This dissertation consists of three standalone articles that contribute to the economics literature concerning technology adoption, information diffusion, and network economics in one way or another, using a couple of primary data sources from Ethiopia. The first empirical paper identifies the main behavioral factors affecting the adoption of brand new (radical) and upgraded (incremental) bioenergy innovations in Ethiopia. The results highlight the importance of targeting different instruments to increase the adoption rate of the two types of innovations. The second and the third empirical papers of this thesis, use primary data collected from 3,693 high school students in Ethiopia, and shed light on how we should select informants to effectively and equitably disseminate new information, mainly concerning environmental issues. There are different well-recognized standard centrality measures that are used to select informants. These standard centrality measures, however, are based on the network topology---shaped only by the number of connections---and fail to incorporate the intrinsic motivations of the informants. This thesis introduces an augmented centrality measure (ACM) by modifying the eigenvector centrality measure through weighting the adjacency matrix with the altruism levels of connected nodes. The results from the two papers suggest that targeting informants based on network position and behavioral attributes ensures more effective and equitable (gender perspective) transmission of information in social networks than selecting informants on network centrality measures alone. Bifidobacteria is amongst one of the health promoting bacteria. The role of this important probiotic genera can be elucidated by understanding its genome. Comparative analysis of the whole genus of these bacteria can reveal their adaptation to a diverse host range. This study comprises of four research projects. In the first study, a reference library for genus Bifidobacterium was prepared. The core genes in each genus were selected based on a newly proposed statistical definition of core genome. Comparative analysis of Bifidobacterium with another probiotic genus Lactobacillus revealed the metabolic characteristics of genus Bifidobacterium. The second study investigated the immunomodulatory role of a B. bifidum strain TMC3115. The analysis of TMC3115 provided insights into its extracellular structures which might have their role in host interaction and immunomodulation. The study highlighted the variability among these genomes just not on species level but also on strain level in terms of host interaction. The last two studies aim to inspect the relationship between bifidobacteria and its host diet. Bifidobacteria, are both host- and niche-specific. Such adaptation of bifidobacterial species is considered relevant to the intestinal microecosystem and hosts’ oligosaccharides. Many species should have co-evolved with their hosts, but the phylogeny of Bifidobacterium is dissimilar to that of host animals. The discrepancy could be linked to the niche-specific evolution due to hosts’ dietary carbohydrates. The distribution of carbohydrate-active enzymes, in particular glycoside hydrolases (GHs) that metabolize unique oligosaccharides was examined. When bifidobacterial species were classified by their distribution of GH genes, five groups arose according to their hosts’ feeding behaviour. The distribution of GH genes was only weakly associated with the phylogeny of the host animals or with genomic features such as genome size. Thus, the hosts’ dietary pattern is the key determinant of the distribution and evolution of GH genes.

In colorectal cancer (CRC), two carbohydrate structures are modulated: the Sda antigen, synthesized by B4GALNT2, and sLex antigen, mainly synthesized by FUT6. sLex antigen is often overexpressed and associated with worse prognosis; B4GALNT2/Sda antigen are dramatically downregulated but their role in tumor progression and development is not fully clear. TCGA interrogation revealed a dramatic down-regulation of B4GALNT2 mRNA in CRC, compared with normal samples. Patients with higher B4GALNT2 mRNA in CRC samples displayed longer survival. Yet, methylation and miRNA expression play a relevant role in B4GALNT2 downregulation in CRC. To clarify the mechanisms linking the B4GALNT2/Sda expression level to CRC phenotype, three different CRC cell lines were modified to express B4GALNT2: LS174T cell line, in which the constitutively expressed sLex antigen was partially replaced by Sda; SW480/SW620 pair, both lacking Sda and sLex antigens. In LS174T cells, the expression of B4GALNT2 reduced the ability to grow in poor adherence conditions and the expression of ALDH, a stemness marker. In SW620 cells, B4GALNT2 expression impacted on the main aspects of malignancy. In SW480 cells the expression of B4GALNT2 left unchanged the proliferation rate and the wound healing ability. To clarify the impact of sLex on CRC phenotype, the SW480/SW620 pair were permanently transfected to express FUT6 cDNA. In both cell lines, overexpression of FUT6/sLex boosted the clonogenic ability in standard growth conditions. Conversely, the growth in soft agar and the capacity to close a wound were enhanced only in SW620 cells. Transcriptome analysis of CRC cell lines transfected either with B4GALNT2 or FUT6 showed a relevant impact of both enzymes on gene expression modulation. Overall, current data may help to personalize therapies for CRC patients according to the B4GALNT2 levels and support a causal effect of this glycosyltransferase on reducing malignancy independently of sLex inhibition.

