Chemoprevention opens new perspectives in the prevention of cancer and other degenerative diseases. Use of target-organ biological models at the histological and genetic levels can markedly facilitate the identification of such potential chemopreventive agents. Colon cancer is one of the highest incidence rates throughout the world and some evidences have indicated carotenoids as possible agents that decrease the risk of colorectal cancer. In the present study, we evaluate the activity of annatto (Bixa orellaria L.), a natural food colorant rich in carotenoid, on the formation of aberrant crypt foci (ACF) induced by dimethy1hydrazine (DMH) in rat colon. Further, we investigate, the effect of annatto on DMH-induced DNA damage, by the comet assay. Male Wistar rats were given s.c. injections of DMH (40 mg/kg body wt.) twice a week for 2 weeks to induce ACE They also received experimental diets containing annatto at 20, 200 or 1000 ppm for five 5 weeks before (pre-treatment), or 10 weeks after (post-treatment) DMH treatment. In both protocols the rats were sacrificed on week 15th. For the comet assay, the animals were fed with the same experimental diets for 2 weeks. Four hours before the sacrifice, the animals received an s.c. injection of DMH (40 mg/kg body wt.). Under such conditions, dietary administration of 1000 ppm annatto neither induce DNA damage in blood and colon cells nor aberrant crypt foci in rat distal colon. Conversely, annatto was successful in inhibiting the number of crypts/colon (animal), but not in the incidence of DMH-induced ACF, mainly when administered after DMH. However, no antigenotoxic effect was observed in colon cells. These findings suggest possible chemopreventive effects of annatto through their modulation of the cryptal cell proliferation but not at the initiation stage of colon carcinogenesis. To evaluate the dry matter, neutral detergent fiber and crude protein in situ degradation of elephant grass silages with five levels (0; 4; 8; 12 and 16%, on a fresh matter basis) of annatto grain by-product (AGBP), this work was carried out. A split-plot design, consisting on the addition levels the plots and the incubation times the sub-plots, with four replicates, was adopted. For the silages making, the elephant grass was cut 70 days-old and mixed to the by-product, in 20 plastic drums. The samples were dried, grid in a bolter of 5 mm mesh, being used 3 g of samples in each nylon bag, for incubation in the rumem for 0; 6; 48 e 96 hours; 2 ram without defined breed were used. Was observed increased the disappearance of the DM until the level 13.37% of addition of (AGBP) the elephant grass silages for the incubation time 48 hours, this level was that presented the most disappearance DM. For the NDF the disappearance increased until 16% of addition. For the whole of parameters of degradation, was observed that the level of addition of 16% of (AGBP) presented the most values for the potential and effective degradability of variables in question. The inclusion of the annatto grain by-product for elephant grass silage results in greater potential and effective degradability.

Annatto (Bixa orellana L.) is a natural food colorant extensively used in many processed foods, especially dairy products. The lower cost of production and the low toxicity, make annatto a very attractive and convenient pigment in substitution to the many synthetic colorants. In the present study we investigate the carcinogenic and anticarcinogenic effects of dietary annatto in Wistar rat liver using the preneoplastic glutathione S-transferase (GST-P) foci and DNA damage biomarkers. Annatto, containing 5% bixin, was administered in the diet at concentrations of 20, 200, and 1000 ppm (0.07; 0.80 and 4.23 bixin/kg body wt/day, respectively), continuously during 2 weeks before, or 8 weeks after DEN treatment (200 mg/kg body wt, i.p.), to evaluate its effect on the liver-carcinogenesis medium-term bioassay. The comet assay was used to investigate the modifying potential of annatto on DEN (20 mg/kg body wt)-induced DNA damage. The results showed that annatto was neither genotoxic nor carcinogenic at the highest concentration tested (1000 ppm). No protective effects were also observed in both GST-P foci development and comet assays. In conclusion, in such experimental conditions, annatto shows no hepatocarcinogenic effect or modifying potential against DEN-induced DNA damage and