Standard Test Methods (e.g. ASTM, DIN) for materials characterization in general, and for fatigue in particular, do not contemplate specimens with complex geometries, as well as the combination of axial and in-plane bending loads in their methodologies. The present study refers to some patents and the new configuration or configurations of specimens (non-standardized by the status quo of test methods) and a device developed to induce axial and bending combined forces resultants from axial loads applied by any one test equipment (dynamic or monotonic) which possesses such limitation, towards obtaining more realistic results on the fatigue behavior, or even basic mechanical properties, from geometrically complex structures. Motivated by a specific and geometrically complex aeronautic structure (motor-cradle), non-standardized welded tubular specimens made from AISI 4130 steel were fatigue-tested at room temperature, by using a constant amplitude sinusoidal load of 20 Hz frequency, load ratio R = 0.1 with and without the above referred auxiliary fatigue apparatus. The results showed the fatigue apparatus was efficient for introducing higher stress concentration factor at the welded specimen joints, consequently reducing the fatigue strength when compared to other conditions. From the obtained results it is possible to infer that with small modifications the proposed apparatus will be capable to test a great variety of specimen configurations such as: squared tubes and plates with welded or melted junctions, as well as other materials such as aluminum, titanium, composites, polymeric, plastics, etc. © 2009 Bentham Science Publishers Ltd.

Soil CO2 efflux is the primary source of CO2 emissions from terrestrial ecosystems to the atmosphere. The rates of this flux vary in time and space producing hot moments (sudden temporal high fluxes) and hot spots (spatially defined high fluxes), but these high reaction rates are rarely studied in conjunction with each other. We studied temporal and spatial variation of soil CO2 efflux in a water-limited Mediterranean ecosystem in Baja California, Mexico. Soil CO2 efflux increased 522% during a hot moment after rewetting of soils following dry summer months. Monthly precipitation was the primary driver of the seasonal trend of soil CO2 efflux (including the hot moment) and through changes in soil volumetric water content (VWC) it influenced the relationship between CO2 efflux and soil temperature. Geostatistical analyses showed that the spatial dependence of soil CO2 efflux changed between two contrasting seasons (dry and wet). During the dry season high soil VWC was associated with high soil CO2 efflux, and during the wet season the emergence of a hot spot of soil CO2 efflux was associated with higher root biomass and leaf area index. These results suggest that sampling designs should accommodate for changes in spatial dependence of measured variables. The spatio-temporal relationships identified in this study are arguably different from temperate ecosystems where the majority of soil CO2 efflux research has been done. This study provides evidence of the complexity of the mechanisms controlling the spatio-temporal variability of soil CO2 efflux in water-limited ecosystems. (C) 2014 Elsevier Ltd. Paracoccidioides brasiliensis infections have been little studied in wild and/or domestic animals, which may represent an important indicator of the presence of the pathogen in nature. Road-killed wild animals have been used for surveillance of vectors of zoonotic pathogens and may offer new opportunities for eco-epidemiological studies of paracoccidiodomycosis (PCM). The presence of P. brasiliensis infection was evaluated by Nested-PCR in tissue samples collected from 19 road-killed animals; 3 Cavia aperea (guinea pig), 5 Cerdocyon thous (crab-eating-fox), 1 Dasypus novemcinctus (nine-banded armadillo), 1 Dasypus septemcinctus (seven-banded armadillo), 2 Didelphis albiventris (white-eared opossum), 1 Eira barbara (tayra), 2 Gallictis vittata (grison), 2 Procyon cancrivorus (raccoon) and 2 Sphiggurus spinosus (porcupine). Specific P. brasiliensis amplicons were detected in (a) several organs of the two armadillos and one guinea pig, (b) the lung and liver of the porcupine, and (c) the lungs of raccoons and grisons. P. brasiliensis infection in wild animals from endemic areas might be more common than initially postulated. Molecular techniques can be used for detecting new hosts and mapping 'hot spot' areas of PCM.

Plasmon-enhanced spectroscopic techniques have expanded single-molecule detection (SMD) and are revolutionizing areas such as bio-imaging and single-cell manipulation. Surface-enhanced (resonance) Raman scattering (SERS or SERRS) combines high sensitivity with molecularfingerprint information at the single-molecule level. Spectra originating from single-molecule SERS experiments are rare events, which occur only if a single molecule is located in a hot-spot zone. In this spot, the molecule is selectively exposed to a significant enhancement associated with a high, local electromagnetic field in the plasmonic substrate. Here, we report an SMD study with an electrostatic approach in which a Langmuir film of a phospholipid with anionic polar head groups (PO 4 -) was doped with cationic methylene blue (MB), creating a homogeneous, two-dimensional distribution of dyes in the monolayer. The number of dyes in the probed area of the Langmuir-Blodgett (LB) film coating the Ag nanostructures established a regime in which single-molecule events were observed, with the identification based on direct matching of the observed spectrum at each point of the mapping with a reference spectrum for the MB molecule. In addition, advanced fitting techniques were tested with the data obtained from micro-Raman mapping, thus achieving real-time processing to extract the MB single-molecule spectra. © 2013 Society for Applied Spectroscopy.

