Visando a alcançar alta eficiência na indução de calos a partir de explantes foliares de plantas matrizes de C. arabica com alta heterozigose, por meio da embriogênese somática indireta, foram instalados três experimentos. O primeiro experimento foi conduzido em esquema fatorial 2x5, constituído de dois meios de cultura (BOXTEL & BERTHOULY, 1996 e TEIXEIRA et al., 2004) e cinco genótipos de C. arabica. No segundo experimento, foi avaliado o potencial de produção de calos embriogênicos em 10 genótipos, sendo cada genótipo considerado como um tratamento e, no terceiro experimento, foram avaliadas as variações nas concentrações de 2.4-D (2,5 e 20µM) e 2-iP (2,5 e 20µM) nos meios primário e secundário de TEIXEIRA et al. (2004). As culturas foram mantidas a 25oC, sob obscuridade. Para os genótipos 2.2 e 7.2, verificou-se a superioridade do meio de cultura Teixeira et al. (2004) em relação ao meio BOXTEL & BERTHOULY (1996). No genótipo 4.2, observou-se o comportamento inverso, ou seja, a superioridade do meio BOXTEL & BERTHOULY (1996). Os genótipos 3.0 e 5.0 apresentaram o mesmo comportamento em ambos os meios de cultura estudados, evidenciando que a produção de embriões somáticos é fortemente dependente do genótipo. A indução de calos depende da relação de 2-iP e 2.4-D. A combinação de 20.0µM of 2.4-D e 20.0µM of 2-iP promoveu a maior porcentagem de indução de calos embriogênicos.

In this work we study the photosynthetic induction in Eucalyptus urograndis leaves using the Open Photoacoustic Cell Technique. In vivo and in situ measurements were performed in leaves of four months-old E. urograndis seedlings and C041 cuttings previously dark-adapted for at least 10 h. Experimental results for the gas exchange component of the photoacoustic (PA) signal are interpreted considering that a gas uptake component would have a phase angle nearly opposite to that of the oxygen evolution component. Analysis of the photosynthetic induction data shows that seedlings present a net oxygen evolution before cuttings, but cuttings reach a higher steady-state photosynthetic activity.

Photosynthetic induction in leaves of four-month-old Eucalyptus urograndis seedlings and of cuttings obtained from adult trees that were previously dark-adapted was studied by the in vivo and in situ Open Photoacoustic Cell Technique, Results for the gas exchange component of the photoacoustic (PA) signal were interpreted considering that the gas uptake component would have a phase angle nearly opposite to that of the oxygen evolution component. By subtracting the thermal component from the total PA signal, we studied the competition between gas uptake and oxygen evolution during the photosynthetic induction. Seedlings presented a net oxygen evolution prior to cuttings, but cuttings reached a higher steady-state photosynthetic activity. The chlorophyll (Chl) a/b ratio and the Chl fluorescence induction characteristic F-v/F-m were significantly higher for cuttings, while there was no difference between samples in stomata density and leaf thickness. Thus the differences in PA signals of seedlings and cuttings are associated to differences between the photosystem 2 antenna systems of these samples.

