The scientific question addressed in this work is: what hides beneath first order kinetic constant k (s(-1)) measured for hybridization of a DNA target on a biosensor surface. Kinetics hybridization curves were established with a 27 MHz quartz microbalance (9 MHz, third harmonic) biosensor, constituted of a 20-base probe monolayer deposited on a gold covered quartz surface. Kinetics analysis, by a known two-step adsorption-hybridization mechanism, is well appropriate to fit properly hybridization kinetics curves, for complementary 20-base to 40-base targets over two concentration decades. It was found that the K-1 (M-1) adsorption constant, relevant to the first step, concerns an equilibrium between non hybridized targets and hybridized pre-complex and increases with DNA target length. It was established that k(2) (s(-1)), relevant to irreversible formation of a stable duplex, varies in an opposite way to K-1 with DNA target length. (C) 2012 Published by Elsevier B.V.

In this work, we synthesized a novel series of hydrogels composed of polyacrylamide (PAAm), methylcellulose (MC), and calcic montmorillonite (MMt) appropriate for the controlled release of fertilizers, where the components presented a synergistic effect, giving very high fertilizer loading in their structure. The synthesized hydrogel was characterized in relation to morphological, hydrophilic, spectroscopic, structural, thermal, and kinetic properties. After those characterizations, the application potential was verified through sorption and desorption studies of a nitrogenated fertilizer, urea (CO(NH2)2). The swelling degree results showed that the clay loading considerably reduces the water absorption capability; however, the hydrolysis process favored the urea adsorption in the hydrogel nanocomposites, increasing the load content according to the increase of the clay mass. The FTIR spectra indicated that there was incorporation of the clay with the polymeric matrix of the hydrogel and that incorporation increased the water absorption speed (indicated by the kinetic constant k). By an X-ray diffraction technique, good nanodispersion (intercalation) and exfoliation of the clay platelets in the hydrogel matrix were observed. Furthermore, the presence of the montmorillonite in the hydrogel caused the system to liberate the nutrient in a more controlled manner than that with the neat hydrogel in different pH ranges. In conclusion, excellent results were obtained for the controlled desorption of urea, highlighting the hydrolyzed hydrogels containing 50% calcic montmorillonite. This system presented the best desorption results, releasing larger amounts of nutrient and almost 200 times slower than pure urea, i.e., without hydrogel. The total values of nutrients present in the system show that this material is potentially viable for application in agriculture as a nutrient carrier vehicle. © 2013 American Chemical Society.

An investigation was made on the adsorption and kinetics of photodegradation of potassium hydrogenphthalate in an aqueous suspension of TiO2. Two models, Langmuir and Freundlich, were used to describe the adsorption process and the model proposed by Langmuir-Hinshelwood (L-H) was employed to describe the kinetics of the photodecomposition reactions of hydrogenphthalate. The results of the adsorptions were fitted to the models proposed by Langmuir and Freundlich. Adsorption was found to be a function of the temperature, with adsorption capacity increasing from 2.4 to 4.5 mg/g when the temperature rose from 20 to 30 degrees C. The kinetic model indicates that the rate constant, k, of the first order reaction, is high in the 10.0 to 100 mg/l interval, which is coherent with the low value of the adsorption constant, K. The results fitted to the L-H model led to an equation that, within the range of concentrations studied here, theoretically allows one to evaluate the photodegradation rate. (c) 2005 Elsevier Ltd. A study was conducted to investigate the thermal constant in an experimental population of Muscina stabulans (Fallén 1817). A natural population was obtained from a livestock and their F1 generations were maintained at four constant temperatures (16°C, 20°C, 26°C and 31°C). The thermal constant was calculated by the hyperbole method using the base temperature, tb = 4.4°C and the thermal constant K = 35.3 GD. Ricker's geometric regression, considering tb = 7.8°C and K = 28.9 GD was also calculated.

