A população de cervo-do-pantanal (Blastocerus dichotomus) está drasticamente reduzida no Brasil. O nosso objetivo foi o de estimar a abundância do cervo-do-pantanal na bacia do Rio Paraná e discutir a metodologia aplicada. Os resultados darão suporte para uma análise do impacto do enchimento da represa de Porto Primavera sobre essa população. Sessenta e nove animais foram registrados através de sobrevôo utilizando-se a metodologia de transecção linear com amostragem das distâncias. Os dados não corrigidos resultaram em uma densidade estimada de 0,0035ind/ha e uma população de 636 indivíduos. A correção de g para os animais que não foram vistos apresentou uma densidade de 0,0049 ind/ha e uma abundância de 896 (CV=0,27) indivíduos. A metodologia foi aplicada com sucesso na estimativa de cervo-do-pantanal. Esse resultado é importante para avaliarmos a população do cervo-do-pantanal na área e para futuramente analisarmos o impacto do enchimento da represa.

A presente pesquisa teve como objetivo estudar os efeitos fitotóxicos de fungicidas, acaricidas e inseticidas e algumas associações entre eles, em plantas de mamoeiros (Carica papaya L.) cv. Sunrise Solo Improved Line 72/12, em condições de campo, no município de São Mateus -- ES, pertencente à maior região produtora do Estado. O experimento foi arranjado em delineamento de blocos casualizados, com 4 repetições e 03 plantas úteis por parcela. Foram utilizados os seguintes produtos, com as respectivas doses, para cada 100 L de água: chlorothalonil (Daconil PM-200g); mancozeb (Dithane PM -- 200g); oxicloreto de cobre (Reconil -- 400g); thiabendazole (Tecto 450 -- 100ml); dicofol + tetradifon (Carbax -- 200ml); triazophós (Hostathion 400 BR -- 150ml); óxido de fenbutatina (Torque 500 SC -- 60ml); e abamectin (Vertimec 18 CE -- 50ml): Analisou-se a fitotoxicidade dos produtos testados, em relação à altura da planta, nº de folhas, número de flores e frutos ; diâmetro do caule e queimaduras ou injúrias foliares. As datas das avaliações foram: 01 dia antes das pulverizações, 15 dias e 30 dias após as mesmas. Os fungicidas Daconil BR, Reconil e Tecto 450; o fungicida acaricida Dithane PM; os acaricidas Carbax e Torque 500 SC; e o inseticida-acaricida Vertimec 18 CE, aplicados isoladamente, não afetaram o crescimento e a produção das plantas, nem causaram injúrias nas folhas das mesmas. A associação de fungicidas e fungicida-acaricida, com os acaricidas, ou inseticida-acaricida, não mostrou nenhum efeito fitotóxico sobre os parâmetros de crescimento avaliados, nem causaram queimaduras ou injúrias foliares.

Foram utilizados os seguintes produtos, com as respectivas doses, para cada 100 l de água: abamectin (Vertimec 18 CE - 50 ml); dicofol + tetradiphon (Carbax - 200 ml), fenbutatin oxide (Torque 500 SC - 60 ml); mancozeb (Dithane PM - 200g); oxicloreto de cobre (Reconil - 400g) e thiabendazole (Tecto 450 - 100ml). Analisou-se a fitotoxicidade dos produtos em relação à altura da planta, nº de folhas, número de flores e frutos ; diâmetro do caule e queimaduras ou injúrias foliares. As medições e contagens foram feitas um dia antes das pulverizações, 15 e 30 dias após. Constatou-se que o Vertimec 18 CE, associado ao Reconil ou ao Tecto 450, ocasionou leves injúrias foliares, detectadas aos 15 dias após as pulverizações, que se tornaram praticamente imperceptíveis, aos 30 dias após as pulverizações; e que Dithane PM, Reconil, Tecto 450, Carbax, Torque 500 SC, Dithane PM + Carbax, Dithane PM + Torque 500 SC, Dithane PM + Vertimec 18 CE, Reconil + Carbax, Reconil + Torque 500 SC, Tecto 450 + Carbax e, Tecto 450 + + Torque 500 SC não interferiram nos parâmetros de desenvolvimento e de produção estudados, bem como não causaram injúrias ou queimaduras nas folhas dos mamoeiros.

