The Topliss method was used to guide a synthetic path in support of drug discovery efforts toward the identification of potent antimycobacterial agents. Salicylic acid and its derivatives, p-chloro, p-methoxy, and m-chlorosalicylic acid, exemplify a series of synthetic compounds whose minimum inhibitory concentrations for a strain of Mycobacterium were determined and compared to those of the reference drug, p-aminosalicylic acid. Several physicochemical descriptors (including Hammett's sigma constant, ionization constant, dipole moment, Hansch constant, calculated partition coefficient, Sterimol-L and -B-4 and molecular volume) were considered to elucidate structure-activity relationships. Molecular electrostatic potential and molecular dipole moment maps were also calculated using the AM1 semi-empirical method. Among the new derivatives, m-chlorosalicylic acid showed the lowest minimum inhibitory concentration. The overall results suggest that both physicochemical properties and electronic features may influence the biological activity of this series of antimycobacterial agents and thus should be considered in designing new p-aminosalicylic acid analogs.

The study of the H+ concentration at the micellar interface is a convenient system for modeling the distribution of H+ at interfaces. We have synthesized salicylic acid derivatives to analyze the proton dissociation of both the carboxylic and phenol groups of' the probes, determining spectrophotometrically the apparent pK(a)'s (pK(ap)) in sodium dodecyl Sulfate, SDS, micelles with and without added salt. The synthesized probes were 2-hydroxy-5-(2-trimethylammoniumacetyl)benzoate; 2-hydroxy-5-(2-dimethylhexadecylammoniumacetyl)benzoate- 2-hydroxy-5-(2-dimethylhexadecylammoniumhexanoyl)benzoate-, 2-hydroxy-5-(2-diniethylhexadecylammoniumundecanoyl)betizoate; 2-hydroxy-5-acetylbenzoic acids and 2-hydroxy-5-dodecanoylbenzoic acid. Upon incorporation into SDS micelles the pK(ap)'s of both carboxylic and phenol groups increased by ca. 3 pH units and NaCl addition caused a decrease in the probe-incorporated pKap. The experimental results were fitted with a cell model Poisson-Boltzmann (P-B) equation taking in consideration the effect of salt on the aggregation number of SDS and using the distance of' the dissociating group as a parameter. The conformations of the probes were analyzed theoretically using two dielectric constants, e.g., 2 and 78. Both the P-B analysis and conformation calculations can be interpreted by assuming that the acid groups dissociate very close to, or at, the interface. Our results are consistent with the assumption that the intrinsic pK(a)'s of both carboxylic and phenol groups of the salicylic acid probes used here can be taken as those in water. Using this assumption the micellar and salt effects on the pKap's of the (trialkylammonium)benzoate probes were described accurately using a cell model P-B analysis. (c) 2005 Elsevier B.V. This study evaluated the effects of application of 0.5 µl L-1 1-methyl-cyclopropene (1-MCP), 1000 mg L-1 salicylic acid (SA) and the interaction between the two products at room temperature and pre-exposure at 9±2°C for 24 h in maintaining postharvest quality of lisianthus flowers. After applying the treatment, the flowers were kept in jars with water, stored at 24±2°C. The SA treatments proved ineffective, presenting symptoms of phytotoxicity, with high rate of bent neck and yellowing of petals, both at room temperature and in cold storage, and propitiate the emergence of pathogens. The association between 1-MCP in pre-exposure to 9±2°C for 24 h increased the durability of the stems in six days compared to the control treatment, with less symptoms of bent neck and swelling of stems.

O objetivo deste trabalho foi avaliar as alterações nos níveis de ácido chiquímico e ácido salicílico em plantas de cana-de-açúcar submetidas à aplicação de maturadores. Aplicou-se glyphosate nas doses de 400 e 200 mL ha-1 e na dose de 150 mL ha-1 em mistura com sulfumeturon-methyl a 12 e 20 g ha-1 e sulfumeturon-methyl a 20 g ha-1. As avaliações foram realizadas aos 15 e 30 dias após a aplicação (DAA) e aos 30, 60, 90, 120 e 150 dias após a colheita da cana-de-açúcar. Os teores de ácido chiquímico e salicílico nas plantas de canade-açúcar foram determinados por cromatografia líquida e espectrometria de massas. Os resultados mostraram que as doses de glyphosate correlacionaram-se diretamente com as concentrações de ácido chiquímico na planta, sendo superiores à da testemunha. Aos 30 DAA, houve aumento na concentração de ácido salicílico em todos os tratamentos estudados, revelando um processo de senescência da planta. Maiores doses de glyphosate promoveram aumento na concentração de ácido chiquímico e ácido salicílico antes da colheita da canade-açúcar. No período de crescimento da planta, aumentos nos teores dos ácidos chiquímico e salicílico revelaram dependência da aplicação dos produtos e também dos fatores abióticos e bióticos a que a cultura foi exposta.

