Propagation by hardwood cuttings is an important technique for producing plant of many species. The technique is usually easy and inexpensive and makes it possible to high plants. But it is not commonly used for peach propagation in Brazil. The objective of this study was to evaluate the effects of different substrates, container types and cutting diameters on rooting of peach hardwood cuttings. The cuttings (two size groups, diameters of 2-6 mm and 6-10 mm) were inserted in six substrates (carbonized rice husk, vermiculite, washed sand, carbonized rice husk + vermiculite, carbonized rice husk + washed sand and vermiculite + washed sand, each 1:1 v/v), each in three types of containers (plastic bag, plastic tray and polystyrene tray). The best rooting occurred with smaller cuttings (47%) and larger cuttings (38%) in plastic bag with vermiculite, and with smaller cuttings in plastic bag with washed sand + vermiculite (33%). While these rooting percentages were unsatisfactory for commercial propagation, the results are encouraging and suggest need for more investigation to improve rooting.

The objective of this paper is to show a methodology to estimate transmission line parameters. The method is applied in a single-phase transmission line using the method of least squares. In this method the longitudinal and transversal parameters of the line are obtained as a function of a set of measurements of currents and voltages (as well as their derivatives with respect to time) at the terminals of the line during the occurrence of a short-circuit phase-ground near the load. The method is based on the assumption that a transmission line can be represented by a single circuit π. The results show that the precision of the method depends on the length of the line, where it has a better performance for short lines and medium length. © 2012 IEEE.

Understanding the biological activity profile of the snake venom components is fundamental for improving the treatment of snakebite envenomings and may also contribute for the development of new potential therapeutic agents. In this work, we tested the effects of BthTX-I, a Lys49 PLA2 homologue from the Bothrops jararacussu snake venom. While this toxin induces conspicuous myonecrosis by a catalytically independent mechanism, a series of in vitro studies support the hypothesis that BthTX-I might also exert a neuromuscular blocking activity due to its ability to alter the integrity of muscle cell membranes. To gain insight into the mechanisms of this inhibitory neuromuscular effect, for the first time, the influence of BthTX-I on nerve-evoked ACh release was directly quantified by radiochemical and real-time video-microscopy methods. Our results show that the neuromuscular blockade produced by in vitro exposure to BthTX-I (1 μM) results from the summation of both pre- and postsynaptic effects. Modifications affecting the presynaptic apparatus were revealed by the significant reduction of nerve-evoked [3H]-ACh release; real-time measurements of transmitter exocytosis using the FM4-64 fluorescent dye fully supported radiochemical data. The postsynaptic effect of BthTX-I was characterized by typical histological alterations in the architecture of skeletal muscle fibers, increase in the outflow of the intracellular lactate dehydrogenase enzyme and progressive depolarization of the muscle resting membrane potential. In conclusion, these findings suggest that the neuromuscular blockade produced by BthTX-I results from transient depolarization of skeletal muscle fibers, consequent to its general membrane-destabilizing effect, and subsequent decrease of evoked ACh release from motor nerve terminals. © 2012 Elsevier Ltd.

Photoacoustics as a tool for the diagnosis of radicular stress: Measurements in eucalyptus seedlings

In reforesting companies (cellulose industry), eucalyptus is usually cultivated in small plastic containers (50 mL). As seedlings remain for about 120 days in these containers-until transplantation-their roots become space restricted, with consequent limitations in water and nutrient absorption. These restrictions may lead to plant stress, decreasing productivity. In this work, we used the photoacoustic technique to evaluate the photosynthetic activity of Eucalyptus grandis, E. urophylla and E. urograndis seedlings subjected to this limited space availability, seeking a correlation with morphological parameters and fluorescence measurements in these seedlings. Photoacoustic, fluorescence, and morphological analysis were conducted every 15 days, from 45 to 120 days after sowing. Fluorescence and photosynthetic rate were evaluated in vivo and in situ, the latter one using the open photoacoustic technique. Data show that root dry matter diminished markedly at 90 and 120 days after sowing; this behavior showed a high correlation with the gas exchange component of the photoacoustic signal, as well as with the fluorescence ratio Fv/Fm. These results indicate that the soil volume of the container becomes insufficient for the roots after 90 days, probably leading to a nutritional deficiency in plants, which explains the decrease observed in the photosynthetic rate of seedlings. A resistência de 30 genótipos de milho ao ataque de Sitophilus zeamais Mots. foi avaliada em teste sem chance de escolha, sendo efetuada uma seleção de 10 deles (M 9560, XHT 12, A 952, AG 5011, X 9557, Z 8452, C 615, C 435, DINA 888, C 606), os quais foram submetidos ao teste com chance de escolha, avaliando-se a atratividade e a preferência para oviposição. Pelos resultados concluiu-se que os genótipos menos atrativos foram Z 8452 (3,1%) e M 9560 (3,2%), enquanto o mais atrativo foi C 615 (24,7%). Os genótipos que apresentaram resistência do tipo não-preferência para oviposição foram A 952, M 9560 e XHT 12 com números médios de 2,0; 3,0 e 3,4 ovos por recipiente. A resistência do tipo não-preferência para alimentação e/ou antibiose foi observada nos genótipos M 9560, A 952 e Z 8452. Os genótipos com alta suscetibilidade ao gorgulho foram C 435, C 444, C 606 e AG 8010.

