The effect of the concentration of sucrose solutions on the cellular structure of potato tissue in equilibrium at 27 degreesC was Studied. Two different methods of investigation were used to determine the volume of the different phases composing the cellular tissue of the potato when in equilibrium with the solutions. one based on data of the concentration itself and the overall volume of 2 mm slices after 48 h at equilibrium, and the other on microscopic images of cells in thin slices of fresh tissue stained with neutral red after an hour in equilibrium to show protoplasts, vacuoles and plasmolysis spaces. The results of these methods were compared with those obtained by a predictive thermodynamic approach considering the semipermeability of cell membranes. Phase volume data obtained from microscopic analysis were more similar to what was predicted by the theoretical model than those obtained by means of composition measurement. where the long equilibrium time apparently led to the loss of semi permeability of the cell membranes, since total volumes calculated without consideration of the cell membranes were similar to those measured. This suggests that the length of time of osmotic dehydration brings about a change in cell structure and the consequent involvement of a different mechanism in mass transfer. (C) 2002 Elsevier B.V. Mentha piperita L. is an aromatic and medicinal species of the family Lamiaceae, known as mint or peppermint, and its leaves and branches produce essential oil rich in menthol. This study aimed to evaluate physiological indexes, macro- and micronutrients inthe shootsand essential oil of Mentha piperita L. grown in nutrient solution number 2 of Hoagland and Arnon (1950) with different N, P, K and Mg levels. Shoot length, dry mass of the different organs, total dry mass, leaf area, essential oil yield and composition, and macronutrient (N, P, K, Mg, Ca, S) and micronutrient (Mn, Cu, Fe, Zn) contents in the shoot were evaluated. Plants treated with 65%N/50%P/25%K/100%Mg had a tendency towards longer shoot, greaterroot and leaf blade dry masses, higher essential oil yield, higher menthol levels and lower menthone levels. The results showed that Mentha can be grown in nutrient solution by reducing 65% N, 50% P, 25% K and 100% Mg. This solution had better development compared to the other tested treatments. Therefore,we recommendMentha piperita L. to be grown with such nutrient levels.

Aluminum (Al3+) toxicity is a major limiting factor to crop productivity in acid soils. The effects of aluminum on root and shoot growth of physic nut (Jatropha curcas L.) young plants and, the uptake and distribution of phosphorus, calcium, magnesium and aluminum in the roots and shoots were investigated in the present study. Plants were grown in 2.5L pots in a greenhouse. After fourteen days of adaptation to nutrient solution, plants were exposed to Al concentrations of 0, 370, 740, 1,100 and 1,480 mu mol L-1, corresponding to an active Al3+ solution of 13.3, 35.3, 90.0, 153.3 and 220.7 mu mol L-1, respectively. The dry matter partitioning between roots, stems and leaves, and the concentrations of P, Ca, Mg and Al in plant tissue, were measured after 75 days exposure to Al. The increasing level of Al3+ activity in solution progressively decreased the growth of the shoot and root of physic nut plants, and at the two highest active Al3+ levels, plants showed morphological abnormalities typical of the toxicity caused by this metal. Higher Al3+ activity reduced P concentrations in leaves and Ca and Mg in leaves and roots of physic nut, demonstrating the effect of Al on the uptake, transport and use of these nutrients by plants. The Al accumulated preferentially in the roots of physic nut, whereas only a small amount was transported to shoots.

The Vale do Ribeira, SP, main agricultural activity is the banana crop, which accounts for most of this fruit production in the State of São Paulo. The nutritional balance of the plant is one of the most important factors for the banana plant can complete the cycle and achieve high productivity. Aiming to evaluate the seasonal variation of leaf nutrient concentration in banana plants in Vale do Ribeira-SP, we used the results of 252 chemical analyses of plant tissue, collected from August 2009 to September 2010, in the 18 representative properties for the region, ten cultivated with subgroup Cavendish banana plant and eight of subgroup Prata banana plant. The largest variation between the macronutrient occurred for K and S, and among the micronutrients, especially for Fe and B. In some dates of evaluation, there was a higher leaf concentration of P, K, Ca and Zn, in subgroup Cavendish banana plants, while the subgroup Prata banana plants showed higher leaf concentration, especially of Mn, B and N. Climatic conditions, especially rain, influenced the leaf nutrient content, especially for K, N, S, B and Fe.

