Streptomyces clavuligerus é uma importante espécie industrial que produz múltiplos compostos de interesse clínico, destacando-se o antibiótico cefamicina C (CefC) e o ácido clavulânico (AC), um potente inibidor de b-lactamases produzidas por bactérias resistentes a penicilinas e cefalosporinas. Sabe-se que situações limite como subculturas sucessivas, forma inadequada de armazenamento das células, variações bruscas de condições de cultura, podem causar deleções de mais de 25% dos genes de Streptomyces sp que, embora muitas vezes não comprometam a sobrevivência do micro-organismo, acarretam perdas na capacidade produtiva. Neste trabalho, investigou-se a estabilidade da linhagem selvagem S. clavuligerus ATCC 27064 quanto à produção de CefC variando-se condições do inóculo utilizado nos cultivos submersos de obtenção do antibiótico. As fermentações foram realizadas em frascos agitados (28ºC, 260 rpm) e as amostras coletadas em 48 e 72 horas do caldo fermentativo foram analisadas quanto ao pH, biomassa (massa seca a 105ºC por 24 h) e CefC foi determinada por bioensaio de difusão em ágar (bactéria-teste Escherichia coli ESS 2235). Foi selecionado o micro-organismo melhor produtor por meio de screening, e o que apresentou a melhor produção de CefC e os menores desvios com relação à média foi utilizado para preparar novos lotes de células: ampolas de micélios liofilizados e criotubos contendo 108 esporos.mL-1 . Observou-se que a capacidade produtiva foi afetada quando foram utilizadas subculturas do micro-organismo original. A realização do screening mostrou-se eficiente na seleção de colônias superiores em termos de produção volumétrica, sendo recomendada a realização de tal procedimento periodicamente para manutenção de uma linhagem com maior potencial produtivo... Since 1988 to 1992, a study about susceptibility to antimicrobial drugs of bacterias isolated from hospitalized patients was performed. The compared susceptibility to important drugs (ampicillin, cephalotin, cefoxitin, ceftaxizime, ceftriaxone, aztreonam, gentamicin, amikacin, pefloxacin, ciprofloxacin, imipenem, oxacillin and vancomycin) was investigated in 1200 strains (300 of each specie) of the prevalent bacterias: E. coli, Klebsiella pneumoniae, Pseudomonas aeruginosa and S. aureus. Minimal inhibitory concentration (MIC) was determined by agar dilution method, using from 0.05 to 256 mcg of each drug per ml of culture medium (Mueller-Hinton). Ranges of MIC, MIC(50%), MIC(90%) and the proportion of resistant strains were determined and permitted to know the 4 drugs that were found to be more active against bacterias; the CIM(90%) values are: E. coli - aztreonam (0.1 mcg/ml), pefloxacin (0.1), ceftazidime (0.25) and ceftriaxone (0.05); K. pneumoniae-aztreonam (0.25) ceftriaxone (0.25), ceftazidime (0.5) and pefloxacin (2.0); P. aeruginosa-imipenem (4.0), aztreonam (16), ceftazidime (16) and ciprofloxacin (16); S. aureus-vancomycin (1.01, ciprofloxacin (8, 0), amikacin (128) and cephalothin (128 mg/ml). The better 'in vitro' antibacterial activity observed was related to: aztreonam (77-100% of the sensitive strains), ceftazidime (50-99,7%), pefloxacin (73-99,7%), ciprofloxacin (80%), imipenem (93%) and vancomycin (100%).

Some photosensitizers (PSs) used for PACT (Antimicrobial Photodynamic Therapy) show an affinity for bacterial walls and can be photo-activated to cause the desired damage. However, on dentine bacterias may be less susceptible to PACT as a result of limited penetration of the PS. The aim of this study was to evaluate the diffusion of one PS based on hematoporphyrin on dentine structures. Twelve bovine incisors were used. Class III cavities (3 x 3 x 1 mm) were prepared on the mesial or distal surfaces using a diamond bur. Photogem (R) solution at 1 mg/mL (10 uL for each cavity) was used. The experimental Groups were divided according to thickness of dentine remaining and etched or no-etched before the PS application. The fluorescence excitation source was a VelScope (R) system. For image capture a scientific CCD color camera PixelFly (R) was coupled to VelScope. For image acquisition and processing, a computational routine was developed at Matlab (R). Fick's Law was used to obtain the average diffusion coefficient of PS. Differences were found between all Groups. The longitudinal temporal diffusion was influenced by the different times, thickness and acid etching.

"Antimicrobial activity of crude extratcs of Petiveria alliacea L.". Petiveria alliacea L. (Phytolaccacea) is an herbaceous plant of great importance in traditional medicine. This species have been widely used in several applications such as antirheumatic, anticarcinogenic, anti-flu, antitussive, analgesic, insecticidal, acaricidal, as well as bactericide and fungicide. Currently, the pathogenic microorganisms are acquiring resistence against the traditional antibiotics, and the search for new herbal antimicrobial agents has been intensified. The objective of this study was to evaluate the antifungal and antibacterial activity of several leaf crude extracts of P. alliacea against several strains of bacterias and yeasts namely Bacilus subtilis, Pseudomonas aeruginosa, Escherichia coli, Streptococcus mutans, Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis, Candida parapsilosis, Candida kefyr and Candida albicans, using microdilution method. Promising results were observed for the 70% v/v ethanolic extract which presented minimum inhibitory concentration (MIC) from 250 to 760 mu g/mL for yeast. For the bacteria strains tested the MIC ranged between 240 to 3960 mu g/mL, depending of the extractive solution tested.

Salting have been a very utilized fish conservation method, however only in the last years the basic mechanism involved in salting fish has been understood. The objectives of this study were determine the addition in brine salt of rosemary leaves, rosemary extracts and tocopherol, and your action in the followed parameters: water activity (Aw), moisture, ash, salt content and TBARS. The results showed that the addition of antioxidants was difficulted the salt absorption, however didn't have differences between rosemary or tocopherol use. In the salting time of 3 hours the values of Aw and salt levels, was respectively: 0.77±0.01 and 14.42±1.69. for control treatment; 0.85±0.02 and 9.09±1.39for rosemary filtrate; 0.78±0.03 and 10.63±0.69 rosemary without filtrating and 0.85±0.02 and 11.96±1.78 tocopherol, showed that didn't grow indigenous bacterias. Lipid oxidation was evaluated by TBARS and the results showed the oxidative effect of salt and the pro oxidant effects of alls antioxidants used in brine salting.

In the Experiment 1, 50 pigs weaned at the 21 days were fed with either of five diets: T1 - basal diet; T2 - basal diet + antibiotic; T3 - basal diet + probiotic; T4 - basal diet + prebiotic; T5 - basal diet + simbiotic. The variables studied were body weight, feed intake and feed conversion in the Phase 1 (21 to 43 days), Phase 2 (44 to 57 days), Phase 3 (58 to 70 days) and Total Phase (21 to 70 days). During the performance experiment, a fecal survey score was conducted to verify diarrhea incidence. In the Experiment 2, 44 pigs weaned at 21 days were fed with the same diets of Experiment 1. Pigs were slaughtered at three differents ages (at weaning, at seven and 14 days after weaning). A segment of the small intestine was collected for analisys of total coliformes. The results showed that the best performance was obtained with the utilization of prebiotic and simbiotic. There were not differences in relation to diarrhea incidence among the treatments studied. The addition of probiotic and/or prebiotic in the diet prevented increase of colonization by pathogenic bacterias from seven to 14 days after weaning.
