Flotation is a process of cell separation based on the affinity of cells to air bubbles. In the present work, flotability and hydrophobicity were determined using cells from different yeasts (Hansenulla polymorpha, Saccharomyces cerevisiae, Candida albicans), which were propagated in different media and at different temperatures. Alterations to the supernatant of the cells were also carried out before the flotation assays. The results described here indicate that supernatants of the yeast cells can play a more important role on flotation than cell-wall hydrophobicity. For example, wall-hydrophobicity of strain FLT-01 of S. cerevisiae was high but flotation did not occur when their washed cells were resuspended in water. Additions of neopeptone to cultures of S. cerevisiae and H. polymorpha repressed flotation and increased the volume of foam. An additional task of the present work was to show that the relationship between cell-wall hydrophobicity and flotation performance was dependent on the method used for the measurement of hydrophobicity. Based on the assay procedure, two types of hydrophobicity were distinguished: (a) the apparent hydrophobicity for cells suspended in the medium and expressed by the degree of cell affinity to the organic solvent in the two-phase system supernatant/hexane; (b) the standard hydrophobicity, which was determined for cells suspended in a standard solution (acetate buffer, in the present work) within the acetate buffer/hexane system. Flotation of cells of S. cerevisiae and C albicans were best related to the degree of apparent hydrophobicity (varying with the supernatant composition at the cell/medium interface) rather than to the degree of standard hydrophobicity (varying with the alterations in the wall components, since the liquid phase was constant in the assay). However, depending on the yeast unpredictable results can be obtained. For example, cells of H. polymorpha exhibited good flotation associated to a high degree of standard hydrophobicity while having a lower degree of apparent hydrophobicity. Concerning growth temperature, flotation of cells of C albicans was strongly repressed when the temperature was raised from 30 to 38 degreesC while a similar effect was not observed in cultures of S. cerevisiae and H. polymorpha. It is difficult to understand and predict flotation of yeast cells but simple modifications made to the supernatant of cultures can activate or repress flotation. (C) 2003 Elsevier B.V. B.V. Com objetivo de determinar o consumo e a digestibilidade in vivo, bem como a composição química de dietas com níveis crescentes (0, 50 e 100%) de feno de catingueira (FC), utilizaram-se 15 ovinos Morada Nova, durante um período de 22 dias. Foi usado delineamento inteiramente casualizado com três tratamentos e cinco repetições. O FC apresentou 91,5% de matéria seca (MS), 93,89% de matéria orgânica (MO), 11,25% de proteína (PB), 4,31% de extrato etéreo (EE), 45,47% de fibra em detergente neutro (FDN), 37,08% de fibra em detergente ácido (FDA), 8,39% de hemicelulose (HE), 24,37% de celulose (CE), 12,52% de lignina e 6,30% de tanino condensado. Os consumos de MO e MS, em g/dia, g/kg0,75 e %PV; energia bruta (EB), em kcal/kg/dia e kcal/kg0,75; e dos componentes da parede celular foram reduzidos linearmente pelo aumento de FC na dieta. Os consumos de PB, proteína digestível (PD) e EE aumentaram linearmente com o aumento do FC na dieta. Os coeficientes de digestibilidade da MS, MO, PB e EB das dietas não foram afetados pela inclusão do FC. No entanto, observou-se redução linear para os coeficientes de digestibilidade da FDN, FDA, CE e EE e efeito quadrático para o coeficiente de digestibilidade da HE. Os consumos médios de MS e PB verificados nos níveis intermediários de FC atenderam as exigências de mantença dos animais.

X-rays were initially used for the inspection of special-purpose wood pieces for quantitative evaluation properties of different species. X-ray densitometry has had its use expanded ill dendroclimatology of Picea engelmannii trees. Subsequent laboratories developed applications of X-ray densitometry for environmental, wood science and technology, and related areas. This paper describes the basic methodology of X-ray densitometry applied to the eucalypt wood analysis, as well its presenting the results of applications in three areas: (i) evaluation of wood biodegradation by white rot fungi, (ii) detection of sapwood and heartwood, and (iii) determination of the effect of management oil wood properties. The wood decayed by white rot fungi was detected by X-ray densitometry with it decreasing wood density due to the biodegradation of cell wall components. The sapwood and heartwood of eucalypts were separated in response to the attenuation of X-rays, reflected by the wood anatomical structure and chemical composition. Also, Ill eucalypt trees after the application of irrigation and i characteristic wood density profiles were detected. Ill addition, the significant potential of X-ray densitometry for eucalypt wood research and analysis is discussed.

