Investigou-se a relação entre as características da água e a infestação de protozoários parasitos, Icthyophthirius multifiliis e Trichodina sp., em peixe espada, Xiphophorus helleri e em plati, Xiphophorus maculatus, coletados em uma piscicultura de peixes ornamentais no Estado de São Paulo, Brasil. Os peixes foram coletados mensalmente, durante um ano, dos viveiros e das caixas de estocagem. A prevalência da infestação nos peixes das caixas e dos viveiros foram, respectivamente, 34,2% e 22,5% para I. multifiliis e 13% e 54% para Trichodina sp. A elevada condutividade elétrica e o pH da água reduziram a infestação por I. multifiliis. A baixa concentração de oxigênio resultou em aumento na infestação por Trichodina sp. O uso do sal, para aumentar a condutividade elétrica da água, consistiu em um método de controle de I. multifiliis. A redução do oxigênio dissolvido e a adição de fertilizante orgânico favoreceram a reprodução de Trichodina sp.

The aim of this study was to evaluate the occurrence of potentially zoonotic intestinal protozoan infections in exotic and wildlife Brazilian birds. Fecal samples from 207 birds of 45 species were examined. Infections by Balantidium sp., Entamoeba sp., and Blastocystis sp. were observed in 17 individuals (8.2%) of Gnorimopsar chopi, Oryzoborus angolensis, Sporophila caerulescens, Ramphastos toco, Aratinga leucophtalmus, and Pavo cristatus.

In the poultry industry, the use of water with adequate physical, chemical and microbiological quality it is of fundamental importance. Since many birds have access to the same water source, quality problems will affect a great number of animals. The drinking water plays an important role in the transmission of some bacterial, viral and protozoan diseases that are among the most common poultry diseases. Important factors to prevent waterborne diseases in broiler production are the protection of supply sources, water disinfection and the quality control of microbiological, chemical and physical characteristics. Water is an essential nutrient for birds and therefore quality preservation is fundamental for good herd performance. The farmer may prevent many diseases in bird flocks by controlling the quality of the ingested water, will certainly result in decreased costs and increased profit, two essential aims of animal production nowadays.

Inquiries about toxoplasmosis are essential because its zoonotic potential and pathogenicity in pets and production animals. The dog can be involved in the mechanical transmission of the infection by Toxoplasma gondii and in spite of not being the definitive host, it has an epidemiological role in this disease. The high rate of dogs naturally infected by T gondii and the great possibility of adquiring this protozoan, justify the importance of attempting against for the occurrence of this illness. The aim of this article was to demonstrated through literature revision, some clinical aspects, epidemic and pathological of the toxoplasmosis in the canine species.

The present work studied protozoan parasites of cultivated fishes (N = 433) from two feefishing farm situated in Franca, São Paulo, Brazil, during a period of April 1997 through March 1999. Specimens of piauçú Leporinus macrocephalus Garavello & Britski, 1988 (Anostomidae), pacu Piaractus mesopotamicus Holmberg, 1887 (Characidae) carp Cyprinus carpio Linnaeus, 1758 (Cyprinidae), Tillapia rendalli Boulenger, 1896 (Cichlidae), nile-tilapia Oreochromis niloticus Linnaeus, 1758 (Cichlidae), matrinxã Brycon cephalus Günther, 1869 (Characidae) and tambacu hybrid (male of P. mesopotamicus x female of Colossoma macropomum Cuvier, 1818) were collected. The fishes were parasitized with protozoans Ichthyophthirius multifiliis Fouquet, 1876 (Protozoa), Trichodina sp. and Piscinoodinium pillulare (Schaperclaus, 1954), Lom, 1981 (Protozoa). In the cold season (autumn and/or winter) all species of fish were infected with I. multifiliis. Higher susceptibility to Trichodina sp. was observed in L. macrocephalus, C. carpio and P. mesopotamicus compared to tambacu, B. cephalus, T. rendalli and 0. niloticus. It was not observed significant difference (P > 0.05) in the seasonal variation of Trichodina sp. and P. pillulare infection of all species. A great number of P. pillulare without significant difference (P > 0.05) was reported to L. macrocephalus, P. mesopotamicus and tambacu.

