From field observations on Drimys brasiliensis, principally in the Botucatu region of São Paulo State, Brazil, new data on the reproductive biology, the rhythm of growth, and the development of lateral cymose inflorescences, flowers and fruits are presented. Pollination accelerates the rate of flower-development for about 4-6 days. Pollination experiments show that D. brasiliensis is not self-sterile; because of mechanical devices the sticky pollen grains do not normally come into contact with the stigmata unless an animal pollen vector is involved. The pollinators are diurnal Coleoptera, Diptera and Thysanoptera which eat from the pollen, lick from the stigmatic exudates and (in case of the flies) probably also from the staminal glands. Fruit- and seedeaters are birds which seem to be the main dispersal agents. Establishment of new individuals normally is through seedlings, but also by vegetative propagation through plagiotropous branches which may root and separate from the mother plant. The morphological, developmental and reproductive aspects in D. brasiliensis are discussed in a wider context, compared with data from other Magnoliidae, and related to aspects of early Angiosperm evolution. © 1980 Springer-Verlag.

Over 40,000 sugarcane (Saccharum officinarum) consensus sequences assembled from 237,954 expressed sequence tags were compared with the protein and DNA sequences from other angiosperms, including the genomes of Arabidopsis and rice (Oryza sativa). Approximately two-thirds of the sugarcane transcriptome have similar sequences in Arabidopsis. These sequences may represent a core set of proteins or protein domains that are conserved among monocots and eudicots and probably encode for essential angiosperm. functions. The remaining sequences represent putative monocot-specific genetic material, one-half of which were found only in sugarcane. These monocot-specific cDNAs represent either novelties or, in many cases, fast-evolving sequences that diverged substantially from their eudicot homologs. The wide comparative genome analysis presented here provides information on the evolutionary changes that underlie the divergence of monocots and eudicots. Our comparative analysis also led to the identification of several not yet annotated putative genes and possible gene loss events in Arabidopsis.

The establishment of plant species depends crucially on where the seeds are deposited. However, since most studies have been conducted in continuous forests, not much is known about the effects of forest fragmentation on the maintenance of abiotic and biotic characteristics in microhabitats and their effects on seed survival. in this study, we evaluated the effects of forest fragmentation on the predation upon the seeds of the palm Syagrus romanzoffiana in three microhabitats (interior forest, forest edge and gaps) in eight fragments of semi-deciduous Atlantic forest ranging in size from 9.5 ha to 33,845 ha in southeastern Brazil. Specifically, we examined the influence of the microhabitat structure, fauna and fragment size on the pattern of seed predation. Fragments < 100 ha showed similar abiotic and biotic characteristics to those of the forest edge, with no seed predation in these areas. Forest fragments 230-380 ha in size did not present safe sites for S. romanzoffiana seed survival and showed high seed predation intensity in all microhabitats evaluated. In fragments larger than 1000 ha, the seed predation was lower, with abiotic and biotic differences among gaps, interior forests and forest edges. In these fragments, the survival of S. romanzoffiana seeds was related to squirrel abundance and interior forest maintenance. Based on these results, we concluded that there are no safe sites for S. romanzoffiana seed establishment in medium- and small-sized fragments as result of the biotic and abiotic pressure, respectively We suggest that on these forest fragments, management plans are needed for the establishment of S. romanzoffiana, such as interior forest improvement and development in small-sized sites in order to minimize the edge effects, and on medium-sized fragments, we suggest post-dispersal seed protection in order to avoid seed predation by vertebrates. our findings also stress the importance of assessing the influence of forest fragmentation on angiosperm reproductive biology as part of the effective planning for the management of fragmented areas. The histamine releasing properties of glucose (mannose)-specific lectins isolated from Brazilian beans was examined. The Canavalia brasiliensis, Dioclea rostrata, and Dioclea virgata lectins induced histamine release in rat peritoneal mast cells similar to concanavalin A. Less potency and efficacy was observed for Canavalia maritima, Dioclea guianensis, and Dioclea violacea while very low activities were seen for the lectins from Dioclea grandiflora, Canavalia bonariensis, and Cratylia floribunda. The histamine releasing effect was quenched by higher doses of D. virgata lectin similar to what was reported for concanavalin A. This effect was abrogated by increasing the concentration of calcium in the incubating medium. As these above proteins have sites that bind calcium, higher doses of the lectins might withdraw the calcium which is essential for the mast cell secretion.

