The objective of this study was to evaluate the methodology to establish the hemolytic activity of alternative complement pathway as an indicator of the innate immunity in Brazilian fish pacu (Piaractus mesopotamicus), in addition to verifying the influence of β-glucan as an immunostimulant. Fish were fed with diets containing 0, 0.1 and 1% β-glucan, during seven days, and then inoculated with Aeromonas hydrophila. Seven days after the challenge, they were bled for serum extraction. The methodology consisted of a kinetic assay that allows calculating the required time for serum proteins of the complement to promote 50% lysis of a rabbit red blood cell suspension. The method developed in mammals was successfully applied for pacu and determined that the hemolytic activity of the proteins of the complement system (alternative pathway) increased after the pathogen challenge, but was not influenced by the β-glucan treatment.

Frequentemente, os indivíduos com perda auditiva têm dificuldade de entender a fala no ambiente ruidoso. OBJETIVO: O objetivo deste estudo foi avaliar clinicamente o desempenho dos indivíduos adultos com deficiência auditiva neurossensorial, com relação à percepção da fala, utilizando o aparelho de amplificação sonora individual digital com o algoritmo de redução de ruído denominado Speech Sensitive Processing, ativado e desativado na presença de um ruído. MATERIAL E MÉTODO: Este estudo de casos foi realizado em 32 indivíduos com deficiência auditiva neurossensorial de graus leve, moderado ou leve a moderado. Foi realizada a avaliação por meio de um teste de percepção de fala, onde se pesquisou o reconhecimento de sentenças na presença de um ruído, para obter a relação sinal/ruído, utilizando o aparelho auditivo digital. RESULTADOS: O algoritmo pôde proporcionar benefício para a maioria dos indivíduos deficientes auditivos, na pesquisa da relação sinal/ruído e os resultados apontaram diferença estatisticamente significante na condição em que o algoritmo encontrava-se ativado, comparado quando o algoritmo não se encontrava ativado. CONCLUSÃO: O uso do algoritmo de redução de ruído deve ser pensado como alternativa clínica, pois observamos a eficácia desse sistema na redução do ruído, melhorando a percepção da fala.

The capture web of N. clavipes presents viscous droplets, which play important roles in web mechanics and prey capture. By using scanning and transmission electron microscopy, it was demonstrated that the web droplets are constituted of different chemical environments, provided by the existence both of an aqueous and a lipid layer, which, in turn, present a suspension of tenths of vesicles containing polypeptides and/or tipids. GC/EI-MS Analysis of the contents of these vesicles led to the identification of some saturated fatty acids, such as decanoic acid, undecanoic acid, dodecanoic acid, tetradecanoic acid, octadecanoic acid, and icosanoic acid, while other components were unsaturated fatty acids, such as (Z)-tetradec-9-enoic acid, (Z)-octadec-9-enoic acid, and (Z)-icosa-11-enoic acid; and polyunsaturated fatty acids like (9Z,12Z)-octadeca-9,12-dienoic acid, (9Z,12Z,15Z)-octadeca-9,12,15-trienoic acid, and (11Z,14Z)-icosa-11,14-dienoic acid. Toxic proteins such as calcium-activated proteinase and metalloproteinase jararhagin-like precursor were also identified by using a proteomic approach, indicating the possible involvement of these enzymes in the pre-digestion of spiders' preys web-captured. Apparently, the mixture of fatty acids are relatively toxic to insects by topical application (LD50 64.3 +/- 7.6 ng mg(-1) honeybee), while the proteins alone present no topical effect; however, when injected into the prey-insects, these proteins presented a moderate toxicity (LD50 40.3 +/- 4.8 ng mg(-1) honeybee); the mixture of fatty acids and proteins is very toxic to the preys captured by the web droplets of the viscid spiral of Nephila clavipes when topically applied on them (LD50 14.3 +/- 1.8ng mg(-1) honeybee).