The cotton growth occupies about 1.1 million hectares in Brazil and the midwest, especially the state of Mato Grosso, is the largest producer in the country. This work aimed to establish the sufficiency ranges for macronutrients in cotton crops by the method of mathematical chance. The work was carried out from 2000 to 2003 in the cities of Costa Rica (MS), Diamantino and Sapezal (MT), totaling 152 samples of plant tissue and yield. The chance mathematics method was adequate for to establish cotton macronutrients references values, can be to help parameters were the conventional research cannot to prove in the small time space. The use of different yields for the calculation of mathematical chance (4,000 and 4,500kg ha(-1)) in the population studied, no difference between the sufficiency ranges for macronutrients. The macronutrients sufficiency range was, in the mg kg(-1), 33-45; 3.8-5.3; 15-21; 20-36; 4.0-7.2 e 2.8-8.6 for N, P, K, Ca, Mg e S, respectly. The indicated range, however to similar with the recommendations published, they demonstrate improvement about to obtainment of increases productivities.

Crop species with the C-4 photosynthetic pathway are more efficient in assimilating N than C-3 plants, which results in different N amounts prone to be washed from its straw by rain water. Such differences may affect N recycling in agricultural systems where these species are grown as cover crops. In this experiment, phytomass production and N leaching from the straw of grasses with different photosynthetic pathways were studied in response to N application. Pearl millet (Pennisetum glaucum) and congo grass (Brachiaria ruziziensis) with the C-4 photosynthetic pathway, and black oat (Arena Strigosa) and triticale (X Triticosecale), with the C-3 photosynthetic pathway, were grown for 47 days. After determining dry matter yields and N and C contents, a 30 mm rainfall was simulated over 8 t ha(-1) of dry matter of each plant residue and the leached amounts of ammonium and nitrate were determined. C-4 grasses responded to higher fertilizer rates, whereas N contents in plant tissue were lower. The amount of N leached from C-4 grass residues was lower, probably because the C/N ratio is higher and N is more tightly bound to organic compounds. When planning a crop rotation system it is important to take into account the difference in N release of different plant residues which may affect N nutrition of the subsequent crop.

A pesquisa da composição do meio de cultura mais adequado à espécie vegetal e ao tipo de explante empregado é o fator de maior relevância da cultura de tecidos. O cultivo de ápice caulinar com recuperação da planta matriz é uma técnica de grande impacto para a propagação de plantas in vitro, regeneração de plantas livres de vírus, conservação de germoplasma e modificação genética. Objetivou-se, neste trabalho, avaliar composições do meio de cultivo para organogênese direta in vitro a partir de ápices caulinares pertencentes à população FCA-UNESP-PB de mamoneira (Ricinus communis L.), com vistas à propagação clonal de genótipos elite. Foram testadas quatro formulações: MS básico (T1), MS modificado 1 (T2), MS modificado 2 (T3) e WPM (T4), em delineamento experimental inteiramente casualizado, com 20 repetições em cada tratamento, sendo a repetição 1 ápice caulinar/frasco. O T3 apresentou-se superior e diferiu significativamente dos outros tratamentos apresentando 35% dos ápices caulinares diferenciados e desenvolvidos; seguiu-se o T2 com 10% e os tratamentos T1 e T4 não apresentaram diferenciação de tecidos. Os resultados permitiram concluir que os balanceamentos de sais minerais nos meios de cultura avaliados, especialmente a relação NO3 / NH4 e ausência de FeSO4.7H2O, indicaram grande influência no desenvolvimento de ápices caulinares de mamoneira.

Entre as maiores aplicações da cultura de tecidos de plantas está a propagação massal de mudas de plantas ornamentais. O objetivo deste trabalho foi avaliar o cultivo in vitro de um híbrido de orquídea Brassocattleya em diferentes concentrações de NH4NO3, KNO3 e BAP. Foram utilizadas sementes do híbrido de orquídea Brassocattleya 'Pastoral' e as plantas foram cultivadas em meio MS com redução pela metade das fontes de P, Mg e Ca e adição de 25 g L-1 de sacarose, 100 mg L-1 de mio-inositol, 1,5 g L-1 de carvão ativo e 6,5 g L-1 de ágar-ágar, sendo o pH ajustado para 5,8. Como tratamentos foram usados quatro concentrações dos sais NH4NO3 e KNO3 (2x; 1x; ½ e ¼ do meio MS) e três concentrações de BAP (0,0; 0,5 e 1,0 mg L-1). Avaliou-se a multiplicação, o crescimento em altura, massa fresca e seca, além dos teores de açucares redutores na massa seca das mudas. Observou-se grande influência das doses de NH4NO3 e KNO3 sobre o crescimento em altura das mudas, massa fresca e seca e teores de açucares redutores em Bc. ('Pastoral' x Auto). A dose de ¼ da utilizada no meio MS promoveu aumento significativo do crescimento das plantas. Para multiplicação, houve melhor resultado com a dose de ½ dos sais NH4NO3 e KNO3 utilizados no meio MS e 1,0 mg L-1 de BAP.