Objetivo: Analisar os efeitos das soluções de aspirina e de ácido acético, in vivo, em fígado de coelhos sadios, verificando o efeito histolítico e o resultado anatomo-patológico das lesões e eventuais alterações bioquímicas hepáticas. Métodos: Utilizou-se 80 coelhos, divididos em 2 protocolos experimentais (1 e 2), subdivididos em 5 grupos cada, sendo os mesmos submetidos a laparotomia mediana, com injeção de 0,4 ml da solução de aspirina (2,5 e 5,0%), de ácido acético (2,5 e 5,0%) e solução salina; o sacrifício ocorreu apos 24 horas (protocolo1) e 14 dias (protocolo 2); avaliou-se o peso, evolução clinica, dosagens bioquímicas, cavidade abdominal e torácica e microscopia do fígado. Resultados: Não foram observadas alterações na evolução clinica, peso e nas dosagens bioquímicas, apenas elevação da AST e ALT no grupo 24 horas(Protocolo 1). À macroscópica mostrou que nos animais tratados, em ambos os grupos, a presença de lesão hepática localizada na área infiltrada, correspondente a necrose (24 horas) e fibrose (14 dias). Conclusão: Ambas as soluções (aspirina e ácido acético) acarretaram destruição localizada do órgão substituída por fibrose apos 14 dias.

O bloqueio de eventos dependentes da sinalização do etileno pode afetar de maneira positiva ou negativa a qualidade de frutos tropicais após o armazenamento refrigerado. Dessa forma, os objetivos do presente trabalho foram estudar o envolvimento do etileno no desenvolvimento de injúrias de frio em tangor 'Murcott' e avaliar as respostas envolvidas no processo de resistência às injúrias. Os frutos foram expostos a 500nL L-1 de 1-metilciclopropeno (1-MCP) durante 12 horas ou imersos em soluções contendo 2000nL L-1 de ethephon ou ácido salicílico durante cinco minutos antes de serem armazenados a 1°C, por 90 dias. Como controle, parte dos frutos foi armazenada a 1°C. O tratamento de frutos com ethephon ou ácido salicílico antecipou e intensificou as injúrias de frio. Por outro lado, a inibição do etileno pelo 1-MCP retardou o surgimento dos sintomas e resultou em menor índice de injúrias e percentual de frutos podres ao final do armazenamento. A atividade da superóxido dismutase (SOD) foi intensificada aos 45 dias, contudo em menor intensidade nos frutos tratados com ácido salicílico. Nas avaliações subsequentes, houve decréscimo na atividade da SOD em todos os tratamentos, porém aos 90 dias a intensidade manteve-se levemente superior à observada nos primeiros 30 dias de armazenamento. Os teores de putrescina (Put) e espermina (Spm), no flavedo dos frutos, não sofreram significativa alteração durante o armazenamento. em contrapartida, os teores de espermidina (Spd) foram mais afetados pelo estresse ocasionado pelo frio.

Rhizoctonia foliar blight (RFB) of soybean [Glycine max (L.) Merrill] occurs in many tropical and subtropical regions, causing yield reductions of up to 70% and in Brazil, up to 60%. The disease is caused by Rhizoetonia solani AG1-IA and AG1-IB, and by AG2-3 in Japan. RFB occurs in the North, Northeast and Mid-west regions of Brazil. Chemical control remains the only effective method of controlling RFB, but its efficiency depends upon environmental conditions. In this study, 18 fungicides, salicylic acid (SA) and acibenzolar-s-methyl (ASM) were evaluated on R. solani AG1-IA in vitro, by mycelial growth rating and estimating effective concentration for 50% (EC 50) and 90% (EC 90) inhibition of mycelial growth, and in vivo by reduction of disease severity on soybean plants in greenhouse conditions. Mycelial growth was strongly inhibited by the fungicides pyraclostrobin + boscalid and fludioxonil. Preventive fungicide applications were the most effective. Strobilurins were more efficient both in preventive and curative applications. Best results with plant resistance activators were obtained with SA (2.5 mM) sprayed at 20 d before inoculation and with ASM (12.5 mg a.i. l(-1)) 10 d before inoculation. (c) 2005 Elsevier Ltd. A critical revision of literature as regards to the drug stability in the presence of surfactants were realized. The functional groups envolved in the drug decomposition were used to the development of the discussion. The analysis indicated that the detergent effect can be used to control the rates and mechanisms of drug decomposition and to obtain specific information about the drug reactivity in the environment of pharmacological action.

