Given its strategic position at the center of the Mediterranean Basin, and its unique history of contacts and migrations, Calabria is an ideal region to decipher the genetic traces of at least some of the numerous demographic prehistoric events in Southern Italy. This thesis focuses on the genetic and social changes of ancient inhabitants of Calabria, covering a timeline of approximately 7000 to 3300 years ago, ranging from the Middle Neolithic to the Middle Bronze Age. We generated the first genome-wide data from Calabria, by focusing on the single inhumation of “Grotta di Pietra Sant’Angelo” (San Lorenzo Bellizzi, Cosenza) and on the vast community found buried in “Grotta della Monaca” (Sant’Agata di Esaro, Cosenza). Supported by archaeological evidence, the primary objective of this research was to employ paleogenomic evidence to decipher funerary customs, social organization, family ties, and demographic shifts in Southern Italy over a period extending beyond three millennia. The possibility of gender-related burial practices and kinship ties among the deceased was also explored. Subsequently, the biogeographical origin and ancestry of prehistoric people of Calabria was contextualized within the broad landscape of existing data on Mediterranean populations. By generating the first genomic evidence from prehistoric Calabria, unresolved questions were addressed, related to the appearance and persistence of distinct genomic components, such as the Iran-related and the Steppe-related ancestry, whose impact on ancient Southern Italian genomes remains uncharted.

Per comprendere le vicende di una famiglia illustre e nobile il cui ruolo politico e sociale in Sicilia si data alle soglie del XIV secolo, non possiamo astenerci dal ricordare i fatti e gli eventi che hanno dominato la storia siciliana e determinato l’ascesa di Castelvetrano come centro signorile per eccellenza. E’ necessario, quindi, collocare geograficamente e storicamente l’isola per inserirla all’interno di un preciso quadro socio-politico. All’origine della sua storia sono sicuramente da individuare sia il legame intercorso nei secoli tra l’Asia e l’Europa, in particolare tra l’Asia Minore bizantina e l’area mediterranea unificata proprio dall’impero di Bisanzio, sia le lotte per l’egemonia tra Chiesa e Impero, (che abbastanza presto sarà impero d’Occidente) lotte che vedono entrambe le parti impegnate a contendersi il ruolo di guida politica, morale e spirituale dell’intera cristianità medievale, ritenendo ogni altro potere subordinato al proprio.

Fondato nel 1423 per decisione del Maggior Consiglio di Venezia, il Lazzaretto Vecchio, è la prima struttura permanente in Europa destinata al ricovero degli ammalati di peste e alla quarantena preventiva di coloro che erano entrati in contatto con infetti o che provenivano da zone epidemiche. Questo studio si pone come obiettivo quello di analizzare antropologicamente una parte dei resti umani rinvenuti e comparare i risultati ottenuti con le fonti edite riguardanti la composizione dei ricoverati all’interno della struttura. Sono stati selezionati 110 individui provenienti da 9 comuni appartenenti a due diversi archi cronologici, il 1575-1577 e il 1630-1631. Gli individui sono stati sottoposti a diverse analisi antropologiche tradizionali e indagini più dettagliate di stampo biomolecolare. Parallelamente, è stata vagliata la letteratura prodotta da studi popolazionistici editi. Le analisi antropologiche hanno rivelato un quadro popolazionistico con normale presenza di maschi e femmine, e attestazione di tutte le classi di età, dagli 0 agli oltre 50 anni. Le analisi chimico-fisiche hanno rivelato la presenza di sette individui non locali. Con riguardo all’alimentazione non vi sono particolari differenze in termini di periodi cronologici o di sesso degli individui, quanto invece si possono individuare schemi relativi alla qualità del cibo ingerito in relazione all’inserimento nel mondo del lavoro. Per quanto riguarda le indagini di carattere documentale, nonostante alcuni degli studi popolazionistici individuati non siano considerabili recenti, ci forniscono un quadro dell’andamento della popolazione a Venezia a partire dal 1500 fino al 1700, coprendo in questo modo le cronologie d’interesse del progetto, legate agli episodi di maggiore mortalità della peste. Si riscontra una sostanziale coerenza tra i dati riportati nei diversi resoconti e la sex ratio e le fasce di età alla morte riscontrate, anche se non mancano casi specifici di forte discostamento tra i dati.

