This work shows the chemical characterization of a dye processing plant effluent that was contributing to the mutagenicity previously detected in the Cristais river, São Paulo, Brazil, that had an impact on the quality of the related drinking water. The mutagenic dyes Disperse Blue 373, Disperse Orange 37 and Disperse Violet 93, components of a Black Dye Commercial Product (BDCP) frequently used by the facility, were detected by thin layer chromatography (TLC). The blue and orange dyes were quantified by high performance liquid chromatography (HPLC/DAD) in a raw and treated effluent samples and their contribution to the mutagenicity was calculated based on the potency of each dye for the Salmonella YG1041. In the presence of S9 the Disperse Blue 373 accounted for 2.3% of the mutagenic activity of the raw and 71.5% of the treated effluent. In the absence of S9 the Disperse Blue 373 accounted for 1.3% of the mutagenic activity of the raw and 1.5% of the treated effluent. For the Disperse Orange 37, in the presence of S9, it contributed for 0.5% of the mutagenicity of the raw and 6% of the treated effluent. In the absence of S9; 11.5% and 4.4% of the raw and treated effluent mutagenicity, respectively. The contribution of the Disperse Violet 93 was not evaluated because this compound could not be quantified by HPLC/DAD. Mutagenic and/or carcinogenic aromatic amines were also preliminary detected using gas chromatograph/mass spectrometry in both raw and treated and are probably accounting for part of the observed mutagenicity. The effluent treatment applied by the industry does not seem to remove completely the multagenic compounds. The Salmomella/microsome assay coupled with TLC analysis seems to be an important tool to monitor the efficiency of azo dye processing plant effluent treatments. (c) 2006 Elsevier B.V. In this study, micronucleus (MN) and chromosome aberration (CA) tests in Allium cepa (onion) were carried out in order to make a preliminary characterization of the water quality of the Atibaia River in an area that is under the influence of petroleum refinery and also to evaluate the effectiveness of the treatments used by the refinery. For these evaluations, seeds of A. cepa were germinated in waters collected in five different sites related with the refinery in ultra-pure water (negative control) and in methyl methanesulfonate solution (positive control). According to our results, we can suggest that even after the treatments (physicochemical, biological and stabilization pond) the final refinery effluent could induce chromosome aberrations and micronucleus in meristematic cells of A. cepa and that the discharge of the petroleum refinery effluents in the Atibaia River can interfere in the quality of this river. Studies to determine suitable levels of intensification are essential for developing sustainable aquaculture. The objective of this study was to evaluate the quality of effluents discharged from ponds stocked with 10 (D10), 20 (D20), 40 (D40), and 80 (D80) postlarvae of Macrobrachium amazonicum/m2. Intake and effluent water samples were taken throughout a 5.5-mo grow-out cycle. In that study, twelve 0.01-ha earthen ponds were stocked postlarvae with 0.01g. Average water exchange rate was 15%/d; water was discharged from the bottom of the ponds. Prawns were fed a commercial feed with 38% crude protein according to their biomass (3-10%) and the concentration of dissolved oxygen (DO). In our research, temperature, turbidity, total suspended solids, conductivity, DO, pH, biochemical oxygen demand (BOD), chemical oxygen demand (COD), N-ammonia, N-nitrite, N-nitrate, N-Kjeldahl nitrogen, total phosphorus, and soluble orthophosphate were measured every 15d throughout the experiment in the early morning (0630 to 0730h). Turbidity was lower in D10 than in D20 and D40 and total phosphorus was higher in D80 than in D10 and D20. An analysis of principal components comparing treatments and intake water showed three groups: intake, D10 and a cluster of D20, D40, and D80. On the basis of the water characteristics found in our study it appears that the farming of M. amazonicum is likely to have a low environmental impact, at least up to a stocking density of 80prawns/m2. © by the World Aquaculture Society 2013.

