Il carcinoma epatocellulare (HCC) è il più frequente tumore maligno del fegato e rappresenta il sesto tipo di tumore più comune nel mondo. Spesso i pazienti con HCC vengono diagnosticati a stadi piuttosto avanzati, quando le uniche opzioni terapeutiche in grado di migliorarne la sopravvivenza sono la chemoembolizzazione dell'arteria epatica ed il trattamento con l'inibitore multi-cinasico, Sorafenib. In questo contesto, la scoperta del ruolo centrale dei microRNA (miRNA) nella tumorigenesi umana risulta di fondamentale importanza per lo sviluppo di nuovi marcatori diagnostici e bersagli terapeutici. I microRNA (miRNA) sono delle piccole molecole di RNA non codificante, della lunghezza di 19-22 nucleotidi, filogeneticamente molto conservati, ed esercitano un ruolo cruciale nella regolazione di importanti processi fisiologici, quali sviluppo, proliferazione, differenziamento, apoptosi e risposta a numerosi segnali extracellulari e di stress. I miRNA sono inoltre responsabile della fine regolazione dell'espressione di centinaia di geni bersaglio attraverso il blocco della traduzione o la degradazione dell'mRNA target. Studi di profiling hanno evidenziato l'espressione aberrante di specifici miRNA in numerosi tipi di tumore umano. Lo scopo del presente lavoro è stato quello di individuare un pannello di miRNA deregolati nell'epatocarcinoma umano e di caratterizzare il ruolo biologico di tre miRNA deregolati nell'HCC, al fine di individuare alcuni dei meccanismi molecolari alla base della trasformazione maligna miRNA-associata. La nostra ricerca è stata inoltre focalizzata nell'individuazione di nuovi bersagli e strumenti terapeutici, quali i microRNA, per il trattamento combinato di HCC in stadio intermedio-avanzato.

Aim of the present study was to evaluate the accuracy of transrectal ultrasound biopsy (TRUS-biopsy) directed to regions with abnormal MRI and/or MRSI (magnetic resonance spectroscopic imaging ) for both the transition (TZ) and the peripheral (PZ) zones in patients who presented with persistent suspect for prostate cancer and with prior negative biopsy. We also evaluated relationship between MRSI results and histopathological findings of biopsy. 54 patients with the aforementioned characteristics underwent MRI/MRSI at least 6 months after prior negative biopsy; interval between MRI/3D-MRSI and the further TRUS-biopsy was less than 3 months. The prostate was divided in 12 regions both for imaging interpretation and biopsy. Moreover one to three cores more were taken from each region with abnormal MRI and/or 3D-MRSI. Twenty-two out of 54 patients presented cancer at MRI/MRSI-directed-TRUS-biopsy. On a patient basis the highest accuracy was obtained by assigning malignancy on a positive finding with MRSI and MRI even though it was not significantly greater than that obtained using MRI alone (area under the ROC curve, AUC: 0.723 vs. 0.676). On a region (n=648) basis the best accuracy was also obtained by considering positive both MRSI and MRI for PZ (0.768) and TZ (0.822). Twenty-eight per cent of cores with prostatitis were false positive findings on MRSI, whereas only 2.7% of benign prostatic hyperplasia was false positive. In conclusion the accuracy of MRI/MRSI-directed biopsies in localization of prostate cancer is good in patient and region analyses. The combination of both MRI and MRSI results makes TRUS-biopsy more accurate particularly in the TZ (0.822) for patients with prior negative biopsies. Histopathological analysis showed that the main limitation of MRSI is the percentage of false positive findings due to prostatitis.

