The anatomy and ultrastructure of root nodules of Anadenanthera peregrina var. falcata (Leguminosae-Mimosoideae) were analysed, as was plant growth. To ensure that nodules developed, seedlings were inoculated with a mixture of six strains of rhizobia. Nodules were produced that differed in appearance-and probably also effectiveness-but their structure was similar and they showed characteristics typical of indeterminate nodules, such as persistent meristematic tissue and a gradient of cells at different stages of development. Many starch grains were present in inner cortex cells and interstitial cells of infected tissue. Infected cells were densely packed with bacteroids, which contained many poly-beta-hydroxybutyrate granules. The high incidence of these granules, together with high levels of starch accumulation in interstitial cells, suggested low N-2-fixation efficiency of the rhizobia isolates used for inoculation. In the symbiosomes of early-senescent infected cells, reticulum-like structures, small vesicles and a fibrillar material were observed; these may be related to bacteroid degradation. In the cytoplasm of late-senescent infected cells, many vesicles and membrane-like structures were observed, probably associated with membrane degradation of bacteroids and peribacteroids. The total biomass of plants inoculated with rhizobia was low and their xylopodia and shoots had low levels of N compared with non-inoculated plants fertilized with ammonium nitrate. However, inoculated plants did not show N-deficiency symptoms and grew better than non-inoculated plants without N fertilization. These growth results, together with ultrastructural observations of nodules, suggest that nitrogen fixation of rhizobia isolates associated with Anadenanthera peregrina var. falcata roots is poor. The effects of anaerobic digestion and initial pH on the bioleaching of metals from sewage sludge were investigated in shake flask experiments. A strain of Acidithiobacillus thiooxidans was employed in the assays using secondary and anaerobic sludges, which resulted in similar solubilization yields of the metals chromium, copper, lead, nickel, and zinc for both the sludges investigated. The effect of initial pH (7.0 and 4.0) on metal bioleaching was assayed by using the anaerobic sludge inoculated with indigenous sulfur-oxidizing thiobacilli. Although the time required to reach the end of the experiment (final pH close to 1.0) was shortened at initial pH of 4.0, final metal solubilization was not significantly different for both initial pH values, resulting in higher solubilization yields for copper, nickel, and zinc (higher than 80%). Chromium and lead presented solubilization yields close to 50%. The results obtained in this work showed that the metal bioleaching process can be applied to sewage sludge regardless of the type of sludge and without the requirement of pH adjustment.

A new platinum(II) complex with deoxyalliin was synthesized and characterized by chemical and spectroscopic techniques. Elemental and mass spectrometry analyses of the solid complex fit to the composition [Pt(C6H11NO2S)Cl-2] center dot H2O. C-13 NMR, N-15 NMR and infrared spectra of the complex are consistent with coordination of deoxyalliin to Pt( II) through the nitrogen and sulfur atoms forming a square-planar geometry. The complex is soluble in dimethylsulfoxide. Biological analysis for evaluation of a potential cytotoxic effect of the complex was performed using HeLa cells, a human cervix adenocarcinoma-derived cell line. The results were compared with those of a palladium( II) complex previously described.

Introduction. Rambutan is a tropical fruit species with recalcitrant seeds. Despite the expansion of exotic fruit cultivation in Brazil, lots of which fruit species, including rambutan, need basic information, especially in relation to propagation and storage of seeds, which are important for genetic improvement studies, maintenance of genetic sources and seedling production. Materials and methods. A completely randomized design was adopted with treatments distributed in a factorial arrangement, 3 x 4, referring to three seed storage conditions [room temperature conditions; a dry chamber with (18 +/- 2) degrees C and 60% relative humidity; and a cold chamber with (10 +/- 2) degrees C and 70% relative humidity] and four storage times ( 0, 7, 14 and 21 d). Each treatment of 10 seeds was replicated five times. Data on seedling emergence, emergence rate, plant height, number of leaves and length of main root were submitted to variance analysis and means were separated using Tukey's test. Correlation analysis between seed moisture and seedling emergence was performed. Results and discussion. Our results indicated that dry chamber conditions promoted the statistically significantly highest seedling emergence after 7 d of storage. Cold chamber conditions promoted an extremely low seedling emergence independently of time. Conclusion. Rambutan seeds can be stored in a dry chamber for 7 d without losing viability; after 14 d of storage the loss of emergence is 60%.

Dictyosperma album, commonly known as Princess Palm, due to its graceful appearance and its ability to withstand strong hurricane force winds is largely used in landscaping areas. The aim of this work was to describe the diaspores (seeds with the stuck endocarp) morphology and the stages of the D. album germination process. Therefore we concluded that the species germination is of the tubular remote type and begins with the opening of a circular operculum in the fruit endocarp, through which emerges a bulbous and hollow structure, named cotyledonal petiole or embryophore. With the embryo growth, the endosperm is being gradually consumed. The embryo grows approximately up to 10 cm, when the dilation begins in its extremity. Starting from this extensive area, it begins the growth of the primary root and the opening of a longitudinal rift, through which emerges the aerial part that is composed by two sheaths that cover the first juvenile leaf. In this phase, the emergence of secondary roots is observed. The first leaves are simple and lance-shaped.

Realizaram-se dois experimentos para avaliar a eficiência da bohemina e roscovitina associadas à ionomicina para ativação partenogenética e desenvolvimento embrionário inicial de bovinos. No primeiro, foram testadas diferentes concentrações (0, 50, 75 ou 100µM) e diferentes tempos de exposição (2, 4 ou 6 horas) à bohemina ou à roscovitina na ativação de oócitos bovinos maturados in vitro (MIV) pré-expostos à ionomicina. Os melhores tratamentos, bohemina 75µM e roscovitina 50µM, ambos por seis horas, foram utilizados no segundo experimento, no qual oócitos bovinos MIV foram expostos à ionomicina seguido ou não pelo tratamento com inibidores específicos das quinases dependentes de ciclina (CDKI), e avaliados quanto à configuração nuclear, taxa de ativação e desenvolvimento até blastocisto. Os tratamentos combinados (ionomicina+CDKI) apresentaram melhor taxa de ativação (77,3%) e desenvolvimento embrionário inicial (35,2%) do que a ionomicina sozinha (69,4% e 21,9%, respectivamente), e também promoveram ativação mais uniforme (aproximadamente 90% de formação de um pronúcleo). Estes resultados demonstram que os CDKIs potencializam o efeito da ionomicina na ativação e desenvolvimento embrionário inicial e podem auxiliar na obtenção de protocolos de ativação mais eficientes, aumentando a capacidade de desenvolvimento de embriões produzidos por meio de biotécnicas reprodutivas.
