Quantitative variations in heterotrophic plate count (HPC) and in the presence of indicator microorganisms in 0.5, 1.5 and 20-L bottles of different brands of Brazilian mineral water were analyzed during their shelf life. No variations were identified in the presence of indicator microorganisms, but quantitative variations in HPC were observed in some brands, which suggests that changes may be occurring in the water quality during storage. The aim of this study was also to evaluate the quality of the bottled mineral waters and the presence of enterococci and Pseudomonas aeruginosa were verified in six and two bottles, respectively, which is in disagreement with the microbiological quality criteria established in the current legislation. Although no limit is set for HPC in mineral water, this study relies on the limit of 500 colony-forming units per mL of sample (CFU/mL). Seventy-two bottles presented levels above 500 CFU/mL and up to 560,000 CFU/mL. This study showed that the control of HPC (<500 CFU/mL) for non-returnable packaging seems to be adequate to ensure the quality of mineral water during storage. The high values of HPC and its variations detected during storage seem to fully justify the need for a reevaluation of the use of HPC in bottled mineral water quality management. More detailed studies on the potential health risk of HPC and its variations in mineral water are also needed. © 2012 Elsevier Ltd.

The microbiological quality of bottled mineral water of various domestic brands sold in Brazil was investigated, with particular focus on the heterotrophic plate count (HPC). Neither total coliforms nor Escherichia coli were found in any 1.5 L bottle samples. Total coliforms were found in 2.9% of the small bottles, while in 20 L bottles the presence of total coliforms and E. coli was demonstrated in 15.5 and 2.4% of samples, respectively. Pseudomonas aeruginosa was detected in 4.3, 4.5 and 9.5% of small, 1.5 and 20 L bottles, respectively. In 36.4% of the samples of 1.5 L bottles, the HPC was above 500 cfu/mL. This percentage of samples with an HPC above 500 cfu/mL increased to 52.0 and 61.9% in small and 20 L bottles, respectively. Higher contamination by total coliforms, E. coli, P. aeruginosa and HPCs occurred in 20 L bottles. In conclusion, several samples in this study were outside the international quality standard for mineral water and the large number of samples with high HPCs shows that more work must be done on the use of HPC in mineral water and the damaging effects that these microorganisms may cause to humans. The bottled mineral water was confirmed as a particularly important public health problem, due to the poor microbiological quality of the products that are marketed. © IWA Publishing 2012.

Water treatment uses chlorine for disinfection causing formation of trihalomethanes. In this work, an electrolytic water pre-treatment was studied and applied to the water from a fountainhead. The action against microorganisms was evaluated using cast-iron and aluminum electrodes. Assays were made in laboratory using the electrolytic treatment. After 5 min of electrolysis the heterotrophic bacteria count was below 500 cfu/mL and complete elimination of total and fecal coliforms was observed. Using electrolytic treatment as a pretreatment of conventional tap water treatment is proposed.

Estudou-se o reuso de água de um sistema composto por estação de tratamento de esgoto (ETE) com aeração prolongada e lodo ativado, e em uma estação compacta de tratamento de água (ECTA) de uma indústria metalúrgica. Os processos para obtenção da água de reuso foram: microbiológico e físico-químico. O esgoto doméstico foi bombeado para a ETE, onde houve formação de flocos biológicos e água clarificada. Avaliou-se a eficiência do processo microbiológico da ETE mediante a remoção de demanda bioquímica de oxigênio (DBO), demanda química de oxigênio (DQO) e sólidos sedimentáveis (SS). A eficiência do processo físico-químico de clarificação e desinfecção foi avaliada mediante análises de pH, turbidez, cor, contagem de bactérias heterotróficas aeróbias, cloro livre, dureza, alcalinidade, cloretos, sulfatos, sólidos totais dissolvidos (STD). Na água de reuso além desses parâmetros avaliou-se a toxicidade aguda ao microcrustáceo Daphnia similis.

A proteólise do leite UAT/UHT durante a estocagem à temperatura ambiente é um dos fatores limitantes de sua vida de prateleira. Neste trabalho, dois lotes de leite cru contendo 10 amostras cada e, posteriormente ao processamento, dois lotes de leite UAT/UHT contendo 25 amostras cada foram colhidos em um laticínio para a contagem de microrganismos psicrotróficos (leite cru) e para o estudo do comportamento reológico e o índice proteolítico (leite UAT/UHT durante 120 dias de estocagem). Para a contagem de microrganismos psicrotróficos, foi utilizada a técnica da contagem padrão em placas. Para a determinação do índice proteolítico, foi determinada a presença de glicomacropeptídeo livre por espectrofotometria a 470 nm. A determinação dos parâmetros reológicos foi efetuada à temperatura ambiente, em quintuplicata em um reômetro de cone e placa. Houve aumento da proteólise no decorrer do armazenamento e aumento da viscosidade aparente após 60 dias de estocagem, provavelmente relacionados à presença de proteases de bactérias psicrotróficas do leite cru.

The aims of this study were to estimate the changes in total bacterial counts (TBC) in poultry litter samples, consisting of rice hulls, after storage, and to identify pathogenic bacteria. For the countings Plate Count agar (Difco) was used. Enrichment and selective media such as blood agar, MacConkey, Baird Parker, brain and heart agar, and egg yolk solid media, and cooked meat and brain and heart infusion, incubated under aerobic or anaerobic conditions were used to isolate Staphylococcus aureus, Salmonella sp, Clostridium perfringens, C. botulinum, C. chauvoei, Campylobacter sp, Escherichia coli, and Corynebacterium sp. Litter samples were collected from the houses of the Veterinary School experimental aviary. A fully randomized experimental design was used with four treatments and four replications, for a total of 16 samples. A decrease in TBC was detected when treatment T1 (zero days of storage) was compared with treatments T2 (14 days of storage). on the other hand the treatments T3 (28 days of storage) and T4 (42 days of storage) presented significantly superior counting in relation to treatment T1. Some pathogenic bacteria of Enterobacteriaceae such as Escherichia coil, Proteus, Arizona, Providencia, Edwardsiella, as well as Staphylococcus aureus, S. epidermidis, different species of genus Clostridium as C. perfringens, C. sordelli, C. chauvoei, C. tetani and C. novyi as well as some strains of Corynebacterium pyogenes were isolated.

The effectiveness of a new management tool for screening refrigerated raw milk quality in a dairy industry is provided. Three hundred and seventy-four samples of raw milk from individual producers and 125 samples from bulk milk carriers from the same producers were analyzed. Producers' samples underwent a fast tube reduction test, or Tetra-Test, to estimate total bacteriological charge and their predominant microbiota. Bulk milk samples underwent standard plate count (SPC) as reference method. Tetra-test was effective in screening and assessing the microbiological quality of refrigerated raw milk, since 82.4% of samples with more than 106UFC/mL were readily detected (5 minutes to 2h). The test may be employed as a management tool either in screening or in interventions on the detected problem. In fact, its results provided complementary information on the characteristics of dominant microbiota. Most samples failed in compliance to IN 51 requirements. Further, 211 samples (56.4%) showed high microbial charge and, consequently, poor quality raw milk. Psychrotrophic groups were not yet the main degrading factors of cooled raw milk, albeit present in 76.8% of samples.
