Heart failure (HF) is a complex syndrome that involves changes in behavioral, neural and endocrine regulatory systems. Dietary salt restriction along with pharmacotherapy is considered an essential component in the effective management of symptomatic HF patients. However, it is well recognized that HF patients typically have great difficulty in restricting sodium intake. We hypothesized that under HF altered activity in systems that normally function to regulate body fluid and cardiovascular homeostasis could produce an increased preference for the taste of salt. Therefore, this study was conducted to evaluate the perceived palatability (defined as salt preference) of food with different concentrations of added salt in compensated chronically medicated HF patients and comparable control subjects. Healthy volunteers (n = 25) and medicated, clinically stable HF patients (n = 38, NYHA functional class II or III) were interviewed and given an evaluation to assess their preferences for different amounts of saltiness. Three salt concentrations (0.58, 0.82, and 1.16 g/100 g) of bean soup were presented to the subjects. Salt preference for each concentration was quantified using an adjective scale (unpleasant, fair or delicious). Healthy volunteers preferred the soup with medium salt concentration (p = 0.042), HF patients disliked the low concentration (p < 0.001) and preferred the high concentration of salted bean soup (p < 0.001). When compared to healthy volunteers, HF patients demonstrated a significantly greater preference for the soup with a high salt concentration (p = 0.038). The adaptive capacity of bean (Phaseolus vulgaris L.) calluses (cultivars IAC-carioca, JALO EEP-558, BAT-93 and IAPAR-14) to salt stress (0-80 mM) was verified to determine the existence of biochemical markers such as organic and inorganic compounds, and metabolism of polyamines. The results obtained demonstrate that salt (NaCl) interfered with all the parameters analyzed and its intensity ranged due to the salt concentration and the cultivars used.

The present study evaluated stress indicators in pacu exposed to ammonia in water under the following conditions: without NH4Cl (0.00 g/L); with 0.0078 g NH4Cl/L; and with 0.078 g NH4Cl/L (pH 8.3 and 27 ºC). After the salt dilution the water flow was interrupted and reestablished in 24 hours. Sampling occurred prior to the addition of NH4Cl (control) and after 12, 24 and 48 hours. Glycaemia increased only in fish with the highest salt concentration when compared with group control, regardless of time, and at 24 hours, regardless of treatment. Plasma ammonia, highest in fish exposed to the highest NH4Cl concentration, decreased progressively up to 48 hours. Plasma chloride only decreased in fish not exposed to salt when compared with control and osmolality increased after 24 hours. Hematocrit (Ht), number and volume of erythrocytes and hemoglobin did not change when NH4Cl was added; Ht decrease was reported after 12 hours, but it was not followed by the other blood parameters. The results show tolerance of the pacu to ammonia in the environment.

The aim of this study was to evaluate the growth and survival of pacu, Piaractus mesopotamicus, larvae reared in different salinities and to determine the Artemia nauplii life span in freshwater and in saline water. First feeding 5-d-old pacu larvae were reared in freshwater or at 2, 4, 6, 8, 10, 12, and 14 ppt salinities. The larvae were reared in 1.5-L aquaria at a density of 10 larvae/L with three replicates per treatment. After 10 d of rearing, significant differences (P < 0.05) were observed for growth and survival. Larval growth was higher at 2 and 4 ppt, and survival at 2 ppt was 100%. In freshwater and at 4, 6 and 8 ppt, the survival was 91.1, 93.3, 73.3, and 39.9%, respectively. At higher salinities, there was 100% mortality after 2 h (12 and 14 ppt) and 8 h (10 ppt) of exposure. The slightly saline water of at least 2 ppt increased the Artemia nauplii life span compared to the life span in freshwater. Later, in a second trial, 5-d-old pacu larvae were reared in freshwater and at 2 and 4 ppt salinities during the first 5 or 10 d of active feeding, and then the fish were transferred to freshwater. At the end of 15 d, larval growth was lower in freshwater (42 mg) than in treatments 2 and 4 ppt (5963 mg). The abrupt transfer of fish from freshwater to slightly saline water and the return to freshwater did not affect the survival rates (8997%). The larvae were able to adapt to these saline environments and handle abrupt changes in salt concentration. We concluded that salinity concentration of 2 ppt can be used for pacu larval rearing, allowing the Artemia nauplii lifetime to last longer and cause faster fish growth.

