cycle system so that it will have the same structure of the dominant instabilities of the system. The unstable subspace is estimated by Breeding on the Data Assimilation System (BDAS). AUS- BDAS has already been tested in realistic models and observational configurations, including a Quasi-Geostrophicmodel and a high dimensional, primitive equation ocean model; the experiments include both fixed and“adaptive”observations. In these contexts, the AUS-BDAS approach greatly reduces the analysis error, with reasonable computational costs for data assimilation with respect, for example, to a prohibitive full Extended Kalman Filter. This is a follow-up study in which we revisit the AUS-BDAS approach in the more basic, highly nonlinear Lorenz 1963 convective model. We run observation system simulation experiments in a perfect model setting, and with two types of model error as well: random and systematic. In the different configurations examined, and in a perfect model setting, AUS once again shows better efficiency than other advanced data assimilation schemes. In the present study, we develop an iterative scheme that leads to a significant improvement of the overall assimilation performance with respect also to standard AUS. In particular, it boosts the efficiency of regime’s changes tracking, with a low computational cost. Other data assimilation schemes need estimates of ad hoc parameters, which have to be tuned for the specific model at hand. In Numerical Weather Prediction models, tuning of parameters — and in particular an estimate of the model error covariance matrix — may turn out to be quite difficult. Our proposed approach, instead, may be easier to implement in operational models.

The vast majority of known proteins have not yet been experimentally characterized and little is known about their function. The design and implementation of computational tools can provide insight into the function of proteins based on their sequence, their structure, their evolutionary history and their association with other proteins. Knowledge of the three-dimensional (3D) structure of a protein can lead to a deep understanding of its mode of action and interaction, but currently the structures of <1% of sequences have been experimentally solved. For this reason, it became urgent to develop new methods that are able to computationally extract relevant information from protein sequence and structure. The starting point of my work has been the study of the properties of contacts between protein residues, since they constrain protein folding and characterize different protein structures. Prediction of residue contacts in proteins is an interesting problem whose solution may be useful in protein folding recognition and de novo design. The prediction of these contacts requires the study of the protein inter-residue distances related to the specific type of amino acid pair that are encoded in the so-called contact map. An interesting new way of analyzing those structures came out when network studies were introduced, with pivotal papers demonstrating that protein contact networks also exhibit small-world behavior. In order to highlight constraints for the prediction of protein contact maps and for applications in the field of protein structure prediction and/or reconstruction from experimentally determined contact maps, I studied to which extent the characteristic path length and clustering coefficient of the protein contacts network are values that reveal characteristic features of protein contact maps. Provided that residue contacts are known for a protein sequence, the major features of its 3D structure could be deduced by combining this knowledge with correctly predicted motifs of secondary structure. In the second part of my work I focused on a particular protein structural motif, the coiled-coil, known to mediate a variety of fundamental biological interactions. Coiled-coils are found in a variety of structural forms and in a wide range of proteins including, for example, small units such as leucine zippers that drive the dimerization of many transcription factors or more complex structures such as the family of viral proteins responsible for virus-host membrane fusion. The coiled-coil structural motif is estimated to account for 5-10% of the protein sequences in the various genomes. Given their biological importance, in my work I introduced a Hidden Markov Model (HMM) that exploits the evolutionary information derived from multiple sequence alignments, to predict coiled-coil regions and to discriminate coiled-coil sequences. The results indicate that the new HMM outperforms all the existing programs and can be adopted for the coiled-coil prediction and for large-scale genome annotation. Genome annotation is a key issue in modern computational biology, being the starting point towards the understanding of the complex processes involved in biological networks. The rapid growth in the number of protein sequences and structures available poses new fundamental problems that still deserve an interpretation. Nevertheless, these data are at the basis of the design of new strategies for tackling problems such as the prediction of protein structure and function. Experimental determination of the functions of all these proteins would be a hugely time-consuming and costly task and, in most instances, has not been carried out. As an example, currently, approximately only 20% of annotated proteins in the Homo sapiens genome have been experimentally characterized. A commonly adopted procedure for annotating protein sequences relies on the "inheritance through homology" based on the notion that similar sequences share similar functions and structures. This procedure consists in the assignment of sequences to a specific group of functionally related sequences which had been grouped through clustering techniques. The clustering procedure is based on suitable similarity rules, since predicting protein structure and function from sequence largely depends on the value of sequence identity. However, additional levels of complexity are due to multi-domain proteins, to proteins that share common domains but that do not necessarily share the same function, to the finding that different combinations of shared domains can lead to different biological roles. In the last part of this study I developed and validate a system that contributes to sequence annotation by taking advantage of a validated transfer through inheritance procedure of the molecular functions and of the structural templates. After a cross-genome comparison with the BLAST program, clusters were built on the basis of two stringent constraints on sequence identity and coverage of the alignment. The adopted measure explicity answers to the problem of multi-domain proteins annotation and allows a fine grain division of the whole set of proteomes used, that ensures cluster homogeneity in terms of sequence length. A high level of coverage of structure templates on the length of protein sequences within clusters ensures that multi-domain proteins when present can be templates for sequences of similar length. This annotation procedure includes the possibility of reliably transferring statistically validated functions and structures to sequences considering information available in the present data bases of molecular functions and structures.

