Over 40,000 sugarcane (Saccharum officinarum) consensus sequences assembled from 237,954 expressed sequence tags were compared with the protein and DNA sequences from other angiosperms, including the genomes of Arabidopsis and rice (Oryza sativa). Approximately two-thirds of the sugarcane transcriptome have similar sequences in Arabidopsis. These sequences may represent a core set of proteins or protein domains that are conserved among monocots and eudicots and probably encode for essential angiosperm. functions. The remaining sequences represent putative monocot-specific genetic material, one-half of which were found only in sugarcane. These monocot-specific cDNAs represent either novelties or, in many cases, fast-evolving sequences that diverged substantially from their eudicot homologs. The wide comparative genome analysis presented here provides information on the evolutionary changes that underlie the divergence of monocots and eudicots. Our comparative analysis also led to the identification of several not yet annotated putative genes and possible gene loss events in Arabidopsis.

More sensitive methodologies are necessary to improve strongyloidiasis diagnosis. This study compared the sensitivities of the McMaster modified technique and polymerase chain reaction (PCR) assays, both performed in faecal samples. Lewis rats were subcutaneously infected with 4,000, 400 or 40 infective third-stage larvae, considered as high, moderate or low infection, respectively. Seven days later, they were euthanized to count adult nematodes recovered from the small intestine. Stool samples were used to count the number of eggs per gram (EPG) of faeces and to detect parasite DNA by PCR performed with a species and a genus primer pair. The sensitivity of these assays depended upon parasite burden and the primer specificity. All assays presented 100% sensitivity at the highest parasite load. In the moderate infection, EPG and PCR with the genus primer maintained 100% specificity, whereas PCR sensitivity with the species primer decreased to 77.7%. In low infection, the sensitivity was 60% for EPG, 0% for PCR with the species primer and 90% for PCR done with the genus primer. Together, these results suggest that PCR with a genus primer can be a very sensitive methodology to detect Strongyloides venezuelensisin faeces of Lewis rats infected with very low parasite burden.

We aimed to evaluate whether the occurrence of cryptic species of Paracoccidioides brasiliensis, S1, PS2, PS3 and Paracoccidioides lutzii, has implications in the immunodiagnosis of paracoccidioidomycosis (PCM). Small quantities of the antigen gp43 were found in culture filtrates of P. lutzii strains and this molecule appeared to be more variable within P. lutzii because the synonymous-nonsynonymous mutation rate was lower, indicating an evolutionary process different from that of the remaining genotypes. The production of gp43 also varied between isolates belonging to the same species, indicating that speciation events are important, but not sufficient to fully explain the diversity in the production of this antigen. The culture filtrate antigen AgEpm83, which was obtained from a PS3 isolate, showed large quantities of gp43 and reactivity by immunodiffusion assays, similar to the standard antigen (AgB-339) from an S1 isolate. Furthermore, AgEpm83 was capable of serologically differentiating five serum samples from patients from the Botucatu and Jundiaí regions. These patients had confirmed PCM but, were non-reactive to the standard antigen, thus demonstrating an alternative for serological diagnosis in regions in which S1 and PS2 occur. We also emphasise that it is not advisable to use a single antigen preparation to diagnose PCM, a disease that is caused by highly diverse pathogens.