Toxoplasma gondii is an intracellular obligate protozoan, which infects humans and warm-blooded animals. The aim of the present study was to clone the rop2, gra5 and gra7 genes from T. gondii RH strain and to produce recombinant proteins. The rop2, gra5 and gra7 gene fragments produced by polymerase chain reaction were cloned into the pET102/D-TOPO(R) vector which contains thioredoxin and polyhistidine tags at the C-and N-ends, respectively, and is expressed in Escherichia coli BL21(DE-3). The expression fusion proteins were found almost entirely in the insoluble form in the cell lysate. These recombinant proteins were purified with an Ni-NTA column. Concentrations of the recombinant antigens produced in the E. coli BL21-star ranged from 300 to 500 mu g/mL growth media, which was used to immunize rabbits. We observed an identity ranging from 96 to 97% when nucleotide sequences were compared to GenBank database sequences. Immunocharacterization of proteins was made by indirect immunofluorescence assay. These proteins will be used for serodiagnosis and vaccination.

A criptosporidiose humana é uma infecção causada pelo Cryptosporidium sp, um protozoário coccídio de patogenicidade emergente e responsável por severa e prostrante diarréia aquosa em humanos, principalmente em indivíduos imunodeprimidos. O diagnóstico, feito através da utilização de esfregaços de fezes submetidos a técnicas de concentração e coloração específica pela fucsina-carbólica tem oferecido bons resultados em nosso laboratório. Tendo em vista o longo tempo despendido para a observação dos esfregaços considerando-se a pequenez das formas e o contraste da coloração, realizamos modificações no procedimento técnico da coloração ácido-resistente que resultaram em sensível melhoria das preparações: a fucsina-carbólica passou a ser deixada sobre o esfregaço por período de 3 minutos (LENNETTE et al., 1985) e procedeu-se a substituição da solução de álcool-ácido sulfúrico a 5% (HENRIKSEN & POHLENZ, 1981) por solução de ácido clorídrico a 0,5% em álcool etílico 70%, por cerca de 2 minutos (contribuição original). Estas alterações promoveram melhor remoção do excesso de fucsina-carbólica, aumentando a eficiência da etapa de descoloração e consequentemente otimizando o contraste do processo de coloração. Nestas condições, as lâminas examinadas em microscópio óptico em aumentos de 250x e 1000x tiveram a visualização dos oocistos do protozoário facilitada, sendo os mesmos observados em contraste destacado com pigmentação intensa de cor rosa-avermelhada contra coloração de fundo azulada. Vale destacar que estas modificações oferecem vantagens de rápido processamento do material e facilidade de visualização do protozoário, diminuindo o tempo de microscopia, tornando a análise das lâminas mais rápida e menos cansativa, agilizando o diagnóstico.

Blastocrithidia culicis is a protozoan of the family Trypanosomatidae. It is a parasite of insects, but the presence of bacteriumlike endosymbionts in its cytoplasm led some investigators to study this protozoan. This trypanosomatid does not infect humans and although it is phylogenetically distant from Trypanosoma cruzi, it presents many morphological characteristics, which are similar. In previous studies our group showed the presence of a L27 ribosomal protein in T cruzi (named TcrL27) using a RT-PCR, which also resulted in the cloning, sequencing and expression of an unexpected ribosomal protein, L17, in Blastocrithidia culicis (BcL17). In this paper, Western blot analysis demonstrated that the anti-BcL17 antibody recognizes the presence of the same ribosomal protein either in Blastochritidia culicis and T. cruzi nuclear extracts. Besides, two similar bands (40 and 47 kDa) appeared also in T. cruzi isolated ribosomal proteins and B. culicis nuclear extract corroborating with the findings showed in the phylogenetic reconstruction. With respect to their localization within the ribosome, both the L17 and L27 ribosomal proteins appear to belong to the peptidyl-transferase site, and are therefore part of the key step in protein synthesis. Both ribosomal proteins bind spiramycin derivatives, being therefore compounds of the macrolides connection sites in the ribosome. These findings would open a possibility to better evaluate this issue.
