Correlative fractography is a new expression proposed here to describe a new method for the association between scanning electron microscopy (SEM) and light microscopy (LM) for the qualitative and quantitative analysis of fracture surfaces. This article presents a new method involving the fusion of one elevation map obtained by extended depth from focus reconstruction from LM with exactly the same area by SEM and associated techniques, as X-ray mapping. The true topographic information is perfectly associated to local fracture mechanisms with this new technique, presented here as an alternative to stereo-pair reconstruction for the investigation of fractured components. The great advantage of this technique resides in the possibility of combining any imaging methods associated with LM and SEM for the same observed field from fracture surface. © Microscopy Society of America 2013.

Pathogens in seeds imply quarantine constraints for exportation. This research aimed to quantify nematodes and fungus populations in seed samples of forage grasses from the main Brazilian producing states, and to multiply the nematodes in vitro, as well to study the taxonomy of the nematodes detected. Seed samples of 237 lots of different forage grasses from São Paulo, Minas Gerais, Mato Grosso do Sul, Mato Grosso, Bahia, and Goias States were collected and shipped for analyses in the Nematology and Plant Pathology Laboratories at FCAV[long dash]UNESP [long dash] Jaboticabal( SP) by Comercio e Industria Matsuda Imp., Exp. Ltd. Nematodes were extracted from 10 g of seeds. To detect the fungus, the Blotter-test was applied. The identifications were done by using a photonic microscope and a stereomicroscope. For the study of in vitro multiplication of the nematodes, the following parthenogenetic species were selected: Aphelenchoides sexlineatus, Aphelenchus sp. and Ditylenchus montanus. Cultures of the fungi Fusarium sp. and Didymella brioniae were used as substrate to multiply the nematodes in Petri dishes. Each plate was inoculated with 10 mature females, then incubated in B.O.D. at 25 [plus or minus]1 [degree]C, in the dark. Thirty days after inoculation, the nematodes were extracted. The populations obtained in the suspensions were estimated in the microscope using Peters counting chamber, and the reproduction factor estimated. For the taxonomic study of the nematodes, morphological characters of specimens were recorded under the light and scanning electron microscopes. The results indicated a large distribution of nematodes and fungus in seeds of forage grasses in Brazil. The nematodes identified in the present study were: Aphelenchoides besseyi, A. bicaudatus, A. fragariae, A. sexlineatus, Ditylenchus myceliophagus, D. dipsaci, D. montanus, and Aphelenchus sp. In addition, species of the fungi Fusarium, Helminthosporium and Phoma were recovered.

A set of NIH Image macro programs was developed to make qualitative and quantitative analyses from digital stereo pictures produced by scanning electron microscopes. These tools were designed for image alignment, anaglyph representation, animation, reconstruction of true elevation surfaces, reconstruction of elevation profiles, true-scale elevation mapping and, for the quantitative approach, surface area and roughness calculations. Limitations on time processing, scanning techniques and programming concepts are also discussed.

The stretch zone width (SZW) data for 15-5PH steel CTOD specimens fractured at -150 degrees C to + 23 degrees C temperature were measured based on focused images and 3D maps obtained by extended depth-of-field reconstruction from light microscopy (LM) image stacks. This LM-based method, with a larger lateral resolution, seems to be as effective for quantitative analysis of SZW as scanning electron microscopy (SEM) or confocal scanning laser microscopy (CSLM), permitting to clearly identify stretch zone boundaries. Despite the worst sharpness of focused images, a robust linear correlation was established to fracture toughness (KC) and SZW data for the 15-5PH steel tested specimens, measured at their center region. The method is an alternative to evaluate the boundaries of stretched zones, at a lower cost of implementation and training, since topographic data from elevation maps can be associated with reconstructed image, which summarizes the original contrast and brightness information. Finally, the extended depth-of-field method is presented here as a valuable tool for failure analysis, as a cheaper alternative to investigate rough surfaces or fracture, compared to scanning electron or confocal light microscopes. Background and Aims the occurrence of stomata in seed coats is uncommon and there is limited information about their function(s). The aim of this study was to verify the distribution of stomata in seed coats of Swietenia macrophylla and to relate it to the imbibition process and aspects of the structure of the outer integument layers.Methods For the structural and ultrastructural studies, the seeds were processed using the usual techniques and studied under light and scanning electron microscopes. Histochemical tests were employed to identify the cell wall composition in the different seed coat portions. To assess the role of the stomata in the imbibition, non-impervious seeds were compared with partially impervious ones, in which only the embryo, median or hilar regions were left free. Further, the apoplastic pathway marker was employed to confirm the role of the stomata as sites of water passage during imbibition.Key Results A positive relationship was observed between seed coat thickness and stomata density. The stomata were devoid of movement, with a large pore. They occurred in large numbers in the embryo region and extended with lower frequency towards the wing. Imbibition rates were related to stomata density, suggesting that the stomata act as preferential sites for water entry in the S. macrophylla seeds.Conclusions At maturity, the stomata in the seed coat play a significant role in seed imbibition. The data may also infer that these permanently opened stomata have an important role in gas exchange during seed development, aiding embryo respiration.