Ex-situ conservation and the in-situ conservation of natural habitats are the tools to conserve biodiversity. Habitats and ecosystems have been becoming altered by human activities and a growing number of species requires form of management to ensure their survival. Conservation queries become more complex and urgent. Developing scientifically based and innovative approaches to ex-situ conservation is necessary. Recent studies underline importance of gut microbiome in animal health with implications for animal conservation and management. Animal and human studies have demonstrated that environmental factors can impact gut microbiome composition. Within this scenario, the present work focused on species belonging to different taxa, reptiles and mammals: Aldabrachelys gigantea, the giant tortoise of the Seychelles islands and Indri indri, the greatest leaving lemur of Madagascar. The Seychelles giant tortoise is vulnerable species with declining population, whereas the indri is a critically endangered species that could reach the extinction within 25 years. Both need research to help them to survive. Tortoises live for very long time and to observe how they can afford the environmental changes is very difficult. Indris, instead, are able to survive only in a small area of the Madagascar forest, with a very strong link between the species’ survival and the environment. The obtained results underline importance of environmental factors, both in-situ and ex-situ, for species conservation. Microbiome could help the organisms to respond on a short timescale and cope with, environmental changes. However, species with long generation time might not be able to adapt to fast changes but bacteria with a short generation time can adapt on a shorter timescale allowing the host to cope with fluctuating environment. Gut microbiome plays an important role in an animal’s health and has the potential to improve the management of individuals under human care for conservation purposes.

Strawberry (Fragaria × ananassa) is an important soft fruit but easily to be infected by pathogens. Anthracnose and gray mold are two of the most destructive diseases of strawberry which lead to serious fruit rot. The first chapter introduced strawberry anthracnose caused by Colletotrichum acutatum. The infection strategy, disease cycle and management of C. acutatum on strawberry were reported. Likewise, the second chapter summarized the infection strategy of Botrytis cinerea and the defense responses of strawberry. As we already know white unripe strawberry fruits are more resistant to C. acutatum than red ripe fruits. During the interaction between strawberry white/red fruit and C. acutaum, a mannose binding lectin gene, FaMBL1, was found to be the most up-regulated gene and induced exclusively in white fruit. FaMBL1 belongs to the G-type lectin family which has important roles in plant development and defense process. To get insight into the role of FaMBL1, genome-wide identification was carried out on G-type lectin gene family in Fragaria vesca and the results were showed in chapter 3. G-type lectin genes make up a large family in F. vesca. Active expression upon biotic/abiotic stresses suggested a potential role of G-lectin genes in strawberry defenses. Hence, stable transgenic strawberry plants with FaMBL1 gene overexpressed were generated. Transformed strawberry plants were screened and identified. The results were showed in chapter 4, content of disease-related phytohormone, jasmonic acid, was found decreased in overexpressing lines compared with wild type (WT). Petioles inoculated by C. fioriniae of overexpressing lines had lower disease incidence than WT. Leaves of overexpressing lines challenged by B. cinerea showed remarkably smaller lesion diameters compared with WT. The chitinase 2-1 (FaChi2-1) showed higher expression in overexpressing lines than in WT during the interaction with B. cinerea, which could be related with the lower susceptibility of overexpressing lines.

