We observed 82 healthy subjects, from both sexes, aged between 19 and 77 years. All subjects performed two different tests: for being scientifically acknowledged, the first one was used as a reference and it was a stress test (CPX). During the entire test, heart rate and gas exchange were recorded continuously; the second, the actual object of this study, was a submaximal test (TOP). Only heart rate was recorded continuously. The main purpose was to determinate an index of physical fitness as result of TOP. CPX test allowed us to individuate anaerobic threshold. We used an incremental protocol of 10/20 Watt/min, different by age. For our TOP test we used an RHC400 UPRIGHT BIKE, by Air Machine. Each subject was monitored for heart frequency. After 2 minutes of resting period there was a first step: 3 minutes of pedalling at a constant rate of 60 RPM, (40 watts for elder subjects and 60 watts for the younger ones). Then, the subject was allowed to rest for a recovery phase of 5 minutes. Third and last step consisted of 3 minutes of pedalling again at 60 RPM but now set to 60 watts for elder subjects and 80 watts for the young subjects. Finally another five minutes of recovery. A good correlation was found between TOP and CPX results especially between punctua l heart rate reserve (HRR’) and anaerobic threshold parameters such as Watt, VO2, VCO2 . HRR’ was obtained by subtracting maximal heart rate during TOP from maximal theoretic heart rate (206,9-(0,67*age)). Data were analyzed through cluster analysis in order to obtain 3 homogeneous groups. The first group contains the least fit subjects (inactive, women, elderly). The other groups contain the “average fit” and the fittest subjects (active, men, younger). Concordance between test resulted in 83,23%. Afterwards, a linear combinations of the most relevant variables gave us a formula to classify people in the correct group. The most relevant result is that this submaximal test is able to discriminate subjects with different physical condition and to provide information (index) about physical fitness through HRR’. Compared to a traditional incremental stress test, the very low load of TOP, short duration and extended resting period, make this new method suitable to very different people. To better define the TOP index, it is necessary to enlarge our subject sample especially by diversifying the age range.

Machines with moving parts give rise to vibrations and consequently noise. The setting up and the status of each machine yield to a peculiar vibration signature. Therefore, a change in the vibration signature, due to a change in the machine state, can be used to detect incipient defects before they become critical. This is the goal of condition monitoring, in which the informations obtained from a machine signature are used in order to detect faults at an early stage. There are a large number of signal processing techniques that can be used in order to extract interesting information from a measured vibration signal. This study seeks to detect rotating machine defects using a range of techniques including synchronous time averaging, Hilbert transform-based demodulation, continuous wavelet transform, Wigner-Ville distribution and spectral correlation density function. The detection and the diagnostic capability of these techniques are discussed and compared on the basis of experimental results concerning gear tooth faults, i.e. fatigue crack at the tooth root and tooth spalls of different sizes, as well as assembly faults in diesel engine. Moreover, the sensitivity to fault severity is assessed by the application of these signal processing techniques to gear tooth faults of different sizes.

A systematic characterization of the composition and structure of the bacterial cell-surface proteome and its complexes can provide an invaluable tool for its comprehensive understanding. The knowledge of protein complexes composition and structure could offer new, more effective targets for a more specific and consequently effective immune response against a complex instead of a single protein. Large-scale protein-protein interaction screens are the first step towards the identification of complexes and their attribution to specific pathways. Currently, several methods exist for identifying protein interactions and protein microarrays provide the most appealing alternative to existing techniques for a high throughput screening of protein-protein interactions in vitro under reasonably straightforward conditions. In this study approximately 100 proteins of Group A Streptococcus (GAS) predicted to be secreted or surface exposed by genomic and proteomic approaches were purified in a His-tagged form and used to generate protein microarrays on nitrocellulose-coated slides. To identify protein-protein interactions each purified protein was then labeled with biotin, hybridized to the microarray and interactions were detected with Cy3-labelled streptavidin. Only reciprocal interactions, i. e. binding of the same two interactors irrespective of which of the two partners is in solid-phase or in solution, were taken as bona fide protein-protein interactions. Using this approach, we have identified 20 interactors of one of the potent toxins secreted by GAS and known as superantigens. Several of these interactors belong to the molecular chaperone or protein folding catalyst families and presumably are involved in the secretion and folding of the superantigen. In addition, a very interesting interaction was found between the superantigen and the substrate binding subunit of a well characterized ABC transporter. This finding opens a new perspective on the current understanding of how superantigens are modified by the bacterial cell in order to become major players in causing disease.

Igor Stravinskij (1882-1971) utilizzò di sovente fonti preesistenti come parte integrante del proprio artigianato compositivo. In questa tesi dottorale ho studiato il processo creativo di Stravinskij negli anni Venti sulle musiche di Pëtr Il'ič Čajkovskij (1840-1893). Nella prima parte della dissertazione ho indagato la Sleeping Princess (1921) e il successivo Mariage d’Aurore (1922-1929), entrambi allestiti dai Ballets russes di Sergej Djagilev (1872-1929). Dopo aver localizzato e contestualizzato le fonti manoscritte e i materiali d’uso, ho ricostruito le ri-orchestrazioni effettuate da Stravinskij della Danse russe (Coda del Pas de deux n. 28) e del Presto del Finale (n. 30), che erano a tutt’oggi inedite. La ricerca sulla Sleeping Princess si è rivelata fondamentale per la conseguente analisi del Baiser de la Fée (1928, Ballets de Mme Ida Rubinstein), balletto basato su pezzi pianistici e romanze per voce e pianoforte di Čajkovskij. Grazie allo studio dello Skizzenbuch VIII, della partitura pianistica manoscritta e di tutte le fonti rinvenute, ho gettato ulteriore luce sul processo compositivo di Stravinskij sulle fonti čajkovskiane. Ho rinvenuto nuove appropriazioni che finora non erano note.

La tesi ha per oggetto la cultura ebraica cretese nei secoli XIV-XVI e, in particolare, l’influsso esercitato su di essa dalla cultura e dalle tradizioni degli ebrei sefarditi e ashkenaziti che cominciarono a stabilirsi sull’isola a partire dalla metà del Trecento. La tesi si basa da un lato su fonti amministrative e notarili e, dall’altro, sui manoscritti ebraici prodotti o portati a Candia nel periodo considerato. Il primo capitolo tratta della comunità ebraica nel primo Cinquecento e porta nuove notizie a proposito della geografia della zudeca, delle sue sinagoghe, della sua composizione sociale, dell’entità della sua popolazione e della biografia del principale leader spirituale e culturale attivo a Candia a quell’epoca: Elia Capsali. Il secondo capitolo offre una panoramica sull’immigrazione ebraica a Candia nei secoli XIV-XV. Il terzo capitolo esplora alcune particolarità della liturgia sinagogale elaborata dagli ebrei candioti sotto l’influsso della tradizione ashkenazita. Il quarto capitolo tratta di due liste di libri databili alla seconda metà del Quattrocento (Bologna, Biblioteca Universitaria, ms. 3574 B) e suggerisce di considerarle come indicative del peso che ebbero alcuni immigrati ebrei catalani nella diffusione della cultura medica sefardita a Candia. Il quinto capitolo è dedicato al medico, filosofo e astronomo Mosheh ben Yehudah Galiano, il quale visse a Candia tra la seconda metà degli anni Venti del Cinquecento e il 1543. L’ultimo capitolo tratta degli effetti provocati dall’epidemia di peste del 1592-95 all’interno della zudeca di Candia.
