The nuclear poly(A)-binding protein 1 (PABPN1) is a ubiquitously expressed protein that plays a critical role in polyadenylation. Short expansions of the polyalanine tract in the N-terminus of PABPN1 lead to oculopharyngeal muscular dystrophy (OPMD), which is an adult onset disease characterized by eyelid drooping, difficulty in swallowing and weakness in the proximal limb muscles. Although significant data from in vitro biochemical assays define the function of PABPN1 in control of poly(A) tail length, little is known about the role of PABPN1 in mammalian cells. To assess the function of PABPN1 in mammalian cells and specifically in cells affected in OPMD, we examined the effects of PABPN1 depletion using siRNA in primary mouse myoblasts from extraocular, pharyngeal and limb muscles. PABPN1 knockdown significantly decreased cell proliferation and myoblast differentiation during myogenesis in vitro. At the molecular level, PABPN1 depletion in myoblasts led to a shortening of mRNA poly(A) tails, demonstrating the cellular function of PABPN1 in polyadenylation control in a mammalian cell. In addition, PABPN1 depletion caused nuclear accumulation of poly(A) RNA, revealing that PABPN1 is required for proper poly(A) RNA export from the nucleus. Together, these experiments demonstrate that PABPN1 plays an essential role in myoblast proliferation and differentiation, suggesting that it is required for muscle regeneration and maintenance in vivo.

Oslerus osleri is a small nematode that infects the respiratory tract of domestic and wild canids and is responsible for causing chronic nodular tracheobronchitis. This paper aims to report a case of parasitism by O. osleri in a free-living maned wolf (Chrysocyon brachyurus) that was struck by a motor vehicle. Fecal samples were collected, and the presence of spiral larvae, with S-shaped tails, was observed on flotation. This characteristic was compatible with the Filaroididae Family larvae of O. osleri. Although the animal did not show clinical signs of respiratory system impairment, a tracheobronchoscopy was performed. Semitransparent nodules, 5 mm in diameter, containing adult parasites were observed in the third distal portion of the trachea, cranial to the carina. Larval morphological characteristics and the nodular locations were compatible with an O. osleri respiratory tract infection.

The objective of this study was to determine morphological and functional characteristics of semen retrieved from the feline epididymis before and after cooling. Sixteen adult male cats were orchiectomized. The distal portion of the epididymis and proximal part of the deferent ducts were dissected and squeezed to obtain their content. After centrifugation, the supernatant was removed, sperm were resuspended in a 0.9 mL Tris-fructose-citric acid extender containing 20% egg yolk, aliquoted into three 0.3 mL samples, placed in a refrigerator (4.8 degrees C) and cooled (0.5 degrees C/min). Semen evaluations were performed on four occassions: immediately after epididymal sperm retrieval (TO), and at 24 h (T-1), 48 h (T-2) and 72 h (T-3) after cooling. on each occasion, progressive motility, vigor and sperm morphology were determined. Mean motility and vigor decreased (P < 0.05) between each successive examination. Although the majority of sperm cell damage occurred within the first 24 h, there was a decrease (P < 0.05) in mean percentage of morphologically normal sperm between To and each evaluated time (T-1, T-2, T-3) after cooling, due to an increase in coiled and bent sperm tails. Further studies are needed to evaluate the effects of cooling on the fertilizing capacity of cat epididymal spermatozoa in assisted reproduction programs. (c) 2006 Elsevier B.V. All rights reserved.

The South American water snake Helicops leopardinus is very abundant in the Pantanal wetlands of Central Brazil. We studied the biology of this species based on specimens collected by local workers in the Paraguay River. Females attained greater body lengths and had larger heads than males, but the latter had longer tails. Helicops leopardinus fed on a wide variety of fishes (70% of the total prey items) and frogs, with fishes of the order Gymnotiformes (33.34%) and frogs of the family Hylidae (23.34%) representing the most common prey items. There was relatively lows diet overlap between the sexes and between juveniles and adults. Mean litter size was 6.6 and ranged from 4-11. The reproductive cycle was seasonal, with birth occurring late in the wet season (simultaneous with the flooding of the Paraguay River), facilitating dispersal of young snakes by floating vegetation. Floating vegetation may be a critical habitat for H. leopardinus, and management plans for their maintenance need to be developed.

Coral snakes in the genus Micrurus are widely distributed in the Neotropics, with more than 50 species already described in this region. They are primarily ground or leaf litter dwellers, feed on snakes or other elongate vertebrates, and tend to reproduce during the rainy season. We present data on the biology of Micrurus pyrrhocryptus from two semideciduous forests of Mato Grosso do Sul state, western Brazil. Two snake species were consumed: the dipsadid Sibynomorphus lavillai and the typhlopid Typhlops brongersmianus. Sexual dimorphism is marked in M. pyrrhocryptus. with males attaining larger sizes and having longer tails. Activity was concentrated in the wet season, when reproduction occurs. The defensive behavior in this species is similar to that displayed by other Micron's, although less pronounced.

