Avaliou-se o comportamento pós-colheita de pêssegos da cv. Aurora-1 armazenados sob refrigeração. Os frutos foram colhidos em dois estádios de maturação, verde maduro (de vez) e maduro. Os lotes foram armazenados em três temperaturas (2°C; 6°C e 12°C), por 35 dias, e avaliados a cada sete dias: quanto à coloração da casca, perda acumulada de massa fresca (PMF), firmeza (FIR), aparência, teores de acidez titulável (AT), sólidos solúveis (SS), açúcares solúveis (AS) e redutores (AR), pectina solúvel (PS) e total (PT), além da porcentagem de solubilização de pectinas (SOL). A menor temperatura de armazenamento elevou o tempo de prateleira dos pêssegos, e os frutos de vez apresentaram melhor aparência. A PMF demonstrou um gradiente em função do aumento da temperatura, e os frutos de vez apresentaram menor perda ao final do armazenamento sob todas as temperaturas, quando comparados aos maduros. A coloração da casca dos frutos de vez, a 2°C, teve pouca alteração, conferindo-lhes mudança de coloração de verde-amarelada para amarelo-clara; enquanto nas temperaturas de 6°C e 12°C esse gradiente foi mais intenso. O mesmo efeito foi verificado nos pêssegos maduros. A FIR sofreu efeito da temperatura, pois temperaturas menores sofreram redução mais lenta e AT dos pêssegos maduros foi superior aos de vez. Não houve influência dos tratamentos nos teores de SS, AS e AR. Os pêssegos 'Aurora-1' não demonstraram sensibilidade ao frio, e os de vez, armazenados a 2°C, tiveram vida útil de 35 dias.

A fim de promover a conservação pós-colheita de frutas e hortaliças, as embalagens biodegradáveis à base de amido, pectinas, celulose e outros polímeros, revelam resultados variáveis a depender dos vegetais tratados. Assim, o presente trabalho teve como objetivo avaliar o efeito de biofilme à base de fécula de mandioca na conservação dos frutos de pimentão 'Magali R', armazenados à temperatura ambiente e sob refrigeração. Os frutos foram selecionados, lavados, desinfetados, revestidos com biofilme de fécula de mandioca nas concentrações de 3%, 4% e 5% e armazenados por até 20 dias a 24,5 ± 3 ºC e 60,5 ± 12% UR e a 10 ± 1 ºC e 90 ± 5% UR. Os efeitos dos tratamentos foram avaliados por meio da perda de massa, da firmeza do fruto e do teor de sólidos solúveis, em delineamento inteiramente casualizado, com três repetições e dois frutos por parcela, no esquema fatorial 4 x 6 x 2. Pelos resultados, verifica-se que o uso de biofilme de fécula de mandioca nas concentrações utilizadas não foi eficiente em retardar o metabolismo pós-colheita e prolongar a conservação de pimentões 'Magali R' refrigerados ou não; o uso de refrigeração a 10 ± 1 ºC e 90 ± 5% UR, sem associação com biofilme, foi eficiente em manter os pimentões Magali R com perda de massa inferior aos 15%, estabelecido como limite para a vida útil, por até 20 dias de armazenamento, enquanto em temperatura ambiente a vida útil foi de apenas por oito dias.

Seventy-five fungal strains from different groups of basidiomycetes, newly isolated from rotten wood, were screened for pectinolytic activity. Despite the fact that basidiomycetes are scarcely referred to as pectinase producers, the polygalacturonase (PG) activity was detected in 76 % of the strains; 16 % with activity higher than 40 nkat/g, 40 % between 13.3 and 40 nkat/g, and 44 % with activity lower than 13.3 nkat/g. The highest productions were obtained among the fungi from order Aphyllophorales, family Polyporaceae. The characterization of the enzymes from the highest PG producers (Lentinus sp., Gloeophyllum striatum, Pycnoporus sanguineus, Schizophyllum commune) showed optimum temperature for catalytic activity at 60-70°C and two peaks of pH optimum (3.5-4.5 and 8.5-9.5). The enzymes exhibited high pH stability (3.0-11.0) but after incubation at 40°C for 1 h their activity dropped by 18-73 %.

Polymers blends represent an important approach to obtain materials with modulated properties to reach different and desired properties in designing drug delivery systems in order to fulfill therapeutic needs. The aim of this work was to evaluate the influence of drug loading and polymer ratio on the physicochemical properties of microparticles of cross-linked high amylose starch-pectin blends loaded with diclofenac for further application in controlled drug delivery systems. Thermal analysis and X-ray diffractograms evidenced the occurrence of drug-polymer interactions and the former pointed also to an increase in thermal stability due to drug loading. The rheological properties demonstrated that drug loading resulted in formation of weaker gels while the increase of pectin ratio contributes to origin stronger structures. © 2012 Elsevier Ltd.