This work investigates some factors affecting the inactivation of common bean trypsin inhibitor and phytohemagglutin. Trypsin inhibitor activity was totally stable to heat treatment (30 min, 97C) in the total protein extract, albumin or globulin fraction. Heat treatment of the whole beans easily inactivated the inhibitor. Heat resistance of trypsin inhibitor was intermediate in the bean flour which received the same heat treatment. Independent of sample, the inhibitor was very stable to heat treatment at neutral and acidic pH and labile under strong alkaline conditions. Heating for 30 min in boiling water at pH 12 resulted in complete inactivation of the trypsin inhibitor. Autoclaving (121C) soaked whole beans and flour for 5 min inactivated 55% of the trypsin inhibitor activity in the soaked flour and 75% in the whole beans. After autoclaving 20 min, inactivation of trypsin inhibitor was about 65% in the flour and 80% in the whole beans. The phytohemagglutinin (lectin) activity was totally destroyed in the autoclaved beans after 5 min and in the flour after 15 min.

The effect of Mg levels in nutrient solution upon root, stem, leaf, fruit and total dry matter production of common bean (Phaseolus vulgaris L. cv. Carioca) was studied. Bean plants were grown in Hoagland & Arnon n. 2 solution modified to obtain 2.4, 24.3, 48.6, 72.9 and 97.2 ppm of Mg. The experimental design was a split-plot factorial replicated three times with five Mg levels and five samplings which were done fortnightly. Results were compared with those observed in plants submitted to a nutrient solution with 48.6 ppm of Mg concentration, considered the ideal Mg content. Only a mild prejudicial effect was observed with Mg excessive levels. Stem, leaves and total dry matter were higher when 2.4 ppm were used, suggesting this level as the concentration chosen for the culture of common bean.

Magnesium, calcium and potassium concentration in the seeds and cotyledons of Phaseolus vulgaris L. cv. Carioca and its translocation from the cotyledons to the plant were evaluated. For such an evaluation plants were cultivated during the cotyledons phase and were irrigated with deionized water. Three samplings of cotyledons completely randomized were done and replicated ten times, before its senescence and fall, at 8, 12 and 15 days after the germination of the seeds. Fresh and dry matter of the cotyledons and its magnesium, calcium and potassium content were determinated in every sample. These parameters were also determined in the seeds from the same lot. Potassium concentration was higher than magnesium in both seeds and cotyledons. Calcium presented the lesser concentration both in seeds and cotyledons. These minerals were translocated from the seeds and cotyledons to the young plants. Eight days after germination 59% of the magnesium, 78% of the calcium and 71% of the potassium were translocated to the plants. Fifteen days after germination these percentages were respectivaley 73%, 82% and 91%. Thus, magnesium was the less and potassium was the most translocated mineral at the end of the study.

The objective of this research was to evaluate the effectiveness of the extraction of Xanthomonas campestris pv. phaseoli from naturally infected dry bean seeds. Extraction methods tested included soaking whole seeds in sterilized saline phosphate buffer and crushing seeds after soaking in sterilized saline phosphate buffer. The bacterium was isolated on a semiselective agar medium. The seed crushing method was found to be more effective. The bacterium strains isolated were pathogenic to bean leaves, reacted with X. campestris pv. phaseoli antiserum, and had morphological and physiological/biochemical characteristics typical of the X. campestris pv. phaseoli.

Numerous natural compounds have a potential for therapeutic applications, but may have to be chemically modified to alter toxic side effects. We investigated structural parameters that could affect the cytotoxicity of isocoumarins similar to 9,10-dihydroxy-5,7-dimethoxy-1H-naphtho(2,3c)pyran-1-one (paepalantine 1). Paepalantine 1 has antimicrobial activity, as well as significant in vitro cytotoxic effects in the McCoy cell line. Two other natural and two semi-synthetic isocoumarins with similar structures obtained from the capitula of Paepalanthus bromelioides were tested on the same cell line by the neutral red assay. Substitution of the 9 and/or 10-OH group made these compounds less cytotoxic.