preneoplastic foci in the rat liver. Most manufactured foods contain chemicals added as a deliberate part of the manufacturing process. The aims of the present study were to evaluate the mutagenicity and antimutagenicity of annatto, a natural pigment extracted from the Bixa orellana L. and widely used as a colorant in foods. The micronucleus test was performed in bone marrow cells from Swiss male mice treated with one of the three concentrations of annatto (1330, 5330 and 10,670 ppm), incorporated into the diet. The animals were fed with the diets for 7 days and sacrificed 24 h after the last treatment. For the evaluation of the antimutagenic potential of annatto, at day 7, the animals received an intraperitoneal injection of cyclophosphamide (50 mg/kg body weight). Under the concentrations tested annatto did not present mutagenic or antimutagenic activities on the mice bone marrow cells. However, an increased frequency of micronucleated cells was observed when the highest concentration (10,670 ppm) was administered simultaneously with cyclophosphamide. In conclusion, the data indicate that annatto colour, for the conditions used, is neither mutagenic nor an inhibitor of induced mutations, although it should be used carefully since high doses may increase the effect of a mutagen. © 2003 Elsevier Science Ltd. Este trabalho teve como objetivo avaliar a capacidade de colonização do Brevipalpus phoenicis (Geijskes) em espécies de cercas-vivas, quebra-ventos e plantas invasoras mais freqüentes em pomares cítricos. O ensaio foi conduzido em laboratório e casa-de-vegetação, utilizando-se 11 espécies vegetais (tratamentos): Hibiscus sp., Malvaviscus mollis, Grevillea robusta, Mimosa caesalpiniaefolia, Bixa orellana, Euphorbia splendens, Bidens pilosa, Commelina benghalensis, Sida cordifolia e Ageratum conyzoides e Citrus sinensis. Adotou-se o delineamento estatístico de blocos casualizados, com cinco repetições. Para cada unidade experimental, constituída de uma planta envasada, foram transferidas 100 fêmeas de B. phoenicis provenientes de uma criação estoque. Decorridos 45 dias após a transferência dos ácaros para as plantas invasoras e 60 dias para as cercas-vivas e os quebra-ventos, estas tiveram suas folhas destacadas e seus ramos repicados para avaliação dos totais de estágios de desenvolvimento. Os dados relativos às contagens foram transformados em ln(x+5) e analisados pelo teste F. As médias foram comparadas pelo teste de Tukey (P < 0,05). As cercas-vivas e quebra-ventos mais favoráveis ao ácaro, em ordem decrescente, foram: M. mollis (6.342 ácaros), Hibiscus sp. (2.546), B. orellana (2.097), G. robusta (1.485) e M. caesalpiniaefolia (776). Nenhum ácaro foi encontrado em E. splendens. Com relação às plantas invasoras, foram: B. pilosa (2.238), C. benghalensis (920), S. cordifolia (738) e A. conyzoides (684). em C. sinensis foram contados 3.116 ácaros. Com exceção de E. splendens, todas as plantas mostraram-se favoráveis ao crescimento populacional de B. phoenicis. A presença destes nos pomares cítricos pode representar uma ameaça à cultura.

Objetivou-se avaliar o valor nutritivo de silagens de capim-elefante contendo subproduto do processamento do urucum. Vinte ovinos machos, não-castrados, foram distribuídos em delineamento inteiramente casualizado com cinco dietas (0; 4; 8; 12 e 16% de subproduto de urucum na silagem) e quatro repetições, e utilizados para estimar o consumo, a digestibilidade aparente de nutrientes, o teor de nutrientes digestíveis totais e o balanço de nitrogênio. A adição de subproduto de urucum elevou os consumos de matéria seca (MS), matéria orgânica (MO), proteína bruta (PB), fibra em detergente neutro (FDN), fibra em detergente ácido (FDA), extrato etéreo (EE), carboidratos não-fibrosos (CNF), carboidratos totais e nutrientes digestíveis totais (NDT), tanto em relação ao peso vivo (%PV) quanto em relação ao peso metabólico (g kg-1PV0,75). As digestibilidades de MS (55,95%), MO (58,19%), PB (45,34%), FDN (52,79%), FDA (45,79%), EE (34,96%) e CNF (99,86%) não foram influenciadas pela adição de subproduto de urucum. A inclusão de subproduto de urucum na silagem de capim-elefante promoveu aumento na digestibilidade dos carboidratos totais, no teor de NDT e no balanço de nitrogênio. A adição de subproduto de urucum em níveis de até 16% da matéria natural na ensilagem de capim-elefante melhora o consumo de nutrientes e o balanço de nitrogênio.