Seyfert galaxies are the closest active galactic nuclei. As such, we can use them to test the physical properties of the entire class of objects. To investigate their general properties, I took advantage of different methods of data analysis. In particular I used three different samples of objects, that, despite frequent overlaps, have been chosen to best tackle different topics: the heterogeneous BeppoS AX sample was thought to be optimized to test the average hard X-ray (E above 10 keV) properties of nearby Seyfert galaxies; the X-CfA was thought the be optimized to compare the properties of low-luminosity sources to the ones of higher luminosity and, thus, it was also used to test the emission mechanism models; finally, the XMM–Newton sample was extracted from the X-CfA sample so as to ensure a truly unbiased and well defined sample of objects to define the average properties of Seyfert galaxies. Taking advantage of the broad-band coverage of the BeppoS AX MECS and PDS instruments (between ~2-100 keV), I infer the average X-ray spectral propertiesof nearby Seyfert galaxies and in particular the photon index (~1.8), the high-energy cut-off (~290 keV), and the relative amount of cold reflection (~1.0). Moreover the unified scheme for active galactic nuclei was positively tested. The distribution of isotropic indicators used here (photon index, relative amount of reflection, high-energy cut-off and narrow FeK energy centroid) are similar in type I and type II objects while the absorbing column and the iron line equivalent width significantly differ between the two classes of sources with type II objects displaying larger absorbing columns. Taking advantage of the XMM–Newton and X–CfA samples I also deduced from measurements that 30 to 50% of type II Seyfert galaxies are Compton thick. Confirming previous results, the narrow FeK line is consistent, in Seyfert 2 galaxies, with being produced in the same matter responsible for the observed obscuration. These results support the basic picture of the unified model. Moreover, the presence of a X-ray Baldwin effect in type I sources has been measured using for the first time the 20-100 keV luminosity (EW proportional to L(20-100)^(−0.22±0.05)). This finding suggests that the torus covering factor may be a function of source luminosity, thereby suggesting a refinement of the baseline version of the unifed model itself. Using the BeppoSAX sample, it has been also recorded a possible correlation between the photon index and the amount of cold reflection in both type I and II sources. At a first glance this confirms the thermal Comptonization as the most likely origin of the high energy emission for the active galactic nuclei. This relation, in fact, naturally emerges supposing that the accretion disk penetrates, depending to the accretion rate, the central corona at different depths (Merloni et al. 2006): the higher accreting systems hosting disks down to the last stable orbit while the lower accreting systems hosting truncated disks. On the contrary, the study of the well defined X–C f A sample of Seyfert galaxies has proved that the intrinsic X-ray luminosity of nearby Seyfert galaxies can span values between 10^(38−43) erg s^−1, i.e. covering a huge range of accretion rates. The less efficient systems have been supposed to host ADAF systems without accretion disk. However, the study of the X–CfA sample has also proved the existence of correlations between optical emission lines and X-ray luminosity in the entire range of L_(X) covered by the sample. These relations are similar to the ones obtained if high-L objects are considered. Thus the emission mechanism must be similar in luminous and weak systems. A possible scenario to reconcile these somehow opposite indications is assuming that the ADAF and the two phase mechanism co-exist with different relative importance moving from low-to-high accretion systems (as suggested by the Gamma vs. R relation). The present data require that no abrupt transition between the two regimes is present. As mentioned above, the possible presence of an accretion disk has been tested using samples of nearby Seyfert galaxies. Here, to deeply investigate the flow patterns close to super-massive black-holes, three case study objects for which enough counts statistics is available have been analysed using deep X-ray observations taken with XMM–Newton. The obtained results have shown that the accretion flow can significantly differ between the objects when it is analyzed with the appropriate detail. For instance the accretion disk is well established down to the last stable orbit in a Kerr system for IRAS 13197-1627 where strong light bending effect have been measured. The accretion disk seems to be formed spiraling in the inner ~10-30 gravitational radii in NGC 3783 where time dependent and recursive modulation have been measured both in the continuum emission and in the broad emission line component. Finally, the accretion disk seems to be only weakly detectable in rk 509, with its weak broad emission line component. Finally, blueshifted resonant absorption lines have been detected in all three objects. This seems to demonstrate that, around super-massive black-holes, there is matter which is not confined in the accretion disk and moves along the line of sight with velocities as large as v~0.01-0.4c (whre c is the speed of light). Wether this matter forms winds or blobs is still matter of debate together with the assessment of the real statistical significance of the measured absorption lines. Nonetheless, if confirmed, these phenomena are of outstanding interest because they offer new potential probes for the dynamics of the innermost regions of accretion flows, to tackle the formation of ejecta/jets and to place constraints on the rate of kinetic energy injected by AGNs into the ISM and IGM. Future high energy missions (such as the planned Simbol-X and IXO) will likely allow an exciting step forward in our understanding of the flow dynamics around black holes and the formation of the highest velocity outflows.

