The dynamics of hydration of meso and thermo citrate synthases has been investigated using the EEF1 methodology implemented with the CHARNM program. The native enzymes are composed of two identical subunits, each divided into a small and large domain. The dynamics behavior of both enzymes at 30 degrees C and 60 degrees C has been compared. The results of simulations show that during the hydration process, each subunit follows a different pathway of hydration, in spite of the identical sequence. The hydrated structures were compared with the crystalline structure, and the root mean square deviation (RMSD) of each residue along the trajectory was calculated. The regions with larger and smaller mobility were identified. In particular, helices belonging to the small domain are more mobile than those of the large domain. In contrast, the residues that constitute the active site show a much lower displacement compared with the crystalline structure. Hydration free energy calculations point out that Thermoplasma acidophilum citrate synthase (TCS) is more stable than chicken citrate synthase (CCS), at high temperatures. Such result has been ascribed to the higher number of superficial charges in the thermophilic homologue, which stabilizes the enzyme, while the mesophilic homologue denatures. These results are in accord with the experimental found that TCS keeps activity at temperatures farther apart from the catalysis regular temperature than the CCS. (c) 2005 Wiley Periodicals, Inc.

The condition of certain neglected structures and their possible importance to the classification of the family Tabanidae are studied. The front coxae are considered as belonging to two types: 1) short and robust; 2) long and slender. The posterior edge of the meso-katepimeron presents two conditions: 1) without projection, 2) with backward projection. Similar conditions are found in the meso-Katepisternum, but with intermediate conditions. These characters are discussed in relation to the system proposed for the family by Mackerras. Conditions 1 and 2 of the former two structures separate, respectively, the Pangoniinae and Scepsidinae from the Chrysopsinae and Tabaninae. Condition 2 of the third structure seems to be developing independently in different groups of Tabaninae, with varying degrees of development.

The mafic/ultramafic Ipanema Layered Complex (ILC), Minas Gerais Brazil, consists of seven individual bodies. These units crosscut polyphase orthogneisses and interlayered paragneisses of the Paleoproterozoic Juiz de Fora Complex. Intrusive granitoids tectonically related to [lie Neoproterozoie Aracuai orogen are also present in the study area.A Sm-Nd whole-rock linear array for seven samples metapyoxenites, metaperidotiles, metagabbro. and meta-anorthosite) from the Santa Cruz massif, the largest body of the ILC. suggest that it was emplaced at 1104 +/- 78 Ma the original magma was derived from a depleted mantle source (epsilon(Ndt)= +3.8). U-Pb single-grain zircon stud of a meta-anorthosite yields all upper intercept age of 1719 +/- 4 Ma, which is interpreted to represent inheritance. The lower intercept at 630+/-3 Ma indicates (hat a Neoproterozoic tectonothermal episode overprinted the ILC, this event occurred under upper-amphiolite-, to granulite-facies conditions. The 630 Ma episode is consistent with the timing of regional metamorphism and deformation of the adjacent Aracuai orogen (Brasiliano collage). Emplacement of the ILC and other coeval metamafies and meta-ultramafics (of alkaline affinity) in the re, oil is attributed to early extension tectonics, accompanying accretion of the Rodinia super- continent during the Mesoproterozoic-Neoproterozoic time boundary.

An optimization study of the reaction conditions of Fe(TDCPP)Cl when it is used as catalyst in the hydroxylation of cyclohexane by iodosylbenzene (PhIO) has been carried out, It was found that Fe(TDCPP)Cl follows the classical PhIO mechanism described for Fe(TPP)Cl, which involves the monomeric active species Fe-IV(O)P-+. (I). In the optimized condition ([Fe(TDCPP) = 3.0 X 10(-4) mol l(-1) in 1,2-dichloroethane (DCE); ultrasound stirring at 0 degrees C; PhIO/FeP molar ratio = 100), this FeP led to a yield of cyclohexanol (C-ol) of 96% and a turnover number of 96, Therefore, Fe(TDCPP)Cl may be considered a good biomimetic model and a very stable, resistant and selective catalyst, which yields C-ol as the sole product. DCE showed to be a better solvent than dichloromethane (DCM), 1 DCE:1 MeOH mixture or acetonitrile (ACN). Since the Fe-IV(O)P-+. is capable of abstracting hydrogen atom from DCM, MeOH or ACN, the solvent competes with the substrate. Presence of O-2 lowers the yield of C-ol, as it can further oxidize this alcohol to carboxylic acid in the presence of radicals, Presence of H2O also causes a decrease in the yield, since it converts the active species I into Fe-IV(OH)P, which cannot oxidize cyclohexane. Addition of excess imidazole or OH- to the system results in a decrease in the yield of C-ol, due to the formation of the hexacoordinated complexes Fe(TDCPP)Im(2)(+) (low-spin, beta(2) = 2.5 X 10(8) mol(-2) l(2)) and Fe(TDCPP)(OH)(2)(-) (high-spin, beta(2) = 6.3 X 10(7) mol(-2) l(2)), the formation of both Fe(TDCPP)Im(2)(+) and Fe(TDCPP)(OH)(2)(-) complexes were confirmed by EPR studies. The catalytic activities of Fe(TDCPP)C and Fe(TFPP)Cl were compared, the unusually high yields of C-ol with Fe(TFPP)Cl obtained when ultrasound, DCM and O-2 atmosphere were used, suggest that a parallel mechanism involving the mu-oxo dimer form, O-2 and radicals may also be occurring with this FeP, besides the PhIO mechanism.

