Foram utilizados quatro cavalos castrados sem raça definida, distribuídos em blocos casualizados. Objetivou-se estudar a viabilização dos indicadores internos, celulose (CELi) e lignina indigestíveis (LIGi), para predizer a digestibilidade em cavalos. Os tratamentos consistiram na determinação da digestibilidade por método direto com a coleta total de fezes (CT) e indireto pelo uso CELi e LIGi obtidos pelas técnicas in situ (IS) na cavidade ruminal de bovinos e in vivo (IV) nos equinos por meio do saco de náilon móvel (SNM). A produção fecal e taxa de recuperação (p > 0,05) médios encontrados foram de 2,80 kg na MS e 101%, respectivamente. As estimativas dos CD dos nutrientes (p > 0,05) foram adequadamente preditos pela CELi e LIGi, obtidos in situ e in vivo, no qual os valores médios observados foram de 52,63, 54,17, 64,90, 43,73 e 98,28% para MS, MO, PB, FDN e Amido, respectivamente. Concluiu-se que a CELi e LIGi podem ser obtidas in vivo por meio do SNM em equinos, para predizer os coeficientes de digestibilidade de nutrientes, consumindo dieta mista.

Experiments were performed to (1) verify the inhibitory effect of bovine trophoblast protein-1 (bTP-1) on uterine prostaglandin synthesis, (2) evaluate whether other interferon-alpha (IFN-alpha) molecules also inhibit prostaglandin secretion, and (3) test whether the enzyme 2',5'-oligoadenylate synthetase (2-5A synthetase) can be induced in endometrium by interferon-alpha. In experiment 1, all interferon molecules (bTP-1, oTP-1, bIFN-alpha and hIFN-alpha) equally inhibited secretion of PGF and PGE2 from endometrial explant cultures obtained at day 17 of the estrous cycle. In experiment 2, endometrial explants obtained from day 17 of the cycle were cultured with and without bovine serum albumin (BSA; 50-mu-g/ml) and bIFN-alpha (0, 0.84, 4.2, and 42 nM). Addition of BSA to the culture medium greatly enhanced the accumulation of PGF into the medium. The bIFN-alpha inhibited accumulation of PGF and PGE2 in both the presence or absence of BSA by 12 h. All three concentrations of bIFN-alpha were equally effective in inhibiting prostaglandin accumulation. Additionally, all concentrations of bIFN-alpha increased the amounts of 2-5A synthetase in endometrium. In conclusion, these results confirm the inhibitory effect of bTP-1 on PGF release from endometrium and demonstrate that bTP-1 can also inhibit PGE2 secretion. Furthermore, other interferon-alpha molecules, including bIFN-alpha, hIFN-alpha, and oTP-1, also reduced PGF and PGE2 secretion in culture. It is likely, therefore, that conceptus and other interferon-alpha molecules exert similar effects on endometrium in vitro and that the antiluteolytic effects of bIFN-alpha in vivo are mediated in part by changes in endometrial prostaglandin synthesis.

Bovine interferon-alpha I1 (bIFN-alpha) may be useful for enhancing fertility in sheep and cattle because it has extensive sequence homology with ovine and bovine trophoblast protein-1 and, like those proteins, extends corpus luteum lifespan. To test the effectiveness of bIFN-alpha to enhance fertility, several experiments were performed in which inseminated heifers were given i.m. injections of bIFN-alpha approximately at the time of embryo-mediated signals that result in maintenance of the corpus luteum. In Exp. 1, heifers given 20 mg of bIFN-alpha daily from d 14 to 17 tended (P less than .07) to have lower pregnancy rates at d 110 to 112 of gestation (36/75; 48% vs 43/72; 60%). Similar results were obtained in Exp. 2 when heifers received a single injection of 40 mg of bIFN-alpha or placebo at d 13 after estrus; pregnancy rates at d 42 were 39/104 (38%) for bIFN-alpha and 47/98 (48%) for placebo. In Exp. 3, heifers were given gradually increasing doses of bIFN-alpha or placebo from d 11 to 19, because such a regimen had been shown to reduce the number of heifers experiencing hyperthermia after bIFN-alpha injection. Pregnancy rates were 42/95 (44%) for bIFN-alpha and 62/111 (56%) for placebo. Across all three experiments, pregnancy rates were lower (P less than .01) for heifers treated with bIFN-alpha (117/274; 43%) than for heifers treated with placebo (152/281; 54%). In conclusion, these results demonstrate that, under the administration systems used, bIFN-alpha does not increase pregnancy rate, but rather tends to reduce it.