Descrizione, tema e obiettivi della ricerca La ricerca si propone lo studio delle possibili influenze che la teoria di Aldo Rossi ha avuto sulla pratica progettuale nella Penisola Iberica, intende quindi affrontare i caratteri fondamentali della teoria che sta alla base di un metodo progettuale ed in particolar modo porre l'attenzione alle nuove costruzioni quando queste si confrontano con le città storiche. Ha come oggetto principale lo studio dei documenti, saggi e scritti riguardanti il tema della costruzione all'interno delle città storiche. Dallo studio di testi selezionati di Aldo Rossi sulla città si vuole concentrare l'attenzione sull'influenza che tale teoria ha avuto nei progetti della Penisola Iberica, studiare come è stata recepita e trasmessa successivamente, attraverso gli scritti di autori spagnoli e come ha visto un suo concretizzarsi poi nei progetti di nuove costruzioni all'interno delle città storiche. Si intende restringere il campo su un periodo ed un luogo precisi, Spagna e Portogallo a partire dagli anni Settanta, tramite la lettura di un importante evento che ha ufficializzato il contatto dell'architetto italiano con la Penisola Iberica, quale il Seminario di Santiago de Compostela tenutosi nel 1976. Al Seminario parteciparono numerosi architetti che si confrontarono su di un progetto per la città di Santiago e furono invitati personaggi di fama internazionale a tenere lezioni introduttive sul tema di dibattito in merito al progetto e alla città storica. Il Seminario di Santiago si colloca in un periodo storico cruciale per la Penisola Iberica, nel 1974 cade il regime salazarista in Portogallo e nel 1975 cade il regime franchista in Spagna ed è quindi di rilevante importanza capire il legame tra l'architettura e la nuova situazione politica. Dallo studio degli interventi, dei progetti che furono prodotti durante il Seminario, della relazione tra questo evento ed il periodo storico in cui esso va contestualizzato, si intende giungere alla individuazione delle tracce della reale presenza di tale eredità. Presupposti metodologici. Percorso e strumenti di ricerca La ricerca può quindi essere articolata in distinte fasi corrispondenti per lo più ai capitoli in cui si articola la tesi: una prima fase con carattere prevalentemente storica, di ricerca del materiale per poter definire il contesto in cui si sviluppano poi le vicende oggetto della tesi; una seconda fase di impronta teorica, ossia di ricerca bibliografica del materiale e delle testimonianze che provvedono alla definizione della reale presenza di effetti scaturiti dai contatti tra Rossi e la Penisola Iberica, per andare a costruire una eredità ; una terza fase che entra nel merito della composizione attraverso lo studio e la verifica delle prime due parti, tramite l'analisi grafica applicata ad uno specifico esempio architettonico selezionato; una quarta fase dove il punto di vista viene ribaltato e si indaga l'influenza dei luoghi visitati e dei contatti intrattenuti con alcuni personaggi della Penisola Iberica sull'architettura di Rossi, ricercandone i riferimenti. La ricerca è stata condotta attraverso lo studio di alcuni eventi selezionati nel corso degli anni che si sono mostrati significativi per l'indagine, per la risonanza che hanno avuto sulla storia dell'architettura della Penisola. A questo scopo si sono utilizzati principalmente tre strumenti: lo studio dei documenti, le pubblicazioni e le riviste prodotte in Spagna, gli scritti di Aldo Rossi in merito, e la testimonianza diretta attraverso interviste di personaggi chiave. La ricerca ha prodotto un testo suddiviso per capitoli che rispetta l'organizzazione in fasi di lavoro. A seguito di determinate condizioni storiche e politiche, studiate nella ricerca a supporto della tesi espressa, nella Penisola Iberica si è verificato il diffondersi della necessità e del desiderio di guardare e prendere a riferimento l'architettura europea e in particolar modo quella italiana. Il periodo sul quale viene focalizzata l'attenzione ha inizio negli anni Sessanta, gli ultimi prima della caduta delle dittature, scenario dei primi viaggi di Aldo Rossi nella Penisola Iberica. Questi primi contatti pongono le basi per intense e significative relazioni future. Attraverso l'approfondimento e la studio dei materiali relativi all'oggetto della tesi, si è cercato di mettere in luce il contesto culturale, l'attenzione e l'interesse per l'apertura di un dibattito intorno all'architettura, non solo a livello nazionale, ma europeo. Ciò ha evidenziato il desiderio di innescare un meccanismo di discussione e scambio di idee, facendo leva sull'importanza dello sviluppo e ricerca di una base teorica comune che rende coerente i lavori prodotti nel panorama architettonico iberico, seppur ottenendo risultati che si differenziano gli uni dagli altri. E' emerso un forte interesse per il discorso teorico sull'architettura, trasmissibile e comunicabile, che diventa punto di partenza per un metodo progettuale. Ciò ha reso palese una condivisione di intenti e l'assunzione della teoria di Aldo Rossi, acquisita, diffusa e discussa, attraverso la pubblicazione dei suoi saggi, la conoscenza diretta con l'architetto e la sua architettura, conferenze, seminari, come base teorica su cui fondare il proprio sapere architettonico ed il processo metodologico progettuale da applicare di volta in volta negli interventi concreti. Si è giunti così alla definizione di determinati eventi che hanno permesso di entrare nel profondo della questione e di sondare la relazione tra Rossi e la Penisola Iberica, il materiale fornito dallo studio di tali episodi, quali il I SIAC, la diffusione della rivista "2C. Construccion de la Ciudad", la Coleccion Arquitectura y Critica di Gustavo Gili, hanno poi dato impulso per il reperimento di una rete di ulteriori riferimenti. E' stato possibile quindi individuare un gruppo di architetti spagnoli, che si identificano come allievi del maestro Rossi, impegnato per altro in quegli anni nella formazione di una Scuola e di un insegnamento, che non viene recepito tanto nelle forme, piuttosto nei contenuti. I punti su cui si fondano le connessioni tra l'analisi urbana e il progetto architettonico si centrano attorno due temi di base che riprendono la teoria esposta da Rossi nel saggio L'architettura della città : - relazione tra l'area-studio e la città nella sua globalità, - relazione tra la tipologia edificatoria e gli aspetti morfologici. La ricerca presentata ha visto nelle sue successive fasi di approfondimento, come si è detto, lo sviluppo parallelo di più tematiche. Nell'affrontare ciascuna fase è stato necessario, di volta in volta, operare una verifica delle tappe percorse precedentemente, per mantenere costante