No presente estudo procedeu-se ao isolamento e caracterização da fração globulina majoritária (11 S) de grão-de-bico, var. IAC-Marrocos. A globulina majoritária extraída foi isolada por cromatografia de filtração em gel e de troca-iônica mostrando apenas uma banda de proteína na eletroforese em gel de poliacrilamida. A globulina majoritária, após passagem em coluna de Sephadex, revelou duas bandas protéicas de 55 e 52,5kDa e três bandas menores em gel de poliacrilamida dodecilsulfato de sódio. Na presença de 2-mercaptoetanol 6 polipeptídios na faixa de 18 a 42kDa foram revelados na eletroforese. A globulina isolada foi submetida à ação da tripsina e quimotripsina onde a forma nativa mostrou-se resistente à ação enzimática enquanto o aquecimento (96 e 121°C/15min) não foi suficiente para aumentar a susceptibilidade à hidrólise, significativamente. Adição de NaCl 0,3M levou a um aumento da estabilidade estrutural com menor susceptibilidade à digestão proteolítica, fato em parte perdido com o aquecimento. As hidrólises foram acompanhadas por eletroforese em gel de poliacrilamida dodecilsulfato de sódio.

Objetivou-se nesse estudo avaliar o papel atenuador da espermidina exógena sobre a germinação, vigor de sementes e crescimento de plântulas de cultivares das forrageiras alfafa, guandu e labe-labe submetidas ao estresse salino. A semeadura foi realizada em caixas de plástico tipo gerbox forradas com papel de filtro umedecidos com soluções salinas nas concentrações de 0, 20, 40, 60, 80 e 120 mM de NaCl contendo 0 ou 0,5 mM de espermidina. O delineamento experimental foi inteiramente casualizado em esquema fatorial 3 x 6 x 2 (forrageiras x salinidade x espermidina) com cinco repetições de 25 sementes. As avaliações da germinação foram realizadas no quarto e décimo dias, juntamente com o índice de velocidade de germinação (IVG), avaliado até o nono dia após a semeadura. Determinou-se a concentração salina que reduz em 50% a germinação e a massa seca da parte aérea e raiz no décimo dia após a semeadura. A aplicação de espermidina exógena proporcionou maior acúmulo de massa seca das raízes de labe-labe cv. Rongai, além de ter possibilitado a germinação de 50% das sementes de guandu cv. Caqui até a concentração salina de 72,09 mM de NaCl, assim como maior IVG nas sementes das leguminosas alfafa cv. Crioula e labe-labe cv. Rongai.

Uma das utilizações da técnica de cultura de tecidos para o melhoramento vegetal é a identificação de linhas de células que apresentem tolerância ao estresse salino. Para se estudar os mecanismos bioquímicos envolvidos na expressão genética da tolerância a salinidade, calos oriundos de eixos embrionários de quatro cultivares de feijão (Phaseolus vulgaris L.; cultivares IAC - carioca, IAPAR 14, JALO-EEP 558, BAT - 93), foram cultivados em meio sólido Murashige & Skoog (1962), suplementado com NaCl nas concentrações de 0, 20, 40, 60 e 80 mM. Após 14 dias de incubação, os calos foram coletados e analisados quanto aos padrões isoenzimáticos e de atividade das peroxidases. Os cultivares BAT e IAPAR apresentaram duas zonas de atividade em comum na região anódica e apenas uma zona enzimática específica a cada um deles (migração mais rápida).Possivelmente as duas zonas anódicas intermediárias sejam produtos do mesmo loco enzimático, porém com alelos diferentes, consequentemente diferentes mobilidades eletroforéticas. O cv. JALO apresentou duas zonas anódicas de atividade em comum com os cultivares IAC e IAPAR com uma zona anódica exclusiva de migração mais lenta, a qual apresentou atividade mais intensa de todos os cultivares analisados. Este cultivar revelou ainda uma zona catódica provavelmente dimérica e heterozigota nos indivíduos de todos os tratamentos aplicados. Provavelmente, esta é a mesma zona que ocorre em homozigose com fixação do alelo lento para os indivíduos de todos os tratamentos efetuados nos cultivares BAT e IAPAR. O cv. IAC apresentou duas bandas anódicas em comum com os cv. IAPAR e JALO. Apresentou também a banda anódica mais rápida em comum com o cv. IAPAR e uma banda anódica exclusiva de migração mais lenta. Curiosamente, os indivíduos deste cv. mantidos em meio suplementado com 20 mM de NaCl não apresentaram atividade nas três zonas anódicas mais lentas. Ocorreu no cv. IAC uma única zona de atividade catódica, dimérica e heterozigota para os indivíduos provenientes de todos os tratamentos, composta provavelmente de dois alelos diferentes da zona correspondente ao cv. JALO. Amostras provenientes dos tratamentos 40 e 60 mM de NaCl, desta zona catódica, apresentaram maior atividade enzimática. A análise da atividade da peroxidase no extrato bruto, revelou que os cultivares responderam diferentemente ao aumento da concentração salina no meio de cultura, com aumento pronunciado dessa atividade nos cultivares IAC e JALO.

