Crude extracts of new and old sesame leaves (Sesamum indicum, Linaeus) obtained by a sequential process of extraction were tested through contact experiments to investigate their toxicity to Atta sexdens rubropilosa (Forel) workers. Methanol extract of old leaves was toxic to leaf-cutting ants at the lower tested concentration (20mg.ml(-1)). This extract was fractioned and their fractions were tested showing a toxic effect of the methanol fraction. The methanol fraction was fractioned again and the sub fraction responsible for worker's toxicity is composed of some sugars.

Para avaliar um esquema de fracionamento de zinco em Argissolo arenoso e suas relações com a disponibilidade de Zn para Cynodon spp cv. Tifton-85, realizou-se um experimento em casa de vegetação, em esquema fatorial 5 x 2 x 2 (5 doses de Zn, 2 doses de calcário e 2 épocas de amostragem), em delineamento inteiramente casualizado, com três repetições. As doses de Zn foram de 0; 2,5; 5,0; 7,5 e 10 mg dm-3; metade dos vasos não recebeu calagem (V = 42 %) e metade recebeu a calagem com vistas em elevar o índice de saturação por bases a 70 %. As épocas de amostragem foram 30 e 150 dias após aplicação de Zn, respectivamente, antes do plantio e depois do 3º corte de Tifton-85. A aplicação de Zn resultou em aumento significativo do elemento nas frações: trocável, óxidos de Mn, matéria orgânica e óxidos de Fe. O Zn ligado aos óxidos de Mn aumentou significativamente com a calagem. Após 150 dias de experimentação, houve diminuição do Zn trocável, ligado aos óxidos de Mn, à matéria orgânica e aos óxidos de Fe, e aumento na fração residual. A distribuição de Zn nas frações do solo foi: residual > óxidos de Fe > óxidos de Mn > trocável > matéria orgânica. As relações entre as características estudadas mostraram que tanto o Zn-DTPA quanto o Zn trocável, ligado à M.O. e aos óxidos de Mn, foram eficientes para representar o Zn absorvido pela planta.

The partitioning of Mn, Al, Zn, Cu and Ti ions in municipal sewage sludge was investigated before and after bioleaching processes effectuated by Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans. Oxidation reduction potential increase and pH decrease were obtained as a result of bacterial activity. A less pronounced and constant decrease was obtained with A. ferrooxidans, whereas A. thiooxidans presented a lag phase before a steep pH decrease. Metal solubilization was accomplished in experimental systems supplemented with energy source, Fe2+ for A. ferrooxidans and S-0 for A. thiooxidans. Solubilization efficiency differed for each metal except for Al, and was relatively similar for either organism. Metal partitioning was conducted using five-step sequential extraction procedure before and after the bioleaching. The results indicated that Zn and Mn ions were mostly associated with the organic fraction, whereas Cu, Al and Ti ions with the sulphide/ residue fraction. The bioleaching process caused prompt solubilization of metals mostly associated with the more labile fractions (exchangeable, adsorbed and organically bound metals), whereas those associated to the less labile ones (EDTA and sulphide/residue fractions) were exchanged towards more labile fractions.

The chemical fractionation and bioleaching of Mn, At, Zn, Cu and Ti in municipal sewage sludge were investigated using Thiobacillus ferrooxidans as leaching microorganism. As a result of the bacterial activity, ORP increase and pH reduction were observed. Metal solubilization was accomplished only in experimental systems supplemented with energy source (Fe(II)). The solubilization efficiency approached similar to80% for Mn and Zn, 24% for Cu, 10% for At and 0.2% for Ti. The chemical fractionation of Mn, At, Zn, Cu and Ti was investigated using a five-step sequential extraction procedure employing KNO3. KF, Na4P2O7, EDTA and HNO3. The results show that the bioleaching process affected the partitioning of Mn and Zn, increasing its percentage of elution in the KNO3 fraction while reducing it in the KF, Na4P2O7 and EDTA fractions. No significant effect was detected on the partitioning of Cu and Al. However, quantitatively the metals Mn, Zn, Cu and At were extracted with higher efficiency after the bacterial activity. Titanium was unaffected by the bioleaching process in both qualitative and quantitative aspects. (C) 2002 Elsevier B.V. Ltd. All rights reserved.

