Il tema dello sviluppo sostenibile, e i numerosi ambiti di ricerca e di applicazione a esso connessi, è oramai coralmente accettato dalla comunità scientifica, intellettuale e politica internazionale. Tuttavia, una certa tendenza a una sua iper-utilizzazione può comportare svuotamento di significati e perdita di valore. Obiettivo della presente ricerca, dunque, è quello di ripensare il concetto della sostenibilità tenuto conto delle trasformazioni strutturali che hanno interessato le regioni alpine tra il XIX e il XX secolo e sulla base di esperienze realizzate, in atto o in potenza che ben ne evidenzino le possibili applicazioni a livello regionale e locale. A tal fine, nel corso della trattazione si propone un’analisi delle buone pratiche incontrate lungo il cammino compiuto sui sentieri della sostenibilità.

Introduction: Apoptotic cell death of cardiomyocytes is involved in several cardiovascular diseases including ischemia, hypertrophy and heart failure, thus representing a potential therapeutic target. Apoptosis of cardiac cells can be induced experimentally by several stimuli including hypoxia, serum withdrawal or combination of both. Several lines of research suggest that neurohormonal mechanisms play a central role in the progression of heart failure. In particular, excessive activation of the sympathetic nervous system or the renin-angiotensin-aldosterone system is known to have deleterious effects on the heart. Recent studies report that norepinephrine (NE), the primary transmitter of sympathetic nervous system, and aldosterone (ALD), which is actively produced in failing human heart, are able to induce apoptosis of rat cardiomyocytes. Polyamines are biogenic amines involved in many cellular processes, including apoptosis. Actually it appears that these molecules can act as promoting, modulating or protective agents in apoptosis depending on apoptotic stimulus and cellular model. We have studied the involvement of polyamines in the apoptosis of cardiac cells induced in a model of simulated ischemia and following treatment with NE or ALD. Methods: H9c2 cardiomyoblasts were exposed to a condition of simulated ischemia, consisting of hypoxia plus serum deprivation. Cardiomyocyte cultures were prepared from 1-3 day-old neonatal Wistar rat hearts. Polyamine depletion was obtained by culturing the cells in the presence of α-difluoromethylornithine (DFMO). Polyamines were separated and quantified in acidic cellular extracts by HPLC after derivatization with dansyl chloride. Caspase activity was measured by the cleavage of the fluorogenic peptide substrate. Ornithine decarboxylase (ODC) activity was measured by estimation of the release of 14C-CO2 from 14C-ornithine. DNA fragmentation was visualized by the method of terminal transferase-mediated dUTP nick end-labeling (TUNEL), and DNA laddering on agarose gel electophoresis. Cytochrome c was detected by immunoflorescent staining. Activation of signal transduction pathways was investigated by western blotting. Results: The results indicate that simulated ischemia, NE and ALD cause an early induction of the activity of ornithine decarboxylase (ODC), the first enzyme in polyamine biosynthesis, followed by a later increase of caspase activity, a family of proteases that execute the death program and induce cell death. This effect was prevented in the presence of DFMO, an irreversible inhibitor of ODC, thus suggesting that polyamines are involved in the execution of the death program activated by these stimuli. In H9c2 cells DFMO inhibits several molecular events related to apoptosis that follow simulated ischemia, such as the release of cytochrome c from mitochondria, down-regulation of Bcl-xL, and DNA fragmentation. The anti-apoptotic protein survivin is down-regulated after ALD or NE treatement and polyamine depletion obtained by DFMO partially opposes survivin decrease. Moreover, a study of key signal transduction pathways governing cell death and survival, revealed an involvement of AMP activated protein kinase (AMPK) and AKT kinase, in the modulation by polyamines of the response of cardiomyocytes to NE. In fact polyamine depleted cells show an altered pattern of AMPK and AKT activation that may contrast apoptosis and appears to result from a differential effect on the specific phosphatases that dephosphorylate and switch off these signaling proteins. Conclusions: These results indicate that polyamines are involved in the execution of the death program activated in cardiac cells by heart failure-related stimuli, like ischemia, ALD and NE, and suggest that their apoptosis facilitating action is mediated by a network of specific phosphatases and kinases.