We studied the effects of Amphotericin B (AmB) on Cryptococcus neoformans using different viability methods (CFUs enumeration, XTT assay and propidium iodide permeability). After 1 h of incubation, there were no viable colonies when the cells were exposed to AmB concentrations >= 1 mg/L. In the same conditions, the cells did not become permeable to propidium iodide, a phenomenon that was not observed until 3 h of incubation. When viability was measured in parallel using XTT assay, a result consistent with the CFUs was obtained, although we also observed a paradoxical effect in which at high AmB concentrations, a higher XTT reduction was measured than at intermediate AmB concentrations. This paradoxical effect was not observed after 3 h of incubation with AmB, and lack of XTT reduction was observed at AmB concentrations higher than 1 mg/L. When stained with dihydrofluorescein, AmB induced a strong intracellular oxidative burst. Consistent with oxidative damage, AmB induced protein carbonylation. Our results indicate that in C. neoformans, Amphotericin B causes intracellular damage mediated through the production of free radicals before damage on the cell membrane, measured by propidium iodide uptake. Isolated mitochondria may undergo uncoupling, and in presence of Ca2+ at different conditions, a mitochondrial permeability transition (MPT) linked to protein,thiol oxidation, and demonstrated by CsA-sensitive mitochondrial swelling; these processes may cause cell death either by necrosis or by apoptosis. Isocoumarins isolated from the Brazilian plant Paepalanthus bromelioides (Eriocaulaceae) paepalantine (9,10-dihydroxy-5,7-dimethoxy-1H-naptho(2,3c)pyran-1-one), 8,8'-paepalantine dimer, and vioxanthin were assayed at 1-50 mu M on isolated rat liver mitochondria, for respiration, MPT, protein thiol oxidation, and interaction with the mitochondrial membrane using 1,6-diphenyl-1,3,5-hexatriene (DPH). The isocoumarins did not significantly affect state 3 respiration of succinate-energized mitochondria; they did however, stimulate 4 respiration, indicating mitochondrial uncoupling. Induction of MPT and protein thiol oxidation were assessed in succinate-energized mitochondria exposed to 10 mu M Ca2+; inhibition of these processes was assessed in non-energized organelles in the presence of 300 mu M t-butyl hydroperoxide plus 500 mu M Ca2+. Only paepalantine was an effective MPT/protein thiol oxidation inducer, also releasing cytochrome c from mitochondria; the protein thiol oxidation, unlike mitochondrial swelling, was neither inhibited by CsA nor dependent on the presence of Ca2+. Vioxanthin was an effective inhibitor of MPT/protein thiol oxidation. All isocoumarins inserted deeply into the mitochondrial membrane, but only paepalantine dimer and vioxantin decreased the membrane's fluidity. A direct reaction with mitochondrial membrane protein thiols, involving an oxidation of these groups, is proposed to account for MPT induction by paepalantine, while a restriction of oxidation of these same thiol groups imposed by the decrease of membrane fluidity, is proposed to account for MPT inhibition by vioxanthin. Chlorhexidine, even at low concentrations, is toxic for a variety of eukaryotic cells; however, its effects on host immune cells are not well known. We evaluated in vitro chlorhexidine-induced cytotoxicity and its effects on reactive oxygen/nitrogen intermediate induction by murine peritoneal macrophages. Thioglycollate-induced cells were obtained from Swiss mice by peritoneal lavage with 5 ml of 10 mM phosphate-buffered saline, washed twice and resuspended (10(6) cells/ml) in appropriate medium for each test. Cell preparations contained more than 95% macrophages. The cytotoxicity was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay and the presence of hydrogen peroxide (H2O2) and nitric oxide (NO) by the horseradish peroxidase-dependent oxidation of phenol red and Griess reaction, respectively. The midpoint cytotoxicity values for 1- and 24-h exposures were 61.12 ± 2.46 and 21.22 ± 2.44 µg/ml, respectively. Chlorhexidine did not induce synthesis or liberation of reactive oxygen/nitrogen intermediates. When macrophages were treated with various sub-toxic doses for 1 h (1, 5, 10, and 20 µg/ml) and 24 h (0.5, 1, and 5 µg/ml) and stimulated with 200 nM phorbol myristate acetate (PMA) solution, the H2O2 production was not altered; however, the NO production induced by 10 µg/ml lipopolysaccharide (LPS) solution varied from 14.47 ± 1.46 to 22.35 ± 1.94 µmol/l and 13.50 ± 1.42 to 20.44 ± 1.40 µmol/l (N = 5). The results showed that chlorhexidine has no immunostimulating activity and sub-toxic concentrations did not affect the response of macrophages to the soluble stimulus PMA but can interfere with the receptor-dependent stimulus LPS.

Induction motors are largely used in several industry sectors. The selection of an induction motor has still been inaccurate because in most of the cases the load behavior in its shaft is completely unknown. The proposal of this article is to use artificial neural networks for torque estimation with the purpose of best selecting the induction motors rather than conventional methods, which use classical identification techniques and mechanical load modeling. Since proposed approach estimates the torque behavior from the transient to the steady state, one of its main contributions is the potential to also be implemented in control schemes for real-time applications. Simulation results are also presented to validate the proposed approach.

In this paper, a factor referred to as k(f) for linear induction motor end effect analysis is presented. The mathematical model takes into account the longitudinal entry end effect. The entry end effect produces considerable distortion in magnetic field distribution. It is shown how this influence is derived from the machine-developed force that is calculated through the application of the I-D theory. The k(f) factor establishes the relationship between the longitudinal end effect and machine parameters, mainly the number of magnetic poles, secondary resistivity, and frequency.

Condition monitoring is used to increase machinery availability and machinery performance, reducing consequential damage, increasing machine life, reducing spare parts inventories, and reducing breakdown maintenance. An efficient real time vibration measurement and analysis instruments is capable of providing warning and predicting faults at early stages. In this paper, a new methodology for the implementation of vibration measurement and analysis instruments in real time based on circuit architecture mapped from a MATLAB/Simulink model is presented. In this study, signal processing applications such as FIR filters and fast Fourier transform are treated as systems, which are implemented in hardware using a system generator toolbox, which translates a Simulink model in a hardware description language - HDL for FPGA implementations.

Many factors such as the sunlight, intensity of radiation, temperature, and moisture may influence the degradation process of geosynthetics. UV stabilizers are used especially in polyolefin geomembrane to prevent the degradation process. In these geomembranes the service lifetime is initially governed by the consumption of antioxidants. Tests like MFI and OIT are a alternative to detect the oxidative degradation in polyolefins. This article evaluates HDPE geomembrane degradation after UV exposure through the results of MFI and OIT tests. Two kinds of geomembranes were evaluated: a black and smooth (0.8, 1.0, 1.5, 2.5 mm) and a white and textured (1.0 mm). MFI test showed some levels of superficial degradation (crosslink) in HDPE geomembrane.