Polymers mixtures as well as cross-linking reactions are approaches that have been used successfully to modulate the polymers characteristics in order to improve the control over drug release rate. High amylose and pectin are polysaccharides frequently used to prepare drug delivery systems. Since the drying technique can strongly influence the properties of such systems, the aim of this work was to characterize high amylose/pectin mixtures cross-linked with sodium trimetaphosphate and dried by different techniques-oven and lyophilization. The results showed that samples dried by lyophilization presented reduced particle size, higher porosity and higher swelling ability than the samples dried in oven. Besides, lower thermal stability and different diffraction patterns showed by the former particles should reflect the structural changes as a function of drying technique. © 2013 Informa Healthcare USA, Inc.

The present work evaluated the effect of strobilurins, boscalid, plant growth regulators and vegetal extracts on the physico-chemical quality of tomato fruits (Solanum lycopersicum L.), hybrid Giuliana. The fruits from each treatment were selected and separated in 4 repetitions: control, pyraclostrobin, boscalid, pyraclostrobin + boscalid, IBA + GA(3) + kinetin, GA(4+7) + benzylaminopurine and vegetal extract. The first application was carried out at 30 days after transplant and the following at every 15 days. The evaluations were: weight loss, titratable acidity (TA), soluble solids (SS), SS/TA relation, pH, ascorbic acid content, texture, total soluble sugars (TSS), activity of pectin methyl esterase (PME) and polygalacturonase (PG). The SS content, responsible for fruit flavor, varied accordingly to the treatment, being highest for GA(4+7) + benzylaminopurine and lowest for the control. The same occurred regarding the SS/AT relation. The fruits were kept on the shelf for 9 days, at room temperature, being that at the end of this period several treatments still presented fruit in optimal consumption conditions, notably the boscalid treatment, which presented lowest weight loss, followed by pyraclostrobin. Therefore, it can be concluded that the application of the treatments did not modify the values for pH, AT and AST of the fruits. The highest PME activity was observed for the treatments with boscalid and the mixture of boscalid and pyraclostrobin, while the lowest PG activity occurred in the control and the pyraclostrobin treatment, indicating that some products accelerated the process of demethylation of pectins by PME, facilitating the action of PG.

A pectin lyase, named PLIII, was purified to homogeneity from the culture filtrate of Aspergillus giganteus grown in submerged culture containing orange peel waste as carbon source. PLIII was able to digest apple pectin and citrus pectins with different degrees of methyl esterification. Interestingly, the PLIII activity was stimulated in the presence of some divalent cations including Pb(2+) and was not significantly affected by Hg(2+). Like other pectin lyases, PLIII is stimulated by but is not dependent on Ca(2+). The main soluble product released during the degradation of pectic substances promoted by the PLIII is compatible with an unsaturated monogalacturonate. PLIII is a unique enzyme able to release unsaturated monogalacturonate as the only soluble product during the degradation of pectic substances; therefore, PLIII was classified as an exo-pectin lyase. To our knowledge, this is the first characterization of an exo-pectin lyase. The PLIII described in this work is potentially useful for ethanol production from pectin-rich biomass, besides other common applications for alkaline pectinases like preparation of textile fibers, coffee and tea fermentation, vegetable oil extraction, and the treatment of pulp in papermaking.

In the postharvest management, the fruits can be exposed to injuries that depreciated the quality and the shelf life. Thus, it was evaluated the modified atmosphere effects on guavas var. Paluma subjected to different mechanical damages. Once harvested, the fruits were selected, sanitized and submitted to the treatments T1 (control) - without injuries or packaging in bags of low density polyethylene (LDPE); T2 - without injuries + LDPE bags; T3 - damage by fall of 1 m + LDPE bags; T4 - damage by compression of 9 N + LDPE bags; T5 - damage by fall of 1 m + damage by compression of 9 N + LDPE bags and T6 - damage by fall of 1 m + damage by compression of 9 N without LDPE bags. The treatments were kept in cold storage at 10 ± 1 o C and 94 ± 2% de R.H. The analysis of CO2/ethylene production, enzymatic activity, total and soluble pectins, pulp firmness, titratable acidity (TA), soluble solids (SS), reducing sugars and ascorbic acid were performed every 10 days of refrigeration, and an additional day outside cold storage (22 ± 1o C and 75 ± 3% R.H.) for 30 days. Guavas packed in LDPE bags, not subject to mechanical damage, presented the best quality standards. The fruits suffered only one kind of damage, when packaged, presented satisfactory pattern compared to the fruits without package and not exposed to any mechanical damages. Applying the two kinds of damages, the LDPE packaging was not adequate to decrease the metabolic rate of these fruits, making them unfit for marketing.