O objetivo deste trabalho foi avaliar a produção de matéria seca e a composição bromatológica de forrageiras em sistema de integração lavoura-pecuária, em diferentes datas de semeadura. Foi usado delineamento experimental de blocos ao acaso, em esquema fatorial 4x3, com quatro forrageiras - braquiária, Urochloa brizantha; capim-moa, Setaria italica; milheto, Pennisetum glaucum; e sorgo forrageiro, Sorghum bicolor - semeadas em três datas nas estações inverno/primavera e verão/outono, em sucessão à soja. O milheto e o sorgo forrageiro produziram maior quantidade de matéria seca em todas as datas de semeadura, nas duas estações de cultivo e, ao contrário da braquiária e do capim-moa, não diminuíram o teor de nutrientes digestíveis totais no inverno/primavera. Os teores de proteína bruta reduziram com o avanço das datas de semeadura no inverno/primavera e aumentaram no verão/outono, exceto para braquiária e milheto. O sorgo apresentou os menores teores de fibras em detergente neutro nas segunda e terceira datas de semeadura no inverno/primavera, e na primeira data de semeadura no verão/outono. Nas segunda e terceiras datas do inverno/primavera, o sorgo apresentou menor teor de fibras em detergente ácido. Quanto aos constituintes da parede celular, apenas os teores de hemicelulose não se alteram nas forrageiras em virtude das datas de semeadura, em ambas as estações.

beta-glucan, one of the major cell wall components of Saccharomyces cerevisiae, has been found to enhance immune functions. This study investigated in vivo and in vitro effects of beta-glucan on lymphoproliferation and interferon-gamma (IFN-gamma) production by splenic cells from C57BL/6 female mice. All experiments were performed with particulate beta-glucan derived from S. cerevisiae. Data demonstrated that both, i.p administration of particulate beta-glucan (20 or 100 µg/animal) and in vitro stimulation of splenic cells (20 or 100 µg/ml of culture) decreased lymphoproliferation and IFN-gamma production induced by concanavalin A. These results suggest that beta-glucan can trigger a down-modulatory effect regulating a deleterious immune system hyperactivity in the presence of a strong stimulus.

Fatores de virulência em fungos de micoses sistêmicaFungos patogênicos causadores de micoses sistêmicas possuem vários fatores que permitem seu crescimento nas condições adversas oferecidas pelo hospedeiro, propiciando o estabelecimento da relação parasitária e contribuindo no processo de doença. Esses fatores são conhecidos como fatores de virulência auxiliando no desenvolvimento da infecção e interferindo com a patogênese das micoses. O presente trabalho avalia os fatores de virulência em fungos patogênicos como Blastomyces dermatitidis, Coccidioides immitis, Cryptococcus neoformans, Histoplasma capsulatum e Paracoccidioides brasiliensis, em relação à termotolerância, dimorfismo, componentes da parede celular ou cápsula, bem como a produção de enzimas. Os fatores de virulência auxiliam na aderência, colonização, disseminação e habilidade do fungo para resistir a ambientes hostis e escapar dos mecanismos da resposta imune do hospedeiro.Tanto os fatores de virulência apresentados por diferentes fungos, como os mecanismos de defesa oferecidos pelo hospedeiro requerem ação e interação de processos complexos, cujo conhecimento permitirá a melhor compreensão da patogenia das micoses sistêmicas.

The objective of this research was to evaluate the dry mass yield and chemical composition of four Brachiaria species in different options for sowing, exclusively or in intercrop with corn crop, under a no-tillage system. The experiment was carried out during the growing seasons of 2006 at FEPE (FE/Unesp, Ilha Solteira Campus) located in Selvíria, Mato Grosso do Sul State, Brazil. The soil of the experimental area was classified as distroferric Red Latosol (Oxisol). The experimental design was in randomized blocks, in a factorial scheme (4 × 4), with five replications. The treatments consisted of four Brachiaria species (Brachiaria brizantha cv. Marandu, Brachiaria decumbens, Brachiaria ruziziensis and Mulato II grass) grown in rows and spread on total area, exclusively or intercropped simultaneously with corn crop sowing. The study evaluated the dry mass yield and total digestible nutrients, crude protein, ash, neutral detergent fiber, acid detergent fiber, hemicelluloses, cellulose and lignin content of forage. The spread on total area intercrop of forages with corn crop proved to be viable by presenting similar dry mass yield to exclusive sowing arrangements, conversely to what happened with intercrop in row of corn crop, which decreased such yield. Brachiaria ruziziensis showed superior chemical composition and the intercrops increased energy and crude protein contents, and decreased cell wall components.

