Membrane integrity, as measured by electrical conductivity (EC), is suggested as an indicator of seed vigor in soybean [Glycine max (L.) Merrill] seeds. This study evaluated the effect of storage time and temperature on EC of six soybean seed lots (two lots each of high, medium and low vigor). All seed lots were adjusted to 120 g kg(-1) seed moisture, sealed in aluminum foil packets and placed in storage at 10 and 20 degreesC or stored unsealed in multi-wall paper bags in warehouse (WH) conditions at Lexington, KY, USA for 486 days. Four of the six seed lots were also stored unsealed at 10 degreesC. All seed lots were sampled at 3-month intervals and evaluated for seed moisture (SMC), standard germination (SG) and vigor [accelerated aging (AA) and EC]. After 91 and 204 days in storage, samples initially stored at 20 degreesC and WH were moved to 10 degreesC and sampled at the same intervals. Seed moisture content for unsealed samples equilibrated at 107 g kg(-1) (+/-9 g kg(-1)) in both the WH and 10 degreesC environments. No change in SG occurred for seeds stored sealed (120 g kg(-1)) at 10 degreesC, except for the low vigor seed lots which declined significantly at the last sample date. The AA germination declined significantly for all, seed lots stored sealed at 10 degreesC, however the EC did not change during the same storage period. Seeds stored sealed at 20 degreesC and unsealed in the WH showed rapid declines in AA and SG and significant increases in EC. When these seeds were moved to 10 degreesC, however, the AA continued to decline while the EC remained at the same level (no significant change) for the remainder of the seed storage period. Thus whilst the AA declined in all environments, the EC only increased at higher temperatures (20 degreesC, WH) but showed little change during storage at 10 degreesC. Thus, precautions must be taken if using EC to measure soybean seed vigor following storage at 10 degreesC.

Determinaram-se a degradabilidade potencial (DP) e a digestibilidade intestinal da proteína não degradada no rúmen (DIPNDR) do capim-elefante em diferentes idades de rebrote (30, 45 e 60 dias) e comparou-se a técnica do saco de náilon móvel (in situ) com o método de três estádios (in vitro). Para tanto, utilizaram-se seis novilhos mestiços canulados no rúmen e duodeno alimentados exclusivamente com capim-elefante picado. O ensaio de degradabilidade foi realizado com amostras do capim incubadas no rúmen por 3, 6, 9, 12, 24, 48, 72, 96 e 120h. A digestibilidade intestinal foi determinada utilizando-se os resíduos de incubação por 24 horas. Na técnica in situ os resíduos em sacos de náilon foram colocados no duodeno e recuperados nas fezes. No método in vitro, os resíduos foram submetidos à digestão com HCl-pepsina-pancreatina. em amostras de capim com idades de 30, 45 e 60 dias foram observados valores de DP da proteína de 87,5; 87,8 e 83,8%, respectivamente. A DIPNDR variou com a idade do capim e foi semelhante entre os métodos in situ e in vitro somente para o capim com 60 dias. O método in situ apresentou estimativa de digestibilidade intestinal mais coerente com as mudanças na composição química do capim-elefante decorrentes do envelhecimento.

Objetivou-se avaliar o consumo de nutrientes e o desempenho de novilhas Nelore alimentadas com dietas contendo cana-de-açúcar in natura (CN) ou hidrolisada (CH) com 0,5% de Ca(OH)2 armazenada por 24, 48 e 72 horas. Foram utilizadas 24 novilhas com nove meses de idade e 119,6±8,1kg de peso corporal inicial. O delineamento experimental utilizado foi o inteiramente casualisado com quatro tratamentos e seis repetições. O consumo de matéria seca foi reduzido em 29% pela adição de Ca(OH)2, não sendo alterado pelo período de armazenamento da CH. O consumo de cálcio foi incrementado (P<0,05) pela utilização de CH nas dietas, mantendo níveis de ingestão aceitáveis. Novilhas alimentadas com CH armazenada por 24, 48 e 72 horas apresentaram, respectivamente, ganhos de peso 41, 30 e 35% inferior (P<0,05) àquelas alimentadas com CN. O tratamento da cana-de-açúcar com Ca(OH)2 não é recomendado para alimentação de novilhas Nelore, em virtude de limitar a ingestão e reduzir o ganho de peso.