The citrus mealybug, Planococcus citri is an important pest attacking several crops. Biotic and abiotic factors, as well as the substrate they feed on influence its population. The present study was aimed to study the effect of some feeding substrates on mealybug development in laboratory conditions. Leaf disks of guava, fig and cherimoya were placed individually on an agar film inside a Petri dish. One egg of the insect was confined inside each container, replicated 60 times for each treatment. Containers were kept in a climatized chamber at 25[degree] C, 70 [plus or minus] 10% RH and 12 hours photophase. Insects reared on guava leaves showed a longer nymphal development, while the female longevity was longer on fig leaves. However, no statistical differences were found in the survival index, which showed values higher than 83%, suggesting that all substrates studied here were adequate for the development of P. citri.

Acacia polyphyla (Mimosaceae) é uma espécie arbórea nativa do Brasil, importante para a recuperação de áreas degradadas. Sementes dessa espécie foram armazenadas em condições naturais e artificiais, com os objetivos de avaliar a longevidade das sementes no solo, conservar a sua qualidade fisiológica pelo período correspondente a duas colheitas e verificar o requerimento fotoblástico das sementes armazenadas. em condições naturais, as sementes foram enterradas em clareira, sob dossel ralo e sob dossel denso. As sementes deterioraram rapidamente, revelando-se incapazes de compor o banco de sementes do solo. em condições artificiais, as sementes foram acondicionadas em diferentes embalagens e armazenadas por dois anos em ambiente não controlado e em câmara fria. Periodicamente, as sementes foram colocadas para germinar a 25ºC, na ausência de luz e sob fotoperíodo de oito horas sob luzes branca e de sombreamento. Durante todo o período de armazenamento, a germinação no escuro foi inferior à constatada sob luzes branca e de sombreamento. A qualidade fisiológica foi conservada por dois anos, quando as sementes foram acondicionadas em embalagem impermeável e armazenadas em câmara fria. O comportamento germinativo das sementes armazenadas por dois anos foi comparado com o de sementes recém-colhidas, em temperaturas constante e alternada, não sendo constatado efeito da idade e do regime de temperatura no requerimento fotoblástico das sementes.

'Paluma' guavas, after internal quality evaluation using magnetic resonance tomography, were used to produce fresh-cut product. Fruits were peeled or not, cut in halves and seed removed, and they were packaged in polystyrene trays covered with PVC film or in a PET container with a lid. These packages were stored for 12 days at 5 degrees C, 10 degrees C and ambient temperature (22.6 degrees C). Tomography evaluation verified that impacts produced internal bruising with loss of cellular integrity and liquefication of the placenta tissues. Compression was more evident on the pericarp and cutting promoted superficial deformation. Storage temperature affected the weight loss, with fruit packaged in the polystyrene tray having a greater weight loss. The peeling did not influence weight loss. Product stored at 5 degrees C and 10 degrees C for 8 days had low microbial growth (< 10(3) UFC.g(-1)) and no coliforms. Rapid spoilage and a short shelf life (3-4 days) occurred when the product was stored at ambient temperature. Peeling reduced ascorbic acid concentration and total soluble solids. Use of calcium to protect fresh-cut products was not efficient. Calcium absorption capacity of 'Pedro Sato' guava was tested using Ca-45. Fruits treated with 2% CaCl2, with or without the radioisotope, were divided in four layers (epicarp, mesocarp, endocarp and seed) and analyzed for the total and Ca-45 calcium. It was observed that the applied calcium remained in superficial layers of fruits, which was confirmed by autoradiography. Internal layers did not contain Ca-45, indicating that calcium was not distributed into different parts of the fruit.