Two-year field trials were conducted in northern Italy with the aim of developing a trapcrop-based agroecological approach for the control of flea beetles (Chaetocnema tibialis (Illiger), Phyllotreta spp. (Chevrolat) (Coleoptera: Chrysomelidae)) and Lygus rugulipennis Poppius (Hemiptera: Miridae), key pests of sugar beet and lettuce, respectively. Flea beetle damage trials compared a trap cropping treatment, i.e., a sugar beet plot with a border of Sinapis alba (L.) and Brassica juncea (L.) with a control treatment, i.e., a sugar beet plot with bare soil as field border. Sugar beets grown near trap crops showed a significant decrease (≈40%) in flea beetle damage compared to control. Moreover, flea beetle damage varied with distance from the edge of the trap plants, being highest at 2 m from the edge, then decreasing at higher distances. Regarding L. rugulipennis on lettuce two experiments were conducted. A semiochemical-assisted trap cropping trial was supported by another test evaluating the efficacy of pheromones and trap placement. In this trial, it was found that pheromone baited traps caught significantly more specimens of L. rugulipennis than unbaited traps. It was also found that traps placed at ground level produced larger catches than traps placed at the height of 70 cm. In the semiochemical-assisted trap cropping experiment, a treatment where lettuce was grown next to two Alfa-Alfa borders containing pheromone baited traps was compared with a control treatment, where lettuce was grown near bare soil. This experiment showed that the above-mentioned strategy managed to reduce L. rugulipennis damage to lettuce by ≈30%. From these studies, it appears that trap crop-based strategy, alone or with baited traps, made it possible to reduce crop damage to economically acceptable levels and to minimize the need for insecticide treatments, showing that those strategy could be implemented in organic farming as a means of controlling insect pests.

This research aims to discover the virome diversity and composition in Fusarium poae and Fusarium proliferatum collections, characterize the mycovirus that may have an effect on host pathogenicity to provide potential materials for the biological control of Fusarium spp. pathogens. Next-Generation Sequencing (NGS) analysis of 30 F. poae isolates revealed an extreme diversity of mycoviruses. Bioinformatic analysis shows that contigs associated with viral genome belong to the families: Hypoviridae, Mitoviridae, Partitiviridae, Polymycoviridae, proposed Alternaviridae, proposed Fusagraviridae, proposed Fusariviridae, proposed Yadokariviridae, and Totiviridae. The complete genomes of 12 viruses were obtained by assembling contigs and overlapping cloning sequences. Moreover, all the F. poae isolates analyzed are multi-infected. Fusarium poae partitivirus 1 appears in all the 30 strains, followed by Fusarium poae fusagravirus 1 (22), Fusarium poae mitovirus 2 (18), Fusarium poae partitivirus 3 (16), and Fusarium poae mitovirus 2 and 3 (11). Using the same approach, the virome of F. proliferatum collections resulted in lower diversity and abundance. The identified mycoviruses belong to the family Mitoviridae and Mymonaviridae. Interestingly, most F. proliferatum isolates are not multi-infected. The complete genomes of four viruses were obtained by assembling contigs and overlapping cloning sequences. By multiple liner regression of the virome composition and growth rate of 30 F. poae, Fusarium poae mitovirus 3 is significantly correlated with the growth rate among F. poae collection. Furthermore, the principal component analysis of the virome composition from 30 F. poae showed that the presence of Fusarium poae mitovirus 3 and other two viruses could increase the F. poae growth rate. The curing experiment and pathogenicity test in Petri indicated that Fusarium poae hypovirus 1 might be associated with the host hypovirulence phenotype, while Fusarium poae fusagravirus 1 and Fusarium poae partitivirus 3 may have some beneficial effect on host pathogenicity.

