We report the results of a transcript finishing initiative, undertaken for the purpose of identifying and characterizing novel human transcripts, in which RT-PCR was used to bridge gaps between paired EST Clusters, mapped against the genomic sequence. Each pair of EST Clusters selected for experimental validation was designated a transcript finishing unit (TFU). A total of 489 TFUs were selected for validation, and an overall efficiency of 43.1% was achieved. We generated a total of 59,975 bp of transcribed sequences organized into 432 exons, contributing to the definition of the structure of 211 human transcripts. The structure of several transcripts reported here was confirmed during the course of this project, through the generation of their corresponding full-length cDNA sequences. Nevertheless, for 21% of the validated TFUs, a full-length cDNA sequence is not yet available in public databases, and the structure of 69.2% of these TFUs was not correctly predicted by computer programs. The TF strategy provides a significant contribution to the definition of the complete catalog of human genes and transcripts, because it appears to be particularly useful for identification of low abundance transcripts expressed in a restricted Set of tissues as well as for the delineation of gene boundaries and alternatively spliced isoforms.

DNA barcoding facilitates the identification of species and the estimation of biodiversity by using nucleotide sequences, usually from the mitochondrial genome. Most studies accomplish this task by using the gene encoding cytochrome oxidase subunit I (COI; Entrez COX1). Within this barcoding framework, many taxonomic initiatives exist, such as those specializing in fishes, birds, mammals, and fungi. Other efforts center on regions, such as the Arctic, or on other topics, such as health. DNA barcoding initiatives exist for all groups of vertebrates except for amphibians and nonavian reptiles. We announce the formation of Cold Code, the international initiative to DNA barcode all species of these 'cold-blooded' vertebrates. The project has a Steering Committee, Coordinators, and a home page. To facilitate Cold Code, the Kunming Institute of Zoology, Chinese Academy of Sciences will sequence COI for the first 10 specimens of a species at no cost to the steward of the tissues. © 2012 Blackwell Publishing Ltd.

It has become evident that policies aimed at mitigating the growing water resources and water use conflicts in Brazil are crucial. The municipality of Extrema in Minas Gerais state in Brazil pioneered the first Brazilian municipal PES initiative (Conservador das Aguas program), based on the relationship between forests and the benefits they provide. This study aimed to assess soil loss in the Posses sub-basin, where the Conservador das Aguas program began. Additionally, we aimed to determine the potential that this PES initiative has for soil conservation, as well as to minimize the soil losses as a function of forest area size and location in order to propose a technical approach for implementing PES. In this sense, considering the prescribed conservation practices, land use situation, and soil cover in the Posses sub-basin, we analyzed the effectiveness of the Conservador das Aguas program before and after implementation in relation to reduced soil loss under 36 different land use and soil cover scenarios. We used a geographic information system (GIS) for spatializing and producing different information plans and the Revised Universal Soil Loss Equation (RUSLE) for estimating soil loss. As a result, we found that minimization of soil loss may be obtained by adopting pasture conservation practices. Additionally the expected average soil loss in the Posses sub-basin under conditions of land use and soil cover, before and after implementing the water conservation program was 30.63 and 7.06 Mg ha(-1) year(-1), respectively. This paper is the result of discussions held during the initial research on ways of reading and reading activities, to obtain the title of the Ph.D. graduate program in Education at UNESP - Marilia. The paper seeks to rethink the practice followed the reality historically constructed as reference, creating a dialectic movement, and a constant construction, and the overcoming of it. In this perspective, we approach possible practices that can enhance the student's prior knowledge, and from it, to propose ways of action that lead to the development of the subject through the processes of teaching and learning of reading in school. From the study of actions that are agreed as reading practices, the study proposes to work with the Reading Strategies to promote the development of the individual. Among the different results that have been announced in the research, we highlight the need for conscious mediation of the teacher in the act of teaching reading and conscious participation of the student in the process. Partial results give evidence to confirm the hypothesis that: intentional actions are driving the learning of children in activity in the classroom, considering their experiences and choices of the teacher as mediator in the process. Students in third grade of elementary school are the subjects of research, theory and practice. The researcher is the mediator and proposer of the actions based on microgenetic research.