The Neotropical pitviper genus Bothrops comprises about 40 species, which occur in all main ecosystems of cis-Andean South America. We explored the relationships of body size and form (tail length and stoutness) with macrohabitat use in 20 forms of Bothrops. Sen-ii-arboreal habits appeared only in forest forms. Semi-arboreals are significantly more slender and have longer tails than terrestrials; body size is not significantly different between terrestrials and semi-arboreals. Within Bothrops, independent contrasts for macrohabitat use were significantly correlated with contrasts of tail size (positively) and stoutness (negatively); thus, the more arboreal the species, the longer its tail and the more slender its body. Contrasts of adult body size seems to remain constant over the lower range of macrohabitat use, but to decrease in species of Bothrops which are more arboreal. Reconstructions of character states indicate that: (1) the ancestor of Bothrops was a small, stout, terrestrial species; (2) semi-arboreal habits appeared one to three times in the genus; (3) a decrease in stoutness and an increase in tail length occurred along with an increase in arboreality in some clades. Although macrohabitat use seems to be important in determining body form in Bothrops, our results also indicate that tail size, stoutness and body size may also be affected by selective agents other than macrohabitat use. The selective agents responsible for the shifts in macrohabitat use in Bothrops are still uncertain, although they may have included prey availability and/or predation pressure. The plasticity of macrohabitat use, morphology and body size described in this study may have been key features that facilitated the highly successful ecological diversification of Bothrops in South America.

An analysis of testicular morphology and spermatogenesis in the Heteroptera species Antileuchus tripterus (Pentatomidae) revealed that these traits differ from that recorded for other species of the same family. The testes of A. tripterus have only six lobes, while other species of the same family typically have seven lobes, including a compound lobe, i.e., a lobe containing another in its lumen. A study of meiosis and sperm structure in the different lobes of A. tripterus revealed a conventional meiosis in the lobes numbered one to three. In lobe four, however, prophase I spermatocytes exhibit the morphological characteristics of the so called diffuse stage and are larger than those in lobes 1-3 in this and the remaining phases of meiosis. Thus, the resulting spermatids are not only larger but also their head is morphologically different. Lobe 5 exhibits characteristies known from other Pentatomidae species such as an uneven distribution of chromatin to the daughter cells, which give rise to cells of different sizes. This lobe forms spermatozoa of different sizes. Lobe 6, contained within lobe 5, differs from it by having larger cells at a stage similar to the so called diffuse and spermatid tails with cross divisions, which are not found in other lobes and unreported in the literature.

Differential scanning calorimetry (DSc) and dynamic light scattering (DLS) were used to obtain the gel to liquid-crystalline phase transition temperature (T-m) and the apparent hydrodynamic radius (R-h) of spontaneously formed cationic vesicles of dialkyldimethylammonium bromide salts (CnH2n+1)(2)(CH3)(2)N+center dot Br-, with varying chain lengths. The preparation of cationic vesicles from aqueous solution of these surfactants, for n = 12, 14, 16 and 18 (DDAB, DTDAB, DHDAB and DODAB, respectively), requires the knowledge of the surfactant gel to liquid-crystalline phase transition temperature, or melting temperature (T-m) since below this temperature these surfactants are poorly or not soluble in water. That series of cationic surfactants has been widely investigated as vesicle-forming surfactants, although C-12 and C-18, DDAB and DODAB are by far the most investigated from this series. The dependence of T-m of these surfactants on the number n of carbons in the surfactant tails is reported. The T-m obtained by DSC increases non-linearly with n, and the vesicle apparent radius R-h is about the same for DHDAB and DODAB, but much smaller for DDAB. (c) 2006 Elsevier B.V.. We studied the statistical distribution of student's performance, which is measured through their marks, in university entrance examination (Vestibular) of UNESP (Universidade Estadual Paulista) with respect to (i) period of study - day versus night period (ii) teaching conditions - private versus public school (iii) economical conditions - high versus low family income. We observed long ubiquitous power law tails in physical and biological sciences in all cases. The mean value increases with better study conditions followed by better teaching and economical conditions. In humanities, the distribution is close to normal distribution with very small tail. This indicates that these power law tails in science subjects axe due to the nature of the subjects themselves. Further and better study, teaching and economical conditions axe more important for physical and biological sciences in comparison to humanities at this level of study. We explain these statistical distributions through Gradually Truncated Power law distributions. We discuss the possible reason for this peculiar behavior.