Venous rethrombosis following thrombectomy is a common event. The aim of the present study was to verify the action of heparin, heparin plus acetyl salicylic acid (ASA) and dipyridamole, and of an arteriovenous fistula (AVF) in the prevention of this complication. Thrombosis was induced in 48 male rabbits by the injection of thrombin in a segment of the left jugular vein, in which the blood flow was arrested for 10 minutes. After 48 hours, the animals were randomly allocated into one of 4 groups of treatment: (1) control, (2) subcutaneous heparin (600 S.I. Units/kg - 8/8 hours), (3) heparin, in the same dose, plus ASA (10 mg/kg/once a day), and dipyridamole (0.5 mg/kg thrice a day), (4) an AVF was surgically constructed between the left carotid after and the left maxillar vein. After 30 minutes, thrombectomy was performed. The venous blood flow, the hematocrit, activated partial thromboplastin time and thrombin time tests were performed before, right after the thrombectomy and 48 hours after thrombectomy. Venography was performed after thrombectomy and at the end of the experiment. The animals were killed 48 hours after thrombectomy and the veins were examined macroscopically. Venous rethrombosis was significantly prevented only in the AVF group (9/12), when compared to control group (0/12), heparin group (1/12) and heparin plus antiaggregating agents group (2/12). These results validate further clinical and experimental investigations with the use of AVF to prevent rethrombosis after venous thrombectomy, when a reduction of venous flow is present.

The 5-aminosalicylic acid is usually prepared: 1. by the reduction of the azodye from diazotized aniline and salicylic acid and the posterior reduction of the formed compound; 2. for carboxylation with carbon dioxide under high pressure of p-aminophenol. Both do not present high yield, but the carboxylation process of p-aminophenol can easily be used and can reduce the obtainment process of 3 steps to only 2. Another advantage of this process is the fact that the product is produced without contaminants, because only one solid reagent (p-aminophenol) and the agent of carboxylation (CO2) is used.

The purpose of this study was to observe the quality of seal of the glass ionomer cement, Ketac-Endo, after treatment of the root canal wall. The root canals of 140 extracted human teeth were prepared biomechanically. The root canals were treated with either EDTA or received an intracanal dressing of calcium hydroxide or camphorated paramonochlorphenol. The root canals were filled by the lateral condensation technique with gutta-percha points and the sealer Ketac-Endo, or zinc oxide-eugenol cement or Sealapex. The teeth were placed into a 2% methylene blue dye solution inside a flask, which was attached to a vacuum pump. Leakage was measured linearly. Sealapex exhibited significantly less leakage than Ketac-Endo or zinc oxide-eugenol cement (P<0.01). The use of EDTA and intermediary dressings reduced significantly (P<0.01) the leakage observed with the zinc oxide-eugenol sealer and Ketac-Endo.

The purpose of this study was to investigate long-term pH changes in cavities prepared in root surface dentin of extracted teeth after obturation of the root canal with gutta-percha and a variety of sealers containing calcium hydroxide. After cleaning and shaping, root canals in 50 recently extracted, human single-rooted teeth were divided into five groups. Each of four groups was obturated with gutta-percha and either Sealapex, Sealer 26, Apexit, or CRCS, all of which contain calcium hydroxide. The remaining group served as the control and was not obturated with gutta-percha or sealer. Cavities were prepared in the facial surface of the roots in the cervical and middle regions. The pH was measured in these dentinal cavities at the initiation of the experiment, and 3, 7, 14, 21, 28, 45, 60, 90, and 120 days after obturation. Results indicate that the pH at the surface of the root does not become alkaline when calcium hydroxide cements are used as root canal sealers. Regardless of the sealer used, the observed pattern of pH change was not different from that seen in the control group of roots that were not treated with sealer. It is concluded that calcium hydroxide-containing cements, although suitable for use as root canal sealants, do not produce an alkaline pH at the root surface. If such a pH change is related to treatment of root resorption, these sealants do not contribute to this treatment. Copyright © 1996 by The American Association of Endodontists.

The calcium hydroxide ionization of four root canal sealers (Sealapex, CRCS, Sealer 26, and Apexit) was studied by measuring conductivity and pH and by conducting atomic absorption spectrophotometry. Samples 6 mm in diameter and 15 mm long were prepared from these sealers. After setting and 48 h storage in a desiccator, five samples of each material were placed in 50 mL distilled water and analysed after 0,1,2,4, 6 and 24 h and 5, 15 and 30 days. The results showed that Sealapex was the root canal sealer showing the highest pH, ionic calcium and total calcium values (P<0.05) throughout the experimental period, followed by CRCS, Apexit and Sealer 26.
