A Pothomorphe umbellata (Piperaceae), é uma espécie medicinal nativa do Brasil, utilizada na indústria de cosméticos e protetores de pele contra raios UVA e UVB. Com o intuito de gerar informações aplicadas à propagação da espécie, o presente trabalho relacionou a coloração e a massa de sementes de P. umbellata a seu comportamento germinativo. A coloração e a massa de sementes de P. umbellata foram características adequadas para avaliar a homogeneidade fisiológica, o vigor, o potencial e o comportamento germinativo. Assim, conclui-se que, embora de germinação lenta, as sementes de coloração preta e mais densa devem ser as escolhidas quando de coleta ou de processo seletivo.

Nesta espécie ocorrem dois tipos de tricomas secretores: glândula perolada e tricoma saculiforme. Ambos originam-se na protoderme do primórdio foliar e atingem sua maturidade nas folhas jovens presentes no ápice caulinar. A densidade dos dois tipos de tricomas diminui durante a expansão do limbo, sendo raros na folha adulta. As glândulas peroladas são constituídas por uma célula basal e por uma célula apical grande cuja forma varia de semi-globóide a espatulada. A liberação da secreção ocorre após a ruptura da cutícula distendida. Os tricomas saculiformes são constituídos por uma célula basal, 1-2 células colares e uma célula apical cônica, de posição inclinada à superfície foliar. A secreção acumula-se em espaços subcuticulares diminutos, não tendo sido observada ruptura da cutícula.
Diversidade genética de populações naturais de pariparoba [Pothomorphe umbellata (L.) Miq.] por RAPD

O objetivo desse trabalho foi analisar a estrutura genética de populações de Pothomorphe umbellata (L.) Miq. com base em polimorfismos moleculares do tipo RAPD. Foram analisadas quatro populações naturais do estado de São Paulo (Jacareí, Jundiaí, Piquete e Ubatuba) e uma população do Paraná (Adrianópolis). Foram identificados 25 locos polimórficos (96,15%). Elevados índices de diversidade genética foram observados dentro das populações (Hs = 0,2220). Verificou-se que 65,33% da variabilidade genética total encontra-se dentro das populações e 34,67% entre as populações; índices estes, obtidos a partir do cálculo da divergência genética (G ST = 0,3467). Os resultados sugerem que essas populações possuem níveis elevados de variabilidade genética, a qual pode ser fortemente impactada pela ação humana.

The essential oils of leaves of the Pothomorphe peltata (L.) Miq., collected in vegetative and reproductive periods, showed yields of 0.10-0.20%. Analysis of the oil by GC/MS revealed that the predominant components were those of the sesquiterpene class. The major constituents were beta-caryophyllene (39.5-68.0%), germacrene D (6.2-13.3%), (E)-nerolidol (4.8-11.1%) and alpha-humulene (2.1-6.5%).

We evaluated the growth and development of the medicinal species Pothomorphe umbellata ( L.) Miq. under different shade levels ( full sun and 30, 50, and 70 % shade, marked as I(100), I(70), I(50), and I(30), respectively) and their effects on gas exchange and activities of antioxidant enzymes. Photosynthetically active radiation varied from 1 254 mu mol m(-2) s(-1) at I(100) to 285 mu mol m(-2) s(-1) at I(30). Stomatal conductance, net photosynthetic rate, and relative chlorophyll (Chl) content were maximal in I(70) plants. Plants grown under I(100) produced leaves with lower Chl content and signs of chlorosis and necrosis. These symptoms indicated Chl degradation induced by the generation of reactive oxygen species. Stress related antioxidant enzyme activities ( Mn-SOD, Fe-SOD, and Cu/Zn-SOD) were highest in I(100) plants, whereas catalase activity was the lowest. Hence P. umbellata is a shade species ( sciophyte), a feature that should be considered in reforestation programs or in field plantings for production of medicinal constituents.

The present research was aimed at standardizing the protocol of seed disinfection, seed germination and organogenesis via callus of Pothomorphe umbellata (L.) Miq.The germinated seeds were inoculated in different concentrations of BAP (benzylamine purine) and NAA (naphthalene acetic acid) in order to stimulate the callus induction. After 60 days of culture, the calluses with some shoots were taken to an organogenesis medium (GA3 0.1 mg.L -1, BAP 0.5 mg.L-1) during 40 days, to be transferred later to a development medium. Finally, the plantules were acclimatized, presenting a good index of survival.

