Neoplecostomus paranensis Langeam, 1990, from the upper Rio Parana, is the only Neoplecostomus species described in this basin and is distinguished from its congeners by the lack or reduction of the adipose fin. Neoplecostomus specimens with a normal and always present adipose fin were caught in the Rio Corumba, upper Rio Parana basin. In the present study two samples of populations, one from a tributary of Rio Paranapanema (identified as a typical N. paranensis) and the other from the Rio Corumba were compared through allozyme electrophoresis. Six diagnostic loci were found, Acp-A, Adh-A, Est-A, Gpi-A, Ldh-A and Ldh-B. In addition, the locus Gpi-B showed significant differences between allelic frequencies for the two samples. Nei's genetic identity between the populations was 0.731. The expressive genetic divergence together with the presence of an adipose fin show that the sample from the Rio Corumba is distinct from N. paranensis and probably represents a new species. The golden-striped salamander (Chioglossa lusitanica) is an endemic species inhabiting stream-side habitats in mountainous areas in the northwestern Iberian Peninsula. This salamandrid is listed in the IUCN Red Data Book as a threatened species. The combination of bioclimatic modeling of the species distribution and multivariate analysis of genetic and phenotypic data strengthens previous hypotheses concerning the historical biogeography of C. lusitanica: the Pleistocene subdivision of the species' range and a process of postglacial recolonization. Discrepancies between bioclimatic modeling predictions and the present-day distribution suggest that the species may still be expanding its range northwards. We propose the identification of two distinct units for the conservation of the species and suggest that this information should be taken into account in defining key areas for conservation in the Iberian Peninsula.

O sistema de cruzamento da espécie arbórea de dossel da Mata Atlântica brasileira, Cryptocarya moschata, foi estudado a partir de material proveniente do Parque Estadual Carlos Botelho, São Miguel Arcanjo, São Paulo, Brasil. As taxas de cruzamento foram determinadas através de marcadores alozímicos obtidos de plântulas germinadas de coortes de sementes coletadas de 35 árvores. O valor médio da taxa de cruzamento de equilíbrio (estimador indireto) foi t^eq = 0.51. As estimativas das taxas de cruzamento uniloco e multilocos (estimadores diretos) foram t^s = 0.725± 0,041 e t^m = 0,884 ± 0,034, respectivamente, indicando um sistema de cruzamento predominantemente alogâmico. As taxas de cruzamento de árvores individuais variaram de 27 a 100 ( x¯ = 87,8) porcento, a partir de t^m calculado com as freqüências gênicas de pólen mantidas constantes ao nível populacional. A partir do modelo de par de irmãos (modelo de cruzamento correlacionado) de Ritland, a correlação entre duas progênies irmãs oriundas de autofecundação (r^s) e a correlação entre duas progênies irmãs oriundas de paternidade por exocruzamento (r^p) foram 35,7% e 99,0%, respectivamente. Esses resultados corroboram o fato de haver variação nas taxas de autocruzamento entre as diferentes árvores, podendo também indicar que quando há endogamia, a maior parte das sementes nas árvores são provavelmente irmãs-germanas.

The D allozyme of placental alkaline phosphatase (PLAP) displays enzymatic properties at variance with those of the common PLAP allozymes. We have deduced the amino acid sequence of the PLAP D allele by PCR cloning of its gene, ALPP Two coding substitutions were found in comparison With the cDNA of the common PLAP F allele, i.e., 692C>G and 1352A>G, which translate into a P209R and E429G substitution. A single nucleotide primer extension (SNuPE) assay was developed using PCR primers that enable the amplification of a 1.9 kb PLAP fragment. Extension primers were then used on this PCR fragment to detect the 692C>G and 1352A>G substitution. The SNuPE assay on these two nucleotide substitutions enabled us to distinguish the PLAP F and D alleles from the PLAP S/I alleles. Functional studies on the D allozyme were made possible by constructing and expressing a PLAP D cDNA, i.e., [Arg209, Gly429] PLAP, into wildtype Chinese hamster ovary cells. We determined the k(cat) and K-m, of the PLAP S, F. and D allozymes using the non,physiological substrate p-nitrophenylphosphate at an optimal pH (9.8) as well as two physiological substrates, i.e., pyridoxal-5'-phosphate and inorganic pyrophosphate at physiological pH (7.5). We found that the biochemical properties of the D allozyme of PLAP are significantly different from those of the common PLAP allozymes. These biochemical findings suggest that a suboptimal enzymatic function by the PLAP D allozyme may be the basis for the apparent negative selective pressure of the PLAP D allele. The development of the SNuPE assay will enable us to test the hypothesis that the PLAP D allele is subjected to intrauterine selection by examining genomic DNA from statistically informative population samples. Previous analyses of mitochondrial (mt)DNA and allozymes covering the range of the Iberian endemic golden-striped salamander, Chioglossa lusitanica, suggested a Pleistocene split of the historical species distribution into two population units (north and south of the Mondego river), postglacial expansion into the northernmost extant range, and secondary contact with neutral diffusion of genes close to the Mondego river. We extended analysis of molecular variation over the species range using seven microsatellite loci and the nuclear P-fibrinogen intron 7 (beta-fibint7). Both microsatellites and beta-fibint7 showed moderate to high levels of population structure, concordant with patterns detected with mtDNA and allozymes; and a general pattern of isolation-by-distance, contrasting the marked differentiation of two population groups suggested by mtDNA and allozymes. Bayesian multilocus analyses showed contrasting results as populations north and south of the Douro river were clearly differentiated based on microsatellites, whereas allozymes revealed differentiation north and south of the Mondego river. Additionally, decreased microsatellite variability in the north supported the hypothesis of postglacial colonization of this region. The well-documented evolutionary history of C. lusitanica, provides an excellent framework within which the advantages and limitations of different classes of markers can be evaluated in defining patterns of population substructure and inferring evolutionary processes across distinct spatio-temporal scales. The present study serves as a cautionary note for investigations that rely on a single type of molecular marker, especially when the organism under study exhibits a widespread distribution and complex natural history.