Epigenetic variability is a new mechanism for the study of human microevolution, because it creates both phenotypic diversity within an individual and within population. This mechanism constitutes an important reservoir for adaptation in response to new stimuli and recent studies have demonstrated that selective pressures shape not only the genetic code but also DNA methylation profiles. The aim of this thesis is the study of the role of DNA methylation changes in human adaptive processes, considering the Italian peninsula and macro-geographical areas. A whole-genome analysis of DNA methylation profile across the Italian penisula identified some genes whose methylation levels differ between individuals of different Italian districts (South, Centre and North of Italy). These genes are involved in nitrogen compound metabolism and genes involved in pathogens response. Considering individuals with different macro-geographical origins (individuals of Asians, European and African ancestry) more significant DMRs (differentially methylated regions) were identified and are located in genes involved in glucoronidation, in immune response as well as in cell comunication processes. A "profile" of each ancestry (African, Asian and European) was described. Moreover a deepen analysis of three candidate genes (KRTCAP3, MAD1L and BRSK2) in a cohort of individuals of different countries (Morocco, Nigeria, China and Philippines) living in Bologna, was performed in order to explore genetic and epigenetic diversity. Moreover this thesis have paved the way for the application of DNA methylation for the study of hystorical remains and in particular for the age-estimation of individuals starting from biological samples (such as teeth or blood). Noteworthy, a mathematical model that considered methylation values of DNA extracted from cementum and pulp of living individuals can estimate chronological age with high accuracy (median absolute difference between age estimated from DNA methylation and chronological age was 1.2 years).

The present work aims at reconstructing the archaeological contexts and analyzing the material culture of the site of Europos. This archaeological site is located in southern Turkey, at the border with Syria, along the right shore of the Euphrates River. The Classical city rose above the remains of the Hittite Karkemish. The present work collects the results of the archaeological expeditions launched by the British Museum in the late 19th and early 20th century, never published, and the ones of the new Turco-Italian Joint Expedition, started in 2011. Europos had an uninterrupted life from the 3rd century BC to the 10th century AD, throughout the Hellenistic, Roman and Byzantine periods, all examined in the present work.

Cultural heritage is constituted by complex and heterogenous materials, such as paintings but also ancient remains. However, all ancient materials are exposed to external environment and their interaction produces different changes due to chemical, physical and biological phenomena. The organic fraction, especially the proteinaceous one, has a crucial role in all these materials: in archaeology proteins reveal human habits, in artworks they disclose technics and help for a correct restoration. For these reasons the development of methods that allow the preservation of the sample as much as possible and a deeper knowledge of the deterioration processes is fundamental. The research activities presented in this PhD thesis have been focused on the development of new immunochemical and spectroscopic approaches in order to detect and identify organic substances in artistic and archaeological samples. Organic components could be present in different cultural heritage materials as constituent element (e.g., binders in paintings, collagen in bones) and their knowledge is fundamental for a complete understanding of past life, degradation processes and appropriate restauration approaches. The combination of immunological approach with a chemiluminescence detection and Laser Ablation-Inductively Coupled Plasma-Mass Spectrometry allowed a sensitive and selective localization of collagen and elements in ancient bones and teeth. Near-infrared spectrometer and hyper spectral imaging have been applied in combination with chemometric data analysis as non-destructive methods for bones prescreening for the localization of collagen. Moreover, an investigation of amino acids in enamel has been proposed, in order to clarify teeth biomolecules survival overtime through the optimization and application of High-Performance Liquid Chromatography on modern and ancient enamel powder. New portable biosensors were developed for ovalbumin identification in paintings, thanks to the combination between biocompatible Gellan gel and electro-immunochemical sensors, to extract and identify painting binders with the contact only between gel and painting and between gel and electrodes.