We used dynamic light scattering (DLS), a steady-state fluorescence, time resolved fluorescence quenching (TRFQ), tensiometry, conductimetry, and isothermal titration calorimetry (ITC) to investigate the self-assembly of the cationic surfactant cetyltrimethylammonium sulfate (CTAS) in aqueous solution, which has SO42- as divalent counterion. We obtained the critical micelled concentration (cmc), aggregation number (N-agg), area per monomer (a(0)), hydrodynamic radius (R-H), and degree of counterion dissociation (alpha) of CTAS micelles in the absence and presence of up to 1 M Na2SO4 and at temperatures of 25 and 40 degrees C. Between 0.01 and 0.3 M salt the hydrodynamic radius of CTAS micelle R-H approximate to 16 angstrom is roughly independent on Na2SO4 concentration; below and above this concentration range R-H increases steeply with the salt concentration, indicating micelle structure transition, from spherical to rod-like structures. R-H increases only slightly as temperature increases from 25 to 40 degrees C, and the cmc decreases initially very steeply with Na2SO4 concentration up to about 10 mM, and thereafter it is constant. The area per surfactant at the water/air interface, a(0), initially increases steeply with Na2SO4 concentration, and then decrases above ca. 10 mM. Conductimetry gives alpha = 0.18 for the degree of counterion dissociation, and N-agg obtained by fluorescence methods increases with surfactant concentration but it is roughly independent of up to 80 mM salt. The ITC data yield cmc of 0.22 mM in water, and the calculated enthalpy change of micelle formation, Delta H-mic = 3.8 kJ mol(-1), Gibbs free energy of micellization of surfactant molecules, Delta G(mic) = -38.0 kJ mol(-1) and entropy T Delta S-mic = 41.7 kJ mol(-1) indicate that the formation of CTAS micelles is entropy-driven. Charqui meats were prepared in laboratory conditions in order to carry out experiments to observe the possibility of development of enterotoxigenic Staphylococcus aureus and Clostridium bottilinum proteolytic type B spores and their toxins. Results demonstrated that the harsh processing conditions, high salt concentration, relative high temperature, a, values, inhibited the growth of both bacteria. Under our experimental conditions, S. aureus would survive throughout the sequence of salting steps i.e. brine followed by rock salting and the sunshine drying step. However, at final a(w) value of 0.70-0.75 would create conditions to inhibit its development. The other experiment revealed that C. botulinum spores germination also was impaired because of these low a(w) values. Under these conditions, charqui meats revealed to be safe products in relation to toxins from both enterotoxigenic S. aureus and C. botulinum. (C) 2003 Elsevier B.V. Ltd. All rights reserved.

Uniform metal iron ellipsoidal particles of around 200 nm in length were obtained by reduction and passivation of alumina-coated alpha-Fe2O3 (hematite) particles under different conditions of temperature and hydrogen flow rate. The monodispersed hematite particles were prepared by the controlled hydrolysis of ferric sulfate and further coated with a homogeneous thin layer of Al2O3 by careful selection of the experimental conditions, mainly pH and aluminum salt concentration. The reduction mechanism of alpha-Fe2O3 into alpha-Fe was followed by x-ray and electron diffraction, and also by the measurements of the irreversible magnetic susceptibility. The transformation was found to be topotactic with the [001] direction of hematite particles, which lies along the long axis of the particles, becoming the [111] direction of magnetite and finally the [111] direction of metal iron. Temperature and hydrogen flow rate during the reduction have been found to be important parameters, which determine not only the degree of reduction but also the crystallite size of the final particles. Magnetic characterization of the samples shows that the only parameters affected by the crystallite size are the saturation magnetization and magnetic time-dependence effect, i.e., activation volume. In this work the sol-gel process was used to prepare SnO2 supported membranes with an average pore size of 2.5 nm. The effects of salt concentration (NaCl or CaCl2) and of the pH of the aqueous solutions used on the flux and selectivity through the SnO2 membrane were analyzed by permeation experiments and the results interpreted taking account of the zeta potential values determined from the electrophoretic mobility of the SnO2 powder aqueous dispersion. The results show that the ion flux (Na+, Ca2+ and Cl-) throughout the membrane is determined by the electrostatic repulsion among these species and the surface charge at the tin oxide-solution interface.