il filo del discorso col lavoro svolto e ritrovare, durante lo svolgimento stesso della ricerca, gli elementi di connessione tra i diversi episodi analizzati. Tale operazione ha messo in luce talvolta nodi della ricerca rimasti in sospeso che richiedevano un ulteriore approfondimento o talvolta solo una rivisitazione per renderne possibile un più proficuo collegamento con la rete di informazioni accumulate. La ricerca ha percorso strade diverse che corrono parallele, per quanto riguarda il periodo preso in analisi: - i testi sulla storia dell'architettura spagnola e la situazione contestuale agli anni Settanta - il materiale riguardante il I SIAC - le interviste ai partecipanti al I SIAC - le traduzioni di Gustavo Gili nella Coleccion Arquitectura y Critica - la rivista "2C. Construccion de la Ciudad" Esse hanno portato alla luce una notevole quantità di tematiche, attraverso le quali, queste strade vengono ad intrecciarsi e a coincidere, verificando l'una la veridicità dell'altra e rafforzandone il valore delle affermazioni. Esposizione sintetica dei principali contenuti esposti dalla ricerca Andiamo ora a vedere brevemente i contenuti dei singoli capitoli. Nel primo capitolo Anni Settanta. Periodo di transizione per la Penisola Iberica si è cercato di dare un contesto storico agli eventi studiati successivamente, andando ad evidenziare gli elementi chiave che permettono di rintracciare la presenza della predisposizione ad un cambiamento culturale. La fase di passaggio da una condizione di chiusura rispetto alle contaminazioni provenienti dall'esterno, che caratterizza Spagna e Portogallo negli anni Sessanta, lascia il posto ad un graduale abbandono della situazione di isolamento venutasi a creare intorno al Paese a causa del regime dittatoriale, fino a giungere all'apertura e all'interesse nei confronti degli apporti culturali esterni. E' in questo contesto che si gettano le basi per la realizzazione del I Seminario Internazionale di Architettura Contemporanea a Santiago de Compostela, del 1976, diretto da Aldo Rossi e organizzato da César Portela e Salvador Tarragó, di cui tratta il capitolo secondo. Questo è uno degli eventi rintracciati nella storia delle relazioni tra Rossi e la Penisola Iberica, attraverso il quale è stato possibile constatare la presenza di uno scambio culturale e l'importazione in Spagna delle teorie di Aldo Rossi. Organizzato all'indomani della caduta del franchismo, ne conserva una reminescenza formale. Il capitolo è organizzato in tre parti, la prima si occupa della ricostruzione dei momenti salienti del Seminario Proyecto y ciudad historica, dagli interventi di architetti di fama internazionale, quali lo stesso Aldo Rossi, Carlo Aymonino, James Stirling, Oswald Mathias Ungers e molti altri, che si confrontano sul tema delle città storiche, alle giornate seminariali dedicate all’elaborazione di un progetto per cinque aree individuate all’interno di Santiago de Compostela e quindi dell’applicazione alla pratica progettuale dell’inscindibile base teorica esposta. Segue la seconda parte dello stesso capitolo riguardante La selezione di interviste ai partecipanti al Seminario. Esso contiene la raccolta dei colloqui avuti con alcuni dei personaggi che presero parte al Seminario e attraverso le loro parole si è cercato di approfondire la materia, in particolar modo andando ad evidenziare l’ambiente culturale in cui nacque l’idea del Seminario, il ruolo avuto nella diffusione della teoria di Aldo Rossi in Spagna e la ripercussione che ebbe nella pratica costruttiva. Le diverse interviste, seppur rivolte a persone che oggi vivono in contesti distanti e che in seguito a questa esperienza collettiva hanno intrapreso strade diverse, hanno fatto emergere aspetti comuni, tale unanimità ha dato ancor più importanza al valore di testimonianza offerta. L’elemento che risulta più evidente è il lascito teorico, di molto prevalente rispetto a quello progettuale che si è andato mescolando di volta in volta con la tradizione e l’esperienza dei cosiddetti allievi di Aldo Rossi. Negli stessi anni comincia a farsi strada l’importanza del confronto e del dibattito circa i temi architettonici e nel capitolo La fortuna critica della teoria di Aldo Rossi nella Penisola Iberica è stato affrontato proprio questo rinnovato interesse per la teoria che in quegli anni si stava diffondendo. Si è portato avanti lo studio delle pubblicazioni di Gustavo Gili nella Coleccion Arquitectura y Critica che, a partire dalla fine degli anni Sessanta, pubblica e traduce in lingua spagnola i più importanti saggi di architettura, tra i quali La arquitectura de la ciudad di Aldo Rossi, nel 1971, e Comlejidad y contradiccion en arquitectura di Robert Venturi nel 1972. Entrambi fondamentali per il modo di affrontare determinate tematiche di cui sempre più in quegli anni si stava interessando la cultura architettonica iberica, diventando così ¬ testi di riferimento anche nelle scuole. Le tracce dell’influenza di Rossi sulla Penisola Iberica si sono poi ricercate nella rivista “2C. Construccion de la Ciudad” individuata come strumento di espressione di una teoria condivisa. Con la nascita nel 1972 a Barcellona di questa rivista viene portato avanti l’impegno di promuovere la Tendenza, facendo riferimento all’opera e alle idee di Rossi ed altri architetti europei, mirando inoltre al recupero di un ruolo privilegiato dell’architettura catalana. A questo proposito sono emersi due fondamentali aspetti che hanno legittimato l’indagine e lo studio di questa fonte: - la diffusione della cultura architettonica, il controllo ideologico e di informazione operato dal lavoro compiuto dalla rivista; - la documentazione circa i criteri di scelta della redazione a proposito del materiale pubblicato. E’ infatti attraverso le pubblicazioni di “2C. Construccion de la Ciudad” che è stato possibile il ritrovamento delle notizie sulla mostra Arquitectura y razionalismo. Aldo Rossi + 21 arquitectos españoles, che accomuna in un’unica esposizione le opere del maestro e di ventuno giovani allievi che hanno recepito e condiviso la teoria espressa ne “L’architettura della città”. Tale mostra viene poi riproposta nella Sezione Internazionale di Architettura della XV Triennale di Milano, la quale dedica un Padiglione col titolo Barcelona, tres epocas tres propuestas. Dalla disamina dei progetti presentati è emerso un interessante caso di confronto tra le Viviendas para gitanos di César Portela e la Casa Bay di Borgo Ticino di Aldo Rossi, di cui si è occupato l’ultimo paragrafo di questo capitolo. Nel corso degli studi è poi emerso un interessante risvolto della ricerca che, capovolgendone l’oggetto stesso, ne ha approfondito gli aspetti cercando di scavare più in profondità