Il lavoro svolto durante il dottorato di ricerca ha permesso lo sviluppo e la verifica della attendibilità di marcatori molecolari neutrali (loci microsatelliti) specifici per Aristeus antennatus. Tali marcatori sono stati poi utilizzati per studiare la struttura genetica di popolazione della specie del Mediterraneo occidentale e i risultati ottenuti sono stati confrontati con quelli di un progetto di ricerca parallelo su Aristeaomorpha foliacea, analizzando differenze ed analogie fra le due specie. I risultati delle analisi su Aristeus antennatus hanno evidenziato una completa assenza di struttura di popolazione e come i due sessi contribuiscano in modo diverso al flusso genico. La specie infatti presenta un sex-ratio a favore dei maschi oltre gli 800m, mentre tale rappoorto è a favore delle femmine in strati più superficiali, dove sono probabilmente soggette a condizioni oceanografiche più dispersive. Tramite test genetici appropriati è stato possibile valutare indirettamente il grado di dospersione dei sessi dimostrando che nell'area analizzati i maschi erano rappresentati maggiormente da individui stanziali, mentre gli individui di sesso femminile erano migranti. Le femmine appaiono pertanto giocare un ruolo preminente rispetto ai maschi nel determinare l'entità del flusso genico. Il confronto dei risultati ottenuti in Aristeus antennatus con quelli di Aristaeomorpha foliacea ha evidenziato la relazione fra alta capacità dispersiva, sia allo stato larvale che adulto, e completo rimescolamento genetico nei gamberi aristeidi nel Mediterraneo occidentale anche se in quest'ultima specie non ci sono evidenze di dispersione genetica mediata dal sesso. E' pertanto di forte interesse (dato anche il valore economico di questi organismi) come una struttura di popolazione qualitativamente e quantitavamente comporabile venga raggiunta con dinamiche di popolazione molto diverse.

Dopo aver analizzato il conflitto, le sue funzioni e le modalità di gestione, l'autore si sofferma dapprima sulle varie tipologie di mediazione per poi focalizzare l'attenzione sulla mediazione civile e commerciale evidenziando i dati disponibili dall'entrata in vigore del tentativo obbligatorio come condizione di procedibilità della domanda giudiziale per le materie civili, alla fine del 2013.

Until few years ago, 3D modelling was a topic confined into a professional environment. Nowadays technological innovations, the 3D printer among all, have attracted novice users to this application field. This sudden breakthrough was not supported by adequate software solutions. The 3D editing tools currently available do not assist the non-expert user during the various stages of generation, interaction and manipulation of 3D virtual models. This is mainly due to the current paradigm that is largely supported by two-dimensional input/output devices and strongly affected by obvious geometrical constraints. We have identified three main phases that characterize the creation and management of 3D virtual models. We investigated these directions evaluating and simplifying the classic editing techniques in order to propose more natural and intuitive tools in a pure 3D modelling environment. In particular, we focused on freehand sketch-based modelling to create 3D virtual models, interaction and navigation in a 3D modelling environment and advanced editing tools for free-form deformation and objects composition. To pursuing these goals we wondered how new gesture-based interaction technologies can be successfully employed in a 3D modelling environments, how we could improve the depth perception and the interaction in 3D environments and which operations could be developed to simplify the classical virtual models editing paradigm. Our main aims were to propose a set of solutions with which a common user can realize an idea in a 3D virtual model, drawing in the air just as he would on paper. Moreover, we tried to use gestures and mid-air movements to explore and interact in 3D virtual environment, and we studied simple and effective 3D form transformations. The work was carried out adopting the discrete representation of the models, thanks to its intuitiveness, but especially because it is full of open challenges.