The objective of this study was to evaluate the chemical characteristics of the forage and ingestive behavior of Nellore heifers fed hydrolyzed sugarcane in different periods of storage. Twenty-four heifers with initial body weight of 119.6±8.1 kg were utilized. The experimental design was completely randomized, in which the treatments were diets with fresh sugarcane and hydrolyzed sugarcane (5 g of lime kg-1 of chopped sugarcane) stored for 24, 48 or 72 hours as the only roughage. The addition of lime to sugarcane associated with its storage up to 72 hours provided an increase of 20% of the potentially degradable cell wall of carbohydrates, from 382.4 to 458.8 g kg-1 of total carbohydrates. The in vitro digestibility of dry matter was altered by the storage of hydrolyzed sugarcane, increasing 7.08% when the storage time was increased from 24 to 72 hours. Heifers fed fresh sugarcane remained more time consuming compared with heifers fed other diets. The time used for water intake was not influenced by the diet. The rumination time presented a quadratic variation in relation to storage time of the hydrolyzed sugarcane, with higher values for the of hydrolyzed sugarcane diets stored for 48 hours. Heifers fed hydrolyzed sugarcane spent more time on other activities than those fed fresh sugarcane. The supply of hydrolyzed sugarcane stored up to 72 hours in the proportion of 600 g kg-1 of dry matter in the diet, alters the intake patterns, reducing the feed intake in Nellore heifers.

A computer-based sliding mode control (SMC) is analysed. The control law is accomplished using a computer and A/D and D/A converters. Two SMC designs are presented. The first one is a continuous-time conventional SMC design, with a variable structure law, which does not take into consideration the sampling period. The second one is a discrete-time SMC design, with a smooth sliding law, which does not have a structure variable and takes into consideration the sampling period. Both techniques are applied to control an inverted pendulum system. The performance of both the continuous-time and discrete-time controllers are compared. Simulations and experimental results are shown and the effectiveness of the proposed techniques is analysed.

The aim of this study was to evaluate the effects of different litter materials on litter compaction, broiler feathering and the incidence of carcass lesions. In the experiment, 3240 one-day-old Ross® chicks were selected by sex and distributed according to a completely randomized experimental design in a 2 x 6 factorial arrangement (two sex and six litter materials). The following litter materials were used: wood shavings, rice husks, chopped Napier grass, 50% sugar cane bagasse plus 50% wood shavings, 50% sugar cane bagasse plus 50% rice husks, and pure sugar cane bagasse. Litter compaction was weekly assessed using a penetrometer. on days 21, 35 and 42 of the experimental period, feathering on the back and legs was scored according to a 0 - 10 scale. on day 42, birds were slaughtered and the presence of bruises, scratches and footpad lesions was recorded. Litter material had no effect on bird feathering. Carcass lesions (scratches, bruises and footpad lesions) were influenced by the litter material evaluated. Birds reared on sugarcane bagasse and chopped Napier grass presented more scratches, bruises and footpad lesions than the others. Dermatitis was more evident in birds reared on sugarcane bagasse, chopped Napier grass and the combination of litter materials. It was found that males presented higher incidence of dermatitis and footpad lesions than females. Each litter material presented different compaction degrees, which increased along the experimental period. Sugarcane bagasse, chopped Napier grass and the combination of bedding materials presented the highest degree of compaction, compared with wood shavings and rice husks.

A inoculação de forragens com fungos lignocelulolíticos é uma opção para melhorar a qualidade destas sem adição de produtos químicos. O tratamento do substrato influencia a ação do fungo e a qualidade final do produto. Neste experimento, aplicaram-se quatro tratamentos (compostagem do feno inteiro, compostagem do feno picado, hidratação do feno em água fria e hidratação do feno em água quente) a um feno de Brachiaria decumbens. Aos tratamentos seguiu-se inoculação com o fungo Pleurotus ostreatus e incubação por 35 dias, sob temperatura controlada. Usou-se o delineamento inteiramente casualizado, com quatro repetições e medidas repetidas. Amostras foram colhidas semanalmente para acompanhar a degradação do substrato, mediante a análise química do feno. Observou-se aumento linear, com o decorrer do tempo, no teor de proteína bruta (PB) e na proporção de lignina na parede celular (LIG-FDN), e decréscimo linear nos valores de fibra em detergente neutro (FDN), celulose e hemicelulose. Não se observou efeito de tratamento no teor de FDA. Os tratamentos com compostagem apresentaram maiores valores de PB, lignina e LIG-FDN e menores de FDN e hemicelulose. Não se observou diferença entre os tratamentos com hidratação. O tratamento do feno de braquiária com o fungo propiciou degradação da fração fibrosa e aumento no teor de PB, com efeito mais intenso nos tratamentos que usaram compostagem. A ação do fungo foi mais efetiva sobre a hemicelulose que sobre os demais componentes da fibra.

Prostate differentiation during embryogenesis and its further homeostatic state maintenance during adult life depend on androgens. Abundant biological data suggest that androgens play an important role in the development of the prostate cancer and other prostatic diseases. The objective of this work was to evaluate the effects of the testosterone supplementation in gerbil (a new experimental model) at different ages. Tissues from experimental animals were studied by histological and histochemistry procedures, androgen receptor immunohistochemistry assay, morphometric-stereological analysis, and transmission electron microscopy (TEM). After the treatment were observed increase of prostate weight and epithelium height in all ages studied. In some adult and aged treated animals, hyperplasic and displasic process were observed, including prostatic intraepithelial neoplasias and adenocarcinomas. Increase of the thickness of the smooth muscle cell (SMC) layer was observed in pubescent and adult animals and TEM revealed apparent SMC hypertrophy. An apparent increase in the frequency of blood vessels distributed by the subepithelial stroma in the treated animals was noticed. Reversion of the natural effects of aging on the prostate was observed in the aged treated animals in some acini of the gland. These data demonstrate that the gerbil prostate is susceptible to androgenic action at the studied ages and it can serve, for example, as experimental model to studies of prostate neoplasic process induction and hormonal therapy in aged animals.