Uma das aplicações das técnicas da cultura de tecidos no melhoramento é a identificação de linhas de células que apresentam tolerância à salinidade. Vários autores obtiveram linhas de células tolerantes ao estresse salino; e estudo de mecanismos bioquímicos da tolerância a sais em plantas tem demonstrado altas correlações entre estes e o acúmulo de macromoléculas em tecido de plantas superiores. Para verificar essas correlações em feijão (Phaseolus vulgaris cv IAC carioca), calos oriundos de eixos embrionários foram cultivados em meio sólido, suplementado com NaCl nas concentrações de 0 a 60 mM. Após 13 dias de incubação, os calos foram coletados e analisados quanto ao crescimento relativo, teor de proteínas, teor de prolina e atividade da peroxidase. Os parâmetros analisados mostraram decréscimo no crescimento relativo e no de proteínas em resposta ao NaCl. Paralelamente, observou-se aumento significativo no conteúdo de prolina e atividade da enzima peroxidase.

The evaluation of micronutrient availability in agricultural soil treated with sewage sludge was developed in Field with maize cultivation on a Dystrophic Red Latosol. The experiment used a randomized complete block design, with the treatments in a split-plot scheme, in time, with six doses of sewage sludge (main-plot treatment) in relation to the five sampling periods for the soil (subplot treatment), with four replications. The sewage sludge was applied in the superficial layer (0-20 cm). The micronutrients Fe, Cu, Mn and Zn were extracted from the soils using a DTPA (diethylenetriaminepentaacetic acid) solution. The same micronutrients were analyzed in the plant tissue after the nitric-perchloric decomposition of the maize samples by spectrometric atomic absorption. The results showed an increasing amount of Fe, Mn, Zn, Cu, Cation Exchange Capacity (CEC) and a decreasing pH in the soil. The increasing doses of sewage sludge increased the Fe, Mn, Cu and Zn availability in the soil solution, and the uptake amount by maize plants were always greater than the control values.

This work was carried out with Psychotria ipecacuanha, a Brazilian medicinal plant the roots of which contain emetine. The main objective was to develop a protocol for the micro-propagation of these species, by testing different culture techniques, the temporary immersion system, and the semi-solid and liquid media systems. In the semi-solid system, experiments were developed in flasks of two different sizes containing MS, B5, and WP media to which were added different growth regulators. Innoculum density was also evaluated. The liquid medium system consisted of MS medium supplemented with different growth regulators. For the temporary immersion system, the MS medium received an addition of 1.5mg/L BAP and 0.5mg/L GA3, and a reverse digital apparatus and vacuum pump were used. The liquid medium system with MS medium supplemented with 1.5mg/L BAP and 0.5mg/L GA3 presented the best results for shoot proliferation in a period of 30 days in culture (2.37 ± 0.32 shoots/explant). Cultures carried out for 90 days in the semi-solid system, using 8.5 × 5.5cm flasks and 3 explants per flask, developed 1.80 ± 0.20 shoots/explant, achieving 3.06 ± 0.51 cm of height adn presented superior survival ratio (96%). Explants cultured in temporary immersion system for 90 days showed 2.30 ± 1.10 shoots/explant achieving a growth of 2.08 ± 0.12 cm and 52% survival.

Extraction of active principles from medicinal plants generates wastes usually discharged. This process does not extract minerals from plant tissues, which may be considered of great value in organic composting. Wastes of 81 medicinal plant species processed at Centroflora/Anidro industry were analyzed for macro and micronutrients, and according to the results, three different ways to elaborate the compost were proposed: a) wastes of medicinal plants with high content of a specific mineral, aiming to fertilize soils where would be cultivated species that need high quantities of that mineral; b) wastes with mineral contents in order to support specific failures on mineral content of a soil; c) wastes of the eleven richest species in each nutrient that should elevate all mineral contents in soil.

Pyrophosphate-dependent phosphofructokinase (PPi-PFK) has been detected in several types of plant cells, but the gene has not been reported in sugar cane. Using Citrux paradixi PPi-PFK gene (AF095520 and AF095521) sequences to search the sugar cane EST database, we have identified both the α and β subunits of this enzyme. The deduced amino acid sequences showed 76 and 80% similarity with the corresponding α and β subunits of C. paradisi. A high degree of similarity was also observed among the PFK β subunits when the alignment of the sugar cane sequences was compared to those of Ricinus communis and Solanum tuberosum, it appears that α and β are two distinct subunits; they were found at different concentrations in several sugar cane tissues. It remains to be determined if the different gene expression levels have some physiological importance and how they affect sucrose synthesis, export, and storage in vacuoles. A comparison between the amino acid sequences of β PFKs from a variety of organisms allowed us to identify the two critical Asp residues typical of this enzyme's activity site and the other binding sites; these residues are tightly conserved in all members of this protein family. Apparently, there are catalytic residues on the β subunit of the pyrophosphate-dependent enzyme.
