Some factors influencing the growth and production of extracellular lipase by Rhizopus oligosporus were studied. Highest yields of enzyme were obtained when Tweens were the carbon source. Soybean meal extract supported good growth and enzyme production. Carbohydrates, vegetable oils, proteins or amino acids did not stimulate lipase production. The fungus grew well with carbohydrate- or protein-supplemented media but not with oils, unless emulsified with a non-metabolizable gum. The production of biomass in static cultures was maximum at 35-40°C after 4 d at pH 5.5. The yield of lipase was maximum at 25°C after 3 d at pH 6.5. Shaking cultures enhanced growth but decreased lipase production.

Two benomyl-resistant mutants, benD3 tubC41 and benD4 tubC42, of Aspergillus nidulans were isolated after UV treatment. The tubC mutations permitted good conidiation of these strains in culture media containing benomyl and were responsible for increasing their benomyl resistance levels. This implies that β3-tubulin, a product of the tubC gene, in addition to being involved in fungal conidiation, participates in the vegetative growth of the fungus. The tubC gene was located in linkage group I.

A taxonomic survey on fungus-growing ants (Attini) was made at 14 beaches on Santa Catarina Island (SC), Brazil. The samplings were manual, in soil or litterfall, in the following habitats: sandy beach, herbaceous vegetation and shrubby vegetation. From 12 species of Attini (ten of Acromyrmex Mayr and two of Cyphomyrmex Mayr), the most frequent were Cyphomyrmex morschi Emery and Acromyrmex crassispinus Forel, collected, respectively, on eight and ten of the monitored beaches. Altogether, Sorensen's similarity coefficients were high (range: 0.59-0.80), in spite of the lower numbers of ant species on sandy beaches.

The development of Leucoagaricus gongylophorus, the fungus cultured by the leaf-cutting ant Atta sexdens was inhibited in vitro by synthetic compounds containing the piperonyl group. In addition, worker ants that were fed daily on an artificial diet to which these compounds were added had a higher mortality rate than the controls. The inhibition of the fungal growth increased with the size of the carbon side chain ranging from C1 through C8 and decreasing thereafter. 1-(3,4-Methylenedioxybenzyloxy)octane (compound 5) was the most active compound and inhibited the fungal development by 80% at a concentration of 15 μg m1-1. With worker ants the toxic effects started with compound 5 and increased with the number of carbons in the side chain. Thus, for the same concentration (100 μg m1-1) the mortality rates observed after 8 days of diet ingestion were 82%, 66% and 42%, for 1-(3,4-methylenedioxybenzyloxy)decane, 1-(3,4-methylenedioxybenzyloxy)dodecane and compound 5, respectively, whereas with commercial piperonyl butoxide the mortality was 68%. The latter compound, which is known as a synergist insecticide, was as inhibitory to the symbiotic fungus as the synthetic compound 5. The possibility of controlling these insects in the future using compounds that can target simultaneously both organisms is discussed. © 2001 Society of Chemical Industry.

The growth and the extracellular amylase production by Aspergillus ochraceus were studied in a stationary culture medium. Maximum growth rate of this fungus was found after 5 days of incubation at 30° C, but maximum amylase production was obtained after 2 days. The highest amylase production were attained with lactose, maltose, xylose and starch as carbon sources. The extracellular amylase production and mycelial growth were influenced by the concentration of starch. Other carbohydrates supported growth but did not induce amylase synthesis and glucose repressed it, indicating catabolite repression in this microorganism. The presence of both mechanisms of induction and repression suggests that at least these multiple forms of regulation are present in A. ochraceus. Of the nitrogen sources tested, casaminoacids, ammonium nitrate and sodium nitrate stimulated the highest yield of amylase. Optimal amylase production was obtained at pH 5.0, but enzyme activity was found only in the 4.0-6.0 pH range. These results were probably due to the inhibitory effect of NH 4 +-N in the culture medium.

Este trabalho teve como objetivo investigar o efeito da temperatura e do teor de umidade do solo na sobrevivência de Metarhizium anisopliae (Metsch.) Sorok. em três tipos de solos. Foram utilizados o Latossolo Vermelho textura argilosa, Latossolo Vermelho textura média e Argissolo Vermelho Amarelo textura arenosa média. As temperaturas empregadas foram 21,5; 26,8 e 31,5°C, e os teores de umidade foram 35, 65 e 100% de saturação. A sobrevivência do fungo foi avaliada após zero, 20, 40, 60, 80, 100 e 120 dias de incubação em cada temperatura estudada. Na análise do efeito do teor de umidade, a sobrevivência foi avaliada após zero, 14, 28, 42, 56, 70, 84, 98 e 112 dias de incubação à temperatura de 27,0±1,0°C. em ambos os ensaios, foi determinado o número de unidades formadores de colônias (UFC) em placa de Petri. Houve influência significativa da temperatura e do teor de umidade na sobrevivência do fungo. O maior crescimento e a maior sobrevivência ocorreram nas temperaturas de 21,5 e 26,8°C, enquanto que, no solo incubado a 31,5°C, o fungo cresceu pouco, e a população declinou rapidamente. No teor de 65% de umidade, houve rápido crescimento do fungo, mas no 112° dia foi observado um declínio da população nos três tipos de solos. Nos teores de 35 e 100% de umidade, o crescimento foi menor, mas obteve-se maior sobrevivência do fungo no solo.