The cellular and molecular characteristics of a cell line (BME26) derived from embryos of the cattle tick Rhipicephalus (Boophilus) microplus were studied. The cells contained glycogen inclusions, numerous mitochondria, and vesicles with heterogeneous electron densities dispersed throughout the cytoplasm. Vesicles contained lipids and sequestered palladium meso-porphyrin (Pd-mP) and rhodamine-hemoglobin, suggesting their involvement in the autophagic and endocytic pathways. The cells phagocytosed yeast and expressed genes encoding the antimicrobial peptides (microplusin and defensin). A cDNA library was made and 898 unique mRNA sequences were obtained. Among them, 556 sequences were not significantly similar to any sequence found in public databases. Annotation using Gene Ontology revealed transcripts related to several different functional classes. We identified transcripts involved in immune response such as ferritin, serine proteases, protease inhibitors,. antimicrobial peptides, heat shock protein, glutathione S-transferase, peroxidase, and NADPH oxidase. BME26 cells transfected with a plasmid carrying a red fluorescent protein reporter gene (DsRed2) transiently expressed DsRed2 for up to 5 weeks. We conclude that BME26 can be used to experimentally analyze diverse biological processes that occur in R. (B.) microplus such as the innate immune response to tick-borne pathogens. O temperamento de quatro raças bovinas foi avaliado utilizando-se o teste de velocidade de fuga (FT) e o escore de comportamento (BST). FT foi definida como o tempo necessário para animais percorrerem uma distância de 2 m após a pesagem. BST foi baseada no comportamento dos animais na balança, amostrando-se quatro categorias de comportamento: movimentos, intensidade de respiração, vocalizações e coices. Os coeficientes de herdabilidade de FT e BST foram estimados com uso de um modelo de máxima verossimilhança restrita, considerando meio irmãos paternos. Caracu apresentou menores médias para BST do que as demais raças. Nelore apresentou resultados intermediários, seguida por Guzerat e Gyr com médias mais elevadas (p < 0,05). Resultados similares foram observados para FT, mas as médias de Caracu e Nelore não diferiram entre si. Observou-se baixa associação entre FT e BST (r p= -0,36; p < 0,01). A correlação entre rank de touros ordenados pelos seus valores preditos (p) para FT e BST foi moderada e negativa (r s = -0,63; p < 0,001). A herdabilidade de FT e BST foi de 0,35 e 0,34, respectivamente. A comparação de rebanhos Nelore com diferentes critérios de seleção para peso corporal mostrou que linhas de seleção podem modular positivamente o temperamento de Bos indicus.

The Hazard Analysis and Critical Control Point (HACCP) is a preventive system that intends to guarantee the safety and harmlessness of food. It improves the quality of products as it eliminates possible defects during the process, and saves costs by practically eliminating final product inspection. This work describes the typical hazards encountered on the mushroom processing line for fresh consumption. Throughout the process, only the reception stage of mushrooms has been considered a critical control point (CCP). The main hazards at this stage were: the presence of unauthorised phytosanitary products; larger doses of such products than those permitted; the presence of pathogenic bacteria or thermo-stable enterotoxins. Putting into practice such knowledge would provide any industry that processes mushrooms for fresh consumption with a self-control HACCP-based system for its own productions.