The objective of this trial was to evaluate the effect of the addition of chemical and bacterial additive in the ensiling of sugar cane (Saccharum officinarum L.) on chemical composition, pH, kinectic fraction and in situ degradation of nutritions components of silages. Five rumen-cannulated 1/2 Simental + 1/2 Zebu steers were allotted to a completely randomized design. The steers were placed in individual cages and they were fed with diets with 76% forage (%DM). Five silages were evaluated: control - sugar cane, no additives; urea - sugar cane + 0.5% of urea (wet basis); inoculant - sugar cane inoculated with LactoSilo® (390 g/40 t forage); NaOH - sugar cane + 1.0% of sodium hydroxide (wet basis); CaOH - sugar cane + 0.6% of calcium hydroxide (wet basis). The silage additives with sodium hydroxide showed the highest pH values before (11.20) and after (4.87) for silage. No differences were observed among the silages for dry matter (26.85), crude protein (5.25) and acid detergent fiber (57.21). Fractionation of dry matter and organic matter of silages showed similar behavior, with higher values of the soluble fraction (fraction A) for silages with sodium hydroxide (45.86 and 30.95%) and calcium hydroxide (29.47 and 26.13%). The use of sodium hydroxide allowed obtaining higher values for the degradation of cell wall components of silages from cane sugar. The potencial and effective degradability with 3, 5 and 8%/h of passage rate were respectively 88.44, 64.45, 56.73 and 49.83% for NDF and 82.57, 55.51, 46.72 and 38.83% for ADF, indicating that the use of sodium hydroxide as chemical additives can improve the nutritive value of cane sugar silage.

The dimorphic fungus Paracoccidioides brasiliensis is the causative agent of the most frequent systemic mycosis in Latin America. In humans, infection starts by inhalation of fungal propagules, which reach the pulmonary epithelium and differentiate into the yeast parasitic phase. Here we describe the characterization of a Dfg5p ((d) under bar efective for (f) under bar ilamentous (g) under bar rowth) homologue of P. brasiliensis, a predictable cell wall protein, first identified in Saccharomyces cerevisiae. The protein, the cDNA and genomic sequences were analysed. The cloned cDNA was expressed in Escherichia coli and the purified rPbDfg5p was used to obtain polyclonal antibodies. Immunoelectron microscopy and biochemical studies demonstrated the presence of PbDfg5p in the fungal cell wall. Enzymatic treatments identified PbDfg5p as a beta-glucan linked protein that undergoes N -glycosylation. The rPbDfg5p bound to extracellular matrix components, indicating that those interactions could be important for initial steps leading to P. brasiliensis attachment and colonization of host tissues. The P. brasiliensis dfg5 nucleotide and deduced protein, PbDfg5p, sequences reported in this paper had been submitted to the GenBank database under Accession Nos AY307855 (cDNA) and DQ534495 (genomic). Structural features of segmental parts of the aorta of the dog were studied by light microscopy (LM) and scanning electron microscopy (SEM). The variability in the wall architecture composition and vascular thickness of the ascending (T2-3 level), thoracic (T7-8 level) and abdominal (L6-7 infrarenal level) segments of the aorta was analysed. Morphological features such as presence of intimal folds, pattern of the medial myoconnective components with segmental variations in the number of elastic lamellae, whose relative number was higher in the thoracic aorta (ascending and descending parts), compared with the abdominal aorta, and a network of connective (stromal) elements formed by elastic and collagen lamellae and fibres in the adventitia were observed. The results were discussed on a histophysiological basis, because small but significant segmental differences had been characterized in the aortic wall structure of the dog.

This work considers a problem of interest in several technological applications such as the thermal control of electronic equipment. It is also important to study the heat transfer performance of these components under off-normal conditions, such as during failure of cooling fans. The effect of natural convection on the flow and heat transfer in a cavity with two flush mounted heat sources on the left vertical wall, simulating electronic components, is studied numerically and experimentally. The influence of the power distribution, spacing between the heat sources and cavity aspect ratio have been investigated. An analysis of the average Nusselt number of the two heat sources was performed to investigate the behavior of the heat transfer coefficients. The results obtained numerically and experimentally, after an error analysis, showed a good agreement.