Data coming out from various researches carried out over the last years in Italy on the problem of school dispersion in secondary school show that difficulty in studying mathematics is one of the most frequent reasons of discomfort reported by students. Nevertheless, it is definitely unrealistic to think we can do without such knowledge in today society: mathematics is largely taught in secondary school and it is not confined within technical-scientific courses only. It is reasonable to say that, although students may choose academic courses that are, apparently, far away from mathematics, all students will have to come to terms, sooner or later in their life, with this subject. Among the reasons of discomfort given by the study of mathematics, some mention the very nature of this subject and in particular the complex symbolic language through which it is expressed. In fact, mathematics is a multimodal system composed by oral and written verbal texts, symbol expressions, such as formulae and equations, figures and graphs. For this, the study of mathematics represents a real challenge to those who suffer from dyslexia: this is a constitutional condition limiting people performances in relation to the activities of reading and writing and, in particular, to the study of mathematical contents. Here the difficulties in working with verbal and symbolic codes entail, in turn, difficulties in the comprehension of texts from which to deduce operations that, once combined together, would lead to the problem final solution. Information technologies may support this learning disorder effectively. However, these tools have some implementation limits, restricting their use in the study of scientific subjects. Vocal synthesis word processors are currently used to compensate difficulties in reading within the area of classical studies, but they are not used within the area of mathematics. This is because the vocal synthesis (or we should say the screen reader supporting it) is not able to interpret all that is not textual, such as symbols, images and graphs. The DISMATH software, which is the subject of this project, would allow dyslexic users to read technical-scientific documents with the help of a vocal synthesis, to understand the spatial structure of formulae and matrixes, to write documents with a technical-scientific content in a format that is compatible with main scientific editors. The system uses LaTex, a text mathematic language, as mediation system. It is set up as LaTex editor, whose graphic interface, in line with main commercial products, offers some additional specific functions with the capability to support the needs of users who are not able to manage verbal and symbolic codes on their own. LaTex is translated in real time into a standard symbolic language and it is read by vocal synthesis in natural language, in order to increase, through the bimodal representation, the ability to process information. The understanding of the mathematic formula through its reading is made possible by the deconstruction of the formula itself and its “tree” representation, so allowing to identify the logical elements composing it. Users, even without knowing LaTex language, are able to write whatever scientific document they need: in fact the symbolic elements are recalled by proper menus and automatically translated by the software managing the correct syntax. The final aim of the project, therefore, is to implement an editor enabling dyslexic people (but not only them) to manage mathematic formulae effectively, through the integration of different software tools, so allowing a better teacher/learner interaction too.

The emergency of infection by highly pathogenic avian influenza virus (HPAI) subtype H5N1 has focused the attention of the world scientific community, requiring the prompt provision of effective control systems for early detection of the circulation of low pathogenic influenza H5 viruses (LPAI) in populations of wild birds to prevent outbreaks of highly pathogenic (HPAI) in populations of domestic birds with possible transmission to humans. The project stems from the aim to provide, through a preliminary analysis of data obtained from surveillance in Italy and Europe, a preliminary study about the virus detection rates and the development of mathematical models, an objective assessment of the effectiveness of avian influenza surveillance systems in wild bird populations, and to point out guidelines to support the planning process of the sampling activities. The results obtained from the statistical processing quantify the sampling effort in terms of time and sample size required, and simulating different epidemiological scenarios identify active surveillance as the most suitable for endemic LPAI infection monitoring in wild waterfowl, and passive surveillance as the only really effective tool in early detecting HPAI H5N1 circulation in wild populations. Given the lack of relevant information on H5N1 epidemiology, and the actual finantial and logistic constraints, an approach that makes use of statistical tools to evaluate and predict monitoring activities effectiveness proves to be of primary importance to direct decision-making and make the best use of available resources.

Il ginandromorfismo è il fenomeno per il quale un organismo manifesta contemporaneamente caratteristiche fenotipiche maschili e femminili. Per quanto riguarda la Classe degli Insetti, numerose segnalazioni di tale manifestazione sono reperibili in letteratura, ma un’ interpretazione generale sulle origini e sulle cause che la generano non è ancora stata fornita. Lo scopo di questa tesi è stato quello di studiare il fenomeno per quanto riguarda l’Imenottero Diprionide Diprion pini (Linnaeus, 1758) attraverso l’allevamento controllato dell’insetto, esperimenti di inincrocio, studio del cariotipo e la valutazione della comparsa e la distribuzione dei tessuti maschili e femminili negli individui ginandromorfi. Altri parametri biologici (quali i pesi degli individui) sono stati presi in considerazione nel tentativo di fornire una spiegazione riguardo i meccanismi genetici che regolano la determinazione del sesso in questa specie. Gynandromorphism is the phenomenon by which an organism manifests phenotypic characteristics both male and female. For the class of insects, numerous reports of this event can be found in the literature, but a general interpretation of the origins and causes that generate it has not yet been provided. The purpose of this thesis was to study the phenomenon with regard to the Diprionid wasp Diprion pini (Linnaeus, 1758) through the controlled rearing of the insect, inbreeding experiments, study of the karyotype and evaluation of the appearance and distribution of male and female tissue in gynandromorph specimens. Other biological parameters (such as the weights of individuals) were taken into account in an attempt to provide an explanation of the genetic mechanisms that regulate sex determination in this species.

