The extreme use of ethanol causes metabolic and pathologic changes in testes and urogenital system in different animal species. The enzyme alcohol dehydrogenase (ADH) catalyses the conversion of ethanol into carcinogenic metabolite acetaldehyde which is partly excreted into the urine. However, papers relating the chronic ethanol consumption to the urethral morphology are unknown. This work evaluates the toxic effect of the chronic ethanol ingestion on the urethral epithelium of UChA and UChB rats. Conventional techniques of histology, histochemistry, immunohistochemistry and ultrastructural analysis were used. The analysis showed the presence of lipid drops and intercellular spaces in the epithelial cells in the urethra of UChA and UChB rats compared to control rats. Urethral neuroendocrine cell were observed and characterized for presenting vesicles containing electron-dense granules associated with nervous fibers. We conclude that the chronic consumption of ethanol induces the presence lipid drops in the epithelial cells of the urethra of UChA and UChB rats. The NE cells of the urethra of UChA and UChB rats did not show alterations under chronic effect of the ethanol. (C) 2007 Elsevier Ltd. All rights reserved.

1. Intracerebroventricular (I.C.V.) infusion (60 ng h-1) of Isoleu5-angiotensin II (Isoleu5-AngII) and des-amine-angiotensin II (des-amine-AngII) in rats caused increased drinking behaviour and an increase in arterial blood pressure.2. Des-amine-AngII caused similar increases in heart rate and arterial blood pressure as AngII.3. Previous I.C.V. injection of the antagonists [Leu8]-AngII, des-amine-[Leu8]-AngII and octanoyl-[Leu8]-AngII prevented the increases in heart rate and blood pressure produced by I.C.V. infusion of AngII and caused partial reduction of the dipsogenic response.4. The three antagonists had no effect on the increase in arterial blood pressure and heart rate caused by des-amine-AngII. The drinking response was reduced by previous injection of [Leu8]-AngII and des-amine-[Leu8]-AngII but not by octanoyl-[Leu8]-AngII.5. In conclusion, Isoleu5-AngII and des-amine-AngII increase drinking behaviour, arterial blood pressure and heart rate when infused into the cerebral ventricle of rats. The study with the antagonists showed that des-amine-AngII probably binds more strongly to AngII-receptors.

The purpose of this study was to estimate the alcoholic beverage consumption among undergraduate students of a Dentistry Course and compare the effectiveness of screening methods for the detection of binge drinking. A total of 284 undergraduate students participated in the research. We used the full AUDIT and their reduced forms (AUDIT-3 and AUDIT-C). In addition, a ROC curve was created and its area was calculated. Of the students, 91.0% and 90.6% of the male and female genders, respectively, reported alcohol beverage consumption. Bing-drinking behavior occurred in 69.2% of the men and in 52.4% of the women. The 3 methods showed good discriminatory ability. For the women, there was a significant difference between the AUDIT-3 and AUDIT-C. Both the complete version of AUDIT and its reduced forms presented good discriminatory ability

Purpose: To investigate the combined effects of reflux of duodenal contents through the pylorus and treatment with N-methyl-N'-nitro-nitrosoguanidine ( MNNG) on the development of lesions in the glandular stomach, at the gastrojejunal anastomosis and in the forestomach of rats. Methods: Eighty Male Wistar rats were divided into 4 groups: G1: MNNG + Reflux, G2: Reflux, G3: MNNG and G4: Gastrostomy. MNNG was given in the drinking water ( 100 mg/ml) for 12 weeks and then two groups ( G1 and G2) were submitted to a gastrojejunal anastomosis followed by section of the afferent loop and suture of both stumps to allow reflux of duodenal contents through the pylorus. The animals were sacrificed 18 and 36 weeks after surgery. The lesions obtained in the antral mucosa, at the gastrojejunal anastomosis and in the forestomach were analysed histologically. Results: Duodenal reflux induced proliferative lesions at both glandular and squamous mucosa of the stomach. In the antrum, adenomatous hyperplasia (AH) was observed in 20% and 50% of the animals at the 18(th) and 36(th) weeks respectively. Aditionally 85% of the animals presented AH at the gastrojejunal anastomosis and 60% developed squamous hyperplasia at the squamous portion of the stomach. MNNG treatment plus duodenal reflux enhanced the development of malignant tumors at both glandular and squamous mucosa, since there were 30% of antral adenocarcinomas and 45% of squamous carcinomas at the 18th week and the frequency of these malignant tumors rose to 50% in the antrum and 65% in the squamous mucosa at the 36th week. Conclusion: The reflux of duodenal contents through the pylorus enhanced the development of proliferative lesions, benign and malignant, in the glandular stomach and in the forestomach of rats.