Thermally sprayed HVOF coatings are increasingly being used in industrial applications where high wear and corrosion resistance are needed [1,2]. In this paper, electrochemical ac and de experiments were used in order to obtain the corrosion resistance of coated steel with different numbers of Cr3C2-NiCr layers. This work has been performed in order to determine the role of coating thickness in the corrosion behaviour of a steel protected with cermet thermally sprayed coatings. It is known that a thicker layer protects better against corrosion when a metallic coating is evaluated. But cermet coatings, such as Cr3C2-NiCr, contain higher levels of porosity and residual stresses than metallic coatings, which really could influence the corrosion resistance of the deposited layer. Electrochemical measurements, such as Open-Circuit Potential (E-Osubset of), Polarisation Resistance (RP) and Cyclic Voltammetry (CV), were performed in an aerated 3.4 NaCI media (%wt.). Electrochemical Impedance Measurements (EIS) were also done in order to obtain a mechanism that explains the corrosion process. Structural Characterisation was carried out by means of Optical and Scanning Electron Microscopes (OM, SEM) with an Energy Dispersive Spectrometry analyser (EDS). Results show that the corrosion resistance of the complete system is mainly influenced by the substrate behaviour. The application of a higher number of deposited layers did not substantially increase their anticorrosive properties. Stress generation during the spraying deposition process plays an important role in the behaviour of the coated steel against corrosion phenomena. (C) 2002 Elsevier B.V. B.V All rights reserved.

Com o objetivo de estudar a relação entre cristais de oxalato de cálcio e floema, fragmentos de folhas de Peltodon radicans foram fixados e processados, segundo métodos usuais, para estudos ao microscópio de luz e eletrônico de transmissão. Observou-se que os cristais ocorrem nas células da bainha do feixe, lateralmente em relação ao floema. Células intermediárias estabelecem conexão entre elemento crivado e células da bainha, portadoras de cristais, com crescimento intrusivo entre estas. Íons cálcio são abundantes no citoplasma das células da bainha que contém cristais de oxalato de cálcio. Nas células intermediárias a detecção ultra-citoquímica de cálcio também apresentou resultados positivos, enquanto nos elementos crivados a presença deste íon não foi constatada. Há, portanto, um gradiente crescente de concentração de cálcio dos elementos crivados para as células da bainha. Assim, formulamos a hipótese de que a formação de cristais de oxalato de cálcio tem, em P. radicans, o objetivo de controlar os níveis de cálcio citossólico nos elementos crivados.

Spermiogenesis of the domestic sparrow was investigated with the light and electron microscopes and a step by step classification is proposed. Three cell populations corresponding to early, mid and late spermatids were easily divided according to their positions in the seminiferous epithelium. In addition to this initial separation, six steps were recognized, based on nuclear morphology and the degree of chromatin condensation, in association to their acrosomal and flagellar development. Early spermiogenesis is the period previous to chromatin condensation. The first step can be recognized by the extending flagellum and the second by the pro-acrosome development in contact with the nucleus. During the third or intermediate step, chromatin condenses and the cell becomes polarized with the pro-acrosomic vesicle and the tail occupying opposite sides of the nucleus. Late spermiogenesis, including steps IV-VI, is marked by complete chromatin condensation. The final cellular modifications lead to the formation of a spiraled spermatozoon. This shape is due to the twisting of the acrosome and nucleus, as well as the helical arrangement of mitochondria around the axoneme along most of the flagellum, making an exceptionally long middle piece.