To address the request to develop rapid and easy methods for determining the cannabinoids, an HPLC-UV method (8 min) to separate and quantify the 10 main cannabinoids in hemp inflorescences was developed, and in-house validated. Moreover, the antioxidant activity of cannabidiol (CBD) in two oily matrices was investigated and compared to that of α-tocopherol, in relation to the growing market of oily solutions containing cannabidiol. Then, since no univocal legislation on the evaluation of quality and authenticity of hemp seed oil (HSO) exists, the composition and quality of cold-pressed HSOs were also explored, highlighting a great variability in terms of oxidative state minor compounds content. From the sensory point of view, a panel was trained, a specific sensory wheel and a profile sheet were developed. Due to the Covid-19 pandemic, the sensory evaluation was also performed at home. The panel showed a good performance both in the laboratory and remotely. Moreover, a focus group was used to investigate consumers’ attitudes, pointing out that a high-quality HSO has to be cold-pressed and green for them. Then, the evaluation of stability during the storage of HSOs was investigated. The results showed that photo-oxidation did not seem to significantly affect the quality of the oil during the first 3 months of storage. Finally, a study about the evolution of the volatile profile of 9 HSOs, under accelerated oxidation conditions, allowed identifying volatile markers of HSOs oxidation and freshness. This thesis reports five studies that may contribute to understand how weaning affects the immune and intestinal microbiota maturation of the piglet and proposes some possible nutritional strategies to attenuate its negative effects. The first study showed that weaning is associated in Payer’s patches with the activation of MHC response against class I antigens and that related to the stimulation to IFN-γ and showed, for the first time, that their blood at weaning remains dominated by immature blood cells. In the second study we tested if the use of a live vaccine against a conditionally but also genetically based intestinal disease, like PWD, could have an impact on the growth performance of pigs and their intestinal microbiota and if it could provide a model to test the response to nutritional strategies under conditions of an immune and intestinal stimulation for animals susceptible to ETEC type. In this study, we demonstrated how a vaccinal strain of F4/F18 E. coli can affect the gut microbial composition of piglets, regardless of their genetic susceptibility to ETEC infection. In the third study we evidenced how a nucleotide supplementation can favor the proliferation of jejunal Peyer patches and anticipate the maturation of the fecal microbiota. In the fourth study we reported how xylanase can favor the proliferation of Lactobacillus reuteri. Finally, we showed some first results on the muscles fiber development in fast- and slow-growing suckling pigs and the relationship with the intestinal microbiota. Taken together, the results presented in this thesis provide new insight about the interplay between the host-genetics, gut microbial composition, and host physiological status. Furthermore, it provides confirmation that the use of known genetic markers for ETEC F4 and F18 could represent a potential tool to stratify the animals in the trials both in healthy or challenge-based protocols.

Acrylamide (AA) is an undesirable food toxic compound, classified as 'probably carcinogenic to humans' by the International Agency for Research on Cancer due to its toxic effects, including neurotoxicity, genotoxicity, carcinogenicity and reproductive toxicity. AA is mainly formed during the heat treatment of foods (> 120 °C) by the Maillard reaction, an essential reaction that also allows the desired levels of shelf-life and sensory properties of various food products to be achieved. Over the years, authorities and regulations have become more restrictive regarding the maximum levels of AA permitted in foods and beverages. The latest Commission Regulation (EU) 2017/2158 contains reference levels and measures to reduce AA in several food groups that contribute to the highest dietary intake, making necessary the study of promising AA mitigation strategies. The aim of this PhD research project was to identify, characterise and optimise some AA mitigation strategies in the most at-risk widely consumed foods such as potato, coffee and bakery products. Some AA control strategies were selected and investigated for each food category, also considering the main quality characteristics of the final products. The comprehensive results obtained during the three years of research activity have allowed a deeper knowledge of the traditional and innovative AA mitigation strategies, which can be extremely useful for both the food industry and international authorities. The most promising strategies studied in terms of reduction of AA while maintaining the main quality characteristics of the examined foods were: the application of pulsed electric fields and yeast immersion as pre-treatments of chips for frying; the selection of high roasting degrees for coffee products; the selection of static baking conditions for biscuits; the optimisation of alternative biscuit’ formulations by both the use of chickpea legume flour and of flour from bean with intact cotyledon cell walls.