In this study, we evaluated the involvement of rat ventral prostate smooth muscle cells (SMC) in secretory activity and whether this function is modulated after castration. Cell morphology was examined at both light and electron microscopy levels and the organelles involved in secretory function were labeled by the zinc-iodide-osmium (ZIO) method at the ultrastructural level and their volume density was determined by stereology. Castration resulted in marked changes of the SMC, which adopted a spinous aspect and abandoned the layered arrangement observed in the prostates of non-castrated rats. The volume density of ZIO reactive organelles increased progressively after castration, reaching significantly higher levels 21 days after castration, Since previous studies have demonstrated that SMC express SMC markers (even 21 days after castration) and are able to respond to adrenergic stimulation, we concluded that differentiated SMC are able to shift from a predominantly contractile to a more synthetic phenotype without changing their differentiation status. Mongolian gerbils (Meriones unguiculatus) were grouped into two experimental groups: GEx.01 suffered orchiectomy and after 30 days received doses of testosterone cipionate (T), while GEx.02 received weekly and alternated doses of the anti-androgens cyproterone acetate and flutamide for 30 days, and the animals were then euthanized. Structural evaluation reveals a more intense reduction in epithelial height in GEx.02. Smooth muscle cells (SMC) presented a star-shaped aspect after 30 days of hormonal ablation and basal membrane was shown to be more intensely grooved in GEx.01. In both groups, after hormonal replacement, recovery in epithelial height could be noted and the SMC presented its phenotypes, but an increase in RER was seen, characterizing a modulation from its contractile to secreting phenotype. In conclusion, the prostate presented involution capacity after androgen ablation and the ability to reorganize after hormonal replacement, but events resulting from orchiectomy and subsequent T replacement were shown to be more aggressive to the prostate. Prostatic differentiation during embryogenesis and its further homeostatic state maintenance during adult life depend on androgens. Dihydrotestosterone, which is synthesized from testosterone by 5alpha-reductase (5alpha-r), is the active molecule triggering androgen action within the prostate. In the present work, we examined the effects of 5alpha-reductase inhibition by finasteride in the ventral prostate (VP) of the adult gerbil, employing histochemical and electron microscopy techniques to demonstrate the morphological and organizational changes of the organ. After 10 days of finasteride treatment at a dose of 100 mg/kg/day, the prostatic complex (VP and dorsolateral prostate) absolute weight was reduced to about 18%. The epithelial cells became short and cuboidal, with less secretory blebs and reduced acid phosphatase activity. The luminal sectional area diminished, suggestive of decreased secretory activity. The stromal/epithelial ratio increased, the stroma becoming thicker but less cellular. There was a striking accumulation of collagen fibrils, which was accompanied by an increase in deposits of amorphous granular material adjacent to the basal lamina and in the clefts between smooth muscle cells (SMC). Additionally, the periacinar smooth muscle became loosely packed. Some SMC were atrophic and showed a denser array of the cytoskeleton, whereas other SMC had a highly irregular outline with numerous spine-like projections. The present data indicate that 5alpha-r inhibition causes epithelial and stromal changes by affecting intra-prostatic hormone levels. These alterations are probably the result of an imbalance of the homeostatic interaction between the epithelium and the underlying stroma.

Differently graded areas of human prostate adenocarcinoma were examined after Masson's trichrome staining or immunohistochemistry for smooth muscle alpha-actin, type IV collagen and laminin. In addition, the ultrastructure of the prostatic smooth muscle cells (SMC) during glandular proliferation and epithelial invasion in selected tumors was studied. The SMC formed a thick layer below the epithelial structures in unaffected areas and were closely associated with each other in homotypic interactions. As the tumor grade increased, the SMC gradually lost interactions with each other and became atrophic. With the growth of the epithelial compartment, the SMC initially segregated to the tumor periphery and the intercellular spaces increased. In high grade tumors, the epithelial cancer cells invaded the spaces between the SMC. Immunohistochemical analysis of the basal membrane revealed increased disruption of the usually thick basal membrane, which became thinner and faintly stained with each of the antibodies used. We conclude that most SMC become atrophic following epithelial invasion in human tumors and that degradation of the basal membrane is an important factor in this process. At the ultrastructural level, different SMC phenotypes occur in prostatic tissues during epithelial invasion. Interconversion between these phenotypes is suggested and a probable relationship among them is proposed.