Schistosomiasis is still an endemic disease in many regions, with 250 million people infected with Schistosoma and about 500,000 deaths per year. Praziquantel (PZQ) is the drug of choice for schistosomiasis treatment, however it is classified as Class II in the Biopharmaceutics Classification System, as its low solubility hinders its performance in biological systems. The use of cyclodextrins is a useful tool to increase the solubility and bioavailability of drugs. The aim of this work was to prepare an inclusion compound of PZQ and methyl-beta-cyclodextrin (MeCD), perform its physico-chemical characterization, and explore its in vitro cytotoxicity. SEM showed a change of the morphological characteristics of PZQ:MeCD crystals, and IR data supported this finding, with changes after interaction with MeCD including effects on the C-H of the aromatic ring, observed at 758 cm(-1). Differential scanning calorimetry measurements revealed that complexation occurred in a 1:1 molar ratio, as evidenced by the lack of a PZQ transition temperature after inclusion into the MeCD cavity. In solution, the PZQ UV spectrum profile in the presence of MeCD was comparable to the PZQ spectrum in a hydrophobic solvent. Phase solubility diagrams showed that there was a 5.5-fold increase in PZQ solubility, and were indicative of a type A(L) isotherm, that was used to determine an association constant (K(a)) of 140.8 M(-1). No cytotoxicity of the PZQ:MeCD inclusion compound was observed in tests using 3T3 cells. The results suggest that the association of PZQ with MeCD could be a good alternative for the treatment of schistosomiasis.

The functional response between ingestion rate and food concentration was determined for each larval stage of Macrobrachium rosenbergii. Artemia franciscana nauplii were supplied at 2,4, 6, 8, 10 and 12 per milliliter. The nauplii were counted by sight using a Pasteur pipette and transferred to Petri dishes containing 40 ml of brackish water (12 parts per thousand) lying on the top of black plastic. One larva at each stage was individually placed into each Petri dish containing different food density. After 24 h, each larva was removed from the Petri dish and the leftover nauplii were counted. The amount consumed was determined by the difference between the initial and final number of nauplii. Ingestion rate (I) increased as food density (P) increased and was defined by the model I=I-m(1-e(-kP)). The results suggest four levels of ingestion during larval development. The first level includes stages II, III and IV, with average maximum consumption of about 40 nauplii/day; the second level includes stages V and VI, with consumption of approximately 55 nauplii/day; the third level includes stages VII and VIII, with consumption of 80-100 nauplii/day. The fourth level includes stages IX, X and XI, in which the high values for maximum ingestion (Im) exceed the load capacity of the medium. The low values for constant k (that may correspond to the adaptability of the food to prey characteristics, such as, size, mobility, etc.) obtained for stages IX, X and XI indicated that Artemia is not an adequate prey and there is necessity of a supplementary diet. The best relationship between predator and prey seemed to occur during stage IV Results obtained in the present work may subsidize future researches and serve as a guideline for practical considerations of feeding rates. Tillage stimulates soil carbon (C) losses by increasing aeration, changing temperature and moisture conditions, and thus favoring microbial decomposition. In addition, soil aggregate disruption by tillage exposes once protected organic matter to decomposition. We propose a model to explain carbon dioxide (CO2) emission after tillage as a function of the no-till emission plus a correction due to the tillage disturbance. The model assumes that C in the readily decomposable organic matter follows a first-order reaction kinetics equation as: dC(sail)(t)/dt = -kC(soil)(t) and that soil C-CO2 emission is proportional to the C decay rate in soil, where C-soil(t) is the available labile soil C (g m(-2)) at any time (t). Emissions are modeled in terms soil C available to decomposition in the tilled and non-tilled plots, and a relationship is derived between no-till (F-NT) and tilled (F-Gamma) fluxes, which is: F-T = a1F(NT)e(-a2t), where t is time after tillage. Predicted and observed fluxes showed good agreement based on determination coefficient (R-2), index of agreement and model efficiency, with R-2 as high as 0.97. The two parameters included in the model are related to the difference between the decay constant (k factor) of tilled and no-till plots (a(2)) and also to the amount of labile carbon added to the readily decomposable soil organic matter due to tillage (a,). These two parameters were estimated in the model ranging from 1.27 and 2.60 (a(1)) and - 1.52 x 10(-2) and 2.2 x 10(-2) day(-1) (a(2)). The advantage is that temporal variability of tillage-induced emissions can be described by only one analytical function that includes the no-till emission plus an exponential term modulated by tillage and environmentally dependent parameters. Este trabalho teve como objetivo estudar a biologia de Telenomus remus Nixon criado em ovos de Spodoptera frugiperda (J.E. Smith) em diferentes temperaturas. Com base no desenvolvimento do ciclo (ovo-adulto), foram determinados as exigências térmicas e o número de gerações dos parasitóides em temperaturas constantes. Inicialmente, ovos de S. frugiperda, com 24 horas de idade, foram submetidos ao parasitismo por T. remus, durante cinco horas, sendo então transferidos para câmaras climatizadas reguladas para as temperaturas de 15, 20, 25, 28, 31 e 35°C. A duração do ciclo (ovo-adulto) foi influenciada pela temperatura variando de 8,3±0,01 a 47,2±0,01 dias para fêmeas e 8,1±0,01 a 46,8±0,01 dias para machos de T. remus nas temperaturas de 31 e 15°C, respectivamente. A porcentagem de emergência (viabilidade) de T. remus em ovos de S. frugiperda também foi influenciada pela variação da temperatura, apresentando redução significativa apenas nas temperaturas extremas. Foi observada redução na viabilidade a 15°C e não houve desenvolvimento biológico do parasitóide a 35°C. A razão sexual de T. remus não sofreu influência da temperatura. em relação às exigências térmicas, a constante térmica (K) e temperatura base (Tb) foram maiores para as fêmeas de T. remus (158,88 graus dias e 12,52°C) quando comparadas aos machos (154,12 graus dias e 12,59°C). A estimativa do número de gerações anuais para T. remus para machos e fêmeas em condições de laboratório foi respectivamente 5,6 e 5,6; 16,9 e 17,3; 28,3 e 29,0; 35,1 e 36,0; 39,6 e 40,7 nas temperaturas de 15, 20, 25, 28 e 31°C.