Pulsed electric field technology is one of the most attractive new non-thermal technology thanks to its lower energy consumption and short treatment times. It consists of an electric treatment of short duration (from several ns to several ms) with electric field strengths from 0.1 to 80 kV/cm that lead to an increase in the permeability of the cell membrane. In this PhD thesis, PEF technology was investigated with the aim of improving mass transfer in plant and animal foods by using it alone or in combination with conventional food processes. Different methods of evaluating electroporation for optimizing PEF processing parameters were investigated. In this respect, the degree of membrane permeabilization in plant and animal food matrices was investigated using electrical impedance spectroscopy, current-voltage measurements and magnetic resonance imaging. The research findings provided useful insights and calls for critical choice of electroporation assessment methods for the selection of adequate PEF treatment conditions. It was outlined that the effect of electroporation is highly dependent on the properties of the food matrix and secondary phenomena occurring in the cell structure undergoing PEF treatment, such as the water re-distribution in the tissue due to the exchange of fluids between intra- and extra-cellular environments. This study also confirmed the great potential of combining PEF technology with conventional food processes, with the main purpose of improving the quality of the food material and accelerating the kinetics of mass transfers, in both plant and animal tissues. Consistent reduction of acrylamide formation in potato crisps was achieved by monitoring key PEF process parameters and subsequent manufacturing steps. Kiwifruit snacks showed a significant reduction in drying kinetics when pre-treated with PEF, while their quality was well maintained. Finally, the research results showed that PEF pre-treatments can shorten the brine process as well as the rehydration kinetics of fish muscles.

The purpose of this thesis work was the valorization of the main by-products obtained from olive oil production chain (wastewater and pomace) and their utilization in innovative food formulation. In the first part of the thesis, an olive mill wastewater extract rich in phenols were used in the formulation of 3 innovative meat products: beef hamburgers, cooked ham and würstels. These studies confirms that olive mill wastewaters extract rich in phenols could be an alternative for the reduction/total replacement of additives (i.e., nitrites) in ground and cooked meat preparations, which would promote the formulation of healthier clean label products and improve the sustainability of the olive oil industry with a circular economy approach, by further valorizing this olive by-product. In the second part of the thesis, the lipid composition and oxidative stability of a spreadable product obtained from a fermented and biologically de-bittered olive pomace, was assessed during a shelf-life study. This study confirmed that olive pomace represents an excellent ingredient for the formulation of functional foods In the third and last part of the thesis, carried out at the Universidad de Navarra (Pamplona, Spain), during a period abroad (3 months), three extracts obtained from purification of olive mill wastewaters, were subjected to in-vitro digestion and characterized. From the analysis of the three phenolic extracts, it emerged that the most promising extract to be used in the food field is the spry-dried one. Thanks to its formulation containing maltodextrins it manages to maintain its antioxidant capacity even after being underwent to in-vitro digestion. Besides their own adaptation strategies, plants might exploit microbial symbionts for overcoming both biotic and abiotic stresses and increase fitness. The current scenario of rapid climate change is demanding more sustainable agricultural management practices. The application of microbe-based products as a valid alternative to synthetic pesticides and fertilizers and their use to overcome stresses exacerbated by climate change, have been reviewed in the first part of this thesis. Berry fruits are widely cultivated and appreciated for their aromatic and nutraceutical properties. This thesis is focused on the role of plant and fruit microbiome on strawberry and raspberry growth, resistance, fruit quality and aroma. A taxonomical and functional description of the microbiome of different organs of three strawberry genotypes was performed both by traditional cultural dependent method and Next Generation Sequencing technique, highlighting a significant role of plant organs and genotype in determining the composition of microbial communities. Additionally, a selection of bacteria native of strawberry plants were isolated and screened for their plant growth promoting abilities and tested under the biotic stress of Xanthomonas fragariae infection and the abiotic stress of induced salinity. The monitoring of biometric parameters allowed the selection of a more restricted panel of bacterial strains, whose beneficial potential was tested in coordinated inoculations, or singularly. Raspberry plant was used for investigating the effect of cultivation method in determining fruit microbiome, and its consequent influence of berry quality and aroma. Interestingly, the cultivation method strongly influenced fruit nutraceutical traits, aroma and epiphytic bacterial biocoenosis. The involvement of the bacterial microbiota in fruit aroma determination was evaluated by performing GC–MS analysis of VOCs occurring in control, sterile and artificially reinoculated berries and by characterizing control and reinoculated berry microbiome. Differently treated berries showed significantly different aromatic profile, confirming the role of bacteria in fruit aroma development.