Infrared and photoluminescence spectroscopies have been used to investigate the local environment of the Eu3+ ions in luminescent sol-gel derived materials-di-ureasils-based on a hybrid framework represented by U(600). This host is composed of a siliceous backbone grafted, through urea cross-links, to both ends of polymer segments incorporating 8.5 oxyethylene repeat units. The active centers have been introduced as europium perchlorate, Eu(ClO4)3. Samples with compositions n = 232, 62, 23, 12, and 6 (where n denotes the ratio of (OCH2CH2) moieties per lanthanide ion) have been examined. The combination of the information retrieved from the analysis of characteristic bands of the FTIR spectra-the perchlorate and the Amide I/Amide II features-with that obtained from the photoluminescence data demonstrates that at compositions n = 232 and 62 the anions are free, whereas the Eu3+ ions are complexed by the heteroatoms of the polyether chains. At higher salt concentration, the cations are bonded, not only to the ClO4 - ions, but also to the ether oxygen atoms of the organic segments and to the carbonyl oxygen atoms of the urea linkages. The dual behavior of U(600) with respect to cation coordination has been attributed to the presence in this nanohybrid of strong hydrogen-bonded urea-urea structures, which, at low salt content, cannot be disrupted, thus inhibiting the formation of Eu3+-O=C(urea) contacts and promoting the interaction between the lanthanide ions and the (OCH2CH2) moieties. The present work substantiates the claim that the activation of the coordinating sites of the di-ureasil framework can be tuned by varying either the guest salt concentration at constant chain length or the length of the.organic segments at constant salt concentration. This relevant property opens challenging new prospects in the fields of application of this class of hybrids. © 2001 American Chemical Society.

The objectives of this work were to characterize pineapple plants (Ananas comosus L. Merril) cv. Smooth Cayenne, cultured in vitro, in saline medium, in relation to bromelian activity, identifying the parts of the plant with the highest bromelian activity. Also under aim was the study of the influence of saline stress on the enzyme activity. Axillary buds of pineapple were cultivated in vitro in MS medium, supplemented with 2 mg.L-1 BAP and 1 mg.L-1 NAA. The levels of salinity tested were: 0.57 g.L-1 NaCl, 1.15 g.L-1 NaCl, and 2.30 g.L-1 NaCl. Bromelian activity was evaluated in the development of buds, shoots, and roots. The results showed that bromelian activity was higher in buds at the highest salt concentration at 15 days. Cultured shoots showed bromelian activity decreasing in the saline treatments in all the collection, up to 60 days in culture. The roots showed higher bromelian activity in the roots in saline medium.

The effects of salt concentration levels in electrical conductivity (EC) were evaluated in chrysanthemum root, cultivated in substrate using two sampling methods, under greenhouse conditions. The experiment was carried out in Paranapanema, São Paulo using the experimental design in randomized blocks and four replications. The treatments consisted of eight sampling periods of substrate solutions in pots: 7, 14, 21, 28, 35, 42, 49 and 56 days after strike root and five salt concentration levels of applied saline solution: 1.42; 1.65; 1.89; 2.13 and 2.36 dS m -1 in the vegetative period and during the reproduction period of flower budding: 1.71; 1.97; 2.28; 2.57 and 2.85 dS m -1. The substrate solution EC monitoring was done using two methods: solution extractors and 1:2 water diluted solution. The use of solution extractors and 1:2 water diluted solution allowed substrate solution EC monitoring along the culture cycle; the amount of salt concentration applied in the substrate caused the substrate salinity increase; the method using solution extractors presented higher EC values in the substrate.
