A biomimetic sensor is proposed as a promising new analytical method for determination of norfloxacin (NF) in pharmaceuticals. The sensor was prepared by modifying a glassy carbon electrode surface with a Nafion® membrane doped with poly(copper phthalocyanine) complex [poly-CuPc]. Amperometric measurements carried out with the sensor under an applied potential of -0.05 V vs Ag|AgCl in 0.1 mol L-1 acetic acid containing 1.5 × 10-3 mol L-1 hydrogen peroxide showed a linear response range from 2.0 × 10-4 to 1.2 × 10-3 mol L-1. Selectivity and interference studies were also performed. A sensor response mechanism is proposed, based on the experimental evidence. Recovery studies were carried out using environmental samples, in order to evaluate the sensor’s potential for use with these sample classes. Finally, sensor performance was evaluated using analyses of commercial formulations.

A stability-indication high performance liquid chromatographic method has been developed for the determination of norfloxacin in tablet dosage forms. Optimum separation was achieved in less than 7 minutes using Eclipse Plus Zorbax C18 Agilent, 150 mm×4.6 mm i.d., 5 μm particle size column. The analyte was resolved by using a mobile phase 5% acetic acid aqueous solution and methanol (80:20, v/v) at a flow rate 1.0 ml/min on an isocratic high performance liquid chromatographic system at a wavelength of 277 nm. Linearity, system suitability, precision, sensitivity, selectivity, specific, and robustness were established by International Conference Harmonization guidelines. For stress studies the drug was subjected to photolysis, oxidation, acid, alkaline and neutral conditions. The analytical conditions and the solvent developed provided good resolution within a short analysis time and economic advantages. The proposed method not required sophisticated and expensive instrumentation.

The aim of this study was to determine the antimicrobial resistance patterns of Salmonella strains isolated from slaughter-age pigs and environmental samples collected at modern swine raising facilities in Brazil. Seventeen isolates of six serotypes of Salmonella enterica subsp. enterica were isolated out of 1,026 collected samples: Salmonella Typhimurium (1), Salmonella Agona (5), Salmonella Sandiego (5), Salmonella Rissen (1), Salmonella Senftenberg (4), and Salmonella Javiana (1). Resistance patterns were determined to extended-spectrum penicillin (ampicillin), broad-spectrum cephalosporins (cefotaxime and ceftriaxone), aminoglycosides (streptomycin, neomycin, gentamicin, amikacin, and tobramycin), narrow-spectrum quinolone (nalidixic acid), broad-spectrum quinolone (ciprofloxacin and norfloxacin), tetracycline, trimethoprim, and chloramphenicol. Antimicrobial resistance patterns varied among serotypes, but isolates from a single serotype consistently showed the same resistance profile. All isolates were resistant to tetracycline, streptomycin, and nalidixic acid. One isolate, Salmonella Rissen, was also resistant to cefotaxime and tobramycin. All serotypes were susceptible to ceftriaxone, norfloxacin, ciprofloxacin, ampicillin, gentamicin, and chloramphenicol. The high resistance to tetracycline and streptomycin may be linked to their common use as therapeutic drugs on the tested farms. No relation was seen between nalidixic acid and fluoroquinolone resistance.

A descontaminação dos explantes é um dos princípios básicos para o sucesso da cultura de tecidos. Um dos problemas diagnosticados na propagação in vitro da figueira, através de gemas apicais, é a contaminação endógena dos explantes por bactérias. Este trabalho teve como objetivo avaliar a eficiência de alguns antibióticos em meio de cultura para o controle de bactérias endógenas em gemas apicais de figueira. Foram avaliados os seguintes tratamentos: T1(sem adição de antibiótico); T2 (30 mg L-1 de cloranfenicol); T3 (250 mg L-1 de ampicilina sódica); T4 (500 mg L-1 de ácido nalidícico); T5 (150 mg L-1 de cefalotina sódica); T6 (500 mg L-1 de tetraciclina), e T7 (400 mg L-1 de norfloxacina). Após coletados em campo, os segmentos de ramos contendo as gemas foram colocados em recipiente com água corrente. Posteriormente, as gemas apicais foram imersas em álcool etílico a 70% e hipoclorito de sódio a 2,5%. Todo procedimento de desinfestação externa dos explantes foi realizado em câmara de fluxo laminar. Os explantes foram inoculados em tubos de ensaio contendo 15 mL de meio básico MS suplementado, após a autoclavagem, com as doses de antibióticos de acordo com os tratamentos estabelecidos. Após a inoculação, os explantes foram mantidos em sala de crescimento por quatro dias no escuro e, em seguida, sob fotoperíodo de 16 horas de luz branca fria e irradiância de 25 µmol m s-1, na temperatura de 22 ± 3ºC. A assepsia realizada externamente nos explantes foi suficiente para o controle de contaminação fúngica, e a adição de antibióticos ao meio, após autoclavagem, foi eficiente para o controle de bactérias endógenas, cujo antibiótico ampicilina sódica proporcionou mais de 90% de explantes sobreviventes.