Due to the accelerating processes of soil salinization and shortage of fresh water, the practice of saline agriculture is gaining momentum in many areas of the world. However, there are some concerns that using saline water for irrigation may be non-environmentally sustainable, with potential to cause irreversible soil degradation. In addition, there is a lack of information on the morphological, physiological, and biochemical changes that can occur in plants when irrigated with saline water. In light of the above, the major aim of this work was to investigate the effects of a range of water salinity levels and irrigation regimes on the performances of salt tolerant species promising as future crop plants for saline agriculture. The following objectives were addressed: To determine the effects of different water regimes (leaching irrigation vs. no leaching irrigation) with water at increasing salinity concentrations on the growth, ion accumulation and water relations of Sorghum bicolor plants grown under saline soil conditions. To describe the germination response of Salicornia europaea seeds across a wide range of water salinity levels through six reliable indices for screening salinity tolerance at the seed germination stage. To explore the different physiological responses of six wild halophytes commonly found in the Mediterranean area (Artemisia absinthium, Artemisia vulgaris, Atriplex halimus, Chenopodium album, Salsola komarovii, and Sanguisorba minor), and rank their tolerance after exposure to growing levels of water salinity. To identify the main adaptation mechanisms that distinguish C3 from C4 halophytes when exposed to increasing salinity in the growth media, through a comparative study between the C3 species Atriplex hortensis and the C4 species Atriplex halimus. To identify the main adaptation mechanisms that distinguish annual from perennial halophytes when exposed to severe conditions of salinity and drought, through a comparative analysis between two annual Salicornia spp. and the perennial Sarcocornia fruticosa.

Pathogenic fungi are responsible for vine diseases affecting the grapevine yield and the organoleptic quality of the final wine products. Using of biocontrol agents can represent a sustainable alternative to the use of synthetic fungicides whose intense use can have negative effects on the ecosystem and cause increase resistant pathogen population to synthetic agents. The principal aim of my PhD thesis was the isolation and characterization of new yeast strains and Bacillus subtilis SV108 as biocontrol agent and the comprehension of the mechanism of their antimicrobial action. Accordingly, twenty wild yeast and one selected bacterium isolated among 62 samples, isolated from different Italian and Malaysian regions and molecularly identified, were evaluated in a preliminary screening test on agar. Results showed the highest effects on inhibiting mycelial growth by Starmerella bacillaris FE08.05, Metschnikowia pulcherrima GP8 and Hanseniaspora uvarum GM19. On the other side, Bacillus subtilis SV108 showed the ability of inhibit the mycelial growth of selected fungi by producing antimicrobial compounds on Malt Extract Broth medium recovered by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and identified by electrospray ionization (ESI) tandem mass spectrometer Triple TOF 5600. Moreover, in order to analyze the volatile fraction of compounds, the quantitative analysis of the VOCs profiles was performed by GC/MS/SPME. The analysis highlighted the presence of isoamyl and phenylethyl alcohols and an overall higher presence of low-chain fatty acids and volatile ethyl esters. All the data collected suggest that the tested yeasts, found among the epiphytic microbiota associated with grape berries, can be potentially effective for the biological control of pathogenic moulds. On the other hand, the proteomic study conducted on B. subtilis SV108 revealed that there are two cyclic antifungal peptides which can explain the antimicrobial effect of Bacillus subtilis SV108 acting as biocontrol agent against fungal pathogens in grapevine.