Several plants are used in folk medicine to treat gastrointestinal disorders. Mouriri pusa Gardn. (Melastomataceae) is a medicinal plant commonly used in the central region of Brazil against gastric ulcer. Two organic extracts methanolic (MeOH) and dichloromethane (DCM) obtained by sequential extraction from the leaves of Mouriri pusa were evaluated for their ability to protect the gastric mucosa against injuries caused by necrotizing agents (0.3 M HCl/60% EtOH, absolute ethanol, non-steroidal anti-inflammatory drug, stress and pylorus ligature) in mice and rats. The best results were obtained after pretreatment with MeOH extract whereas the DCM extract did not show the same significant antiulcerogenic activity. No acute toxicity was observed in animals treated with 5 g/kg, p.o. of MeOH extract. The mechanism involving the antiulcerogenic action of MeOH extract seemed to be related to NO generation and also suggested the effective participation of endogenous sulfhydryl group in the gastroprotective action. Phytochemical investigation of the MeOH extract of Mouriri pusa yielded tannins, flavonoids and (-)-epicatechin. The presence of these phenolic compounds probably would explain the antiulcerogenic effect of the polar extract of Mouriri pusa leaves. (c) 2006 Elsevier B.V.. All rights reserved.

Three D-glucans were isolated from the mycelium of the fungus Botryosphaeria rhodina MAMB-05 by sequential extraction with hot-water and hot aqueous KOH (2% w/v) followed by ethanol precipitation. Following their purification by gel permeation chrornatography on Sepharose CL-4B, the structural characteristics of the D-glucans were determined by FT-IR and C-13 NMR spectroscopy and, after methylation, by GC-MS. The hot-water extract produced a fraction designated Q(1A) that was a beta-(1 -> 6)-D-glucan with the following structure:[GRAPHICS]The alkaline extract, when subjected to repeated freeze-thawing, yielded two fractions: KIP (insoluble) that comprised a beta-(1 -> 3)-D-glucan with beta-D-glucose branches at C-6 with the structure:[GRAPHICS]and K1SA (soluble) consisting of a backbone chain of alpha-(1 -> 4)-linked D-glucopyranosyl residues substituted at O-6 with alpha-D-glucopyranosyl residues:[GRAPHICS](c) 2008 Elsevier Ltd. The heavy metals when linked to organic matter have a behavior in the soil that is still little known. This study aimed to evaluate the effect of sewage-sludge-based composts when incorporated in the soil, in relation to heavy metals availability. Five composts were incorporated using sugar-cane bagasse, sewage sludge and cattle manure in the respective proportions: 75-0-25, 75-12.5-12.5, 75-25-0, 50-50-0 and 0-100-0 (composts with 0, 12.5, 25, 50 and 100% sewage sludge). The experiment consisted of 6 treatments (5 composts and a control with mineral fertilization) in randomized blocks with a split-plot design. The control and the treatment of 0% sewage sludge received inorganic nitrogen (N). All the treatments received the same amount of N (8.33 g) K (5.80 g) and K (8.11 g) per pot. Tomato plants were cultivated in 24.0 L pots in a greenhouse in Jaboticabal, SP, Brazil. The concentrations of heavy metals were determined in the soil samples at day 0 after compost incorporation. The higher the sewage sludge doses, the higher heavy metal contents in the soil. Among extractants, Melhlich-1 extracted the highest amount of heavy metals, while DTPA extracted the lowest one. The residual fraction presented the highest heavy metal content, followed by Fe oxides crystalline and amorphous to Cu, Cr and Mn, and Mn oxides, and Fe amorphous to Zn, indicating strong associations to oxides and clays. There were significant positive correlations between Mn contents in the plant and Mn linked to Fe oxide amorphous and crystalline.

The objective of this work was to determine the ether extract content of peanut and canola seeds, with extraction in Soxhlet apparatus and with three types of processing: grinding in ball mill type; maceration in mortar and grind into micro mill after maceration in mortar. Data were analyzed in an entirely randomized design with 3 treatments and 12 replicates, submitted to variance analysis and the means were compared using Tukey test (P<0.05) with routines of the SAS statistical package. The largest percentage of ether extract was obtained in the treatment which the peanuts were subjected to a greater number of extractions (56.43%), and all treatments presented different results(P <0.05). In canola grains, it was observed that the extraction with the method of grinding in ball mill and the method of sequential extraction (macerate + ground in micro mill) were statistically similar (43.52 and 42.35% respectively), and these methods were more efficient (P <0.05) to extract the ether extract than the treatment in which the grains were only macerated. For peanut grains it was concluded that the most efficient method was the sequential one and to the canola grain, it can be used only a grinding mill as samples processing witch will be submitted to ether extract extraction.

The sequential extraction procedure of Zinc and lead performed in a Brazilian soil showed that it presents high pollution potential once over 90% of total lead is present in fractions where the metals can be easily mobilized. The fraction contents are as follow: F1 = 174 and 15 mg kg-1; F2 = 3155 and 9.7 mg kg -1; F3 = 99 and 1.6 mg kg -1; Residual fraction = 38 and 5.5 mg kg -1 for lead and zinc, respectively. The comparison with non contaminated soil only Pb 2+ concentration is above its intervention reference concentration, 900 mg kg -1.
