This paper investigates, in light of the theory of Functional Discourse Grammar (HENGEVELD; MACKENZIE, 2008), the concessive clause that is not subordinated to other clauses, i.e., it has no semantic or syntactic relations with clauses before or after it, and which we call Independent Concessive. The aim is to describe the discursive properties and the semantic, morphosyntactic and prosodic structure of this type of clause, showing that its relevance is in the construction and organization of discourse. The results indicate that the Independent Concessive works in speech as a parenthesis which interrupts the discursive course and is then highlighted by a special prosodic contour, combined with the presence of Interactive Acts. It is therefore a Move, the highest layer of the Interpersonal Level, the level that relates to the pragmatic aspects of the grammar of a language. The universe of research used is the Iboruna corpus, a database that records a variety of Portuguese from the Northwest of São Paulo.

eIF5A is a highly conserved putative eukaryotic translation initiation factor that has been implicated in translation initiation, nucleocytoplasmic transport, mRNA decay, and cell proliferation, but with no precise function assigned so far. We have previously shown that high-copy PKCI suppresses the phenotype of tif51A-1, a temperature-sensitive mutant of eIF5A in S. cerevisiae. Here, in an attempt to further understand how Pkc1 functionally interacts with eIF-5A, it was determined that PKCI suppression of tif51A-1 is independent of the cell integrity MAP kinase cascade. Furthermore, two new suppressor genes, ZDS1 and GIC1, were identified. We demonstrated that ZDS1 and ZDS2 are necessary for PKC1, but not for GIC1 suppression. Moreover, high-copy GIC1 also suppresses the growth defect of a PKCI mutant (stt1), suggesting the existence of a Pkc1-Zds1-Gic1 pathway. Consistent with the function of Gic1 in actin organization, the tif51A-1 strain shows an actin polarity defect that is partially recovered by overexpression of Pkc1 and Zds1 as well as Gic1. Additionally, PCL1 and BNI1, important regulators of yeast cell polarity, also suppress tif51A-1 temperature sensitiviiy Taken together, these data strongly Support the correlated involvement of Pkc1 and eIF5A in establishing actin polarity, which is essential for bud formation and G1/S transition in S. cerevisiae.

Bladder carcinoma is one of the most common tumors in the world and, despite the therapy currently available, most of the patients relapse. Better understanding of the factors involved in disease pathogenesis would provide insights for the development of more effective strategies in treatment. Recently, differential miRNA expression profiles in bladder urothelial carcinomas identified miR-100 down-regulation and miR-708 up-regulation among the most common alterations, although the possible influence of these miRNAs in the control of basic mechanisms in bladder tumors has not been addressed. In this context, the present study aimed to evaluate the in vitro effects of miR-100 forced expression and miR-708 inhibition in the bladder carcinoma cell line 5637. Our results showed that overexpression of miR-100 significantly inhibited growth when compared to controls at both times tested (72 and 96 hours, p<0.01) with a maximum effect at 72 hours reducing proliferation in 29.6 %. Conversely, no effects on cell growth were observed after inhibition of miR-708. MiR-100 also reduced colony formation capacity of 5637 cells by 24.4%. No alterations in cell cycle progression or apoptosis induction were observed. The effects of miR-100 on growth and clonogenicity capacity in 5637 cells evince a possible role of this miRNA in bladder carcinoma pathogenesis. Further studies are necessary to corroborate our findings and examine the potential use of this microRNA in future therapeutic interventions.

Two fish species, one top predator (Imparfinis mirini) and one intermediate detritivorous species (Hisonotus depressicauda), were experimentally manipulated to evaluate their relative importance in structuring the periphytic community, as well as their effects on the other trophic levels. An enclosure experiment was conducted in the Potreirinho creek, a second order tributary of Paranapanema River, SE Brazil. Five treatments were used: enclosure of the predator species. enclosure of the detritivorous species, enclosure of both together, exclusion of all fish species (closed control cage), and cage open to all fish community, (open control). Through direct and indirect effects, I. mirini, when alone gave rise to a trophic cascade that resulted in a positive effect on algal resources. Through direct effects, H. depressicauda. when alone, reduced the amount of organic matter, resulting in a positive indirect effect on algae. In addition, when the two species were enclosed together, only the effects determined by the detritivorous species were present. The results indicate the important role of the intermediate detritivorous species in the maintenance of the composition and trophic structure of the analyzed community by reducing the effects caused by the top predator.