Como parte do projeto GENACIS, este artigo visou estimar a prevalência de depressão em amostra urbana de São Paulo, Brasil, assim como a associação entre depressão e padrão de uso de álcool segundo gênero. Para tanto, foi realizado inquérito epidemiológico com amostra probabilística e por conglomerados, com um total de 2083 adultos. Utilizou-se o CIDI SF para identificação de depressão. A análise estatística utilizou o teste de Rao Scott e regressão logística multivariada. A taxa de resposta foi de 74,9%. Predominaram mulheres (58,8%), menores de 40 anos de idade (52%). A prevalência de depressão foi de 28,3% para as mulheres e 12,7% para os homens. Entre os homens, 61,1% são de bebedores no último ano e a depressão esteve associada ao padrão de consumo de álcool, à maior frequência de binge drinking e à presença de problemas decorrentes do álcool. Entre as mulheres, 69,5% são abstinentes e a depressão se associou à convivência com cônjuge com problemas devidos ao álcool. Os resultados ressaltam que a associação entre depressão e consumo de álcool é distinta entre os gêneros.

1. The aim of the present study was to examine the effects of long-term nitric oxide (NO) blockade on contractions of the rat ileum induced by muscarinic agonists.2. Male Wistar rats received the NO synthesis inhibitor N (G) -nitro-l-arginine methyl ester (l-NAME; 20 mg/rat per day) in drinking water for 7, 15, 30 and 60 days. Concentration-responses curves to methacholine and carbachol were obtained and pEC(50) values were calculated. Saturation binding assays were performed in membranes prepared from rat ileum after 60 days of l-NAME treatment and the dissociation constant (K-D ) and maximal number of binding sites (B-max ) were determined by Scatchard analysis.3. The NO synthase activity of the ileum was markedly reduced in all l-NAME-treated groups. At 60 days after l-NAME treatment, a significant increase in the potency of methacholine (fourfold) and carbachol (threefold) was observed. In binding studies, we found a significant increase in B-max for [(3) H]-quinuclidinyl benzilate of approximately 57% in the l-NAME treated group without any significant change in K-D values. The contractile response to methacholine was not modified by the soluble guanylate cyclase inhibitor 1H-[1,2,4]oxadiazolo-[4,3-a]quinoxalin-1-one (3 mumol/L). No morphological alterations in the rat ileum were observed in l-NAME-treated rats.4. Our findings suggest that treatment with l-NAME for 60 days induces a marked increase in the potency of methacholine and carbachol, as well as an increase in receptor number in the rat ileum.

Wistar dams were exposed to 500 ppm of Pb, as Ph acetate, or 660 ppm Na acetate in drinking water during pregnancy and lactation. Male pups at 23 (weaned) or 70 days (adult) of age were submitted to behavioral evaluation and Pb determination. The behaviors evaluated were: locomotor activity (open-field test), motor coordination (rotarod test), exploratory behavior (holeboard test), anxiety (elevated plus maze and social interaction tests), and learning and memory (shuttle box). Ph levels were measured in the blood and cerebral regions (hippocampus and striatum) of dams and pups. The results of the present report demonstrated that exposure to Ph during pregnancy and lactation induces in weaned pups hyperactivity, decreased exploratory behavior, and impairment of learning and memory. These alterations were observed at blood Ph levels in the range that may be attained in children chronically exposed to low levels of Pb (21 +/- 3 mug/dl). Regarding adults, the results demonstrated that the regimen of exposure adopted induces anxiety in these animals at nondetectable blood Ph levels. (C) 2001 Elsevier B.V. All rights reserved.

Activity profiles of excess Pb-210 measured in four sediment cores from the Corumbatai River basin, São Paulo State, Brazil, provided an opportunity to evaluate sedimentation rates that are helpful for defining appropriate management strategies for the hydrological resources in the basin. This is because Rio Claro city and other municipalities make extensive use of surface waters for drinking water supply. The radiochemical analysis of the sediment cores yielded apparent sediment mass accumulation rates of between 406 and 1014 mg cm(-2) year(-1) for secondary drainage lines, whereas an intermediate value of 546 mg cm(-2) year(-1) was found in the Corumbatai River, the main drainage system of the studied area. These values provided estimates of average linear sedimentation rates of between 3.1 and 16.2 mm year(-1) that are compatible with field evidence, with the highest value corresponding with an area characterized by accumulation of sediment.

Four. male Wistar rats were housed in pairs of siblings, on LD 12:12h and 22 degrees C + 2 degrees C. Food and water were provided ad libitum. Behavior was videotaped from the 1st to the 3rd month of life. In each age-bracket the spectral composition of rhythmic expressions of the following behavioral categories was analyzed: rest, eat, drink, cage exploration, self-grooming, and social interaction. Rats maintained a stable rank order of time engaged in different behaviors through development, despite modification of time spent in grooming, drinking and social interaction as they got older. Spectral composition of behaviors followed a general ontogenetic pattern: ultradian frequencies of 12-h and 8-h were the strongest in the 1st month and circadian periodicity was predominant in the 3rd month. The increase of circadian power compared with ultradian power components agrees with literature findings. To our knowledge, self-grooming and social interaction have not been investigated before in this context. The similarities between siblings suggest the mutual influence of partners and/ or the existence of genetic factors. Ongoing studies are examining the importance of the social surroundings in which animals develop to the acquisition of adult rhythmic pattern.

