Costa N.A., Simao L.C.V., Santos R.A., Afonso J.A.B., Fagliari J.J., Cardoso E.C., Soares P.C. & Mendonca C.L. 2010. [Proteinogram and serum concentrations of copper, iron and zinc in Santa Ines ewes with Staphylococcus aureus experimentally induced mastitis.] Proteinograma e teores de cobre, ferro e zinco no soro sanguineo de ovelhas da raca Santa Ines com mastite experimental por Staphylococcus aureus. Pesquisa Veterinaria Brasileira 30(5):435-442. Clinica de Bovinos, Campus Garanhuns, Universidade Federal Rural de Pernambuco (UFRPE), Avenida Bom Pastor s/n, Boa Vista, Caixa Postal 152, Garanhuns, PE 55292-901, Brazil. E-mail: carlalopes.mendonca@gmail.comThe aim of the present study was to evaluate the effect of Staphylococcus aureus experimentally induced mastitis on proteinogram and serum concentrations of cupper, iron and zinc levels of Santa Ines primiparous ewes. The right mammary gland of ten healthy ewes was inoculated with 1,0x10(4) UFC/mL of S. aureus. Clinical examination and determination of serum concentrations of proteins by electrophoresis in polyacrylamide gel (SDS-PAGE), cupper, iron and zinc, as well plasma level of fibrinogen were measured before the inoculation (control) and 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 168h, 180h, 288h and 336h after bacteria inoculation. All animals experimentally infected presented clinical mastitis and subsequent loss of mammary gland function. The electrophoretogram allowed the identification of 23 proteins with molecular weights (MW) ranged from 26.000 to 185.000 daltons (Da) including acute-phase proteins, IgG and IgA. A significant increase (P<0,05) in haptoglobin, ceruloplasmin, IgG and IgA concentrations was observed. Antitrypsin and acid glicoprotein concentrations did not alter. The levels of iron and zinc decreased and the cupper concentration increased. A positive correlation between plasma fibrinogen and serum ceruloplasmin (r=0.74), haptoglobin (r=0.62) and IgA (r=0.62) was also identified. Results showed the importance of ceruloplasmin and haptoglobin as acute-phase proteins in ewes with intramammary infections and confirms fibrinogen as an inflammatory marker because its high correlation with specific proteins. The alterations in the serum levels of Cu, Fe and Zn suggest the action of inflammatory mediators triggered by S. aureus.

The present study aimed to evaluating the nutritional content in leaves and fruits of the fig tree 'Roxo de Valinhos', pruned at different periods corresponding to the months of July, August, September and October in the years of 2004 and 2005, with and without the use of irrigation, in the county of Botucatu, São Paulo State, Brazil. To achieve this objective, the adopted experimental design was in blocks with subdivided plots and 5 replications, in which plots corresponded to treatments with and without irrigation and subplots included prunings done in the above-mentioned four months. The levels of N, P, K, Ca, Mg, S, B, Cu, Mn and Zn in leaves and fruits were evaluated in the two crop cycles. The results indicated no significant differences among macro and micronutrient levels in the leaves subjected to treatments with and without irrigation in the cycle 2004/05, except for cupper which showed higher level with the treatment including irrigation (6 mg kg(-1)). In the fruits, there was no difference, except for Zn, which also showed the highest levels (28 mg kg(-1)) with irrigation. In the crop cycle 2005/06, there were differences for N (40 g kg(-1)) and K (20 g kg(-1)) in the leaves, where the highest levels were observed with the treatment including irrigation. In the fruits, N had significant difference and its highest level was observed without irrigation (21 g kg(-1)). In relation to the pruning periods, significant differences were observed for Ca, Fe and Zn content in the leaves and Ca, K, Mg, S and Zn content in the fruits in the crop cycle 2004/05. In the cycle 2005/06, there were not differences among the levels of the evaluated nutrients in the leaves, and in the fruits there was difference for N, Ca and Cu.

Silica gel chemically modified with 2-aminotiazole groups (SiAT), was used for preconcentration of cupper, zinc, nickel and iron from gasoline, normally used as a engine fuel. Surface characteristics and surface area of the silica gel were obtained before and after chemical modification using FT-IR, Kjeldhal and surface area analysis (B.E.T.). The retention and recovery of the analyte elements were studied by applying batch and column techniques. The experimental parameters, such as shaking time in batch technique, flow rate and concentration of the eluent (HCl-0.25-2.00 mol 1(-1)) and the amount of silica, on retention and elution, have been investigated. Detection limits of the method for cupper, iron, nickel and zinc are 0.8, 3, 2 and 0.1 mug 1(-1), respectively. The sorption-desorption of the studied metal ions made possible the development of a preconcentration method for metal ions at trace level in gasoline using flame AAS for their quantification. The objectives of this work were to determine the micelial growth curve of the pathogen and the sensitivity to some fungicides potencially efficient to disease control. The optimum temperature range for micelial growth of Phyllosticta sp, was between 25 and 27.5 degrees C. The maximum and minimum temperatures for micelial growth were 32.5 degrees C and 10 degrees C. Temperatures of 5 and 35 degrees C completely inhibited the growth of the isolates. Total inhibition of the micelial growth was observed with captan and mancozeb (1000 mg a.i./ml) and triadimenol (100 mg a.i./ml). Partial reduction of the micelial growth was observed with iprodione, methyl tiofanate and chlorothalonil until 1.000 mg/ml. The chemical control of PLS was studied in a commercial area of ginger ''Gigante'', in Morretes, PR, where 18 sprays were carried out, with a break of 7 to 10 days, from December to April. The highest reduction of the area under the disease progress curve standardized (AUDPCs) was observed with the spray of chlorothalonil. With the application of dithianon, cupper oxychloride, folpet, mancozeb and captan it was observed AUDPCs between 15.05 and 18.61 lesions/leaf. Iprodione, benomyl, triadimenol and methyl tiofanate did not control the disease (AUDPCs between 20.03 and 25.04 lesions/leaf). The AUDPCs in the check plot was 35.88 lesions/leaf. There was no significant difference of vigor and of ginger yield between fungicide treatments. The cupper oxichloride was phytotoxic to ginger.

Different kinds of modifiers and coatings on the integrated platform of transversely heated graphite atomizer (THGA) have been tested for the simultaneous determination of two group of elements: the first, the more volatile, formed by arsenic, bismuth, lead, antimony and selenium; the second, the less volatile, formed by cobalt, chromium, cupper, iron and manganese in milk by electrothermal atomic absorption spectrometry. Different Rh-modifiers were studied, such as Rh-coated platforms (Rh), carbide plus rhodium coated platforms (W-Rh, Zr-Rh), carbide-coated platforms (W and Zr) with co-injection of RhCl3, solutions and uncoated platforms with injection of solutions of Pd(NO3)(2), Mg(NO3)(2), and RhCl3. Milk samples were diluted 1:10 in 1.0% HNO3 and injected into the tube. The mass of modifier deposited and co-injected in the tube and the use of end capped tubes were also evaluated in order to improve the electrothermal behavior of analytes. Integrated platform pretreated with W plus co-injection RhCl3 for first group and pretreated with W-Rh for second group were elected. For 20 mu L injected samples the analytical curves in the 5.0-20.0 mu g L-1 concentration range have good linear correlation coefficients (r > 0.998). Relative standard deviations (n = 12) are < 6% and the calculated characteristic masses are between 5 pg and 62 pg.