Preserving large tracts of natural habitats is essential to maintain biodiversity. Nevertheless, even large areas may still suffer from less visible impacts such as loss of ecological processes. Because mapping ecological processes over large scales is not practical, an alternative is to map surrogate species that are key for those processes. In this study, we chose four species of Neotropical large mammals (the largest apex predator: jaguar - Panthera onca; the largest herbivore: tapir - Tapirus terrestris; the largest seed predator: white-lipped peccary - Tayassu pecari; and the largest arboreal seed disperser: muriqui - Brachyteles spp.) in an ecosystem with an old history of human impact (the Atlantic Forest) to test whether areas with native forest still harbor ecological processes that may guarantee long-term ecosystem maintenance. We gathered 94 locations with recent presence of the four species to map current ranges and model suitable areas. Our results reveal that 96% of the remaining Atlantic Forest is depleted of at least one of the four surrogate species and 88% is completely depleted of all four surrogate species. We also found that only 16% is still environmentally suitable for all four, and 55% is completely unsuitable to all four of them. Our study highlights the importance of looking beyond land cover to fully depict intactness of natural areas, and suggests that ecosystems with a long history of human impact (such as the Atlantic Forest) may be suffering from ecological impacts not seen at a first glance. © 2013 Elsevier Ltd.

Topological optimization problems based on stress criteria are solved using two techniques in this paper. The first technique is the conventional Evolutionary Structural Optimization (ESO), which is known as hard kill, because the material is discretely removed; that is, the elements under low stress that are being inefficiently utilized have their constitutive matrix has suddenly reduced. The second technique, proposed in a previous paper, is a variant of the ESO procedure and is called Smooth ESO (SESO), which is based on the philosophy that if an element is not really necessary for the structure, its contribution to the structural stiffness will gradually diminish until it no longer influences the structure; its removal is thus performed smoothly. This procedure is known as "soft-kill"; that is, not all of the elements removed from the structure using the ESO criterion are discarded. Thus, the elements returned to the structure must provide a good conditioning system that will be resolved in the next iteration, and they are considered important to the optimization process. To evaluate elasticity problems numerically, finite element analysis is applied, but instead of using conventional quadrilateral finite elements, a plane-stress triangular finite element was implemented with high-order modes for solving complex geometric problems. A number of typical examples demonstrate that the proposed approach is effective for solving problems of bi-dimensional elasticity. (C) 2014 Elsevier Ltd. All rights reserved.

In the present study we have investigated the effects of heat acclimation on brain and hepatic Hsp70 protein levels and body temperature of broiler chickens in response to gradual heat stress. Two groups of broilers were raised up to 47 days of age under distinct temperature conditions: thermoneutral (TN, according to bird age) or hot environmental (HS, 31-33°C). At 46 days of age, the birds reared at high ambient temperature were transferred to thermoneutrality conditions. After 18 h, these birds and the birds reared at thermoneutral temperature were submitted to gradual heat stress in a climatic chamber so that environment temperature was increased from 28 to 40ºC at a rate of 2ºC/h. Colonic temperature was measured using a thermometer sensor probe at each two hours, and hepatic and brain tissues were collected immediately after slaughter in order to assess Hsp70 protein level by Western blotting analysis. The colonic temperatures of birds reared at high temperature increased steeply during the first 2 h of heat stress (1.06ºC/h) and more slowly thereafter (0.59ºC/h). Broilers reared at thermoneutral temperature showed a small increase in the first 4 h of heat stress (0.18ºC/h) and then colonic temperature increased sharply (0.72ºC/h). Nevertheless, both groups presented similar final colonic temperature by the end of the stress period. Hsp70 levels (ng Hsp70 µg total protein-1) did not change in the liver or brain of the birds reared at high temperature. on the other hand, both liver and brain Hsp70 levels increased significantly during heat stress in the animals reared at thermoneutrality, with a higher expression of this peptide in brain tissue.

Residual stresses play an important role in the fatigue lives of structural engineering components. In the case of near surface tensile residual stresses, the initiation and propagation phases of fatigue process are accelerated; on the other hand, compressive residual stresses close to the surface may increase fatigue life. In both decorative and functional applications, chromium electroplating results in excellent wear and corrosion resistance. However, it is well known that it reduces the fatigue strength of a component. This is due to high tensile internal stresses and microcrack density. Efforts to improve hard chromium properties have increased in recent years. In this study, the effect of a nickel layer sulphamate process, as simple layer and interlayer, on fatigue strength of hard chromium electroplated AISI 4340 steel hardness - HRc 53, was analysed. The analysis was performed by rotating bending fatigue tests on AISI 4340 steel specimens with the following experimental groups: base material, hard chromium electroplated, sulphamate nickel electroplated, sulphamate nickel interlayer on hard chromium electroplated and electroless nickel interlayer on hard chromium electroplated. Results showed a decrease in fatigue strength in coated specimens and that both nickel plating interlayers were responsible for the increase in fatigue life of AISI 4340 chromium electroplated steel. The shot peening pre-treatment was efficient in reducing fatigue loss in the alternatives studied.