Poultry meat and its derivatives are among the foodstuffs considered by environmental health authorities to present the highest risks to the public. A total of 185 samples were collected in five monthly batches, from different processing stages in a sausage plant that uses mechanically-deboned chicken meat (MDCM) and tested for the presence of Salmonella. Enrichment was carried out in both Kauffman tetrathionate broth and Rappaport-Vassiliadis broth and isolation on Salmonella-Shigella agar and brilliant-green agar. Live Salmonella bacteria were isolated from six samples of the raw meat and from the emulsion, in batches three, four and five, but not from any sample in batches one or two. The six isolated strains were all classified as Salmonella Albany, which has not previously been reported in MDCM. of the two enrichment broths, Rappaport-Vassiliadis gave the better results. The Pattern of contamination suggests a probable common source, given that a new supplier was used in the third, fourth, and fifth months. It was also shown that the industrial cooking was effective in preventing Salmonella surviving in the final product.

A high performance liquid chromatography (HPLC) method for extraction and determination of pesticides from raw milk was developed. The method involves direct injection of raw milk samples on a bovine serum albumin-dimethyl-octyl-silica gel (BSA-Si-Cs) column. The mobile phase 0.05 mol.L-1 phosphate buffer pH6.0 in acetonitrile (70:30 v/v) was employed for extraction and separation of bendiocarb, methylparathion, pentachlorophenol, and methomyl pesticides. The method shows good results of recovery in the pesticides studied, higher than 99.6%.
Off line extraction of phenol from human urine sample with isoamyl alcohol and determination by HPLC

This method has been developed for extraction and determination of phenol in a urine sample by high performance liquid chromatography.After acid hydrolysis, the free phenol was extracted with isoamyl alcohol solvent, followed by back extraction with 0.5 mol.L-1 sodium hydroxide solution and analyzed by an isocratic HPLC Varian System, equipped with reverse-phase column (MicroPak-C-18). The mobile phase was acetonitrile in 0.01 mol.L-1 hydrochloric acid solution, (20:80 v/v), and at a now-rate of 1.0 mL.min-1. The chromatogram was monitored at 220 nm in room temperature. The identification was based on retention time and the quantification was performed by automatic peak-area determination, corrected for the external standards method.The recovery was higher than 99.5 % for phenol and reproducibility of method was shown to be 2.3% standard deviation and 5.6% coefficient of variance (n=20). The limit detection was 0.05 mgL(-1) and a range of 0.05 to 20.0 mgL(-1) of phenol for linearity.

In the present study, two alkaloids isolated from Pterogyne nitens, a plant native to Brazil, have been shown to induce apoptosis in human breast cancer cells. These compounds, pterogynine (PGN) and pterogynidine (PGD), were tested for their effect on a human infiltrating ductal carcinoma cell line (ZR-7531). The cell line was treated with each alkaloid at several concentrations. Time-dependence (with or without recuperation time) and concentration-dependence (in the range 0.25-10 mM) were investigated in cytotoxicity and apoptosis assays. The annexin assay indicated an apparently higher percentage of death by necrosis of malignant cells after 24 h exposure to both P. nitens extracts than the Hoechst assay. Thus, our results in the two tests demonstrated that the Hoechst assay can discriminate between late apoptotic cells and necrosis, whereas the flow cytometry-based annexin V assay cannot. We concluded that PGN and PGD have effective antineoplastic activity against human breast cancer cells in vitro, by inducing programmed cell death.

The distribution of natural gas is carried out by means of long ducts and intermediate compression stations to compensate the pressure drops due to friction. The natural gas compressors are usually driven by an electric motor or a gas turbine system, offering possibilities for energy management, one of these consisting in generating energy for use in-plant or to commercialize as independent power producer. It can be done by matching the natural gas demand, at the minimum pressure allowed in the reception point, and the storage capacity of the feed duct with the maximum compressor capacity, for storing the natural gas at the maximum permitted pressure. This allows the gas turbine to drive an electric generator during the time in which the decreasing pressure in duct is above the minimum acceptable by the sink unit. In this paper, a line-pack management analysis is done for an existing compression station considering its actual demand curve for determining the economic feasibility of maintaining the gas turbine system driver generating electricity in a peak and off-peak tariff structure. The potential of cost reduction from the point of view of energy resources (natural gas and electric costs) is also analyzed. (C) 2010 Elsevier Ltd. All rights reserved.
