Reintroduction can be enhanced by data from long-term post-release monitoring, which allows for modeling opportunities such as population viability analysis (PVA). PVA-relevant data were gathered via long-term monitoring of reintroduced red-billed curassows at the Guapiacu Ecological Reserve (REGUA), located in Rio de Janeiro, Brazil, over 25 months. In the present article, we (1) assess the robustness of the reintroduction plan, (2) evaluate the viability of the current reintroduced population, and (3) examine mitigation options to increase the viability of this population. VORTEX indicates that the initial plan, fully implemented, was likely to establish a viable population at REGUA. The current population is unviable; the best mitigation strategies are to eliminate hunting altogether, or at least reduce it by half, and to supplement ten immature pairs in 2015. A positive long-term outcome at REGUA is still possible; we encourage the Brazilian government and private stakeholders to consider population supplementation, both to achieve success at REGUA and to improve the evidence base for future reintroductions. Buccal mucosa (BM) cells have been used in human biomonitoring studies for detecting DNA adducts and chromosomal damage in an epithelial cell population. In the present study, we have investigated if human BM cells are suitable for use in the single-cell gel electrophoresis (SCGE)/Comet assay as an approach for estimating the exposure of epithelial cells to DNA-damaging agents. Our results indicate that only a few cells from BM cell samples yield comets that can be analyzed by current methods, and that the yield of cells with comets is independent of the percentage of viable BM cells in the sample. Data generated after enzymatic enrichment of viable cells and immunomagnetic separation of epithelial cells suggest that most of the BM cells that do form comets are probably leukocytes. Moreover, by reevaluating specific cells after running the Comet assay, we found that viable epithelial BM cells give rise to atypical comets that are not included in the analysis. Comparing DNA migration patterns between small groups of smokers and nonsmokers indicated that long-term smoking had no effect on the subpopulation of cells that yield typical comets. Our results indicate that the SCGE assay, as it is commonly performed, may not be useful for genotoxicity monitoring in human epithelial BM cells.

A population of Loxopagurus loxochelis was studied in terms of seasonal abundance, size frequency distribution, sex ratio, and reproductive period (percentage of ovigerous females). Specimens were collected monthly over a period of two years (from September 1995 to August 1997) in the non-consolidated areas of the Ubatuba, Mar Virado, and Ubatumirim bays (northern coast of São Paulo State, Brazil) with a double-rig trawl net. A total of 1,084 individuals were analysed. Animal size (minimum, maximum, and mean +/- SD shield length) was 2.8, 9.1, and 6.88 +/- 1.13 mm for 625 males; 2.8, 8.2, and 5.78 +/- 0.98 mm for 236 non-ovigerous females; and 4.6, 8.0, and 6.24 +/- 0.68 mm for 223 ovigerous females, respectively. Sexual dimorphism was recorded by the presence of males in the largest size classes. The sex ratio was 1.4 : 1 in favour of males. The highest incidence of ovigerous females occurred during winter and spring (June to October), with a low percentage in summer and fall (November to May), indicating continuity in the reproductive cycle. This strategy of reproduction is related to temperature. The occurrence of L. loxochelis in the Ubatuba region represents the final point of northern distribution of this species as a function of water mass influence, and could be its real limit on the Atlantic coast of South America.

It is crucial for biodiversity conservation that protected areas are large and effective enough to support viable populations of their original species. We used a point count distance sampling method to estimate population sizes of a range of bird species in three Atlantic forest protected areas of size 5600, 22,500, and 46,050 ha. Population sizes were generally related to reserve area, although in the mid-sized reserve, there were many rare species reflecting a high degree of habitat heterogeneity. The proportions of forest species having estimated populations > 500 ranged from 55% of 210 species in the largest reserve to just 25% of 140 species in the smallest reserve. All forest species in the largest reserves had expected populations > 100, but in the small reserve, 28% (38 species) had populations < 100 individuals. Atlantic forest endemics were no more or less likely to have small populations than widespread species. There are 79 reserves (> 1000 ha) in the Atlantic forest lowlands. However, all but three reserves in the north of the region (Espirito Santo and states north) are smaller than 10,000 ha, and we predict serious levels of local extinction from these reserves. Habitat heterogeneity within reserves may promote species richness within them, but it may also be important in determining species loss over time by suppressing populations of individual species. We suggest that most reserves in the region are so small that homogeneity in the habitat/altitude within them is beneficial for maintenance of their (comparatively small) original species compliment. A lack of protection in the north, continued detrimental human activity inside reserves, and our poor knowledge of how well the reserve system protects individual taxa, are crucial considerations in biodiversity management in the region.

A partir de perfis populacionais experimentais de linhagens do díptero forídeo Megaselia scalaris, foi determinado o número mínimo de perfis amostrais que devem ser repetidos, via processo de simulação bootstrap, para se ter uma estimativa confiável do perfil médio populacional e apresentar estimativas do erro-padrão como medida da precisão das simulações realizadas. Os dados originais são provenientes de populações experimentais fundadas com as linhagens SR e R4, com três réplicas cada, e que foram mantidas por 33 semanas pela técnica da transferência seriada em câmara de temperatura constante (25 ± 1,0ºC). A variável usada foi tamanho populacional e o modelo adotado para cada perfíl foi o de um processo estocástico estacionário. Por meio das simulações, os perfis de três populações experimentais foram amplificados, determinando-se, dessa forma, o tamanho mínimo de amostra. Fixado o tamanho de amostra, simulações bootstrap foram realizadas para construção de intervalos de confiança e comparação dos perfis médios populacionais das duas linhagens. Os resultados mostram que com o tamanho de amostra igual a 50 inicia-se o processo de estabilização dos valores médios.

Eucalyptus breeding is typically conducted by selection in open-pollinated progenies. As mating is controlled only on the female side of the cross, knowledge of outcrossing versus selling rates is essential for maintaining adequate levels of genetic variability for continuous gains. Outcrossing rate in an open-pollinated breeding population of Eucalyptus urophylla was estimated by two PCR-based dominant marker technologies, RAPD and AFLP, using 11 open-pollinated progeny arrays of 24 individuals. Estimated outcrossing rates indicate predominant outcrossing and suggest maintenance of adequate genetic variability within families. The multilcous outcrossing rate (t(m)) estimated from RAPD markers (0.93 +/- 0.027), although in the same range, was higher (alpha > 0.01) than the estimate based on AFLP (0.89 +/- 0.033). Both estimates were of similar magnitude to those estimated for natural populations using isozymes. The estimated Wright's fixation index was lower than expected based on t, possibly resulting from selection against selfed seedlings when sampling plants for the study. An empirical analysis suggests that 18 is the minimum number of dominant marker loci necessary to achieve robust estimates of t,. This study demonstrates the usefulness of dominant markers, both RAPD and AFLP, for estimating the outcrossing rate in breeding and natural populations of forest trees. We anticipate an increasing use of such PCR-based technologies in mating-system studies, in view of their high throughput and universality of the reagents, particularly for species where isozyme systems have not yet been optimized.

The immature development of pharaoh's ant, Monomorium pharaonis, workers was s died in a naturally collected Brazilian population. Three larval instars were confirmed by measuring maximal cephalic widths in cohorts of known age. Minimum egg to adult duration was 25 days, while the maximum duration was 54 days, both of which are larger than those reported for European populations. Larval instar development is cyclical at the colony level, suggesting reproductive bursts followed by lower reproductive activity of the queens.
