Ticks are hematophagous ectoparasites which can transmit several diseases to the host during their feeding process. When ticks mechanically damage the tissue, they eventually induce inflammatory responses on the skin spot where they are fixed. One of the alternatives to control these ectoparasites is the use of chemical substances like selamectin - the active principle of Pfizer's antiparasitic Revolution® - a macrocyclic lactone capable of doing neurotoxic damage to the tick and eventually eliminating infestation in dogs and cats. The purpose of this study was to analyze, using histological and histochemical techniques, the occurrence of morphophysiological alterations in the skin of the host rabbits exposed to selamectin and infested with Rhipicephalus sanguineus (Acari: Ixodidae). Histologically, the exposed and infested rabbits showed a partial and/or total decrease in the stratum corneum and the epithelium decreased in the number of cell layers, consequently reducing the stratification (thinning) and quite pronounced formations of sub-epidermal edemas with consequent disorganization of collagen fibers in the dermal layer's connective tissue. Histochemical tests showed strong periodic acid-Schiff-positive reaction in the hair follicle and some regions of the dermis, besides resynthesis of collagen fibers detected by Mallory's trichrome technique. The obtained results showed that selamectin acts like a toxicant agent when in contact with the skin of the rabbit infested with ticks, inducing morphophysiological alterations in the acute inflammatory process in the animal's tegument. Selamectin is a chemical substance which has a dose-dependent action since higher concentrations cause greater morphophysiological damage in the skin of rabbits. © 2013 Springer-Verlag Berlin Heidelberg.

O trabalho teve como objetivo verificar a influência da adubação orgânica em 6 épocas de crescimento na produção de β-ecdisona por plantas de Pfaffia glomerata. O experimento foi conduzido na Fazenda Santo Antonio do Araquá, distrito de Catâneo Ângelo, município de São Manuel, São Paulo, Brasil. Utilizou-se o delineamento de blocos ao acaso, num esquema fatorial 5x6, com quatro repetições, considerando-se 8 plantas úteis por parcela. Os blocos foram constituídos de 6 épocas de crescimento (60, 120, 180, 240, 300 e 360 dias após a germinação) e de 5 doses de esterco de galinha curtido [testemunha (sem adubação), 15, 30, 45 e 60 t ha-1]. Após cada colheita, as raízes das plantas foram secas em estufa com circulação de ar forçada a 40ºC e pesadas para posterior extração do β-ecdisona, seguindo metodologia desenvolvida por Magalhães (2000). Os resultados foram submetidos à análise de variância e ao teste de separação de médias de Scott Knott, todos a 5% de probabilidade. Quando ocorreu interação os resultados foram avaliados usando-se análise de regressão polinominal. O teor de β-ecdisona não foi influenciado pelas doses de adubo e nem pela época do crescimento das plantas. Porém a quantidade total de β-ecdisona por raiz foi influenciada pela época de crescimento, sendo que aos 360 dias após a emergência ocorreu uma maior quantidade do princípio ativo em todos os tratamentos. Apesar de não diferir estatisticamente dos demais tratamentos, aos 360 dias após a emergência das plantas, o tratamento 30 t ha-1 foi o que proporcionou maior quantidade de β-ecdisona.

As raízes de espécies de Pfaffia são utilizadas há muito na medicina popular no Brasil, especialmente como tônicas, afrodisíacas e antidiabéticas. Nas margens e ilhas do Rio Paraná, vegeta naturalmente a espécie Pfaffia glomerata (Spreng.) Pedersen, que corre risco de extinção pela coleta intensiva de suas raízes. Para preservá-la, é preciso desenvolver sistemas que permitam seu uso sustentável. Nesse contexto, foi avaliada a sazonalidade da produção de biomassa e o teor de β-ecdisona em raízes de diferentes acessos de P. glomerata. A coleta dos acessos foi realizada ao longo dos rios Paraná (A1 e A3), Ivaí (A2) e Paranapanema (A4). Mudas produzidas a partir desses acessos foram plantadas experimentalmente em área de ocorrência natural da espécie. O delineamento utilizado foi blocos casualizados, com parcelas subdivididas (acessos nas parcelas; épocas de colheita nas subparcelas), quatro repetições e subparcelas com seis plantas na área útil. Foram realizadas quatro colheitas: oito, dez, doze e quatorze meses após o plantio. As maiores produtividades em massa seca (MS) de raízes foram alcançadas na terceira colheita, com 38,41 g planta-1. Já o acesso A4 foi significativamente mais produtivo que os demais, tanto para massa fresca (94,67 g planta-1), quanto para MS de raízes (26,39 g planta-1). A relação MF/MS das raízes, com média de 26,9% (3,7:1), foi pouco influenciada pela origem dos acessos, embora tenha se alterado significativamente ao longo das colheitas (de 4,1:1, na primeira colheita, a 3,3:1, na terceira colheita, que correspondeu ao valor máximo). O teor de β-ecdisona foi estatisticamente similar nas três primeiras colheitas, variando de 0,26 a 0,38%, com redução drástica na última colheita (0,16%). Os acessos A2 e A3 foram aqueles com teor de β-ecdisona mais altos, respectivamente 0,36 e 0,30%. Considerando os resultados obtidos, sugere-se que a terceira colheita, que corresponde a doze meses após o transplante, como a mais adequada para colheita das raízes.