Acting in the hypothalamus, tumor necrosis factor-alpha (TNF-alpha) produces a potent anorexigenic effect. However, the molecular mechanisms involved in this phenomenon are poorly characterized. In this study, we investigate the capacity of TNF-alpha to activate signal transduction in the hypothalamus through elements of the pathways employed by the anorexigenic hormones insulin and leptin. High dose TNF-a promotes a reduction of 25% in 12 h food intake, which is an inhibitory effect that is marginally inferior to that produced by insulin and leptin. In addition, high dose TNF-a increases body temperature and respiratory quotient, effects not reproduced by insulin or leptin. TNF-alpha, predominantly at the high dose, is also capable of activating canonical pro-inflammatory signal transduction in the hypothalamus, inducing JNK, p38, and NF kappa B, which results in the transcription of early responsive genes and expression of proteins of the SOCS family. Also, TNF-a activates signal transduction through JAK-2 and STAT-3, but does not activate signal transduction. through early and intermediary elements of the insulin/leptin signaling pathways such as IRS-2, Akt, ERK and FOXO1. When co-injected with insulin or leptin, TNF-a, at both high and low doses, partially impairs signal transduction through IRS-2, Akt, ERK and FOXO1 but not through JAK-2 and STAT-3. This effect is accompanied by the partial inhibition of the anorexigenic effects of insulin and leptin, when the low, but not the high dose of TNF-alpha is employed. In conclusion, TNF-alpha, on a dose-dependent way, modulates insulin and leptin signaling and action in the hypothalamus. (c) Published by Elsevier B.V.

OBJETIVO: Investigar as habilidades pragmáticas de alunos com distúrbios do espectro autístico durante a interação com suas professoras em salas comuns de escolas públicas. MÉTODOS: Participaram 14 alunos com distúrbios do espectro autístico, de ambos os gêneros, com idade entre três e oito anos (média=5,9; DP=1,8). Como parte do procedimento, foram realizadas filmagens em situações de sala de aula e para análise dos aspectos pragmáticos da comunicação dos participantes, foi utilizado o Protocolo de Pragmática. Os resultados foram tratados estatisticamente (p<0,05 e em alguns casos p<0,01; foram utilizados os testes de Wilcoxon, Friedman, Kruskal-Wallis, Spearman). RESULTADOS: em média, o número de atos comunicativos expressos por minuto pelas professoras (3,4 atos/min) foi superior (p<0,05) ao apresentado pelos alunos com distúrbios do espectro autístico (2,7 atos/min). Os alunos demonstraram predomínio de utilização do meio gestual, com média de 36,2 ocorrências, e de funções comunicativas menos interativas, com média de 24,1 ocorrências. Além disso, ocorreu correlação positiva entre os atos/min de alunos e professoras (p=0,01). Observou-se, também, que a idade e a escolaridade dos alunos não apresentaram correlações com o estabelecimento da comunicação nas díades. CONCLUSÃO: Foi possível identificar os déficits e as habilidades dos alunos com distúrbios do espectro autístico em sala de aula comum e confirmar a influência exercida pelo professor no desempenho comunicativo de tais alunos. Estes dados podem contribuir para esclarecimentos de lacunas existentes na literatura e para subsidiar intervenções terapêuticas e educacionais nos distúrbios do espectro autístico.

OBJETIVO: caracterizar e comparar o perfil comunicativo de crianças com distúrbios do espectro autístico com dois diferentes interlocutores. MÉTODO: participaram da pesquisa nove sujeitos, sendo quatro crianças do gênero masculino, com faixa etária variando entre cinco anos e oito meses e onze anos, com distúrbios do espectro autístico e cinco terapeutas de linguagem. Foram realizadas análises do perfil comunicativo dessas crianças com os dois diferentes interlocutores (terapeuta de linguagem da própria criança e terapeuta desconhecida). Para análise dos dados foi empregado o Protocolo de Pragmática e tratamento estatístico dos resultados (p < 0,05; Teste dos Postos Sinalizados de Wilcoxon). RESULTADOS: foram levantados o número de atos comunicativos, o meio pelo qual esses atos foram expressos (vocal, verbal e gestual) e as funções comunicativas. em média, o número de atos comunicativos expressos pelos sujeitos tanto com os terapeutas das próprias crianças (5,10 atos/min) como com a terapeuta desconhecida (4,93 atos/min) não foi estatisticamente significante. Observou-se na situação de interação com o próprio terapeuta que a média do percentual da ocorrência do meio comunicativo verbal (31,50%) e a média de funções mais interativas (33,25%) são maiores do que com terapeuta desconhecida. CONCLUSÃO: os diferentes interlocutores, estudados nesta pesquisa, não tiveram uma influência no perfil comunicativo destas crianças. Deste modo, sugerem-se novos estudos, com inclusão de outros interlocutores, a fim de estabelecer quais variáveis interacionais interferem estatisticamente no perfil comunicativo de crianças do espectro autístico, com o intuito de contribuir para a elaboração de melhores propostas terapêuticas para esta população.