The protein complement of the secretion from hypopharyngeal gland of nurse-bees (Apis mellifera L.) was partially identified by using a combination of 2D-PAGE, peptide sequencing by MALDI-PSD/MS and a protein engine identification tool applied to the honeybee genome. The proteins identified were compared to those proteins already identified in the proteome complement of the royal jelly of the honey bees. The 2D gel electrophoresis demonstrated this protein complement is constituted of 61 different polypepides, from which 34 were identified as follows: 27 proteins belonged to MRJPs family, 5 proteins were related to the metabolism of carbohydrates and to the oxido-reduction metabolism of energetic Substrates, I protein was related to the accumulation of iron in honeybee bodies and I protein may be a regulator of MRJP-1 oligomerization. The proteins directly involved with the carbohydrates and energetic metabolisms were: alpha glucosidase, glucose oxidase and alpha amylase, whose are members of the same family of enzymes, catalyzing the hydrolysis of the glucosidic linkages of starch; alcohol dehydrogenase and aldehyde dehydrogenase, whose are constituents of the energetic metabolism. The results of the present manuscript support the hypothesis that the most of these proteins are produced in the hypoharyngeal gland of nurse-bees and secreted into the RJ. (C) 2004 Elsevier Ltd. Considerando a crescente utilização de técnicas de processamento digital de sinais em aplicações de sistemas eletrônicos e ou de potência, este artigo discute o uso da Transformada Discreta de Fourier Recursiva (TDFR) para identificação do ângulo de fase, da freqüência e da amplitude das tensões fundamentais da rede, independente de distorções na forma de onda ou de transitórios na amplitude. Será discutido que, se a freqüência fundamental das tensões medidas coincide com a freqüência a qual a TDF foi projetada, um simples algoritmo TDFR é completamente capaz de fornecer as informações requeridas de fase, freqüência e amplitude. Dois algoritmos adicionais são propostos para garantir seu desempenho correto quando a freqüência difere do seu valor nominal: um deles para a correção do erro de fase do sinal de saída e outro para identificação da amplitude do componente fundamental. Além disto, destaca-se que através dos algoritmos propostos, independentemente do sinal de entrada, a identificação do componente fundamental pode ser realizada em, no máximo, 2 ciclos da rede. Uma análise dos resultados evidenciados pela TDFR foi desenvolvida através de simulações computacionais. Também serão apresentados resultados experimentais referentes ao sincronismo de um gerador síncrono com a rede elétrica, através dos sinais fornecidos pela TDFR.

No problema de minimização de troca de ferramentas procura-se por uma sequência para processar um conjunto de tarefas de modo que o número requerido de trocas de ferramentas seja o menor possível. Neste trabalho propõe-se um algoritmo para resolver este problema baseado em um ordenamento parcial das tarefas. Uma sequência ótima é obtida expandindo-se as sequências parciais enumeradas. Testes computacionais são apresentados.

The bioavailability of calcium from the kale (Brassica oleracea var. acephala) consumed as a complement of a basal rice and bean diet was studied. Three groups of diets, two controls and one experimental were fed to Wistar female rats. The protein source of the first control was casein and of the second, a mixture of bean and rice. To both groups, graded levels of CaCO3 were added. The experimental diet was similar to the second control, except that CaCO3 was substituted for kale supplying the same amount of calcio. At 35 days of experimental period, they were killed and the calcium and phosphorus were determined in the right femur. The results indicated that the calcium from the kale is better utilized than that of the CaCO3. There was no significant difference in the concentration of calcium in the soft tissues studied among three dietary groups. Considering the high concentration and availability of calcium from the kale its consumption as a complement of the basal rice and bean diet may be recommended as a cheap and good source of this mineral.

Aspects of spermatogenesis and nucleolar behavior were analyzed in Brachymetra albinerva, Cylindrostethus palmaris, Halobatopsis platensis, Limnogonus aduncus (Gerridae), Martarega sp (Notonectidae), Rhagovelia whitei, and Rhagovelia sp (Veliidae). The testicles are rounded (Veliidae), elongated (Gerridae) or spiral (Notonectidae) and have a transparent membrane covering them. The complement chromosome was 2n = 23 (22A + X0, L. aduncus and Rhagovelia sp), 25 (24A + X0, B. albinerva and H. platensis), 26 (22A + 2m + XY, Martarega sp), 29 (28A + X0, C. palmaris), or 39 (38A + X0, R. whitei) chromosomes, and the only species with a different sex chromosome system was Martarega sp, which showed an XY system and m-chromosomes. The meiotic behavior of all species was similar: holocentric chromosomes and heteropyknotic material at prophase, interstitial and/or terminal chiasmata, and first reductional division for the autosomes and the reverse for the sex chromosomes. The only difference observed was related to the very large size of Martarega sp cells in all stages of spermatogenesis. With regard to nucleolar behavior, the species did not show differences, except for Martarega sp with larger nucleoli than the other species. The only species in which it was clearly possible to identify the nucleolar organizer region was L. aduncus, in the region of a terminal autosome. It was also confirmed that the telomeric associations do not occur at random. In the other species, specific staining was very discrete, and the nucleolar organizer region location was not at all evident.
