This work shows for the first time that native CSTB polymerizes on addition of Cu2+ and DnaK (Hsp70). Cysteines are involved in the polymerization process and in particular at least one cysteine is necessary. We propose that Cu2+ interacts with the thiol group of cysteine and oxidize it. The oxidized cysteine modifies the CSTB structure allowing interaction with DnaK/Hsp70 to occur. Thus, Cu2+ binding to CSTB exposes a site for DnaK and such interaction allows the polymerization of CSTB. The polymers generated from native CSTB monomers, are DTT sensitive and they may represent physiological polymers. Denatured CSTB does not require Cu2+ and polymerizes simply on addition of DnaK. The polymers generated from denatured CSTB do not respond to DTT. They have characteristics similar to those of the CSTB toxic aggregates described in vivo in eukaryotic cells following CSTB over-expression. Interaction between CSTB and Hsp70 is shown by IP experiments. The interaction occurs with WT CSTB and not with the ïcys mutant. This suggests that disulphur bonds are involved. Methal-cathalyzed oxidation of proteins involves reduction of the metal ion(s) bound to the protein itself and oxidation of neighboring ammino acid residues resulting in structural modification and de-stabilization of the molecule. In this work we propose that the cysteine thyol residue of CSTB in the presence of Cu2+ is oxidized, and cathalyzes the formation of disulphide bonds with Hsp70, that, once bound to CSTB, mediates its polymerization. In vivo this molecular mechanism of CSTB polymerization could be regulated by redox environment through the cysteine residue. This may imply that CSTB physiological polymers have a specific cellular function, different from that of the protease inhibitor known for the CSTB monomer. This hypothesis is interesting in relation to Progressive Myoclonus Epilepsy of type 1 (EPM1). This pathology is usually caused by mutations in the CSTB gene. CSTB is a ubiquitous protein, but EPM1 patients have problems only in the central nervous system. Maybe physiological CSTB polymers have a specific function altered in people affected by EPM1.

We aimed to evaluate the role of anti-TNF-alpha therapy with infliximab and adalimumab in a cohort of pediatric patients followed by our Center from 2002 to 2012. The cohort of patients examined consisted of 40 patients: 34 with Crohn disease (85%), 5 with ulcerative colitis (12.5%), one with chronic pouchitis after IPAA for ulcerative colitis (2.5%). All patients were treated with the anti-TNF-α biologic agents infliximab and adalimumab. Thirty-six received infliximab therapy: 19/36 received only infliximab, 17/36 received infliximab and then adalimumab due to loss of response to infliximab and steroid dependency; 4 patients received only adalimumab (infliximab-naïve). Anti-TNF treatment was started before 18 years of age in 34 patients: 29 received infliximab and 5 started adalimumab during childhood. Medical charts were reviewed and safety and efficacy of anti-TNF-alpha have been determined in this population.

Endodontic therapy consists in the management of several tissues such as pulp tissue, periodontal tissue, periapical bone and dentine. These tissues are often contaminated by blood, periapical exudates and biological fluids. An ideal orthograde or retrograde filling material should be non toxic, noncarcinogenic, nongenotoxic, biocompatible with the host tissues, insoluble in tissue fluids, and dimensionally stable. Calcium-silicate MTA based cements own many of these ideal characteristics, but the long setting time, the non-easy handling and the lack of mechanical properties at early times are few drawbacks which may complicate the clinical application. The aim of this study was to investigate the chemical, physical and biological properties of calcium-silicate MTA cements in order to improve the mechanical properties and the handling keeping the biological characteristics unchanged. Chemical and physical properties such as setting time, solubility, water-uptake, ion release, sealing ability were investigated according the ISO and ADA specifications. The bioactivity (ability to produce apatite nano-sferulities) of MTA cements were evaluated using ESEM/EDX, micro-Raman and ATR/FTIR spettroscopy.

La tesi affronta il concetto di esposizione al rischio occupazionale e il suo scopo è quello di indagare l’ambiente di lavoro e il comportamento dei lavoratori, con l'obiettivo di ridurre il tasso di incidenza degli infortuni sul lavoro ed eseguire la riduzione dei rischi. In primo luogo, è proposta una nuova metodologia denominata MIMOSA (Methodology for the Implementation and Monitoring of Occupational SAfety), che quantifica il livello di "salute e sicurezza" di una qualsiasi impresa. Al fine di raggiungere l’obiettivo si è reso necessario un approccio multidisciplinare in cui concetti d’ingegneria e di psicologia sono stati combinati per sviluppare una metodologia di previsione degli incidenti e di miglioramento della sicurezza sul lavoro. I risultati della sperimentazione di MIMOSA hanno spinto all'uso della Logica Fuzzy nel settore della sicurezza occupazionale per migliorare la metodologia stessa e per superare i problemi riscontrati nell’incertezza della raccolta dei dati. La letteratura mostra che i fattori umani, la percezione del rischio e il comportamento dei lavoratori in relazione al rischio percepito, hanno un ruolo molto importante nella comparsa degli incidenti. Questa considerazione ha portato ad un nuovo approccio e ad una seconda metodologia che consiste nella prevenzione di incidenti, non solo sulla base dell'analisi delle loro dinamiche passate. Infatti la metodologia considera la valutazione di un indice basato sui comportamenti proattivi dei lavoratori e sui danni potenziali degli eventi incidentali evitati. L'innovazione consiste nell'applicazione della Logica Fuzzy per tener conto dell’"indeterminatezza" del comportamento umano e del suo linguaggio naturale. In particolare l’applicazione è incentrata sulla proattività dei lavoratori e si prefigge di impedire l'evento "infortunio", grazie alla generazione di una sorta d’indicatore di anticipo. Questa procedura è stata testata su un’azienda petrolchimica italiana.

The development of High-Integrity Real-Time Systems has a high footprint in terms of human, material and schedule costs. Factoring functional, reusable logic in the application favors incremental development and contains costs. Yet, achieving incrementality in the timing behavior is a much harder problem. Complex features at all levels of the execution stack, aimed to boost average-case performance, exhibit timing behavior highly dependent on execution history, which wrecks time composability and incrementaility with it. Our goal here is to restitute time composability to the execution stack, working bottom up across it. We first characterize time composability without making assumptions on the system architecture or the software deployment to it. Later, we focus on the role played by the real-time operating system in our pursuit. Initially we consider single-core processors and, becoming less permissive on the admissible hardware features, we devise solutions that restore a convincing degree of time composability. To show what can be done for real, we developed TiCOS, an ARINC-compliant kernel, and re-designed ORK+, a kernel for Ada Ravenscar runtimes. In that work, we added support for limited-preemption to ORK+, an absolute premiere in the landscape of real-word kernels. Our implementation allows resource sharing to co-exist with limited-preemptive scheduling, which extends state of the art. We then turn our attention to multicore architectures, first considering partitioned systems, for which we achieve results close to those obtained for single-core processors. Subsequently, we shy away from the over-provision of those systems and consider less restrictive uses of homogeneous multiprocessors, where the scheduling algorithm is key to high schedulable utilization. To that end we single out RUN, a promising baseline, and extend it to SPRINT, which supports sporadic task sets, hence matches real-world industrial needs better. To corroborate our results we present findings from real-world case studies from avionic industry.
