This Thesys reports the study of a HGMS (High GradientMagnetic Separation) process for the treatment of industrialwastewaters that considers an assisted chemical-physical pre-treatment for the removal of heavy metals through the bound by adsorption with added iron-oxide particulate matter (hematite). The considered filter, constituted by ferromagnetic stainless steel wool and permanent magnets, is studied with a new approach based on a statistical analysis that requires the study of the trajectories of the particles. Experimental activity on a laboratory device has been carried out in order to test the model.

The first aims of this study were to demonstrate if mitochondrial biogenesis and senescence can be induced simultaneously in cell lines upon exposure to a genotoxic stress, and if the presence of mtDNA mutations which impair the functionality of respiratory complexes can influence the ability of a cell to activate senescence. The data obtained on the oncocytic model XTC.UC1 demonstrated that the presence of mitochondrial dysfunction is involved in the maintenance of a senescent phenotype induced by γ-rays treatment. The involvement of mTORC1 in the regulation of senescence has been shown in this cell line. On the other hand, in cells which do not present mitochondrial dysfunction it has been verified that genotoxic stress determines the activation of both mitochondrial biogenesis and senescence. Further studies are necessary in order to verify if mitochondrial biogenesis sustains the activation of senescence. The second aim of this thesis was to determine the involvement of mTORC1 in the regulation of PGC-1α expression, in order to verify what is the cause of the development of oncocytoma in patients affected by two hereditary cancer syndromes; Cowden and Birt-hogg-Dubé . The study of oncocytic tumors developed by patients affected by these syndromes suggested that the double heterozigosity of the two causative genes, PTEN and FLCN respectively, induce the activation of mTORC1 and therefore the activation of PGC-1α expression. On XTC.UC1 cell line, the most suitable in vitro model, experiments of complementation of PTEN and FLCN were conducted. To date, these results demonstrated that mTORC1 is not involved in the regulation of PGC-1α expression, and PTEN and FLCN seem to have opposite effect on PGC-1α expression.

Future wireless communications systems are expected to be extremely dynamic, smart and capable to interact with the surrounding radio environment. To implement such advanced devices, cognitive radio (CR) is a promising paradigm, focusing on strategies for acquiring information and learning. The first task of a cognitive systems is spectrum sensing, that has been mainly studied in the context of opportunistic spectrum access, in which cognitive nodes must implement signal detection techniques to identify unused bands for transmission. In the present work, we study different spectrum sensing algorithms, focusing on their statistical description and evaluation of the detection performance. Moving from traditional sensing approaches we consider the presence of practical impairments, and analyze algorithm design. Far from the ambition of cover the broad spectrum of spectrum sensing, we aim at providing contributions to the main classes of sensing techniques. In particular, in the context of energy detection we studied the practical design of the test, considering the case in which the noise power is estimated at the receiver. This analysis allows to deepen the phenomenon of the SNR wall, providing the conditions for its existence and showing that presence of the SNR wall is determined by the accuracy of the noise power estimation process. In the context of the eigenvalue based detectors, that can be adopted by multiple sensors systems, we studied the practical situation in presence of unbalances in the noise power at the receivers. Then, we shift the focus from single band detectors to wideband sensing, proposing a new approach based on information theoretic criteria. This technique is blind and, requiring no threshold setting, can be adopted even if the statistical distribution of the observed data in not known exactly. In the last part of the thesis we analyze some simple cooperative localization techniques based on weighted centroid strategies.

Enterobacteriaceae genomes evolve through mutations, rearrangements and horizontal gene transfer (HGT). The latter evolutionary pathway works through the acquisition DNA (GEI) modules of foreign origin that enhances fitness of the host to a given environment. The genome of E. coli IHE3034, a strain isolated from a case of neonatal meningitis, has recently been sequenced and its subsequent sequence analysis has predicted 18 possible GEIs, of which: 8 have not been previously described, 5 fully meet the pathogenic island definition and at least 10 that seem to be of prophagic origin. In order to study the GEI distribution of our reference strain, we screened for the presence 18 GEIs a panel of 132 strains, representative of E. coli diversity. Also, using an inverse nested PCR approach we identified 9 GEI that can form an extrachromosomal circular intermediate (CI) and their respective attachment sites (att). Further, we set up a qPCR approach that allowed us to determine the excision rates of 5 genomic islands in different growth conditions. Four islands, specific for strains appertaining to the sequence type complex 95 (STC95), have been deleted in order to assess their function in a Dictyostelium discoideum grazing assays. Overall, the distribution data presented here indicate that 16 IHE3034 GEIs are more associated to the STC95 strains. Also the functional and genetic characterization has uncovered that GEI 13, 17 and 19 are involved in the resistance to phagocitation by Dictyostelium d thus suggesting a possible role in the adaptation of the pathogen during certain stages of infection.

E’ stata condotta una analisi su preparati cadaverici in particolare in tre distretti: a) addome e tripode celiaco/mesenterica superiore; b) circolo cerebrale; d) orco aortico.