Foram caracterizados os sorotipos, o perfil de sensibilidade microbiana e os achados clínico-epidemiológicos em 53 linhagens do gênero Salmonella isoladas de 41 cães, nove equinos e três bovinos, acometidos por diferentes manifestações clínicas entre 1997 e 2007. Salmonella Typhimurium (45,3%), Salmonella enterica (22,6%), Salmonella Enteritidis (7,5%), Salmonella enterica subsp enterica 4,5,12i (5,7%), Salmonella Newport (5,7%), Salmonella Dublin (3,8%), Salmonella Agona (3,8%), Salmonella Glostrup (3,8%), Salmonella Saintpaul (1,8%) foram os sorotipos encontrados. Ciprofloxacina (100,0%), norfloxacina (100,0%) e gentamicina (100,0%) foram os antimicrobianos mais efetivos, enquanto a maior resistência das linhagens foi observada para ceftiofur (28,5%) e florfenicol (7,0%). As linhagens foram isoladas de animais com enterite, infecção do trato urinário, septicemia, piometra, pneumonia e conjuntivite. Ressalta-se para o predomínio do sorovar Typhimurium nas diferentes manifestações da salmonelose nos animais. Destaca-se, também, a identificação de sorotipos nos animais que também são observados em casos de salmonelose em humanos

Susceptibility profile and multiple drug resistance in 158 E. coli strains isolated from urinary tract infection (UTI), pyometra, and feces of dogs were studied. Norfloxacin, ciprofloxacin, and enrofloxacin were the most-effective drugs (>60%) for E. coli strains. High rates of resistance to antimicrobials were observed in 60% or more of the isolated strains using sulfametoxazole/trimetoprim. Multiple drug resistance for three or more antimicrobials was observed in two (47.1%) strains isolated from UTI, seven (13.5%) from pyometra, and four (7.3%) from feces. From these, 17 (33.3%), one (1.9%), and three (5.5%), respectively, showed multiple resistance to five or more drugs.

Routine diagnosis methods used in bovine mastitis were studied in 55 mares in lactation. The findings of strip cup test, California Mastitis Test-CMT, electronic somatic cell count-CCS, microbiological culture, and in vitro antimicrobial susceptibility profile of isolates were discussed. Streptococcus spp., Staphylococcus spp, and enterobacteria were the most common microorganisms isolated in health and CMT-positive mammary glands. Staphylococcus aureus and Arcanobacterium pyogenes were identified in two mares presenting clinical mastitis. Mean somatic cell count of eight mares without presence of microorganisms in milk was 247.57x10³/mL and 1.621,86x10³/mL in 47 mares with positive microbiological culture. Moderate concordance (63.8%) between positive reactions in CMT (1 to 3+) and microbiological culture was observed. Amicacin (78.9%), ceftiofur (74.7%), sulpha-trimetoprim (69,0%) and norfloxacin (69.0%), were the most effective drugs, while resistance of isolates was mainly observed against penicillin (64.8%), gentamycin (35.2%), azithromycin (35.2%), enrofloxacin (28.2%), and florfenicol (28.2%).

A retrospective study of etiology, antimicrobial susceptibility profile and multiple drug resistance, and major epidemiological aspects were investigated in 616 cases of canine otitis. Staphylococcus beta hemolitic (26.27%), Malassezia pachydermatis (12.35%), and Pseudomonas aeruginosa (8.8%) were the most common microorganisms identified. The isolates were susceptible mainly to norfloxacin (89.62%), gentamicin (83.25%), and ofloxacin (80.16%). High occurrence of resistance of isolates was observed to neomicin (30.84%) and cephalexin (27.63%). Multiple drug resistance to three or more and five or more of antimicrobials tested was observed in 34.9% and 15.5% of isolates, respectively. The cases of canine otitis occurred predominantly in first years of age, in mixed breeds animals, at autumn season. The presence of itch, bad smell, and secretion in ear conduct were the major signs observed at clinical examination.

Propolis is a honeybee product that has been used since ancient times because of its therapeutic effects. It can be used in the development of alternative therapies for the treatment of many diseases, and because propolis shows antibacterial action, this work was carried out in order to investigate a possible synergism between propolis and antibiotics acting on DNA (ciprofloxacin and norfloxacin) and on the metabolism (cotrimoxazole) against Salmonella Typhi. Propolis samples collected in Brazil and Bulgaria were compared in these assays, and the synergism was investigated by using 1/2 and 1/4 of the minimal inhibitory concentration for propolis and antibiotics, evaluating the number of viable cells according to the incubation time. Brazilian and Bulgarian propolis showed antibacterial activity, but no synergistic effects with the three tested antibiotics were seen. Previous works by our laboratory have revealed that propolis has synergistic effects with antibiotics, acting on the bacterial wall and ribosome, but it does not seem to interact with antibiotics acting on DNA or folic acid, and only a bacteriostatic action was seen in these assay conditions.

Increase in sensitivity of agent using modified method was observed mainly in aminoglycosides (amikacin, gentamicin, tobramycin), quinolones (ofloxacin and norfloxacin) and cephalosporins (cefoperazone and ceftazidime) groups. by standard diffusion disk test and a modified method, by the addition Tris-EDTA, was evaluated.
