Structural insights for fatty acid binding in a Lys49-phospholipase A(2): crystal structure of myotoxin II from Bothrops molojeni complexed with stearic acid

The crystal structure of dimeric Lys49-phospholipase A2 myotoxin-II from Bothrops moojeni (MjTX-II) co-crystallized with stearic acid (C18H36O2) has been determined at a resolution of 1.8 angstrom. The electron density maps permitted the unambiguous inclusion of six stearic acid molecules in the refinement. Two stearic acid molecules could be located in the substrate-binding cleft of each monomer in positions, which favor the interaction of their carboxyl groups with active site residues. The way of binding of stearic acids to this Lys49-PLA(2)s is analogous to phospholipids and transition state analogues to catalytically active PLA(2)s. Two additional stearic acid molecules were located at the dimer interface region, defining a hitherto unidentified acyl-binding site on the protein surface. The strictly conserved Lys122 for Lys49-PLA(2)s may play a fundamental role for stabilization of legend-protein complex. The comparison of MjTX-II/satiric acid complex with other Lys-PLA(2)s structures whose putative fatty acids were located at their active site is also analysed. Molecular details of the stearic acid/protein interactions provide insights to binding in croup I/II PLA(2)s and to the possible interactions of Lys49-PLA(2)s with target membranes. Figurações contemporâneas do espaço na literatura

Neste livro abre-se a uma diversidade de acepções que o conceito de espaço abriga nos objetos e cenários literários contemporâneos. O próprio texto como um espaço espacializante, na sua textualidade como corpo ou na visualidade da escrita, configura um espaço de linguagem. O contexto cultural e histórico envolve referências e dados situados em determinados momentos que articulam um espaço de relações com o texto literário. O espaço da institucionalização, com as representações de poder, o espaço social, o espaço como paisagem, nas configurações natural, regional, física e psicológica, e o espaço do mito com suas imagens e sentidos simbólicos, são outras formas de mobilização do conceito. O espaço pictórico, quer como elemento componente da pintura ou como a própria linguagem na composição da escritura, que se vale de técnicas plásticas, também é contemplado, assim como esse tipo de diálogo abriga outros meios e linguagens, como o cinema, a propaganda, a fotografia, nas relações entre as artes. Há também a possibilidade de se rever o objeto literário, em termos de espacialidade, sob essa perspectiva múltipla do olhar contemporâneo. Ou seja, no lugar daquele sentido restrito de categoria estrutural, o conceito incorpora uma visão ampla e metafórica, dentro da qual o próprio texto faz emergir uma concepção e função da espacialidade por meio de suas estratégias de construção. Diante de tantas possibilidades de leitura do conceito, o presente livro reúne textos que abordam aspectos da simbolização espacial em obras literárias que implicam um olhar contemporâneo para essas figurações.

Classificação dos jogos no ambiente virtual do lazer, na perspectiva de usuários

Esse estudo, de natureza qualitativa, objetivou investigar as diferentes classificações dos jogos virtuais e os mais procurados pelos usuários. A pesquisa foi composta pela união de pesquisas bibliográficas e exploratórias, desenvolvida por intermédio de questionário semi-estruturado, aplicado a usuários de jogos virtuais on-line, participantes de fóruns on-line do Brasil. Os dados foram analisados descritivamente. Dos 17 indivíduos que responderam à pesquisa, 82% são do sexo masculino e 18%, do sexo feminino, com idades entre 21 e 27 anos. O lugar que em que mais acessam os jogos virtuais são suas casas, e a maior freqüência de acesso é de 7 dias por semana (29%), de 3 a 5 horas por dia (76%). O principal interesse pelos games acontece por esses serem uma forma de entretenimento. Dentre os jogos mais citados como seus favoritos estão Metal Gear Solid, Resident Evil, Farmville, Age of Empires e The Legend of Zelda. Quando perguntados o porquê de seu interesse por tal jogo, os participantes responderam que eles promovem socialização, diversão, contém gráficos (imagens e sons) e enredos interessantes, entre outros. A categoria de jogos favorita dos participantes é a de tiro (First Person Shooter), com 35% de citações, seguida por jogos de esportes, com 30%; porém, 23,5% respondeu não ter preferência quanto à categoria do game. Quanto à Classificação Indicativa, 65% dos participantes a consideram um fator importante, enquanto 35% consideram-na desnecessária. Porém, 88% responderam não olhar a Classificação Indicativa do game antes de comprá-lo ou utilizá-lo, citando, dentre os principais fatores, não se importarem ou ofenderem com qualquer conteúdo que o jogo possa apresentar e por serem maiores de idade.

Entre o cinema e a poesia, a memória e a imagem

The aim of this essay is to analyze common aspects between cinema and poetry such as image and memory. Images represent the subject’s perception, whose memory of things and feelings are built artistically (fragmentary sequences, a new idea of time and space, no-narrative style) into the language of cinema and poetry. Homological relations between both languages are put in evidence in this essay in order to bring to discussion those aspects that show the tinny frontiers that separate artistic systems.