Este estudo visou determinar as exigências térmicas do pulgão Myzus persicae (Sulzer), calcular tabelas de vida de fertilidade, e prever a ocorrência de adultos no campo, por meio de modelo de graus-dia. Os pulgões foram mantidos individualmente em gaiolas fixadas em folhas de couve, Brassica oleracea L. var. acephala DC, em laboratório, às temperaturas de 15, 20, 23, 25 e 30ºC. O limite térmico inferior de desenvolvimento (Tb) e a constante térmica (K) foram 2,2ºC e 165,6 graus-dia, respectivamente. O modelo de graus-dia previu a ocorrência de adultos de M. persicae para 0 a1 dia antes da data em que eles foram observados no campo. Os parâmetros das tabelas de vida estimados na escala de tempo em graus-dia evidenciaram que as temperaturas de 23°C e 25°C foram as que proporcionaram as melhores condições térmicas para o crescimento populacional de M. persicae. Nessas temperaturas observou-se o maior valor da capacidade inata de aumentar em número (r m = 0,012), o menor valor da duração média da geração (T = 303,8 graus-dia e 272 graus-dia, respectivamente) e o menor tempo necessário para a população duplicar em número de indivíduos (TD = 57,8 graus-dia).

O desenvolvimento (ovo-adulto) da mosca-branca, Bemisia tabaci (Genn.) biótipo B (=Bemisia argentifolii, Bellows & Perring), foi estudado em condições de campo e em câmaras climatizadas, reguladas nas temperaturas constantes de 15, 20, 25, 30 e 35±1ºC, usando soja, Glycine max (L.) Merrill, cultivar Msoy 8001, como planta hospedeira. O limite térmico inferior de desenvolvimento (Tb) e a constante térmica (K) das fases de ovo, ninfa e ciclo biológico (ovo-adulto) foram 11,1ºC / 98,8 graus-dia, 6,8ºC / 383,8 graus-dia e 8,3ºC / 472,6 graus-dia, respectivamente. Nas temperaturas extremas (15ºC e 35ºC), a viabilidade dos insetos mostrou-se menor. No campo, a emergência de 70% dos adultos ocorreu depois de terem sido acumulados 413,2 graus-dia. O modelo de graus-dia utilizado mostrou-se adequado para a previsão de ocorrência de adultos de Bemisia tabaci biótipo B no campo, podendo também ser utilizado para determinação do número de gerações nas diferentes regiões do País.

O objetivo deste trabalho foi determinar as exigências térmicas do pulgão-da-couve, Brevicoryne brassicae L., Hemiptera: Aphididae, em condições de laboratório e campo, e prever a ocorrência de picos populacionais de adultos, por meio de um modelo de graus-dia. em laboratório, os experimentos foram conduzidos em câmaras climatizadas reguladas a 15, 20, 25, 27 e 30ºC, obtendo-se o limite térmico inferior de desenvolvimento (Tb = 4,5ºC) e a constante térmica (K = 176,1 graus-dia). Na determinação das exigências térmicas sob condições de campo, realizaram-se seis experimentos, a partir dos quais obteve-se o tempo médio de desenvolvimento de ninfas e a temperatura média ambiental. Os resultados estimados de Tb e K foram 3,5ºC e 180,0 graus-dia, respectivamente. Os resultados de Tb e K obtidos em laboratório possibilitam prever com maior precisão a ocorrência de adultos de B. brassicae do que os resultados de Tb e K determinados a partir de experimentos em campo.