The aim of the present work was to compare the content of carotenoids between callus and regenerated plants of Pothomorphe umbellate. Germinated seeds (40 days old) were inoculated in different concentrations and combinations of BAP (benzylaminopurine) and NAA (naphthalene acetic acid) in order to stimulate the callus' production. After 60 days of culture, the callus containing some shoots were transferred to organogenesis medium (GA 3 0.1 mg L -1, BAP 0.5 mg L -1) for 40 days. Next, they were subcultivated in a medium for seedling growth (without regulators) for 40 days. Callus (collected after 60 days) and seedlings (collected after 140 days) were frozen in liquid nitrogen and kept under -80°C for future carotenoids' analysis. The highest concentration of carotenoids was found in plants cultivated in medium without regulators. The callus did not showed difference concerning the culture medium; however, they presented lower content of carotenoids in relation to plants.

The object of this study was assess through pre-clinical tests, the possible diuretic activity and the effects on the mean arterial pressure of the aqueous and ethanolic extracts of branches of “bugre”, Hedyosmum brasiliense Miq., species used in the treatment of renal and urinary disorders The tests were made in males anesthetized Wistar rats and randomly distributed into 4 experimental groups: Group I – Water control, group II – treated with aqueous extract (EA) of “bugre”, Group III – water control + “tween 80”, group IV – treated with ethanol extract (EE) and “bugre”. All groups were subjected to experimental protocol, composed of three periods: Balance (40 minutes), Basal (30 minutes) and Experimental (90 minutes), occurring the urine collection every 30 minutes, from the basal period and measuring blood pressure every 10 minutes. The results presented validate the ethnobotany indication of the use of H. brasiliense tea in the treatment of renal problems, because increased significantly the urinary flow in anesthetized Wistar rats (diuretic effect), without changing significantly (p>0,05) the arterial pressure

Trichophyton rubrum is a dermatophyte, which can cause infections in human skin, hair and nail. Pothomorphe umbellata (L.) Miq. (Piperaceae) is a native Brazilian plant, in which phytochemical studies have demonstrated the presence of steroids, 4-nerolidylcatechol, sesquiterpenes and essential oils. The objective of this study was to analyze the in vitro activity of extracts and fractions of P. umbellata on resistant strains of T. rubrum. The microdilution plate method was utilized to test Tr1, H6 and Delta TruMDR2 strains of T rubrum; Delta TruMDR2 strain was obtained from H6 by TruMDR2 gene rupture, which is involved in multiple drugs resistance. The highest antifungal activity to all strains was observed for dichloromethane and hexane fractions of the 70% ethanolic extract which showed minimal inhibitory concentration (MIC) and minimal fungicide concentration (MFC) of 78.13 mu g/mL. This antifungal activity was also obtained by 70% ethanolic extract, which presented MIC and MFC of 78.13 mu g/mL to Delta TruMDR2, whereas the MIC values for Tr1 and H6 were 78.13 and 156.25 mu g/mL, respectively. Our results suggest the potential for future development of new antifungal drugs from P umbellata, especially to strains presenting multiple resistance. (C) 2012 Elsevier Masson SAS. Supercritical fluid extraction with CO2, Performed in a home-made system, of rhizomes of Dorstenia bryoniifolia Mart. ex Miq. (Moraceae) and of bark roots of Brosimum gaudichaudii Trecul (Moraceae) afforded crude extracts that were analysed by high resolution gas chromatography (HRGC). The D. bryoniifolia extract contained, besides the previously reported pimpinelin and isobergapten, the furocoumarins psoralen, bergapten, isopimpinelin and the triterpenes alpha- and beta-amyrin and the acetate of the latter. The B. gaudichaudii extract contained a number of terpenoids as well as the previously reported psoralen and bergapten. Supercritical fluid extraction gave extracts qualitatively similar to those obtained by Soxhlet extraction with hexane and, together with off-line HRGC, was shown to be a fast and accurate technique to be used in rapid phytochemical examination.