Objetivou-se avaliar o efeito da inclusão de aditivos químicos na ensilagem de resíduos (entrecasca) da produção de palmito pupunha (Bactris gasipaes, Kunth). Utilizaram-se silos experimentais (baldes de 20 litros) providos de aparatos para determinação gravimétrica de perdas por gases e efluentes. Os tratamentos aplicados foram: controle (sem aditivos); ureia (1% da MV) e cal virgem (1% da MV). Decorridos 70 dias de armazenagem, os silos foram pesados, abertos e amostrados. As perdas por efluentes e gases aumentaram com a aplicação de cal virgem na ensilagem. As perdas totais de MS foram de 15,1; 14,4 e 26,6% nas silagens controle, ureia e cal, respectivamente. em todas as silagens, houve redução no teor de FDN e elevação da fração FDA, o que indica desaparecimento da fração hemicelulose. A relação cálcio:fósforo aumentou substancialmente com a adição de cal virgem, de 4,1:1 na silagem controle para 15,6:1 na silagem com cal. O resíduo da extração do palmito pupunha pode ser classificado como alimento de média qualidade e alto teor de umidade. Os aditivos aplicados na ensilagem não são efetivos em reduzir as perdas fermentativas no processo de conservação.

The objective of this trial was to evaluate the nutritional value, fermentation profile and dry matter losses of Palisadegrass silages ensiled with either dried citrus pulp, soybean hulls, chemical or microbial additives. The trial was carried out in a completely randomized experimental design and in a factorial arrangement (3 x 5), with three dry matter levels (wet forage or forage ensiled with pelleted citrus pulp or pelleted soybean hulls) and five additives (without or with the presence of bacterial inoculants or the addition of: sodium benzoate, formic acid in the concentration of 62% or 44%), totalizing 15 treatments and 60 experimental silos. The variables analyzed were: nutritional value, losses due to gases and effluents, and dry matter recovery. The use of dried citrus pulp or soybean hulls at the ensiling time increased the dry matter content (29.4 and 28,9%) and decreased the effluent production (4.1 and 3.8 kg/t of fresh matter), also providing fermentable substrate to microorganisms, resulting in increased fermentation coefficient and digestibility of silages. The use of formic acid resulted in silages with higher digestibility and increased water-soluble carbohydrates and crude protein content. This additive was also effective in reducing the losses due to gases and, as a result, increased the total dry matter recovery. The treatment containing homolactic bacteria showed similar trend of increasing the digestibility and reducing the losses due to gases. The use of sodium benzoate was less effective in altering the fermentation pattern of tropical silages. The nutritional value and total dry matter losses of silages ensiled without additives can be considered satisfactory. However, wet forage ensiled with dried citrus pulp and, mainly, with soybeans hulls showed the best results. Treatments containing formic acid had a beneficial effect on the fermentation profile of tropical grass silages.

O reaproveitamento de nutrientes após a fermentação de resíduos orgânicos em biodigestor, associado à técnica do cultivo hidropônico, é uma alternativa para reduzir custos na agricultura, além de contribuir no menor consumo das reservas naturais de nutrientes do planeta. Com esse enfoque, estudou-se o comportamento da cultura do meloeiro usando a técnica hidropônica e efluente de biodigestor proveniente da fermentação anaeróbica de estrume bovino. Cultivou-se o meloeiro (Cucumis melo L. 'Bônus 2') em condições de ambiente protegido. O delineamento estatístico utilizado foi de blocos casualizados, com 4 tratamentos (cultivo hidropônico em sistema fechado tipo NFT com uso de solução nutritiva organo-mineral; cultivo hidropônico em sistema fechado tipo NFT com uso de solução nutritiva 100% mineral; cultivo em sistema aberto, com substrato e solução nutritiva organo-mineral e cultivo em sistema aberto, com substrato e solução nutritiva 100% mineral) e 6 repetições. Foram realizadas avaliações quanto a altura de plantas aos 50; 56; 63; 70 e 77 dias após a semeadura; tempo de colheita; peso de frutos e produtividade. As melhores respostas foram observadas no cultivo hidropônico em sistema fechado tipo NFT com uso de solução nutritiva 100% mineral. A substituição parcial de adubos minerais por biofertilizante, se mostrou viável para os tratamentos em sistema aberto (com substrato), constituindo-se em masi uma alternativa aos horticultores.