Introduction: The aim of this study was to assess the influence of curing time and power on the degree of conversion and surface microhardness of 3 orthodontic composites. Methods: One hundred eighty discs, 6 mm in diameter, were divided into 3 groups of 60 samples according to the composite used-Transbond XT (3M Unitek, Monrovia, Calif), Opal Bond MV (Ultradent, South Jordan, Utah), and Transbond Plus Color Change (3M Unitek)-and each group was further divided into 3 subgroups (n = 20). Five samples were used to measure conversion, and 15 were used to measure microhardness. A light-emitting diode curing unit with multiwavelength emission of broad light was used for curing at 3 power levels (530, 760, and 1520 mW) and 3 times (8.5, 6, and 3 seconds), always totaling 4.56 joules. Five specimens from each subgroup were ground and mixed with potassium bromide to produce 8-mm tablets to be compared with 5 others made similarly with the respective noncured composite. These were placed into a spectrometer, and software was used for analysis. A microhardness tester was used to take Knoop hardness (KHN) measurements in 15 discs of each subgroup. The data were analyzed with 2 analysis of variance tests at 2 levels. Results: Differences were found in the conversion degree of the composites cured at different times and powers (P < 0.01). The composites showed similar degrees of conversion when light cured at 8.5 seconds (80.7%) and 6 seconds (79.0%), but not at 3 seconds (75.0%). The conversion degrees of the composites were different, with group 3 (87.2%) higher than group 2 (83.5%), which was higher than group 1 (64.0%). Differences in microhardness were also found (P < 0.01), with lower microhardness at 8.5 seconds (35.2 KHN), but no difference was observed between 6 seconds (41.6 KHN) and 3 seconds (42.8 KHN). Group 3 had the highest surface microhardness (35.9 KHN) compared with group 2 (33.7 KHN) and group 1 (30.0 KHN). Conclusions: Curing time can be reduced up to 6 seconds by increasing the power, with a slight decrease in the degree of conversion at 3 seconds; the decrease has a positive effect on the surface microhardness.

Paracoccidioides lutzii, formerly known as 'Pb01-like' strains in the P. brasiliensis complex, is proposed as a new species based on phylogenetic and comparative genomics data, recombination analysis, and morphological characteristics. Conidia of P. lutzii are elongated, different from those of P. brasiliensis. P. lutzii occurs in the central and northern regions of Brazil. Studies comparing P. brasiliensis and P. lutzii may have significant clinical consequences for the diagnosis and treatment of paracoccidioidomycosis.

Espécies do gênero Strongyloides são importantes parasitas de humanos e animais domésticos e aquelas que parasitam roedores são freqüentemente utilizadas como modelos experimentais para o estudo da estrongiloidíase. S. venezuelensis parasita ratos (Rattus norvegicus) e seu estudo tem permitido o entendimento de mecanismos imunológicos envolvidos na relação parasita–hospedeiro da estrongiloidíase humana e animal. O diagnóstico parasitológico de S. venezuelensis pode ser realizado pela detecção de ovos embrionados nas fezes, com o uso do OPG (ovos por grama de fezes), também empregado no monitoramento da infecção. Entretanto, essa técnica se mostra pouco sensível quando a carga parasitária é leve. Os métodos moleculares aumentam a sensibilidade e a especificidade das análises. A PCR tem se mostrado como uma técnica sensível e precisa na detecção de parasitas em tecidos e fezes de hospedeiros infectados. A fim de comparar a sensibilidade das técnicas de OPG e PCR em ratos Lewis infectados por S. venezuelensis, foram realizados dois protocolos no presente estudo. No Protocolo I foi comparada a sensibilidade das duas técnicas em amostras de ratos Lewis com diferentes cargas parasitárias. A PCR com o emprego par de primers descrito por Dorris et al. (2002) obteve melhores resultados que o OPG nos grupos inoculados com 40 L3, considerada como infecção leve. Entretanto, não foi verificada diferença estatística significativa entre as técnicas (P>0,05), provavelmente por terem sido utilizados poucos animais (n=10). O outro par de primers utilizado foi desenhado a partir de uma seqüência parcial do gene do rDNA de S. venezuelensis e não foi capaz de detectar a presença do DNA do helminto em nenhuma amostra desse grupo. Over the last decades, a growing number of scholars from humanities have explored ideas from the postmodern perspective. At the international level, few studies have been undertaken in the public administration field. In Brazil, few articles have been influenced by postmodern ideas. This article presents and discusses theoretical analysis influenced by postmodern ideas in the public administration field, concluding that this appropriation is difficult but worthwhile, especially regarding methodological and analytical issues which may strengthen public administration research.

In recent literature, the concept of policy subsystem has occupied a central place in many theoretical models, it is considered prime unit of analysis in recent approaches to the policy process. Subject of intense theoretical discussion, when compared to related concepts such as policy communities, policy networks and policy issues, these concepts can contribute to the analysis of political participation process, characteristic of democratic governments. This present study aims to present a critical review of the literature on subsystems, policy issues, policy networks and policy communities, as well as discuss these concepts emphasizing the participatory potential of the actors who make up these spaces, seeking to contribute to the dialogue of studies in public policy and participation policy in Brazil.