Electrolyte lesion and ibotenic acid lesion of the lateral preoptic area (LPO) of the rat were used to study the participation of this area in drinking behavior. Drinking was induced by cellular dehydration, hypovolemia, hypotension, and water deprivation. The animals with electrolytic lesion of the LPO showed a significant reduction in water intake in response to cellular dehydration, hypotension, and deprivation. The animals with ibotenic acid lesion of the LPO increased the water consumption produced by subcutaneous (SC) injection of hypertonic saline. The amount of water intake after SC injection of polyethyleneglycol (PEG) or isoprenaline was similar in control and ibotenic acid-lesioned animals. The rats with ibotenic acid lesion of the LPO drank significantly more water than control animals. Fibers of passage may also influence the drinking response, and the LPO may have osmosensitive receptors that facilitate water intake in connection with other areas of the central nervous system (CNS) that are implicated in drinking behavior.

Angiotensin II (ANG II) administered centrally produces drinking by acting on subtype 1 ANG II (AT1) receptors, Carbachol, a cholinergic receptor agonist, also induces drinking behavior by a central action. In the present study we determined whether the response to carbachol also involves AT1 receptors. Male Holtzman rats (250-300 g) with stainless steel cannula implanted into the lateral ventricle (LV) were used. Water intake after injection of 0.15 M NaCl (1.0 mu l) into the LV was 0.2 +/- 0.01 ml/h (N = 8). The AT1 receptor antagonist DUP-753 (50 nmol/mu l) injected into the LV reduced water intake induced by ANG II (10 nmol/mu l) from 9.2 +/- 1.4 to 0.4 +/- 0.1 ml/h (N = 8), and water intake induced by carbachol (2 nmol/mu l) from 9.8 +/- 1.4 ml/h to 3.7 +/- 0.8 ml/h (N = 8), These results suggest that AT1 receptors play a role in the drinking behavior observed after central cholinergic stimulation in rats.

We studied the effect of the alpha(1)- and alpha(2)-adrenergic receptors of the lateral hypothalamus (LH) on the control of water intake induced by injection of carbachol into the medial septal area (MSA) of adult male Holtzman rats (250-300 g) implanted with chronic stainless steel cannulae into the LH and MSA. The volume of injection was always 1 mu l and was injected over a period of 30-60 s. For control, 0.15 M NaCl was used. Clonidine (20 nmol) but not phenylephrine (160 nmol) injected into the LH inhibited water intake induced by injection of carbachol (2 nmol) into the MSA, from 5.4 +/- 1.2 ml/h to 0.3 +/- 0.1 and 3.0 +/- 0.9 ml/h, respectively (N = 26). When we injected yohimbine (80 nmol) + clonidine (20 nmol) and prazosin (40 nmol) + clonidine (20 nmol) into theLH, water intake induced by injection of carbachol into the MSA was inhibited from 5.4 +/- 1.2 ml/h to 0.8 +/- 0.5 and 0.3 +/- 0.2 ml/h, respectively (N = 19). Water intake induced by carbachol (2 nmol) injected into the MSA was decreased by previous injection of yohimbine (80 nmol) + phenylephrine (160 nmol) and prazosin (40 nmol) + phenylephrine (l60 nmol) from 5.4 +/- 1.2 ml/h to 1.0 +/- 0.7 and 1.8 +/- 0.8 ml/h, respectively (N = 16). The cannula reached both the medial septal area in its medial portion and the lateral hypothalamus. It has been suggested that the different pathways for induction of drinking converge on a final common pathway. Thus, adrenergic stimulation of alpha(2),-adrenoceptors ofLH can influence this final common pathway.

Water intake was studied in albino rats with lesions in the lateral preoptic area, in the subfornical organ, and in both the lateral preoptic area and the subfornical organ. Drinking was induced by cellular dehydration, hypovolemia, hypotension (isoproterenol or caval ligation), and water deprivation. The animals with lesions in both areas showed a significant reduction in their water intake in response to cellular dehydration. Drinking due to extracellular dehydration was reduced in the animals that received only subfornical organ lesions, and was reduced even further in the animals with both areas ablated. The lesions in the subfornical organ were sufficient to reduce the thirst induced by caval ligation. The lesions in both areas inhibit water intake induced by caval ligation. Water intake induced by deprivation was reduced when both areas were destroyed. These findings demonstrate that both the lateral preoptic area and the subfornical organ are necessary for normal drinking in response to cellular dehydration, hypovolemia, and hypotension. There is further evidence that the lateral preoptic area and subfornical organ interact in the control of water intake induced by a variety of thirst challenges.