Why some powers manage to coordinate their security efforts while others confront each other as rivals is still one of the most relevant and debated questions in the field of IR theory. The dissertation wants to give a contribution to this important debate. In particular, the main goal of the research is to analyse the dynamics of great power interactions after the end of hegemonic conflicts, that is to understand why, following the defeat of the common enemies, some of the winning allies continue to cooperate, while others begin to engage in political and military competition. In order to understand this difference, the study compares the explanatory value of two rival theoretical perspectives: neorealism, in its main version of the balance of power framework, and a liberal approach focused on domestic politics. The thesis is divided in two sections. In the first, I do summarize the main assumptions and predictions of the theories, from which I derive two different sets of hypotheses on the evolution of post-war great power relations. In the second part, I test the hypotheses by focusing on two cases of post-war alignment dynamics: 1) the relations among Austria, Prussia, Russia, Great Britain and France after the Napoleonic wars; 2) the relations among the US, the UK, France and Italy after the end of WWI. The historical cases disconfirm the logic of the balance of power and confirm the liberal hypotheses, seeing that the results of the analysis show changes in the domestic structures of the great powers had a much larger impact on the emergence of new alliances and rivalries than did the international distribution of power. In the conclusion of the dissertation, I provide the reader with a discussion of the main theoretical implications of the empirical findings.

L’attività della tesi riguarda le protesi mioelettriche, gli arti protesici maggiormente diffusi, le quali sono descrivibili come arti robotici in cui i segmenti artificiali sono attuati da giunti elettromeccanici alimentati da batterie ricaricabili ed attivati mediante segnali elettromiografici (segnali elettrici generati dalla contrazione dei muscoli). Tali protesi di arto superiore attualmente disponibili in commercio potrebbero essere inadeguate per una riabilitazione soddisfacente di alcuni pazienti con una amputazione di alto livello che richiedono una elevata funzionalità nella vita quotidiana. In questo contesto si inserisce l’attività di ricerca del Centro Protesi INAIL di Budrio di Vigorso, Bologna, e dell’Università di Bologna i quali stanno sviluppando nuovi arti protesici con il progetto a lungo termine di rendere disponibili svariate soluzioni di protesi di arto superiore in grado di soddisfare la maggior parte delle richieste degli amputati. Lo scopo di questa tesi è l’introduzione di un nuovo rotatore omerale attivo da integrare alla protesi di arto superiore disponibile presso i nostri laboratori. Per ottenere questo risultato è stata utilizzata una procedura di progettazione già consolidata in attività precedenti per lo sviluppo di una protesi di spalla a due gradi di libertà. Differenti modelli cinematici sono stati studiati tramite analisi cinematiche per determinare l’incremento delle prestazioni a seguito dell’introduzione del nuovo rotatore omerale attivo. Sono state inoltre condotte analisi cinetostatiche per definire le specifiche tecniche di riferimento (in termini di carichi agenti sul rotatore omerale) e per guidare il dimensionamento della catena di trasmissione di potenza del nuovo dispositivo protesico. Ulteriori specifiche tecniche sono state considerate per garantire l’irreversibilità spontanea del moto sotto carichi esterni (quando i giunti attivi della protesi non sono alimentati), per salvaguardare l’incolumità del paziente in caso di caduta, per misurare la posizione angolare del rotatore omerale (in modo da implementare strategie di controllo in retroazione) e per limitare i consumi e la rumorosità del dispositivo. Uno studio di fattibilità ha permesso la selezione della architettura ottimale della catena di trasmissione di potenza per il nuovo rotatore omerale. I criteri di scelta sono stati principalmente la limitazione del peso e dell’ingombro del nuovo dispositivo protesico. Si è quindi proceduto con la progettazione di dettaglio alla quale è seguita la costruzione di un prototipo del nuovo rotatore omerale presso i nostri laboratori. La tesi tratta infine una attività preliminare di sperimentazione che ha permesso di fare considerazioni sulle prestazioni del prototipo ed osservazioni importanti per le successive attività di revisione ed ottimizzazione del progetto del rotatore omerale.