Prevotella is one of the most abundant genera in bovine rumen, although no genome has yet been assembled by a metagenomics approach applied to Brazilian Nelore. We report the draft genome sequence of Prevotella sp., comprising 2,971,040 bp, obtained using the Illumina sequencing platform. This genome includes 127 contigs and presents a low 48% GC.

Foram utilizados quatro cavalos castrados sem raça definida pelo delineamento em blocos casualizados. Objetivou-se viabilizar os indicadores internos indigestíveis, FDNi e FDAi, obtidas pelas técnicas in situ nos bovinos e in vivo nos equinos por meio do saco de náilon móvel (SNM). Os tratamentos consistiram de digestibilidade determinada por método direto com a coleta total de fezes (CT) e indireto pelo uso de FDNi e FDAi obtidos pelas técnicas in situ (IS) na cavidade ruminal de bovinos e in vivo (IV) nos equinos por meio do SNM. As FDNi-IV e FDAi-IV promoveram a melhor taxa de recuperação (TR) do indicador, igualando-se ao grupo CT, em 91,50%, enquanto a técnica in situ resultou nas piores taxas, na média 86,50%, para ambos os indicadores indigestíveis (p < 0,05). As estimativas dos CD dos nutrientes foram adequadamente preditos pela FDAi-IV, para equinos alimentados com feno, no qual os valores observados foram de 46,41; 48,16; 47,92 e 45,51% para MS, MO, FDN e EB, respectivamente. Concluiu-se que o SNM em equinos pode ser usado para obter a FDAi em equinos alimentados exclusivamente com feno de coast-cross e para cavalos consumindo dieta mista, as FDNi e FDAi foram eleitas para predizer CD de nutrientes.

The influence of bovine rumen fluid inoculum during anaerobic treatment of the organic fraction of municipal solid waste (MSW) was studied in this work. The parameters adopted for evaluation were the biostabilization constant of total volatile solids (TVs) and the biostabilization time of the chemical oxygen demand (COD) applied to the reactors. The work was realized in four anaerobic batch reactors of 20 1 capacity each, during a period of 365 days. The proportions between MSW/inoculum loaded in the reactors were Reactor A (100%/0%), Reactor B (95%/5%), Reactor C (90%/10%) and Reactor D (85%/15%). The necessary time for biostabilization of half of the applied COD was 459, 347, 302 and 234 days and the average of methane concentration in the biogas produced was 3.6%, 13.0%, 25.0% and 42.6% for Reactors A, B, C and D, respectively. The organic wastes need to be adequately managed, in order to avoid the environmental pollution and damage to the public health. So, this work aimed to study the composting process using two methods: manual and mechanized, for the treatment of bovine ruminal waste. This residue is generated in large proportions during the bovine slaughter process, and it can lead environmental degradation and contamination, or even damage to the public health, when not treated. For the initial adjustment of the composting parameters, it was incorporated the residue of coffee husks. The manual composting system was done by the manually aerated piles, while the mechanized composting system was done in a reactor coupled to a compressor that enabled the aeration of the system. The proportions used in both systems were: 90% bovine rumen (R) and 10% coffe husks (CC); 85% bovine rumen and 15% coffe husks; 80% bovine rumen and 20% coffe husks. The parameters determined during the monitoring of the composting process were: temperature, pH, moisture, organic matter, ash, organic carbon, Kjeldahl nitrogen and C/N ratio. The results obtained during the monitoring of the piles and reactors presented similar behavior, except for the parameters Kjeldahl nitrogen and C/N ratio. When compared to the “Instrução Normativa no 25 de 23/07/2009 do Ministério da Agricultura, Pecuária e Abastecimento”, the organic produced composts with the best results were: pile 2 (85% R; 15% CC) and reactors 1 (90% R; 10% CC) e 2 (85% R; 15% CC)