The possible development of thermal events in the central portion of São Paulo state was described based on apatite fission track analysis. Using apatites of sedimentary rocks of the Paraná Basin, modeling the thermal history was made possible due to the homogeneity of the data. Every thermal history begins with a total annealing of fission tracks, related to the Serra Geral magmatism, evolving into a cooling period. In addition to cooling after the magmatism (Early Cretaceous) two other periods of cooling were also detected, registered in the Late Cretaceous/ Paleocene and Eocene, driven as much by uplift with tectonic denudation as by faulting. The nearest portion of the edge of the basin (external to the Dome of Pitanga), registered a period of warming over the Paleocene that can be attributed to the increase in the geothermal gradient. The periods of cooling have a regional and temporal relationship with the tectonic events that occurred in the southeastern Brazil and were described in the crystalline basement. The period of warming, registered in the Late Cretaceous/ Paleocene, has local occurrence and can be found only in the southern portion of the studied area.

Hydrocarbon accumulations occur in marine carbonate reservoirs of Quissamã Formation (early to midlle Albian), southwest Campos Basin. We investigated Pampo, Bicudo and Bonito fields, in order to understand the tectono-structural framework of oncolite/bioclast coarse-grained calcarenite reservoir and the calcilutite-marl-shale sealing interval (Late Albian to Turonian). The database of eleven wells from those fields allowed to elaborate structural sections correlating the Macaé Group – both Quissamã reservoir and Outeiro seal, the latter corresponding to the tectonic deepening phase of basin evolution. Based on density and electric logs, it was prepared structural sections of the carbonate reservoirs with consequent identification of porous zones and oil-water contacts. An extensive 3-D seismic database (~300 Km2) allowed to map three reflectors which represent the limiting units of Macaé Group

Neste trabalho, as ninfas de Quesada gigas (Olivier) são descritas e ilustradas. Uma chave para o reconhecimento dos cinco ínstares ninfais é também apresentada. Foram analisados exemplares coletados em cafezais dos municípios de São Sebastião do Paraíso, Monte Santo de Minas e Patrimônio (Minas Gerais), Franca e Casa Branca (São Paulo). As amostras encontram-se preservadas em álcool a 80% e depositadas na Coleção Entomológica do Departamento de Fitossanidade da FCAV-UNESP, Câmpus de Jaboticabal. Os caracteres analisados foram: antenas, tecas alares, perna anterior, ápice da tíbia meso e metatorácicas e ápice abdominal do macho/fêmea. Foi adotada uma fórmula para indicar a configuração que é apresentada pelas estruturas presentes no fêmur anterior. Ninfas de Q. gigas de primeiro ínstar apresentam fórmula femoral 2-1-0, de segundo ínstar 2-1-3, de terceiro ínstar 2-1-5, de quarto ínstar 2-1-6 e as de quinto ínstar 2-2-8. O primeiro número refere-se à somatória dos dentes posteriores e dentes acessórios, o segundo ao número de dentes intermediários e o terceiro aos dentes do pente femoral. As estruturas apresentadas pelo fêmur protorácico constituem caracteres morfológicos adequados para a identificação dos ínstares ninfais de cigarras.

The cellular and molecular characteristics of a cell line (BME26) derived from embryos of the cattle tick Rhipicephalus (Boophilus) microplus were studied. The cells contained glycogen inclusions, numerous mitochondria, and vesicles with heterogeneous electron densities dispersed throughout the cytoplasm. Vesicles contained lipids and sequestered palladium meso-porphyrin (Pd-mP) and rhodamine-hemoglobin, suggesting their involvement in the autophagic and endocytic pathways. The cells phagocytosed yeast and expressed genes encoding the antimicrobial peptides (microplusin and defensin). A cDNA library was made and 898 unique mRNA sequences were obtained. Among them, 556 sequences were not significantly similar to any sequence found in public databases. Annotation using Gene Ontology revealed transcripts related to several different functional classes. We identified transcripts involved in immune response such as ferritin, serine proteases, protease inhibitors,. antimicrobial peptides, heat shock protein, glutathione S-transferase, peroxidase, and NADPH oxidase. BME26 cells transfected with a plasmid carrying a red fluorescent protein reporter gene (DsRed2) transiently expressed DsRed2 for up to 5 weeks. We conclude that BME26 can be used to experimentally analyze diverse biological processes that occur in R. (B.) microplus such as the innate immune response to tick-borne pathogens. A maioria dos estudos pré-clínicos e clínicos aponta a nicotina como o principal agente responsável pelo desenvolvimento da dependência ao tabaco. Muitos trabalhos têm demonstrado que as bases neurais da dependência à nicotina são semelhantes àquelas das outras drogas de abuso. A nicotina induz preferência condicionada por lugar e auto-administração e, portanto, atua como reforçador positivo, esse efeito parece ser mediado pelo sistema dopaminérgico mesolímbico. A nicotina também induz à sensibilização comportamental que é provavelmente resultante de alterações da expressão gênica do núcleo acumbens induzidas pela exposição prolongada a essa substância. A suspensão do uso de nicotina resulta em síndrome de abstinência. As evidências indicam que esses sinais e sintomas sejam mediados por receptores colinérgicos nicotínicos centrais e periféricos. Outros neurotransmissores, como por exemplo a serotonina e os peptídeos opióides, também podem estar envolvidos na mediação da dependência e síndrome de abstinência à nicotina. A revisão da literatura mostra a complexidade dos efeitos da nicotina no organismo. A integração entre as abordagens comportamental, neuroquímica e molecular possibilitará a compreensão dos mecanismos neurais da dependência ao tabaco e fornecerá as bases para o desenvolvimento racional de agentes terapêuticos que possam ser utilizados para o tratamento da dependência e síndrome de abstinência ao tabaco.