The objective was to analyse population structure and to determine genetic diversity of Erysiphe necator (syn. Uncinula necator) populations obtained from some vineyards located in the South-East Po valley (Italy). Powdery mildew is one of the most important fungal diseases of grapes (Vitis vinifera L.) throughout the world. The causal agent is the haploid, heterothallic ascomycete E. necator. It is an obligate biotrophic fungus and it can be found only on green organs of plants belonging to the family Vitaceae. For this pathogen, two sympatric populations (groups A and B) have been described in Europe and Australia. The two genetic groups differ at multiple genetic loci and previous studies reported a lack of interfertility among isolates of the two groups. There are now several well documented examples of plant pathogen species, such as Leptosphaeria maculans, Gaeumannomyces graminis var. tritici, Botrytis cinerea and Erysiphe syringae, which are indeed composed of genetically differentiated clades, that have led to the description of new groups or even new species. Several studies have suggested that genetic E. necator group A and B correlated with ecological features of the pathogen; some researchers proposed that group A isolates over-winter as resting mycelium within dormant buds, and in spring originate infected shoots, known as Flag shoots, while group B isolates would survive as ascospores in overwintering cleistothecia. However, the association between genetic groups and mode of over-wintering has been challenged by recent studies reporting that flag-shoot may be originated indifferently by group A or group B isolate. Previous studies observed a strong association between the levels of disease severity at the end of the growing season and the initial compositions of E. necator populations in commercial vineyards. The frequencies of E. necator genetic groups vary considerably among vineyards, and the two groups may coexist in the same vineyard. This finding suggests that we need more information on the genetics and epidemiology of E. necator for optimize the crop management In this study we monitored E. necator populations in different vineyards in Emilia – Romagna region (Italy), where the pathogen overwinters both as flagshoots and as cleistothecia. During the grape growing season, symptomatic leaves were sampled early in the growing season and both leaves and berries later during the epidemic growth of the disease. From each sample, single-conidial isolate was obtained. Each isolates was grown on V. vinifera leaf cv. Primitivo and after harvesting the mycelium, the DNA was purified and used as template for PCR amplification with SCAR primers (Sequences Characterised Amplified Region ), -tubulin, IGS sequences and Microsatellite markers (SSR). Amplified DNA from b-tubulin and IGS loci was digested with AciI and XhoI restriction enzymes, respectively, to show single-nucleotide polymorphisms specific for the two genetic groups. The results obtained indicated that SCAR primers are not useful to study the epidemiology. of E. necator conversely the b-tubulin IGS sequences and SSR. Summarize the results obtained with b-tubulin, IGS sequences, in treated vineyards we have found individuals of group B along all grape growing season, whereas in the untreated vineyard individuals of the two genetic groups A and B coexisted throughout the season, with no significant change of their frequency. DNA amplified from ascospores of single cleistothecia showed the presence of markers diagnostic for either groups A and B and were seldom observed also the coexistence of both groups within a claistothecium. These results indicate that individuals of the two groups mated in nature and were able to produced ascospores. With SSR we showed the possibility of recombination between A and B groups in field isolates. During winter, cleistothecia were collected repeatedly in the same vineyards sampling leaves fallen on ground, exfoliating bark from trunks, and from soil. From each substrate, was assess the percentage of cleistothecia containing viable ascospores. Our results confirmed that cleisthotecia contained viable ascospores, therefore they have the potential to be an additional and important source of primary inoculum in Emilia-Romagna vineyards.

Recent reports showed that early-interim PET-scan is the only tool predicting treatment outcome in advanced-stage classical Hodgkin lymphoma (asCHL). We evaluated the prognostic impact of a series of immunohistochemical markers, mentioned in literature as prognostic factors, on tissue microarrays assembled from biopsies of 220 patients: STAT1, SAP, TOP2A, PCNA and CD20, both in neoplastic (HRSC) and microenvironment cells (MC); RRM2, MAD2, CDC2, BCL2, P53, BCL11A and EBER in HRSC; ALDH1A1, TIA-1, granzyme B, perforin, FOXP3, and PD-1 in MC. All patients had been treated with standard ABVD ± Rx therapy. Interim-PET after 2 ABVD courses was evaluated according to the criteria indicated by Gallamini in his study (Journal of Clinical Oncology, 2007). The survival analysis has been performed in a subset of 138 patients whose complete clinical information were available: the mean age was 33.3 years (14-79), the stage III-IVB in 98 and IIB in 40, and the mean follow-up 38.1 months (7.6-71.9). Histopathology review showed: NS-I 75, NS-II 22, MC 20, DL 3, and CHL/nos 18 cases. Interim-PET was positive in 30 patients, while treatment failure was recorded in 32. In univariate analysis the factors related to treatment outcome were BCL2 on HRSC (cut-off value 50%), STAT1/SAP on MC, and PET (Log-rank 6.9, 7.9 and 93.9 respectively). The combined expression of STAT1 and SAP was scored in three levels depending on the architectural pattern: score 0 for expression of both with a diffuse/rosetting pattern; score 1 for discordant combination of diffuse/rosetting and scattered patterns; score 2 for both markers with a scattered pattern; the 3y-PFS were 87.4%, 69.9% and 61.9% respectively. In multivariate analysis PET, BCL2 and STAT1/SAP remained significant (HR: 24.8, 4.6, 7.5 and 5.6, respectively; p<.01). The proposed model is able to predict treatment response in AsCHL, even if with a lower efficacy than PET. However, unlike PET, it can be applied upfront therapy.