The tea made with leaves and stems of plant Anchietia salutaris is traditionally used in Brazil to treat allergies. We examined the effects of a crude aqueous extract and of purified fractions of this plant on the histamine release induced in rat and guinea pig tissues. The crude extract (3-10 mu g/ml) inhibits the histamine release induced by compound 48/80 (0.5 mu g/ml) and antigen in rat peritoneal mast cells. The inhibition is significant after 10 s of preincubation and is completed after 3 min. The crude extract dissolved in the perfusion fluid (1-30 mu g/ml) also inhibits the histamine release induced in guinea pig heart by cardiac anaphylaxis and in hearts from pretreated animals (10-100 mg/kg i.p.). In pretreated animals, the effect manifests after 3 h, is maximum after 12 h and disappears after 48 h. The histamine release induced in isolated guinea pig heart by ionophore A23187 is inhibited by similar doses as in antigen-induced histamine release. Extraction with solvents concentrated the active principle (s) in the hexane fractions, as demonstrated by the inhibition of the histamine release induced by antigen in isolated cells from guinea pig heart dispersed with collagenase. In subfractions produced by the fractionation of the hexane fraction, the active principle(s) concentrated in the subfractions obtained by extraction with hexane and ethyl acetate, which shows the low polarity of the compound(s). The same subfractions that inhibit the histamine release induced by antigen in cells from guinea pig heart also inhibit pulmonary cells. Our result show that A. salutaris contains low-polarity compound(s) that inhibit the histamine release induced by three different mechanisms in mast cells from two animal species. These facts suggest that the active principle(s) of A, salutaris could be useful in the treatment of allergies and/or as a tool for the study of mast cell secretions.

Anchietia salutaris tea is traditionally used in Brazil to treat allergies, suggesting it contains compounds with antagonistic activity on the allergic mediators. We have evaluated extracts and semi-purified fractions of Anchietia salutaris as a source of compounds having this type of antagonism on the contraction induced in guinea-pig lung parenchymal strips and on platelet aggregation and shape change. After 10 min pre-incubation dichloromethane extracts containing 30 or 100 μg mL-1 inhibited the contraction induced by prostaglandin D2 (PGD2) in guinea-pig lung parenchymal strips with dose ratios (DR) of 0.76 ± 0.14 and 0.93 ± 0.19, respectively; the amount of inhibition depended both on the concentration and on the time of preincubation (DR after 30 min pre-incubation was 1.21 ± 0.51). The dichloromethane extract and its semi-purified fractions also inhibited the contractions induced by U46619, a more potent, stable, synthetic agonist of thromboxane A2 (TxA2) prostanoid (TP) receptors, the receptors acted upon by PGD2 to produce lung contractions. The dichloromethane extract did not inhibit the lung parenchymal contractions induced by histamine, leukotriene D4 (LTD4) or platelet-activating factor (PAF). Platelet aggregation induced by U46619, adenosine 5'-diphosphate (ADP) or PAF was not inhibited by the dichloromethane extract. Indeed, the extract potentiated platelet aggregation induced by low concentrations of these agonists and also potentiated the shape change induced by U46619. These results imply that the dichloromethane extract of Anchietia salutaris and its semipurified fractions contain an active principle that competitively inhibits TxA2 TP receptors, the stimulation of which causes lung parenchymal contraction. The inhibition seems to be selective for this receptor subtype, because the extract fails to inhibit platelet aggregation or shape change. This provides additional support of earlier reports suggesting the occurrence of TP receptor subtypes.

The objective of this study was to determine the influence of Metarhizium anisopliae on the predatory capacity and functional response of Chrysoperla externa fed Bemisia tabaci B biotype rearing cotton. The stock rearings of B. tabaci B biotype and C. externa were maintained in cages according to preexisting methodologies. Upon hatching, predator larvae were fed on eggs of Ephestia küehniella until their 3rd instar and then fed on whitefly nymphs in density exceeding their intake capacity (150 nymphs/individual). The nymphs were offered on cotton leaf disks (5 cm diameter), 24 hours after the treatments application following: T1: Control (water); T2: M. anisopliae at the concentration of 105 conidia cm-3; T3: 10 8 conidia cm-3 origined from Metharril® formulation of the 109 viable conidia cm-3 M. anisopliae active principle. The predatory capacity was obtained by the difference between the numbers of nymphs supplied and nymphs left. Five different nymphal densities (130, 160, 190, 220, and 250) were selected for the functional response study. The number of nymphs preyed was recorded after 24, 48, 72, and 96 hours. The M. anisopliae entomopathogenic fungi did not influence predatory capacity of C. externa which ranged from 70.6 individuals at the highest concentration to 87.1 in the control. Conclued that the C. externa predatory capacity was not affected by fungi M. anisopliae concentrations. The predator showed a type-II functional response after 24 hours, and a type-I response after 48, 72s, and 96 hours independent from fungi concentrations.

Introduction: Aging skin is a condition that affects (or will affect) all people, and its treatment is considered a clinical challenge. Growth factors and their analogues are emerging as a promising therapeutic option. Objectives: To evaluate the safety profile of some dermocosmetic products with formulations based on growth factors - or their analogs intended for that purpose - using in vitro human skin cell culture models. Methods: Two types of cell cultures were studied, and the effects of the study products on the proliferation of melanoma cells and normal human fibroblasts were evaluated. Results: No significant morphological alterations were found in the cultured human melanoma, and no significant decrease in the number of healthy cells was verified in the normal fibroblasts culture. In some cases there was even a proliferation of those cells. Conclusions: These preliminary data demonstrate that cosmeceutical products containing growth factors as an active principle can be considered safe for topical application.