nell’analisi della reciproca influenza tra la cultura iberica e Aldo Rossi, questa parte, sviscerata nell’ultimo capitolo, La Penisola Iberica nel “magazzino della memoria” di Aldo Rossi, ha preso il posto di quello che inizialmente doveva presentarsi come il risvolto progettuale della tesi. Era previsto infatti, al termine dello studio dell’influenza di Aldo Rossi sulla Penisola Iberica, un capitolo che concentrava l’attenzione sulla produzione progettuale. A seguito dell’emergere di un’influenza di carattere prettamente teorica, che ha sicuramente modificato la pratica dal punto di vista delle scelte architettoniche, senza però rendersi esplicita dal punto di vista formale, si è preferito, anche per la difficoltà di individuare un solo esempio rappresentativo di quanto espresso, sostituire quest’ultima parte con lo studio dell’altra faccia della medaglia, ossia l’importanza che a sua volta ha avuto la cultura iberica nella formazione della collezione dei riferimenti di Aldo Rossi. L’articolarsi della tesi in fasi distinte, strettamente connesse tra loro da un filo conduttore, ha reso necessari successivi aggiustamenti nel percorso intrapreso, dettati dall’emergere durante la ricerca di nuovi elementi di indagine. Si è pertanto resa esplicita la ricercata eredità di Aldo Rossi, configurandosi però prevalentemente come un’influenza teorica che ha preso le sfumature del contesto e dell’esperienza personale di chi se ne è fatto ricevente, diventandone così un continuatore attraverso il proprio percorso autonomo o collettivo intrapreso in seguito. Come suggerisce José Charters Monteiro, l’eredità di Rossi può essere letta attraverso tre aspetti su cui si basa la sua lezione: la biografia, la teoria dell’architettura, l’opera. In particolar modo per quanto riguarda la Penisola Iberica si può parlare dell’individuazione di un insegnamento riferito alla seconda categoria, i suoi libri di testo, le sue partecipazioni, le traduzioni. Questo è un lascito che rende possibile la continuazione di un dibattito in merito ai temi della teoria dell’architettura, della sue finalità e delle concrete applicazioni nelle opere, che ha permesso il verificarsi di una apertura mentale che mette in relazione l’architettura con altre discipline umanistiche e scientifiche, dalla politica, alla sociologia, comprendendo l’arte, le città la morfologia, la topografia, mediate e messe in relazione proprio attraverso l’architettura.

This PhD Thesis is the result of my research activity in the last three years. My main research interest was centered on the evolution of mitochondrial genome (mtDNA), and on its usefulness as a phylogeographic and phylogenetic marker at different taxonomic levels in different taxa of Metazoa. From a methodological standpoint, my main effort was dedicated to the sequencing of complete mitochondrial genomes, and the approach to whole-genome sequencing was based on the application of Long-PCR and shotgun sequences. Moreover, this research project is a part of a bigger sequencing project of mtDNAs in many different Metazoans’ taxa, and I mostly dedicated myself to sequence and analyze mtDNAs in selected taxa of bivalves and hexapods (Insecta). Sequences of bivalve mtDNAs are particularly limited, and my study contributed to extend the sampling. Moreover, I used the bivalve Musculista senhousia as model taxon to investigate the molecular mechanisms and the evolutionary significance of their aberrant mode of mitochondrial inheritance (Doubly Uniparental Inheritance, see below). In Insects, I focused my attention on the Genus Bacillus (Insecta Phasmida). A detailed phylogenetic analysis was performed in order to assess phylogenetic relationships within the genus, and to investigate the placement of Phasmida in the phylogenetic tree of Insecta. The main goal of this part of my study was to add to the taxonomic coverage of sequenced mtDNAs in basal insects, which were only partially analyzed.

The quality of astronomical sites is the first step to be considered to have the best performances from the telescopes. In particular, the efficiency of large telescopes in UV, IR, radio etc. is critically dependent on atmospheric transparency. It is well known that the random optical effects induced on the light propagation by turbulent atmosphere also limit telescope’s performances. Nowadays, clear appears the importance to correlate the main atmospheric physical parameters with the optical quality reachable by large aperture telescopes. The sky quality evaluation improved with the introduction of new techniques, new instrumentations and with the understanding of the link between the meteorological (or synoptical parameters and the observational conditions thanks to the application of the theories of electromagnetic waves propagation in turbulent medias: what we actually call astroclimatology. At the present the site campaigns are evolved and are performed using the classical scheme of optical seeing properties, meteorological parameters, sky transparency, sky darkness and cloudiness. New concept are added and are related to the geophysical properties such as seismicity, microseismicity, local variability of the climate, atmospheric conditions related to the ground optical turbulence and ground wind regimes, aerosol presence, use of satellite data. The purpose of this project is to provide reliable methods to analyze the atmospheric properties that affect ground-based optical astronomical observations and to correlate them with the main atmospheric parameters generating turbulence and affecting the photometric accuracy. The first part of the research concerns the analysis and interpretation of longand short-time scale meteorological data at two of the most important astronomical sites located in very different environments: the Paranal Observatory in the Atacama Desert (Chile), and the Observatorio del Roque de Los Muchachos(ORM) located in La Palma (Canary Islands, Spain). The optical properties of airborne dust at ORM have been investigated collecting outdoor data using a ground-based dust monitor. Because of its dryness, Paranal is a suitable observatory for near-IR observations, thus the extinction properties in the spectral range 1.00-2.30 um have been investigated using an empirical method. Furthermore, this PhD research has been developed using several turbulence profilers in the selection of the site for the European Extremely Large Telescope(E-ELT). During the campaigns the properties of the turbulence at different heights at Paranal and in the sites located in northern Chile and Argentina have been studied. This given the possibility to characterize the surface layer turbulence at Paranal and its connection with local meteorological conditions.