A produção intensiva de alimentos exige manejo adequado do solo para garantir a produtividade e a sustentabilidade ambiental. Uma das alternativas é a utilização de resíduos orgânicos no desenvolvimento das culturas, diminuindo a dependência de adubos minerais. Com o objetivo de avaliar o desenvolvimento da cultura de feijão (Phaseolus vulgaris L.), utilizando biofertilizante e adubação mineral, conduziu-se o experimento com seis tratamentos dispostos ao acaso, em esquema fatorial, em quatro blocos, com parcelas de 8,0 x 5,0 m. Os tratamentos sob solo cultivado com a cultura de feijão caracterizaram-se como: com e sem biofertilizante (CB e SB, respectivamente) e para a adubação mineral foram utilizadas a dose recomendada no plantio, ½ dose de adubação e sem adubação mineral (AM, 1/2AM, SAM). Adotaram-se práticas culturais convencionais para o preparo inicial do solo, e em seguida foi efetuada a aplicação de biofertilizante de origem bovina na dosagem de 100 m³ ha-1, com antecedência de três meses da semeadura. Foram avaliados os parâmetros massa da matéria seca acumulada na parte aérea da planta, área foliar e produtividade da cultura. Os resultados mostraram semelhanças entre as características analisadas, obtendo-se melhor desenvolvimento à cultura que recebeu biofertilizante.

In vitro-matured (IVM) bovine oocytes were activated with single and combined treatments of strontium (S), ionomycin (1) and 6-DMAP (D). Using oocytes IVM for 26 h, we observed that activation altered cell cycle kinetics (faster progression, MIII arrest, or direct transition from MII to pronuclear stage) when compared to in vitro fertilization. The effect of oocyte age on early parthenogenesis was assessed in oocytes IVM for 22, 26 and 30 h. Better results in pronuclear development were obtained in treatments ISD (81.7%) at 22 h; D (66.7%), IS (63.3%), ID (73.3%) and ISD (76.7%) at 26 h; and D (86.7%), IS (85.0%) and ID (78.3%) at 30 h. Higher cleavage occurred on ISD (80.0%) at 22 h; ID (83.3%) and ISD (91.7%) at 26 h; and 1 (86.7%), IS (90.0%), ID (85.0%) and ISD (95.0%) at 30 h. More blastocysts were achieved in ID (25.0%) and ISD (18.3%) at 22h; and in ID at 26h (45.0%) and 30h (50.0%). We also observed that IS allowed higher haploid (77.4%) embryonic development, whilst ID was better for diploid (89.1%) development. It was concluded that association of S and D without I was not effective for blastocyst development; treatments using S were less influenced by oocyte age, but when S was associated with D there was a detrimental effect on aged oocytes; treatment ISD promoted higher activation and cleavage rates in young oocytes and ID protocol was the best for producing blastocysts.

1. The relationship between repeated thermal treatments and hepatic synthesis of Hsp 70 was studied in broiler chickens.2. Sixty broilers were submitted to 5 different treatments (12 birds each) from day 1 to day 42. Four groups were kept in a thermoneutral environment and subjected to 0, 1, 2 and 3 heat stress episodes at 35 degrees C for 4 h per week (TN-0, TN-1, TN-2 and TN-3, respectively). The last group (HT-35) was reared at a room temperature of 35 degrees C.3. From 39 to 42 old, the birds experienced acute heat stress at 41 degrees C. Resistance to heat stress was evaluated by the time taken for rectal temperature to increase by 3 degrees C above the pre-treatment value. Livers were collected (before and after heat stress) and Hsp70 was determined using Western Blot analysis with monoclonal anti-Hsp70 antibody.4. Resistance to heat stress and concentration of Hsp70 were higher in those birds subjected to more heat stress episodes during the experimental period (TN-3) and HT-35. A positive correlation was observed between Hsp70 concentration and the time taken for a 3 degrees C increase in rectal temperature (r=0.42; P<0.01).5. Exposing birds to episodes of heat stress (35 degrees C) during rearing may improve their resistance to acute heat stress, but the previous thermal history did not seem to influence the hepatocyte Hsp70 content after exposure to more severe heat stress (41 degrees C).