Chloroquine, due to its basic properties, has been shown to prevent the release of iron from holotransferrin, thereby interfering with normal iron metabolism in a variety of cell types. We have studied the effects of chloroquine on the evolution of experimental paracoccidioidomycosis by evaluating the viable fungal recovery from lung, liver and spleen from infected mice and H2O2, NO production, tumor necrosis factor-alpha (TNF-alpha), interleukin (IL)-6, IL-10 levels and transferrin receptor (TfR) expression from uninfected and infected peritoneal macrophages. Chloroquine caused a significant decrease in the viable fungal recovery from all organs tested, during all periods of evaluation. Peritoneal macrophages from chloroquine-treated infected mice showed higher H2O2 production and TfR expression, and decreased levels of NO, endogenous and stimulated-TNF-alpha, IL-6 and IL-10 during the three evaluated periods. However, despite its suppressor effects on the macrophage function, the chloroquine therapeutic effect upon murine paracoccidioidomycosis was probably due to its effect on iron metabolism, blocking iron uptake by cells, and consequently restricting iron to fungus growth and survival.

The PKC1 gene in the yeast Saccharomyces cerevisiae encodes protein kinase C that is known to control a mitogen-activated protein (MAP) kinase cascade consisting of Bck1, Mkk1 and Mkk2, and Mpk1. This cascade affects the cell wall integrity but the phenotype of Pkc1 mutants suggests additional targets which have not yet been identified. We show that a pkc1Δ mutant, as opposed to mutants in the MAP kinase cascade, displays two major defects in the control of carbon metabolism. It shows a delay in the initiation of fermentation upon addition of glucose and a defect in derepression of SUC2 gene after exhaustion of glucose from the medium. After addition of glucose the production of both ethanol and glycerol started very slowly. The V max of glucose transport dropped considerably and Northern blot analysis showed that induction of the HXT1, HXT2 and HXT4 genes was strongly reduced. Growth of the pkc1Δ mutant was absent on glycerol and poor on galactose and raffinose. Oxygen uptake was barely present. Derepression of invertase activity and SUC2 transcription upon transfer of cells from glucose to raffinose was deficient in the pkc1Δ mutant as opposed to the wild-type. Our results suggest an involvement of Pkc1p in the control of carbon metabolism which is not shared by the downstream MAP kinase cascade. © 2002 Federation of European Microbiological Societies. Rhizoctonia solani causes pre- and post-emergence damping-off, root and hypocotyl rot and foliar blight in soybean. Foliar blight has resulted in yield losses of 31-60% in north and northeast Brazil. The aim of this study was to characterize isolates of R. solani associated with soybean in Brazil. Among 73 Rhizoctonia isolates examined, six were binucleate and 67 were multinucleate. The multinucleate isolates were characterized according to hyphal anastomosis reaction, mycelial growth rate, thiamine requirement, sclerotia production, and RAPD molecular markers. Four isolates that caused hypocotyl rot belonged to AG-4 and using RAPD analysis they grouped together with the HGI subgroup. Another isolate that caused root and hypocotyl rots was thiamine auxotrophic, grew at 35 °C, and belonged to AG-2-2 IIIB. All 62 isolates that caused foliar blight belonged to AG-1 IA. RAPD analysis of R. solani AG-1 IA soybean isolates showed high genetic similarity to a tester strain of AG-1 IA, confirming their classification. The teleomorph of R. solani, Thanatephorus cucumeris was produced in vitro by one AG-1 IA isolate from soybean. The AG-4 and AG-2-2 IIIB isolates caused damping-off and root and hypocotyl rots of soybean seedlings cv. 'FT-Cristalina', under greenhouse conditions. The AG-2-2 IIIB isolate caused large lesions on the cortex tissue, that was distinct from the symptoms caused by AG-4 isolates. The AG-1 IA isolates caused foliar blight in adult soybean plants cv. 'Xingu' under the greenhouse and also in a detached-leaf assay.