The treatment of the Cerebral Palsy (CP) is considered as the “core problem” for the whole field of the pediatric rehabilitation. The reason why this pathology has such a primary role, can be ascribed to two main aspects. First of all CP is the form of disability most frequent in childhood (one new case per 500 birth alive, (1)), secondarily the functional recovery of the “spastic” child is, historically, the clinical field in which the majority of the therapeutic methods and techniques (physiotherapy, orthotic, pharmacologic, orthopedic-surgical, neurosurgical) were first applied and tested. The currently accepted definition of CP – Group of disorders of the development of movement and posture causing activity limitation (2) – is the result of a recent update by the World Health Organization to the language of the International Classification of Functioning Disability and Health, from the original proposal of Ingram – A persistent but not unchangeable disorder of posture and movement – dated 1955 (3). This definition considers CP as a permanent ailment, i.e. a “fixed” condition, that however can be modified both functionally and structurally by means of child spontaneous evolution and treatments carried out during childhood. The lesion that causes the palsy, happens in a structurally immature brain in the pre-, peri- or post-birth period (but only during the firsts months of life). The most frequent causes of CP are: prematurity, insufficient cerebral perfusion, arterial haemorrhage, venous infarction, hypoxia caused by various origin (for example from the ingestion of amniotic liquid), malnutrition, infection and maternal or fetal poisoning. In addition to these causes, traumas and malformations have to be included. The lesion, whether focused or spread over the nervous system, impairs the whole functioning of the Central Nervous System (CNS). As a consequence, they affect the construction of the adaptive functions (4), first of all posture control, locomotion and manipulation. The palsy itself does not vary over time, however it assumes an unavoidable “evolutionary” feature when during growth the child is requested to meet new and different needs through the construction of new and different functions. It is essential to consider that clinically CP is not only a direct expression of structural impairment, that is of etiology, pathogenesis and lesion timing, but it is mainly the manifestation of the path followed by the CNS to “re”-construct the adaptive functions “despite” the presence of the damage. “Palsy” is “the form of the function that is implemented by an individual whose CNS has been damaged in order to satisfy the demands coming from the environment” (4). Therefore it is only possible to establish general relations between lesion site, nature and size, and palsy and recovery processes. It is quite common to observe that children with very similar neuroimaging can have very different clinical manifestations of CP and, on the other hand, children with very similar motor behaviors can have completely different lesion histories. A very clear example of this is represented by hemiplegic forms, which show bilateral hemispheric lesions in a high percentage of cases. The first section of this thesis is aimed at guiding the interpretation of CP. First of all the issue of the detection of the palsy is treated from historical viewpoint. Consequently, an extended analysis of the current definition of CP, as internationally accepted, is provided. The definition is then outlined in terms of a space dimension and then of a time dimension, hence it is highlighted where this definition is unacceptably lacking. The last part of the first section further stresses the importance of shifting from the traditional concept of CP as a palsy of development (defect analysis) towards the notion of development of palsy, i.e., as the product of the relationship that the individual however tries to dynamically build with the surrounding environment (resource semeiotics) starting and growing from a different availability of resources, needs, dreams, rights and duties (4). In the scientific and clinic community no common classification system of CP has so far been universally accepted. Besides, no standard operative method or technique have been acknowledged to effectively assess the different disabilities and impairments exhibited by children with CP. CP is still “an artificial concept, comprising several causes and clinical syndromes that have been grouped together for a convenience of management” (5). The lack of standard and common protocols able to effectively diagnose the palsy, and as a consequence to establish specific treatments and prognosis, is mainly because of the difficulty to elevate this field to a level based on scientific evidence. A solution aimed at overcoming the current incomplete treatment of CP children is represented by the clinical systematic adoption of objective tools able to measure motor defects and movement impairments. A widespread application of reliable instruments and techniques able to objectively evaluate both the form of the palsy (diagnosis) and the efficacy of the treatments provided (prognosis), constitutes a valuable method able to validate care protocols, establish the efficacy of classification systems and assess the validity of definitions. Since the ‘80s, instruments specifically oriented to the analysis of the human movement have been advantageously designed and applied in the context of CP with the aim of measuring motor deficits and, especially, gait deviations. The gait analysis (GA) technique has been increasingly used over the years to assess, analyze, classify, and support the process of clinical decisions making, allowing for a complete investigation of gait with an increased temporal and spatial resolution. GA has provided a basis for improving the outcome of surgical and nonsurgical treatments and for introducing a new modus operandi in the identification of defects and functional adaptations to the musculoskeletal disorders. Historically, the first laboratories set up for gait analysis developed their own protocol (set of procedures for data collection and for data reduction) independently, according to performances of the technologies available at that time. In particular, the stereophotogrammetric systems mainly based on optoelectronic technology, soon became a gold-standard for motion analysis. They have been successfully applied especially for scientific purposes. Nowadays the optoelectronic systems have significantly improved their performances in term of spatial and temporal resolution, however many laboratories continue to use the protocols designed on the technology available in the ‘70s and now out-of-date. Furthermore, these protocols are not coherent both for the biomechanical models and for the adopted collection procedures. In spite of these differences, GA data are shared, exchanged and interpreted irrespectively to the adopted protocol without a full awareness to what extent these protocols are compatible and comparable with each other. Following the extraordinary advances in computer science and electronics, new systems for GA no longer based on optoelectronic technology, are now becoming available. They are the Inertial and Magnetic Measurement Systems (IMMSs), based on miniature MEMS (Microelectromechanical systems) inertial sensor technology. These systems are cost effective, wearable and fully portable motion analysis systems, these features gives IMMSs the potential to be used both outside