Desempenho de diferentes estádios embrionários no cultivo in vitro de embriões de 'Pêra Rio' x 'Poncã'

Objetivou-se verificar qual o melhor estádio embrionário para o cultivo de embriões imaturos oriundos de frutos provenientes de hibridação entre 'Pêra Rio' x 'Poncã' , bem como o efeito de diferentes concentrações do meio de cultura MT. Os embriões em diferentes estádios de desenvolvimento (globulares, torpedo e cordiforme) foram excisados e inoculados em tubos de ensaio contendo 15 mL do meio MT com diferentes concentrações (0; 50; 100 e 150% da composição original e acrescido de 50 g.L-1 de sacarose). Após a inoculação, os embriões foram incubados à 27±1ºC, fotoperíodo de 16 horas e irradiância de 32 mmol.m-2.s-1. Após 90 dias, avaliou-se o comprimento da parte aérea e do sistema radicular, massa fresca e número de folhas das plântulas. Melhor desenvolvimento dos embriões imaturos foi obtido em estádio cotiledonar e com a concentração de 150% do meio MT.

Female reproductive system of the decapitating fly Pseudacteon wasmanni Schmitz (Diptera : Phoridae)

Pseudacteon wasmanni is a South American decapitating fly that parasitizes workers of Solenopsis fire ants. We used light microscopy (historesin serial-sectioning stained with Haematoxylin/Eosin) and scanning electron microscopy to show and analyze internal and whole external views of the female reproductive system. All specimens analyzed (n = 9) by light microscopy showed post-vitellogenic oocytes inside the ovaries. The lack of typical follicles (oocyte-nurse cell complexes) in all specimens suggests that oogenesis occurs during the pupal stage. The total number of eggs found ranged from 31 to 280 (X = 142 +/- 73, SD). The egg has a slugform or torpedo shape (about 130 by 20 mum) with a pointed apex at the posterior pole as defined by the fly; the micropyle appears to be in a depression or invagination at the anterior pole. An acute hypodermic-like ovipositor is evaginated from the hard sclerotized external genitalia during egg laying. The existence of a muscular bulb associated with the end of the common oviduct suggests that the egg is injected into the ant's body by a strong contraction of the bulb which probably is stimulated by bending of several ventral sensilla. During contraction, the abdomen extends out along a large fold between the sixth and seventh tergites in such a way that the sclerotized genitalia is rotated ventrally into a slightly anterior orientation in preparation for oviposition. CNS-selective noncompetitive cholinesterase inhibitors derived from the natural piperidine alkaloid (-)-spectaline

LASSBio-767 [(-)-3-O-acetyl-spectaline] and LASSBio-822 [(-)-3-O-tert-Boc-spectaline] were recently described as cholinesterase inhibitors derived from the natural piperidine alkaloid (-)-spectaline, obtained from the flowers of Senna spectabilis (Fabaccae). We investigated their mechanism of inhibition of acetylcholinesterase and their efficacy in reversing scopolamine-induced amnesia. Competition assays with the substrate acetylthiocholine showed a concentration-dependent reduction in rat brain cholinesterase V-max without changes in apparent K-m. The kinetic data for LASSBio-767 and LASSBio-822 were best fit by a model of simple linear noncompetitive inhibition with K-i of 6.1 mu M and 7.5 mu M, respectively. A dilution assay showed a fast and complete reversal of inhibition, independent of incubation time. Simulated docking of the compounds into the catalytic gorge of Torpedo acetylcholinesterase showed interactions with the peripheral anionic site, but not with the catalytic triad. Anti-amnestic effects in mice were assessed in a step-down passive avoidance test and in the Morris water maze 30 min after injection of scopolamine (1 mg/kg i.p.). Saline, LASSBio-767, or LASSBio-822 was administered 15 min before scopolamine. Both compounds reversed the scopolamine-induced reduction in step-down latency at 0.1 mg/kg i.p. LASSBio-767 reversed scopolamine-induced changes in water maze escape latency at 1 mg/kg i.p. or p.o., while its cholinergic side effects were absent or mild up to 30 mg/kg i.p. (LD50 above 100 mg/kg i.p.). Thus, the (-)-spectaline derivatives are potent cholinergic agents in vivo, with a unique profile combining noncompetitive cholinesterase inhibition and CNS selectivity, with few peripheral side effects. The work describes actions carried out in colaboration among the National Steel Company - (CSN)-Brazil and the Interdisciplinary Laboratory of Electrochemistry and Ceramic - LIEC of the Federal University of Sao Carlos Brazil (UFSCar-Brazil), in the area of I&D, for integral management and improvement of the quality and performance of the refractory linings in the sintering areas, blast furnace, hot air pipe lines transferency to the blast furnace, pig-iron ladles, running channels, blast furnaces hearths, torpedo car, etc., as well as, the economic impact generated by the installation of the adopted measures.

Discopyge castelloi sp. nov. (Torpediniformes, Narcinidae), una nueva especie de raya eléctrica del Mar Argentino

A new species of electric ray of the family Narcinidae, Discopyge castelloi, is described from the southwestern Atlantic off Argentina. Distinctive characters of the new species are the form of the body, the posterior position of pelvic fins and the manner as pelvic fins join the caudal peduncle, the morphology of the nasal curtain and the presence of small but apparently functional eyes. Discopyge castelloi is the fourth torpediniform species reported from the area. It appears to be very rare on the shelf, though the type locality is separated from that of the paratypes by seven latitude degrees. As in the case of Torpedo puelcha, it is probable that the new species has a normal habitat deeper than that of the available specimens.

Selection of desulfurizing agents and optimization of operational variables in hot metal desulfurization

The growing demand for steels with tighter compositional specifications led the Companhia Siderúrgica Nacional (CSN) to develop more efficient processes. To solve this problem this paper aims to identify the operational variables more impacting in the desulfurization process, specifically in torpedo car, as well as its causes and solutions. Then select and test, with laboratorial and industrial tests, desulfurizing agents based of CaC 2, CaO, CaCO3, and Mg to assess the cost per quantity of product desulfurized. The mixture with best results was not that one with highest content of CaC2. It is believed that this mixture showed better efficiency because of the increased agitation of the bath, produced by the releasing of gas from compound CaCO3 present in this mixture. Corrosion prevention for Al2O3/SiC/C/MgAl2O4 based ceramics by means of impregnation with cerium-carrying solution

A cerium-carrying solution was developed so as to aprtially fill the open porosity of Al2O3/SiC/C/MgAl2O4 based refractory lining microstructure used in torpedo ladles, thereby enhancing wear resistance. The protection mchanism was cleared up and introduced from the impregnation technique using a cerium-carrying solution.