specialized laboratories and to consecutive collect series of tens of gait cycles. The recognition and selection of the most representative gait cycle is then easier and more reliable especially in CP children, considering their relevant gait cycle variability. The second section of this thesis is focused on GA. In particular, it is firstly aimed at examining the differences among five most representative GA protocols in order to assess the state of the art with respect to the inter-protocol variability. The design of a new protocol is then proposed and presented with the aim of achieving gait analysis on CP children by means of IMMS. The protocol, named ‘Outwalk’, contains original and innovative solutions oriented at obtaining joint kinematic with calibration procedures extremely comfortable for the patients. The results of a first in-vivo validation of Outwalk on healthy subjects are then provided. In particular, this study was carried out by comparing Outwalk used in combination with an IMMS with respect to a reference protocol and an optoelectronic system. In order to set a more accurate and precise comparison of the systems and the protocols, ad hoc methods were designed and an original formulation of the statistical parameter coefficient of multiple correlation was developed and effectively applied. On the basis of the experimental design proposed for the validation on healthy subjects, a first assessment of Outwalk, together with an IMMS, was also carried out on CP children. The third section of this thesis is dedicated to the treatment of walking in CP children. Commonly prescribed treatments in addressing gait abnormalities in CP children include physical therapy, surgery (orthopedic and rhizotomy), and orthoses. The orthotic approach is conservative, being reversible, and widespread in many therapeutic regimes. Orthoses are used to improve the gait of children with CP, by preventing deformities, controlling joint position, and offering an effective lever for the ankle joint. Orthoses are prescribed for the additional aims of increasing walking speed, improving stability, preventing stumbling, and decreasing muscular fatigue. The ankle-foot orthosis (AFO), with a rigid ankle, are primarily designed to prevent equinus and other foot deformities with a positive effect also on more proximal joints. However, AFOs prevent the natural excursion of the tibio-tarsic joint during the second rocker, hence hampering the natural leaning progression of the whole body under the effect of the inertia (6). A new modular (submalleolar) astragalus-calcanear orthosis, named OMAC, has recently been proposed with the intention of substituting the prescription of AFOs in those CP children exhibiting a flat and valgus-pronated foot. The aim of this section is thus to present the mechanical and technical features of the OMAC by means of an accurate description of the device. In particular, the integral document of the deposited Italian patent, is provided. A preliminary validation of OMAC with respect to AFO is also reported as resulted from an experimental campaign on diplegic CP children, during a three month period, aimed at quantitatively assessing the benefit provided by the two orthoses on walking and at qualitatively evaluating the changes in the quality of life and motor abilities. As already stated, CP is universally considered as a persistent but not unchangeable disorder of posture and movement. Conversely to this definition, some clinicians (4) have recently pointed out that movement disorders may be primarily caused by the presence of perceptive disorders, where perception is not merely the acquisition of sensory information, but an active process aimed at guiding the execution of movements through the integration of sensory information properly representing the state of one’s body and of the environment. Children with perceptive impairments show an overall fear of moving and the onset of strongly unnatural walking schemes directly caused by the presence of perceptive system disorders. The fourth section of the thesis thus deals with accurately defining the perceptive impairment exhibited by diplegic CP children. A detailed description of the clinical signs revealing the presence of the perceptive impairment, and a classification scheme of the clinical aspects of perceptual disorders is provided. In the end, a functional reaching test is proposed as an instrumental test able to disclosure the perceptive impairment. References 1. The field of complex systems is a growing body of knowledge, It can be applied to countless different topics, from physics to computer science, biology, information theory and sociology. The main focus of this work is the use of microscopic models to study the behavior of urban mobility, which characteristics make it a paradigmatic example of complexity. In particular, simulations are used to investigate phase changes in a finite size open Manhattan-like urban road network under different traffic conditions, in search for the parameters to identify phase transitions, equilibrium and non-equilibrium conditions . It is shown how the flow-density macroscopic fundamental diagram of the simulation shows,like real traffic, hysteresis behavior in the transition from the congested phase to the free flow phase, and how the different regimes can be identified studying the statistics of road occupancy.

Mitochondria are inherited maternally in most metazoans. However, in some bivalves, two mitochondrial lineages are present: one transmitted through eggs (F), the other through sperm (M). This is called Doubly Uniparental Inheritance (DUI). During male embryo development, spermatozoon mitochondria aggregate and end up in the primordial germ cells, while they are dispersed in female embryos. The molecular mechanisms of segregation patterns are still unknown. In the DUI species Ruditapes philippinarum, I examined sperm mitochondria distribution by MitoTracker, microtubule staining and TEM, and I localized germ line determinants with immunocytochemical analysis. I also analyzed the gonad transcriptome, searching for genes involved in reproduction and sex determination. Moreover, I analyzed an M-type specific open reading frame that could be responsible for maintenance/degradation of M mitochondria during embryo development. These transcripts were also localized in tissues using in situ hybridization. As in Mytilus, two distribution patterns of M mitochondria were detected in R. philippinarum, supporting that they are related to DUI. Moreover, the first division midbody concurs in positioning aggregated M mitochondria on the animal-vegetal axis of the male embryo: in organisms with spiral segmentation this zone is not involved in further cleavages, so aggregation is maintained. Moreover, sperm mitochondria reach the same embryonic area where germ plasm is transferred, suggesting their contribution in male germ line formation. The finding of reproduction and ubiquitination transcripts led to formulate a model in which ubiquitination genes stored in female oocytes during gametogenesis would activate sex-gene expression in the early embryonic developmental stages (preformation). Only gametogenetic cells were labeled by in situ hybridization, proving their specific transcription in developing gametes. Other than having a role in sex determination, some ubiquination factors could also be involved in mitochondrial inheritance, and their differential expression could be responsible for the different fate of sperm mitochondria in the two sexes.

En la sociedad europea crece la preocupación por el retorno de tendencias fascistas y neonazis y por la extensión de ideologías xenófobas y antisemitas, algunas de ellas alimentadas a partir de tesis de negacionistas de aquellos trágicos eventos de nuestra historia reciente. La lucha frente a los discursos negacionistas se ha llevado más allá del ámbito social y académico, y se ha propuesto la incorporación en los ordenamientos jurídicos europeos de tipos penales específicos que incriminan este tipo de discurso: negar, banalizar, o justificar el Holocausto u otros genocidios o graves crímenes contra la humanidad. Esta legislación, que encuentra su mayor expresión en la Decisión marco 2008/913/JAI, aunque castiga un discurso socialmente repugnante, sin embargo presenta dudas en cuanto a su legitimidad con un sistema de libertades erigido sobre el pilar del pluralismo propio de los Estados democráticos. Surge así la cuestión de si pueden estar surgiendo «nuevos» delitos de opinión y a ello se dedica entonces la presente tesis. El objetivo concreto de este trabajo será analizar esta política-criminal para proponer una configuración del delito de negacionismo compatible con la libertad de expresión, aunque se cuestionará la conveniencia de castigar penalmente a través de un específico delito este tipo de conductas. En particular se pretende responder a tres preguntas: en primer lugar, ¿el discurso negacionista debe ampararse prima facie por la libertad de expresión en un ordenamiento abierto y personalista y cuáles podrían ser las «reglas» que podrían servir como criterio para limitar este género de manifestaciones? La segunda pregunta sería entonces: ¿Cómo podría construirse un tipo penal respetuoso con los principios constitucionales y penales que específicamente incriminara este género de conductas? Y, como última pregunta: ¿Es conveniente o adecuada una política criminal que lleve a crear un específico delito de negacionismo?

The present thesis focuses on elastic waves behaviour in ordinary structures as well as in acousto-elastic metamaterials via numerical and experimental applications. After a brief introduction on the behaviour of elastic guided waves in the framework of non-destructive evaluation (NDE) and structural health monitoring (SHM) and on the study of elastic waves propagation in acousto-elastic metamaterials, dispersion curves for thin-walled beams and arbitrary cross-section waveguides are extracted via Semi-Analytical Finite Element (SAFE) methods. Thus, a novel strategy tackling signal dispersion to locate defects in irregular waveguides is proposed and numerically validated. Finally, a time-reversal and laser-vibrometry based procedure for impact location is numerically and experimentally tested. In the second part, an introduction and a brief review of the basic definitions necessary to describe acousto-elastic metamaterials is provided. A numerical approach to extract dispersion properties in such structures is highlighted. Afterwards, solid-solid and solid-fluid phononic systems are discussed via numerical applications. In particular, band structures and transmission power spectra are predicted for 1P-2D, 2P-2D and 2P-3D phononic systems. In addition, attenuation bands in the ultrasonic as well as in the sonic frequency regimes are experimentally investigated. In the experimental validation, PZTs in a pitch-catch configuration and laser vibrometric measurements are performed on a PVC phononic plate in the ultrasonic frequency range and sound insulation index is computed for a 2P-3D phononic barrier in the sonic frequency range. In both cases the numerical-experimental results comparison confirms the existence of the numerical predicted band-gaps. Finally, the feasibility of an innovative passive isolation strategy based on giant elastic metamaterials is numerically proved to be practical for civil structures. In particular, attenuation of seismic waves is demonstrated via finite elements analyses. Further, a parametric study shows that depending on the soil properties, such an earthquake-proof barrier could lead to significant reduction of the superstructure displacement.

The study defines a new farm classification and identifies the arable land management. These aspects and several indicators are taken into account to estimate the sustainability level of farms, for organic and conventional regimes. The data source is Italian Farm Account Data Network (RICA) for years 2007-2011, which samples structural and economical information. An environmental data has been added to the previous one to better describe the farm context. The new farm classification describes holding by general informations and farm structure. The general information are: adopted regime and farm location in terms of administrative region, slope and phyto-climatic zone. The farm structures describe the presence of main productive processes and land covers, which are recorded by FADN database. The farms, grouped by homogeneous farm structure or farm typology, are evaluated in terms of sustainability. The farm model MAD has been used to estimate a list of indicators. They describe especially environmental and economical areas of sustainability. Finally arable lands are taken into account to identify arable land managements and crop rotations. Each arable land has been classified by crop pattern. Then crop rotation management has been analysed by spatial and temporal approaches. The analysis reports a high variability inside regimes. The farm structure influences indicators level more than regimes, and it is not always possible to compare the two regimes. However some differences between organic and conventional agriculture have been found. Organic farm structures report different frequency and geographical location than conventional ones. Also different connections among arable lands and farm structures have been identified.

Fluxus è stato definito il più radicale e sperimentale movimento artistico degli anni Sessanta. Dalla prima comparsa ad oggi è stato osannato, analizzato, dimenticato e riscoperto molte volte, tuttora però rimane una delle più grandi incognite critiche della storia dell’arte del Novecento. La ricerca si sviluppa secondo uno schema tripartito: indagare origini, ascendenze e ispirazioni; collocare e contestualizzare il periodo di nascita e sviluppo; esaminare influenze e lasciti. Attraverso un confronto di manifesti, scritti autografi e opere si è cercato di verificare punti di contatto e di continuità tra Fluxus e le Avanguardie Storiche, con particolare riferimento a Futurismo e Dadaismo. Successivamente si è cercato di ricostruire le dinamiche che hanno portato, alla fine degli anni Cinquanta, al definirsi di un terreno fertile dal quale sono germinate esperienze strettamente legate quali Happening, Performance Art e lo stesso Fluxus, del quale si sono ripercorsi i cosiddetti “anni eroici” per evidenziarne le caratteristiche salienti. Nella terza sezione sono state individuate diverse ipotesi di continuazione dell’attitudine Fluxus, dal percorso storico-filologico dei precoci tentativi di musealizzazione, alle eredità dirette e indirette sulle generazioni successive di artisti, fino alla individuazione di idee e concetti la cui attualità rende Fluxus un elemento imprescindibile per la comprensione della cultura contemporanea.
